Summary The baseline separation of 23 dansylated amino acids (including 3 didansylated species) is reported. Lowering of the analysis temperature and fine control of the surfactant concentration were essential in obtaining this result. Calibration graphs with good linearity were obtained.  相似文献

Four novel chiral anionic surfactants having carbohydrate hydrophilic heads, sodium n-dodecyl 1-thio-beta-D-glucopyranoside 6-hydrogen sulfate (6-betaGlcD), sodium n-dodecyl 1-thio-beta-L-glucopyranoside 6-hydrogen sulfate (6-betaGlcL), sodium n-dodecyl 1-thio-beta-L-fucopyranoside 3-hydrogen sulfate (3-betaFucL), and sodium n-dodecyl 1-thio-alpha-L-rhamnopyranoside 3-hydrogen sulfate (3-alphaRhaL), were synthesized by selective sulfation of the corresponding thioglycosides. Their CMC determined by fluorescence using pyrene as a probe in water was 1.3-2.7 mM. These surfactants found to be useful as chiral selectors for enantiomeric separation by MEKC. The enantiomeric separation was optimized with respect to pH, buffer concentration, and surfactant concentration. Under the optimized conditions (50 mM phosphate buffer at pH 6.5, 30 mM surfactant, 20 kV), the enantiomeric separations of five dansylated amino acids (Dns-AAs) were achieved within approximately 20 min with the migration order of Val相似文献

A fast and simple micellar electrokinetic capillary chromatographic method was developed for the analysis of piribedil in pharmaceutical formulations. The effects of buffer concentration, buffer pH, sodium dodecyl sulphate (SDS) concentration, organic modifier, applied voltage and injection time were investigated. Optimum results were obtained with a 50 mM borate buffer at pH 8.0 containing 50 mM SDS by using a fused silica capillary (50 m internal diameter, 72 cm effective length). The sample was injected hydrodynamically for 4 s at 50 mbar pressure and the applied voltage was +30 kV. The detection wavelength was set at 205 nm. Diflunisal was used as an internal standard. The analysis was performed at 25 °C and the total run time was 14 min. The method was suitably validated with respect to linearity range, limit of detection and quantification, precision, accuracy, specificity and robustness. The linear calibration range was 5–100 g mL–1 and the limit of detection was determined as 1 g mL–1. The method developed was successfully applied to the determination of piribedil in pharmaceutical formulations. The results were compared with a spectrophotometric method reported in the literature and no significant difference was found statistically.  相似文献

The limited peak capacity of neutral compounds in micellar electrokinetic chromatography (MEKC) causes peak overlap in a simple 38-compound sample that is predicted by statistical-overlap theory (SOT). The low-concentration sample was prepared in-house from several compound classes to span the entire migration-time range and was resolved partially in a pH=7 phosphate buffer containing 50 mM sodium dodecyl sulfate. Peaks, singlets, doublets, and other multiplets were identified on the basis of known migration times and were counted at 13 voltages spanning 4 – 26 kV. These numbers agreed well with predictions of a simple SOT based on the assumption of an inhomogeneous Poisson distribution of migration times. Because the dispersion theory of MEKC is simple, the standard deviations of single-component peaks were modeled theoretically. As part of a new way to implement SOT, probability distributions of the numbers of peaks, singlets, and so on, were computed by Monte Carlo simulation. These distributions contain all theoretical information on peak multiplicity predictable by SOT and were used to evaluate the agreement between experiment and theory. The peak capacity of MEKC was calculated numerically and substituted into the simplest equations in SOT, affirming that peak overlap arises from limited peak capacity.  相似文献

The use of retention indices in micellar electrokinetic chromatography (MEKC) is evaluated both from a theoretical and a practical point of view. Fundamental equations for the determination of retention indices in MEKC are described, showing that retention indices are independent of the surfactant concentration. Possibilities as well as limitations of different homologous series as reference standards are described. In addition, the practical application of retention indices for identification, investigation of solute-micelle interactions, characterization and classification of pseudo-stationary phases and determination of solute lipophilicity are discussed.  相似文献

A capillary electrophoretic method with UV detection at 278 nm has been developed for analysis of the immunosuppressant rapamycin (sirolimus) in human blood at low microgram per liter levels. Separation has been achieved in an acidic carrier electrolyte containing sodium dodecylsulfate and 30% (v/v) acetonitrile. For sample clean-up and preconcentration, an off-line solid-phase extraction step using a silica-based reversed-phase material and an on-capillary focussing technique were employed. The latter allows the injection of increased sample volumes without excessive band broadening. Although this new method is less sensitive than existing liquid chromatographic procedures combined with mass spectrometry, it is fully suited to routine analysis of rapamycin in blood from patients treated with this drug. Last but not least the low costs make it an attractive alternative to established methods.  相似文献

Summary A micellar electrokinetic chromatographic method is presented which permits quantification of ethinylestradiol and gestodene in pharmaceutical products. Separation was carried out at 25°C and 25 KV, using a 20 mM borate buffer (pH 9.2), 15 mM sodium dodecylsulfate in 30% acetonitrile-water (v/v). Under these conditions analyses were carried out in 7 min. Four different oral contraceptives were analysed and the results compared favourably with those of a reference liquid chromatographic method.  相似文献

The use of micellar electrokinetic capillary chromatography (MECC) for the analysis of the major nucleobases, nucleosides, and nucleotides, and their chemically modified derivatives, has been developed and refined. The dimensions of the separating capillaries, the composition of the buffering systems, and the conditions used for electrophoresis were investigated in order to obtain the best performance. Particular emphasis was placed on the identification of the physiological constituents of nucleic acids and their chemically modified analogs: in vitro studies on calf thymus DNA exposed to genotoxic agents have demonstrated that adducted bases and nucleolides can be identified by MECC.  相似文献

Summary The separation of seven analogous anabolic steroids was studied by micellar electrokinetic capillary chromatography (MECC). The retention order was found to be dependent on polarity. All of these steroids were well separated by the addition of organic modifiers to the separation buffer. Of the organic modifiers tested, 1-propanol gave the best separation, better than methanol or acetonitrile.  相似文献

Micellar electrokinetic capillary chromatography (MEKC) was performed at 25 °C and 30 kV (under a pressure of 15 mbar), using 30 mM borate buffer containing 60 mM sodium dodecysulfate (SDS) and 5% (v/v) methanol as background electrolyte (pH 9.0) to determine doxycycline. UV detection was at 350 nm. The method was shown to be specific, accurate (recovery was 100.3 ± 1.0%), linear over the tested range (correlation coefficient 0.9995) and precise (RSD <1.9%). The method was used to determine doxycycline in tablets, capsules and human urine after oral application.  相似文献

Summary There is no proportional correlation between increasing organic solvent polarity from hexane to methanol and the extractability of propolis solids by the solvents or the bactericidal action of the propolis extracts obtained. Hexane (17.7%) and chloroform (62.1%) were the poorest and the best extractants, respectively, for propolis solids. The antibiotic activity of the extracts againstStaphylococcus strains decreased in the order hexane≥ethanol>methanol. Different capillary GC profiles were obtained for persilyl derivatives of propolis extracted with hexane and methanol and for propolis collected in different Brazilian provinces, suggesting the influence of flora variability on propolis composition. Wavelength-selected DAD detection revealed MEKC to be an innovative and sensitive method for monitoring the occurrence in propolis of the flavonoids and phenolcarboxylic acids thought to be responsible for the antimicrobial activity of propolis.  相似文献

采用胶束电动力学毛细管电泳,在25mmol/L Na2HPO4-NaH2PO4(pH7.0),40mmol/L十二烷基硫酸钠和3mol/L尿素的实验条件下,在28min内实现了牡蛎中8种游离丹酰化氨基酸的分离和测定。在一定浓度范围内,氨基酸的峰面积与其浓度呈良好的线性关系,相关系数0.9955~0.9984之间。对于8种氨基酸,标准加入回收率在99.5%~100.7%之间。  相似文献

建立了以胶束电动色谱为分离模式测定植物中水杨酸的新方法。在一定范围内,随着硼酸和甲醇浓度的升高,苯甲酸内标和水杨酸的分离度以近似线性关系升高;随缓冲液pH的升高分离度呈非线性升高;随十六烷基三甲基溴化铵浓度的升高分离度呈非线性下降。在优化的条件下,两可在12min内分离。测定了苹果和梨样品,并做了回收率试验,回收率在97.1%-102%之间。  相似文献

Summary A sensitive and selective laser-induced luminescence detection scheme for orotic acid in urine, separated by micellar electrokinetic capillary chromatography (MEKC) has been developed. The 325 nm line from a helium cadmium laser is used to excite orotic acid, which transfers its energy to terbium. Resultant luminescence of terbium is linear with orotic acid concentration over more than 2.5 orders of magnitude. This novel and practical system enables the detection of 50 nm orotic acid in urine in less than 1.5 minutes while using only nanoliters of sample. The significant decrease in analysis time over traditional methods (spectrophotometric and chromatographic) comes from the high efficiency of MEKC. A dramatic improvement in sensitivity and selectivity over UV detection in capillary electrophoresis is achieved through the use of laser-induced lanthanide ion energy transfer luminescence detection. Finally, no sample pretreatment is needed and the method is free from any known interferences in urine.  相似文献

A comparison between chiral cyclodextrin‐modified microemulsion electrokinetic chromatography (CD‐MEEKC) and cyclodextrin‐modified micellar electrokinetic chromatography (CD‐MEKC) for the enantiomeric separation of esbiothrin was carried out. For both methods, the separation conditions were optimized by varying CD types and concentration, running buffer pH and compositions, organic modifiers, and temperature. The optimal CD‐MEEKC conditions were 0.8% n‐heptane, 2.3% SDS, 6.6% n‐butanol, 90.3% 10 mM sodium tetraborate containing 3% (w/v, the ratio of CD mass to microemulsion volume) methyl‐β‐cyclodextrin, pH 10, 25°C. The optimized CD‐MEKC conditions were 3.3% SDS, 96.7% 10 mM sodium tetraborate containing 5% (w/v) β‐CD, pH 10, 25°C. The difference in physicochemical properties of the buffer and CDs resulted in different optimal CD type. The competitive distribution between the microemulsion (or micelle) and chiral CD contributed to the chiral separation. Both methods provided excellent separation (Rs ~? 3) with similar migration time (ca. 15 min). CD‐MEEKC provided higher separation efficiencies (>300000) than CD‐MEKC (>200000). The LODs for CD‐MEEKC and CD‐MEKC were 4.7 μg/mL and 3.2 μg/mL, respectively. The RSDs of migration time and peak area for CD‐MEEKC were slightly higher than for CD‐MEKC. Both the demonstrated CD‐MEEKC and CD‐MEKC methods provided high efficiencies, low LODs, and reproducible enantioseparations of esbiothrin.  相似文献
