1, 5, 6, 7-Tetrahydro-2H-[1, 4]diazepin-5, 7-diones from Malonimides and 3-Dimethylamino-2, 2-dimethyl-2H-azirine Reaction of the aminoazirine 1 with malonimides of type 7 in 2-propanol at room temperature leads to the 1,4-diazepine derivatives of type 9 (Scheme 3). The structure of 6, 6-diethyl-3-dimethylamino-2,2-dimethyl-1,5,6, 7-tetrahydro-2H- [1,4] diazepin-5, 7-dione ( 9a ) has been proved by single crystal X-ray analysis (Chapter 4). Reduction of the 7-membered heterocycle 9a with sodium borohydride yields the perhydro-[1,4]diazepin-5, 7-dione 10 , while 9a in ethanol at 60° undergoes a ring contraction to the 4 H-imidazole derivative 11a (Scheme 4): Mechanisms of these two reactions are discussed in comparison with previously reported reactions (Chapter 5).  相似文献

The synthesis of benzodicyclicpyrrolidines 7 and 9 of specific stereochemistry such as N-Substituted-2,3-dimethyl-1,4-endo or exo-(3′,4′-pyrrolidine)-1,4-dihydronaphthalene is described. Dichlorocarbene, generated from ethyl trichloroacetate and sodium ethoxide, reacts with 2,3-dimethyl-5,6-dihydro-1,4-oxathiin to give 7,7-dichloro-1,6-dimethyl-2-oxa-5-thiabicyclo[4,1,0]heptane, and with 2,3-dimethyl-5,6-dihydro-1,4-dithiin to give 6-chloro-2,3-dihydro-7-methyl 5-methylene-5(H)-1,4-dithiepin. TPP-sensitized photooxygenation of tetramethylallene (4) in carbon tetrachloride yields acetone (5), 2,4-dimethyl-4-hydroxy-1-penten-3-one (8) and 2,4-dimethyl-1,4-pentadien-3-one (9) in a ratio of 35:20:45, besides minor amounts of resinous products and carbon dioxide. Isomerization of 4 to 2,4-dimethyl-1,3-pentadiene (6) does not occur under the reaction conditions. DABCO quenches the photooxygenation, whereas 2,4,6-tri-t-butylphenol (10) enhances the oxygen consumption rate but leaves the ratio of 5:8:9 unchanged. These results indicate that 4 is oxygenated by singlet oxygen. A mechanism is proposed according to which acetone is generated via a (2+2) cycloaddition whereas 8 and 9 are formed via an ene-reaction between 4 and singlet oxygen.  相似文献

Reaction of 2-mercapto-3-amino-5,6-dimethyl- and 2-mercapto-3-amino-5,6-diphenylpyrazines withα-halo acid esters gave 2-carbethoxy-3-aminopyrazines, which are converted by the action of sodium ethoxide to 5,6-dimethyl- and 5,6-diphenylpyrazino[2,3-b]-[1,4]thiazin-6-ones. The latter are more conveniently obtained from 2-chloro-3-amino-5,6-dimethyl- and 2-chloro-3-amino-5,6-diphenylpyrazines and thioglycolic acid. 5,6-Dihydropyrazinothiazine is formed by reduction of 5,6-dimethyl pyrazino[2,3-b][1,4]thiazin-6-one, whereas the 2,3-dimethyl-5-amino-6-sulfonic acid and its N-oxide are formed by oxidation.  相似文献

(S)-2-[Benzyl(phenyl)amino]ethyl 5-(5,5-dimethyl-2-oxo-1,3,2-dioxaphosphorinan-2-yl)-1,4-dihydro-2, 6-dimethyl-4-(3-nitrophenyl)-3-pyridinecarboxylate ((S)-NZ-105) and R isomer were synthesized through the fractional crystallization of (S)-2-Methoxy-2-phenylethyl 5-(5,5-dimethyl-2-oxo-1,3,2-dioxaphosphorinan-2-yl)-1,4-dihydro-2, 6-dimethyl-4-(3-nitrophenyl)-3-pyridinecarboxylate. Calcium antagonism activity was found to reside in the S isomer from single crystal X-ray diffraction analysis.  相似文献

4-Aryl-3,5-dialkoxycarbonyl-2,6-dimethyl-1,4-dihydropyridines are brominated by N-bromosuccinimide in methanol at room temperature at the methyl groups at positions 2 and 6 to form mono-, di-, tri-, and tetrabromo derivatives. When the N-unsubstituted bromomethyl-1,4-dihydropyridines are heated they are easily converted to tetrahydrofuropyridines, but in the case of the analogous N-substituted-1,4-dihydropyridines cyclization does not occur. Flash vacuum thermolysis (FVT) of 1-(dimethylamino)pyrrole-2,3-diones 5 causes extrusion of CO with formation of transient hydrazonoketenes 7. The transient ketenes 7 are observable in the form of weak bands at 2130 (7a) or 2115 cm-1 (7b) in the Ar matrix IR spectra resulting from either FVT or photolysis of either 5 or 1,1-dimethylpyrazolium-5-oxides 8, and these absorptions are in excellent agreement with B3LYP/6-31G* frequency calculations. Under FVT conditions the ketenes 7 cyclize to pyrazolium oxides 8, which undergo 1,4-migration of a methyl group to yield 1,4-dimethyl-3-phenylpyrazole-5(4H)-one 9a and 1,4,4-trimethyl-3- phenylpyrazole-5(4H)-one 9b. All three tautomers of 9a have been characterized, viz. the CH form 9a (most stable form in the gas phase, the solid state and solvents of low polarity), the OH form 9a' (metastable solid at room temperature) and the NH form 9a" (stable in aprotic dipolar solvents). The isomeric 1,4-dimethyl-5-phenylpyrazole-3(2H)- one 12 tautomerizes to the 3-hydroxypyrazole 12'. The crystal structure of the hydrochloride 14 of 9a'/9a" is reported, representing the first structurally characterised example of a protonated 5-hydroxypyrazole.  相似文献

A concise formal total synthesis of the cytotoxic bisnaphthazarin derivative hybocarpone has been completed through the development of routes to the synthetic precursor, 3-ethyl-2-hydroxy-5,7,8-trimethoxy-6-methyl-1,4-naphthoquinone. The oxidation of 3-ethyl-1,2,4,5,7,8-hexamethoxy-6-methylnaphthalene under Rapoport conditions gave 3-ethyl-2-hydroxy-5,7,8-trimethoxy-6-methyl-1,4-naphthoquinone in modest yields after basic hydrolysis. In addition, treatment of 3-ethyl-1,2,4,5,7,8-hexamethoxy-6-methylnaphthalene with boron tribromide provided access to the naturally occurring naphthazarin, boryquinone. The analogous oxidative demethylation of 3,6-dimethyl-1,2,4,5,7,8-hexamethoxynaphthalene and 3-ethyl-1,2,4,5,7,8-hexamethoxynaphthalene resulted in the synthesis of 2,5,7,8-tetrahydroxy-3,6-dimethyl-1,4-naphthoquinone (aureoquinone) and 3-ethyl-2,5,7,8-tetrahydroxy-1,4-naphthoquinone, respectively. An alternative selective synthetic route to 3-ethyl-2-hydroxy-5,7,8-trimethoxy-6-methyl-1,4-naphthoquinone was also developed utilizing an intramolecular Claisen condensation of methyl 2-butyryl-3,5,6-trimethoxy-4-methylphenylacetate with concomitant in situ aerial oxidation.  相似文献

Synthetic approaches to 4,4-dimethyl-1,4-thiasilinane and its S-functional derivatives, 4,4-dimethyl-1,4-thiasilinane S-oxide, 4,4-dimethyl-1,4-thiasilinane S,S-dioxide, 4,4-dimethyl- 1-(phenylsulfonylimino)-1,4-thiasilinane, and 1,4,4-trimethyl-1,4-thiasilinan-1-ylium iodide, were studied. The reaction of 2,6-dimethyl- and 2,6-diphenyl-4-dicyanomethylene-4H-pyran with hindered primary amines such as isopropylamine and cyclohexylamine gave 1-alkyl-2-amino-3-cyano-6-rnethyl(or phenyl)-4-acetonylidene (or phenacylidene)-1,4-dihydropyridine derivatives. The 6-methyl-4-acetonylidene examples underwent a facile thermal rearrangement to give 1-alkyl-2,6-dimethyl-4-dicy anomethy lene-1,4-dihydropy ridines. Synthesis of 3,3-Dimethylperhydro-1,4-diazepin-2,5,7-triones from 3-Dimethylamino-2,2-dimethyl-2H-azirine and Malonic Acid Monoamides Reaction of the aminoazirine 1 and malonic acid monoamides 5 in CH3CN yielded triamides of type 6 (Scheme 2), which were transformed to the corresponding phenylthioates 9 by treatment of a solution of 6 and thiophenol in CH3CN with HCl (Scheme 4). Cyclization of 9 to give the 1,4-diazepin-2,5,7-trione of type 10 was achieved with NaH in toluene at about 90°. It has been shown that 2-oxazolin-5-ones are intermediates in the selective cleavage of the therminal amide function of 6 (Scheme 3).  相似文献

Several dibenzodiazepine derivatives were identified as novel retinoid X receptor (RXR) antagonists on the basis of inhibitory activity on retinoid-induced cell differentiation of human promyelocytic leukemia cells HL-60 and transactivation assay using retinoic acid receptors (RARs) and RXRs in COS-1 cells. 4-(5H-2,3-(2,5-Dimethyl-2,5-hexano)-5-n- propyldibenzo[b,e][1,4]diazepin-11-yl)benzoic acid (HX603, 6c) is an N-n-propyl derivative of an RXR pan-agonist HX600 (6a), and exhibited RXR-selective antagonistic activity. Similar RXR-antagonistic activities were observed with 4-(5H-2,3-(2,5-dimethyl-2,5-hexano)-5-methyl- 8-nitrodibenzo[b,e][1,4]diazepin-11-yl)benzoic acid (HX531, 7a) and 4-(5H-10,11-dihydro-5,10-dimethyl-2,3-(2,5-dimethyl- 2,5-hexano)-dibenzo[b,e][1,4]diazepin-11-yl)benzoic acid (HX711, 8b), which also inhibited transactivation of RARs induced by an RAR agonist, Am80. These compounds inhibited HL-60 cell differentiation induced by the combination of a low concentration of the retinoid agonist Am80 with an RXR agonist (a retinoid synergist, HX600). These results indicated that HX603 (6c), and the related RXR antagonists inhibit the activation of RAR-RXR heterodimers as well as RXR homodimers, which is a distinct characteristic different from that of the known RXR antagonist, LG100754 (9).  相似文献

The replacement of 2,3-O-cyclohexylidene-d-glyceraldehyde with (2R,5R,6R)-5,6-dimethoxy-5,6-dimethyl-1,4-dioxane-2-carbaldehyde (Ley’s aldehyde) has led to significant improvements in the isolation of both diastereoisomers of the respective 2,3-O-BDA 1,2,3-trihydroxypropylphosphonates. The triethylamine-catalysed addition of dialkyl phosphites and lithium diethyl phosphonate gave the products in moderate (ca. 1:2) diastereoselectivity while the application of diethyl trimethylsilyl phosphite afforded a 1:9 mixture of diethyl (R)- and (S)-hydroxy-[(2R,5R,6R)-5,6-dimethoxy-5,6-dimethyl-1,4-dioxan-2-yl]methylphosphonates.  相似文献

A new and general synthesis of 2,5-dioxopiperazine condensed with the thiazolidine ring is described. The synthesis involves the use of N-ethoxy carbonyl-2-ethoxy-1-2-dihydroquinoleine (EEDQ) as activating agent to form the 2,5-diketopiperazine (EEDQ) as activating agent to form the 2,5-diketopiperazine ring. By this method 9-carbobutoxy-7,7-dimethyl-2,5-dioxo-8-this-1,4-diazabicyclo[4.3.0] nonane (6) and 9-carboxy-8,8 dimethyl-2,5-dioxo-7-thia-1,4-diazabicy clo [4.3.0] nonane (8) were obtained, with near quantitative yield, from 4-carboxy-2-carbobutoxy-5,5 dimethylthiazolidine (4). The former was transformed into acid 7 by hydrolysis with barium bydroxyde and the latter was esterified with diazornethane thus producing a methylester 9. The use of the ethy lester of N-(2-cabobutoxy-4-carboxy-5,5-dimethyl-thiazolidine)glycine (10) for synthesis of 6 was also successful. The spectromethric data were interpreted and confirm the proposed structure of the new compounds.  相似文献

2,3-Dimethyl-2,3-diphenylbutanediol-1,4-ditosylate (7) was synthesized starting from 2-phenylpropionic acid (1). The acid chloride was brominated and transformed into methyl 2-phenyl-2-bromo-propionate (4) which was coupled with a zinc/copper couple to dimethyl 2,3-dimethyl-2,3-diphenylsuccinate (5). Reduction with lithium aluminum hydride to 2,3-dimethyl-2,3-diphenylbutanediol-1,4 (6) was followed by tosylation. The tosylate 7 a mixture of the meso and racemic compounds, could be separated into the pure isomers,a m. p. 170 °C andb m. p. 121 °C. The mixture of each individual pure compound, when treated with tetraalkyl-ammonium bromide, did not give the expected 2,3-dimethyl-2,3-diphenyl-1,4-dibromobutane (9) but rather 1,4-diphenyl-2,3-dimethylbutadiene-1,3 (8). The identity of the compound was established by independent unequivocal synthesis, the comparison of spectral characteristics, and mixed melting point.  相似文献
