We have explored the scope of the synthetic route towards d-phenylglycyl diketopiperazines, involving a penicillin acylase catalysed formation of d-phenylglycyl dipeptides of l-amino acids with functional groups in the side chain. The synthesis of dipeptides from serine, threonine, glutamic acid, glutamine and methionine was successful. In contrast, aspartic acid, asparagine and cysteine only afforded trace amounts of dipeptides while no dipeptide was detected with arginine, lysine and tyrosine. Isolated dipeptide yields varied from 10% to 76%. The dipeptides were successfully converted into their corresponding enantiopure diketopiperazines by chemical esterification and cyclization under alkaline conditions, in 35–43% yield. In the case of glutamic acid, the procedure yielded the diketopiperazine with an esterified side chain. Remarkably with glutamine, the amide function in the side chain was transformed into an ester moiety, resulting in the same diketopiperazine as with glutamic acid.  相似文献

The nuclear magnetic resonance (NMR) technique was used as analytical tool to determine the complete metabolic profiling of sea bass extracts: water-soluble metabolites belonging to different classes such as sugars, amino acids, dipeptides and organic acids as well as metabolites soluble in organic solvent such as lipids, sterols and fatty acids were identified. The metabolite profiling together with a suitable statistical analysis were used to discriminate between wild and cultured sea bass samples. Preliminary results show that discrimination between wild and cultured sea bass was obtained not only using fatty acid composition but also cholesterol and phosphatidylethanolamine and some water-soluble metabolites such as choline, trimethylamine oxide, glutamine, fumaric and malic acids.  相似文献

Isotachophoretic qualitative indices, RE, for twenty-eight dipeptides were measured in the range pH 7.4-9.6. The absolute mobility, mo, and pKa values were evaluated by the use of the least-squares method, utilizing a simulation of the isotachophoretic steady state. The mo values were newly evaluated and the pKa values were in good agreement with literature values. By comparison of the evaluated mo and pKa values of the dipeptides with those of the constituent amino acids, simple relationships were found which may be used to estimate the mo and pKa values of other dipeptides. The separability of the dipeptides was also evaluated by considering the differences between their simulated effective mobilities. It is concluded that isotachophoresis is very convenient for the separation of dipeptides and their constituent amino acids.  相似文献

报道了以化学合成和生物转化的方法制备光学纯D-谷氨酰胺. 首先在中试规模上用化学方法合成DL-谷氨酰胺. 即以廉价的DL-谷氨酸为原料, 采用邻苯二甲酰基作为保护基保护L-谷氨酸的α-氨基, 醋酐回流15 min, 使其分子内脱水生成N-邻苯二甲酰-DL-谷氨酸酐, 在常温、常压条件下, 分别与2 mol/L氨水反应生成中间产物N-邻苯二甲酰-DL-谷氨酰胺, 中间产物在室温条件下与0.5 mol/L水合肼反应48 h脱除保护基, 以57%总收率得到DL-谷氨酰胺. 在37 ℃, pH 4.8的条件下, 利用大肠杆菌(E. coli. AS 1.505)脱羧酶将底物浓度30 g/L的DL-谷氨酰胺中L型对映体在8 h内完全转化为4-氨基丁酰胺, 分离得到D-谷氨酰胺.  相似文献

Some new and optically active 1,2,4‐triazolo thiadiazoles bearing N‐phthaloyl‐l ‐amino acids were synthesized by reaction of 4‐amino‐5‐(3‐ or 4‐)pyridyl‐3‐mercapto‐(4H)‐1,2,4‐triazoles with N‐phthaloyl‐l ‐amino acids in the presence of phosphorus oxychloride. All the newly synthesized compounds were confirmed by IR, 1H NMR, 13C NMR and elemental analysis.  相似文献

An environmentally benign and inexpensive preparation method is described of some imidoyl chlorides, including the Vilsmeier reagent (VR), by using phthaloyl dichloride. Synthetic applications were demonstrated using the isolated VR or VR prepared in situ for the transformation of acids to acid chlorides, alcohols to chlorides, and the formylation of dimethylaminobenzene.  相似文献

Five organotin(IV) thioesters of N‐phthaloyl amino acids with the general formulae R3SnL (R = Me, Ph) and nBu2SnL2 were synthesized with L = N‐phthaloyl‐thioalanine and N‐phthaloyl‐thioleucine. The structures of trimethyltin(IV) N‐phthaloyl‐thioleucinate ( 1 ), trimethyltin(IV) N‐phthaloyl‐thioalaninate ( 2 ), triphenyltin(IV) N‐phthaloyl‐thioleucinate ( 3 ), triphenyltin(IV) N‐phthaloyl‐thioalaninate ( 4 ), and di‐n‐butyltin(IV) di‐N‐phthaloyl‐thioalaninate ( 5 ) were characterized by means of X‐ray diffractometry. Summary One classical method for quantitation of amino acids in proteins is hydrolysis of the proteins and determination of the free amino acids. Although the drastic experimental conditions necessary for complete hydrolysis always cause degradation of some of the amino acids, if mild hydrolysis conditions are used, a mixture of amino acids and oligopeptides is obtained. If these conditions are adequately tuned, the oligopeptides are almost exclusively dipeptides. For this reason we have initiated a study to find a derivatizing agent suitable for the analysis of amino acids and dipeptides by an absolute method of quantitation already tested for amino acids. FMOC-Cl was found to be a suitable derivatizing agent for this purpose.  相似文献

Conformations of peptides are the basis for their property studies and the predictions of peptide structures are highly important in life science but very complex in practice. Here, thorough searches on the potential energy surfaces of 13 representative dipeptides by considering all possible combinations of the bond rotational degrees of freedom are performed using the density functional theory based methods. Careful analyses of the conformers of the 13 dipeptides and the corresponding amino acids reveal the connections between the structures of dipeptide and amino acids. A method for finding all important dipeptide conformers by optimizing a small number of trial structures generated by suitable superposition of the parent amino acid conformations is thus proposed. Applying the method to another eight dipeptides carefully examined by others shows that the new approach is both highly efficient and reliable by providing the most complete ensembles of dipeptide conformers and much improved agreements between the theoretical and experimental IR spectra. The method opens the door for the determination of the stable structures of all dipeptides with a manageable amount of effort. Preliminary result on the applicability of the method to the tripeptide structure determination is also presented. The results are the first step towards proving Anfinsen's hypothesis by revealing the relationships between the structures of the simplest peptide and its constituting amino acids. It implies that the structures of peptides are not only determined by their amino acid sequences, but also closely linked with the amino acid conformations. © 2009 Wiley Periodicals, Inc. Several high-performance liquid chromatographic (HPLC) methods are described for separation of peptide stereoisomers which are not well resolved by traditional reversed-phase chromatography. These chiral HPLC methods include investigations with a beta-cyclodextrin column, a Pirkle D-Phenyl Glycine column and a Chiral-Pak WH column. A method based on derivatization of dipeptides with a chiral reagent, N-acetyl-L-cysteine and o-phthalaldehyde, is also discussed. A series of linear and cyclic dipeptides and modified amino acids were chromatographed on the four systems. Resolution varied for the four different systems depending on the types of compounds that were chromatographed.  相似文献

从20种天然氨基酸的41个randic molecular profiles非零描述符、44个eigenvalue based indices非零描述符和47个walk and path counts非零描述符分别进行主成分分析,得出一种新的氨基酸描述符-SVREW。将其应用于血管紧张素转化酶(ACE)抑制二肽和ACE抑制三肽、苦味二肽和苦味四肽、后叶催产素类似物、HLA-A*0201限制性CTL表位肽的结构表征,应用多元线性回归(MLR)建立定量构效关系模型,同时采用内部与外部双重验证的方法验证模型的稳定性。所建ACE抑制二肽、ACE抑制三肽、苦味二肽、苦味四肽、后叶催产素类似物、HLA-A*0201限制性CTL表位肽的模型复相关系数(R2cum)分别为0.994,0.797,0.948,0.878,0.686,0.720;留一法交互校验复相关系数(R2cv)分别为0.955,0.859,0.879,0.958,0.796,0.843;外部样本校验相关系数(Q2ext)分别为0.990,0.954,0.890,0.950,0.748,0.773。经研究表明SVREW描述符用于肽分子结构表征所建模型的稳定性与预测能力均较好,有望成为多肽定量构效关系研究中一种有效的结构表征方法,可对新药物的发现和研究提供指导。  相似文献

We have developed a highly efficient and practical approach for palladium‐catalyzed trifluoroacetate‐promoted N‐quinolylcarboxamide‐directed glycosylation of inert β‐C(sp3)?H bonds of N‐phthaloyl α‐amino acids with glycals under mild conditions. For the first time, C(sp3)?H activation for glycosylation was achieved to build C‐alkyl glycosides. This method facilitates the synthesis of various β‐substituted C‐alkyl glycoamino acids and offers a tool for glycopeptide synthesis.  相似文献

Abstract

A method was developed specifically for the determination of glutamine and asparagine in the presence or absence of other amino acids. The amino acids were derivatized by o-phthalaldehyde/ 2-mercaptoethanol and separated by isocratic elution with a mobile phase consisting of acetonitrile and sodium acetate buffer. An application of the method for the analysis of glutamine and asparagine in the enzymatic hydrolysate of cottonseed protein is described.  相似文献

A method for the determination of 3-methylhistidine using an automatic amino acid analyser has been developed. A single column system with lithium buffer (pH 3.950, 0.500 mol/l lithium and 0.067 mol/l citrate) was used for elution. The standard amino acid mixture of basic amino acids and some dipeptides usually present in physiological fluids was analysed for the development of the method. 3-Methylhistidine eluted in 46.7 +/- 0.049 min and the peak area coefficient of variation for the same sample was 1.07%. As 3-methylhistidine is completely resolved from the other basic amino acids and some dipeptides (anserine and carnosine), this method is suitable for the analysis of urine and muscle extracts as well as skeletal muscle protein hydrolysates where this amino acid is present in much lower concentrations than other amino acids.  相似文献

A general and efficient method for the synthesis of spiro-2,5-diketopiperazines (spiro-DKPs) is described. Cyclization of Boc-protected dipeptides containing spiro-amino acids by microwave assisted heating in water furnished the corresponding spiro-DKPs. The spiro-amino acids were prepared by combining stereoselective alkylation reactions using the Schöllkopf methodology for amino acid construction with Grubbs ring-closing metathesis (RCM) methodology using ruthenium complexes. The RCM reactions and all subsequent transformations to the spiro-DKPs were run with microwave assisted heating, resulting in high yields and short reaction times for all steps.  相似文献
