Summary This study deals with the development of a new HPLC method for the determination of 3-methoxy-4-hydroxyphenylglycol (MHPG), the main noradrenaline metabolite in human plasma. A Varian reversed-phase column (C8; 250 mm×4.6 mm i.d.; 5 μm particles) was used as the stationary phase and an aqueous solution of citric acid, 1-octanesulfonic acid, EDTA, and methanol was used as the mobile phase. Coulometric electrochemical detection (ED) was used to obtain the highest sensitivity. Isolation of MHPG from plasma was accomplished by means of a new solid-phase extraction procedure after a protein precipitation step. The extraction yield of MHPG from plasma was very high (>97%). Linearity was observed in the 0.5–25 ng mL−1 concentration range; the limit of detection was 0.2 ng mL−1 and the limit of quantitation was 0.5 ng mL−1. Repeatability (RSD,%) for plasma samples was found to be <3.2% and intermediate precision was <4.3%. The method was applied to the determination of MHPG in the plasma of healthy subjects under experimentally-induced psychological stress.  相似文献

Summary A rapid and simple liquid-chromatographic method has been developed for on-line quantification of amphetamine in biological fluids. Untreated samples (20 μL) are injected directly into the chromatographic system and purified on a 20 mm×2.1 mm i.d. pre-column packed with 30 μm Hypersil C18 stationary phase. After clean-up the analyte is transferred to the analytical column (125 mm×4 mm i.d., 5 μm LiChrospher 100 RP18) for derivatization and separation using a mixture of acetonitrile and the derivatization reagent (o-phthaldialdehyde andN-acetyl-L-cysteine) as the mobile phase. The experimental conditions for on-line derivatization and resolution of the amphetamine have been optimized, and the results have been compared with those obtained by derivatizing the analyte in pre-column mode. The method described has been applied to the determination of amphetamine in plasma and urine. Good linearity and reproducibility were obtained in the 0.1–10.0 μg mL−1 concentration range, and limits of detection were 25 ng mL−1 and 10 ng mL−1 with UV and fluorescence detection, respectively. The procedure described is very simple and rapid, because no off-line manipulation of the sample is required; the total analysis time is approximately 8 min.  相似文献

Summary A reliable and sensitive high-performance liquid chromatographic method for the determination of the recent antidepressant citalopram and two metabolites in human plasma has been developed. Fluorescence detection at 300 nm was used, exciting at 238 nm. Separation was obtained using a reversed-phase column (C18, 250 × 3.0 mm i.d., 5 μm) and a mobile phase. 40% acetonitrile: 60% aqueous tetramethylammonium perchlorate (pH 1.9). Calibration curves were linear over a working range: 5–300 ng mL−1 for citalopram, 2.5–150.0 ng mL−1 for desmethylcitalopram and 2.5–50.0 ng mL−1 for didesmethylcitalopram. The limits of quantitation (LOQ) were 1.5 ng mL−1 for citalopram and desmethylcitalopram and 2.0 ng mL−1 for didesmethylcitalopram. Precision data, as well as accuracy, were satisfactory and no interference from different psychotropic drugs was found. The method was therefore suitable for therapeutic drug monitoring of citalopram and its active metabolites in plasma of depressed patients.  相似文献

A rapid, simple and specific liquid chromatography-electrospray ionization mass spectrometry method has been developed and validated for the determination of hydroxyzine hydrochloride in human plasma. Samples were separated using a Thermo Hypersil-HyPURITYC18 reversed-phase column (150 mm × 2.1 mm i.d., 5 μm). The mobile phase consisted of 50 mM ammonium acetate (pH 4.0)–methanol–acetonitrile (45:36:19, v/v). Hydroxyzine and its internal standard were measured by electrospray ion source in positive selective ion monitoring mode. The method was validated with a linear range of 1.56–200.0 ng mL−1 and the lowest limit of quantification was 1.56 ng mL−1 for hydroxyzine hydrochloride (r 2= 0.9991). The extraction efficiencies were about 70% and recoveries of the method were in the range of 93.5–104.4%. The intra-day relative standard deviation (RSD) was less than 8.0% and inter-day RSD was within 7.4%. QC samples were stable when kept at ambient temperature for 12 h at −20 °C for 30 days and after four freeze–thaw cycles. The method has been successfully applied to the evaluation of pharmacokinetics and bioequivalence of two hydroxyzine hydrochloride formulations in 12 healthy Chinese volunteers after an oral dose of 25 mg.  相似文献

A method for the determination of the fungicide thiabendazole (TBZ) by micellar-stabilized room-temperature phosphorescence is described. It does not require any separation step and allows the direct determination of the fungicide in canned pineapple samples. The effect of various experimental conditions is discussed in detail. The analytical curve of thiabendazole gives a linear dynamic range of 23.8–500.0 ng mL–1 and a detection limit of 23.8 ng mL–1. Recoveries of 103.9 and 89.2% for syrup and canned pineapple pulp, respectively, were obtained for 250 ng mL–1 thiabendazole. Summary An oxalic acid-α-hydroxyisobutyric acid eluent has been used for the separation and determination of rare-earth elements by high-performance ion-exchange chromatography. Fifteen rare-earth elements were separated within less than 25 min on a 150×4.6 mm i.d. column packed with 5-μm sulfonic acid-bonded silica particles by elution with a combined gradient of 0.60–9.0 mM oxalic acid and 19.0–5.0 mM α-hydroxyisobutyric acid at pH 4.6. Detection and quantitation of the separated rare-earth elements was accomplished by visible-absorbance measurements at 600 nm after postcolumn reaction with arsenazo I. The gradient of the two complexing agents was optimized to enable the separation of yttrium(III) without interference from other elements, especially dysprosium(III) and terbium(III). Mass detection limits of the elements were in the range of 2–4 ng. Finally, the chromatographic system was applied to the quantitative analysis of rare-earth elements in monazite and xenotime.  相似文献

Summary A simple and rapid capillary zone electrophoretic method with UV detection has been developed for determination of tosufloxacin and trovafloxacin. The separation was performed in fused-silica capillaries (57 cm length × 75μm i.d.); the running buffer was 35mm borate + 35mm phosphate buffer solution, pH 8.6, containing 6% (v/v) acetonitrile. The applied potential was 15 kV, the temperature 30°C, and detection was at 262 nm. Piromidic acid was used as the internal standard. Response was linearly dependent on concentration in the range 1.0–120.0 μg mL−1 and the detection limit was 0.2 μg mL−1 for both compounds. The analysis was highly reproducible (RSD between 3.41 and 1.25%). The method was applied to the determination of tosufloxacin and trovafloxacin in human and rat urine. The method was validated by using HPLC as a reference method. Recovery was between 96.8 and 102%.  相似文献

A rapid and sensitive method has been developed and validated to determine 6-benzyladenine (6-BA) residues in bean sprout using high-performance liquid chromatography coupled with electrospray ionization ion trap tandem mass spectrometry. Liquid–liquid extraction, using acidified methanol, was used to isolate 6-BA from the sample matrix. The separation was carried out on a Zorbax SB C18 column (150 mm × 4.6 mm i.d., 5 μm) with 0.1% acetic acid/methanol (25:75 v/v) as mobile phase in isocratic mode. The quantitation of 6-BA was based on fragmentation of the molecular ion at m/z 226.1 to a product ion at m/z 91.1. In addition, the recoveries of three extraction solvents by ultrasound extraction and conventional extraction method were compared, and it was found that acidified methanol as the extraction solvent was the best. The calibration curve was linear (r 2 > 0.997) in the concentration range of 0.04–10.0 ng mL−1 with a lower limit of quantification of 0.5 ng g−1 for 6-BA in bean sprout. Intra-day and inter-day relative standard deviations (RSDs) were less than 7.1 and 9.8%. Recoveries of 6-BA were between 85.0 and 88.5%. This assay has been successfully applied to the determination of trace 6-BA residues in bean sprout and monitoring the market in Ningbo, China.  相似文献

A sensitive, simple, and accurate reversed-phase column liquid chromatographic (RP-LC) method with daidzein as an internal standard and UV detection at 348 nm has been developed for determination of linarin in beagle dog plasma. Plasma protein was precipitated by addition of acetonitrile and the remaining solution was evaporated to dryness. The resulting residue was then reconstituted in methanol and injected on to a 250 mm × 4.6 mm i.d., 5 μm particle, ODS analytical column. The mobile phase was a gradient prepared from acetonitrile and an aqueous solution (0.4%) of phosphoric acid; the flow rate was 1.0 mL min−1. Response was a linear function of concentration over the range 3.4–1,373.3 ng mL−1; the correlation coefficient (R 2) was 0.993. Mean recovery was 74.2%. Within-day and between-day precision were better than 8.8% relative standard deviation (RSD). The limit of quantification was 3.4 ng mL−1. This RP-LC method was used successfully in pharmacokinetic studies of linarin in beagle plasma after administration of Yejuhua injection.  相似文献

A sensitive analytical method was established for the determination of Th and U in activated concrete samples. The method combines an anion-exchange separation step with an ICP-MS determination technique. In the ICP-MS measurement, a few μg mL–1 of Al and Ca, a few ng mL–1 of Mn, La, Ce, Nd and Pb and pg mL–1 amounts of Li, Zr, Nb and Ba coexisting in the anion-exchange fraction of Th and U did not interfere. No adverse interference effects were observed in real sample analyses. The obtained detection limits (3σ, n = 10) of Th and U were 2.3 and 1.8 pg mL–1, respectively. The analytical precisions for ca. 5 μg g–1 Th and ca. 1 μg g–1 U in real activated concrete samples were equally less than 7% RSD. The accuracies obtained by the analysis of GSJ rock standard samples were –18.1 to 0.4% for the Th determination and –14.0 to –5.7% for the U determination. Summary An HPLC method with electrochemical detection has been developed for the determination of clozapine and its main metabolites, desmethylclozapine and clozapine N-oxide, in human plasma. An accurate pretreatment of the biological samples was implemented by means of solid phase extraction (SPE) on HLB cartridges. This improved pretreatment, together with a new mobile phase, allows for the accurate determination of clozapine N-oxide, which could not be quantitated by a previous method. The method uses only 100 μL of plasma for one complete analysis and shows good recovery values for all three analytes. The eluates from the SPE procedure were chromatographed in a reversed phase C18 column using a mobile phase composed of phosphate buffer, acetonitrile and methanol. Clozapine, desmethylclozapine and clozapine N-oxide were eluted in less than 10 minutes, without any interference from the biological matrix. Linearity was observed over the 2.50–150 ng mL−1 (clozapine and desmethylclozapine) or 1.25–75 ng mL−1 clozapine N-oxide) range for the three analytes, with satisfactory repeatability values. The limit of detection was 0.3 ng mL−1 for clozapine and desmethylclozapine, samples of patients treated with Leponex gave good results. No interference from other common central nervous system drugs was found. This method seems to be a useful tool for pharmacokinetic studies and for clinical monitoring, because of its need for small plasma samples and its high sensitivity and selectivity.  相似文献

A rapid, sensitive, and specific method for quantification of olmesartan, the prodrug of olmesartan medoxomil, in human plasma, using zidovudine as internal standard, is described. Sample preparation involved a simple solid-phase extraction procedure. The extract was analyzed by high-performance liquid chromatography coupled to electrospray tandem mass spectrometry (LC–MS–MS). Chromatography was performed isocratically on a 5 μm C18 analytical column (50 mm × 4.6 mm i.d.) with water–acetonitrile–formic acid 20:80:0.1 (v/v) as mobile phase. The response to olmesartan was a linear function of concentration over the range 4.82–1,928 ng mL−1. The lower limit of quantification in plasma was 4.82 ng mL−1. The method was successfully applied in a bioequivalence study of an olmesartan formulation after administration as a single oral dose.  相似文献

A new high-performance liquid chromatography assay was developed for the determination of minocycline in plasma and brain. A solid–liquid extraction procedure was coupled with a reversed-phase HPLC system. The system requires a mobile phase consisting of acetonitrile:water:perchloric acid (26:74:0.25, v/v/v) adjusted to pH 2.5 with 5 M sodium hydroxide for elution through a RP8 column (250 × 3.0 mm, i.d.) with UV detection set at 350 nm. The method proved to be accurate, precise (RSD < 20%) and linear between 0.15–20 μg mL−1 in plasma and 1–20 μg mg−1 in brain. The method was successfully applied to a blood-brain barrier minocycline transport study.  相似文献

Summary Capillary GC of metal chelates of diethyl dithiocarbamate (DDTC) was examined on a methylsilicone DB-1 column, (25 meter, 0.2 mm. i.d) with a film thickness of 0.25 μm. Elution was carried out at the initial column temperature of 180°C and programmed at 5°C min−1 to 260°C. Detection was by FID or ECD. Symmetrical peaks with base line separation were obtained with the metal chelates of copper(II), nickel(II), cobalt(III), manganese(II) and chromium(III). The ECD gave better sensitivity than the FID with a linear calibration range of 5–50 μg mL−1 and detection limits 2.0–6.0 μg mL−1, corresponding to 111–333 pg of metal ion reaching the detector. The method was applied to the determination of metal ions in water and pharmaceutical preparations with a coefficient of variation (CV) within 4.0%. When compared with a standard flame AAS method the results revealed no significant difference.  相似文献

A simple, rapid, and precise reversed-phase high-performance liquid chromatographic method has been developed for simultaneous determination of losartan potassium, ramipril, and hydrochlorothiazide. The three drugs were separated on a 150 mm × 4.6 mm i.d., 5 μm particle, Cosmosil C18 column. The mobile phase was 0.025 m sodium perchlorate–acetonitrile, 62:38 (v/v), containing 0.1% heptanesulphonic acid, pH adjusted to 2.85 with orthophosphoric acid, at a flow rate of 1.0 mL min−1. UV detection was performed at 215 nm. The method was validated for linearity, accuracy, precision, and limit of quantitation. Linearity, accuracy, and precision were acceptable in the ranges 35–65 μg mL−1 for losartan, 1.75–3.25 μg mL−1 for ramipril, and 8.75–16.25 μg mL−1 for hydrochlorothiazide.  相似文献

A novel method for the determination of paralytic shellfish poisoning (PSP) toxins using high-performance liquid chromatography with fluorescence detection was developed. The fluorescent derivates of neosaxitoxin (neoSTX), saxitoxin (STX), gonyautoxins 1 and 4 (GTX1+4), and gonyautoxins 2 and 3 (GTX2+3) were separated on a μBondapak NH2 column (300 mm × 3.9 mm, 10 μm) using water and acetate buffer (pH 6.5) as the mobile phase (1.00 mL min−1) in gradient mode with fluorescence detection at 390 nm (excitation at 330 nm). The linear ranges of neoSTX, STX, GTX1+4 and GTX2+3 were 3.31–331, 0.952–95.2, 3.78–378 and 0.124–12.4 ng mL−1, respectively. The detection limits of neoSTX, STX, GTX1+4 and GTX2+3 were 1.10, 0.32, 1.26 and 0.041 ng mL−1, respectively. The method was successfully applied to the determination of PSP toxins in microalgae. The recoveries ranged from 88±2% to 107±4% and the relative standard deviations were 0.16% to 4.4%. The procedure is also environmentally friendly because no organic solvent is used in the mobile phase.  相似文献

Summary An automated microbore, liquid chromatographic method with column-switching was developed for the determination of clomipramine from human plasma samples. After direct injection of samples (60 μL), plasma proteins and clomipramine were separated in size-exclusion mode using 20% acetonitrile in 20 mM phosphate buffer (pH 7.0) on Capcell Pak MF Ph-1 precolumn (10×4 mm I.D.). By valve switching, a fraction containing clomipramine was directed to an intermediate column for subsequent main separation on a microbore C18 column (250×1.5 mm I.D.) using 50% acetonitrile in 20 mM phosphate buffer (pH 2.5) at 0.1 mL min−1. The method was advantageous for rapidity (total analysis time: 15 min), reproducibility (C.V.<4.8%), and increased sensitivity (1 ng mL−1). The linearity of response was good (r 2≥0.999) over the concentration range 1–250 ng mL−1.  相似文献

Summary An HPLC method has been developed for the determination of fat-soluble vitamins. Ten fat-soluble vitamins were separated simultaneously on a 25 cm×4.6 mm i.d. Hypersil C18 column with acetonitrile-dichloromethane-methanol, 60:20:20 (v/v) as mobile phase at 1.0 mL min−1 with wavelength-programmed ultraviolet-visible-absorbance detection. Total analysis time was 12 min. The limits of detection were 0.03, 0.01, 0.55, 1.84, 0.02, 0.02, 0.01, 0.16, 0.33, 0.01, and 0.01 ng mL−1 for retinol, retinyl acetate, retinyl palmitate, β-carotene, ergocalciferol, cholecalciferol, tocopherol, tocopherol acetate, phylloquinone, menatetrenone, and menadione, respectively. Analysis of human serum 2–7 days after ingestion of oral vitamins and Chinese herbs led to the conclusion that the concentration of vitamins was higher than for control serum.  相似文献

Multicommutation implemented with flow-through optosensors is a very promising area of research. This recent approach benefits from the advantages of both methods and results in high sensitivity, selectivity, and speed, and little waste generation. This paper reports the simultaneous determination of furosemide and triamterene, two widely used diuretics, by measurement of their native fluorescence. The system has been proved to be useful for determination of both analytes in pharmaceutical preparations and for determination of triamterene in human urine and serum. A minicolumn filled with Sephadex SPC-25 microbeads was used to achieve separation of both analytes before detection in a flow-through cell filled with the same resin. The sensor is linear in the range 50–1200 and 0.4–8 ng mL−1 with detection limits of 15 and 0.1 ng mL−1 for furosemide and triamterene, respectively.  相似文献

Conditions of the sorption preconcentration of Pt, Pd, Au, Ag, and Hg as colored complexes of azorhodanines, tyrodine, and sulfonitrophenol M from acidic solutions (pH 0–2) on a polyamide membrane in the flow mode are determined. At a flow rate of 10–20 mL/min, concentrations of determined elements of 10-9-10-7 M, and pressure of 10–20 mm Hg, the complexes are nearly quantitatively sorbed on a polyamide membrane disk (d = 1 cm,m = 2.7 mg, andl = 0.1 mm). The influence of the composition of the test solutions on the formation of the analytical signal in the solid phase is studied. Rapid sorption-spectrometric procedures are developed for the determination of Pt, Pd, Au, Ag, and Hg with the detection limit 5–30 ng of the element in the sorbent zone  相似文献
