建立了一种基于超高效液相色谱-离子阱-飞行时间质谱(UPLC-IT-TOF MS)联用的非靶标代谢组学方法,研究了人慢性髓系白血病细胞及其阿霉素耐药细胞(K562和K562-ADM)之间的代谢物差异。考察了细胞破碎方法、提取溶剂体系和溶剂体积对细胞代谢物提取效果的影响,实验发现采用超声破碎法,以800μL的80%甲醇提取时可得到较多的代谢特征峰,方法重复性和稳定性较高。将建立的方法应用于K562及K562-ADM细胞的代谢组差异研究,对所得数据进行主成分分析(PCA)和正交偏最小二乘法判别分析(OPLS-DA)。结果显示2种细胞的代谢特征存在显著差异,并发现40个对分类有显著贡献的代谢物,差异代谢物主要参与氨基酸代谢、鞘脂代谢、嘌呤代谢和嘧啶代谢等通路。该方法可用于K562和K562-ADM细胞的代谢组差异研究,可望为白血病细胞耐药性的代谢组学研究提供帮助。  相似文献

口腔癌的发病率占全身恶性肿瘤的第6位,正确区分正常状态与良性和恶性口腔肿瘤,是恰当选择治疗方案的关键所在。本研究中,首先利用液相色谱-质谱和气相色谱-质谱联用方法分别得到健康人、良性口腔肿瘤患者和恶性口腔肿瘤患者血浆、尿液和唾液的代谢轮廓,然后应用正交信号校正的偏最小二乘法进行多变量统计分析。结果表明健康人、良性肿瘤患者和恶性肿瘤患者在血浆、尿液和唾液等3种体液代谢中都可以被区分开,而且找到和鉴定出19个重要差异代谢物。相关代谢通路分析显示,与健康人相比,良性和恶性口腔肿瘤患者都存在能量代谢紊乱和脂类代谢失衡的现象,但恶性口腔肿瘤患者还表现出三羧酸循环和肌醇代谢异常,这为临床诊断及治疗提供了重要信息。  相似文献

将基于液相色谱-质谱联用(LC-MS)技术的代谢组学分析平台用于薄荷烟对大鼠代谢影响的研究。分析了3组大鼠的尿样,包括对照大鼠、吸食普通烟大鼠和吸食薄荷烟大鼠,并采用主成分分析(PCA)方法对数据进行模式识别。PCA得分图表明吸食薄荷烟大鼠与对照组大鼠尿样的代谢差异要小于吸食普通烟大鼠。从PCA载荷图中找到并鉴定了犬尿喹啉酸等8种重要代谢物。通过考察代谢物在对照大鼠、吸食薄荷烟大鼠和吸食普通烟大鼠尿样中的相对含量变化,进一步说明了烟草中添加薄荷醇可减少烟草对大鼠代谢的影响。  相似文献

利用气相色谱-质谱联用技术(GC-MS)和图模型分析方法,寻找脂代谢异常患者可能的血浆代谢标志物群。采用GC-MS技术对37例脂代谢异常患者和10例健康人的血浆样品进行分析,得到血浆代谢物的表达谱。偏最小二乘-判别分析(Partial least squares-discriminant analysis,PLS-DA)得分图可区分脂代谢异常患者与健康人,运用PLS-DA载荷图及t检验发现有9个代谢物在两组间存在显著性差异。经NIST谱库检索,它们分别为缬氨酸、甘氨酸、丙氨酸、焦谷氨酸、葡萄糖醛酸、半乳糖、甘露糖、亚油酸和甘油。在脂代谢异常患者血浆中,除甘油浓度显著高于健康人外,其余8种代谢物浓度均明显低于健康人。图模型分析结果发现这些代谢物与脂代谢异常临床常用诊断指标之间具有很好的相关性。它们可能是脂代谢异常疾病早期诊断和预后新的特异性代谢标志物群。  相似文献

采用气相色谱-质谱联用技术(GC-MS),对代谢组学研究中血清样本的前处理方法进行考察,研究蛋白去除和衍生化两个步骤对血清代谢物分析的影响。对血清样本和质控(Quality control,QC)样本的图谱进行峰相对标准偏差(RSD)计算,全谱主成分分析(Principal components analysis,PCA)和信息量的计算以及代谢物的定性定量分析。结果表明,QC样本和血清平行样的峰面积RSD均小于10%,PCA分析结果及信息量计算结果都表明这些前处理方法重复性及可靠性良好,适用于代谢组学研究。4种前处理方法所得代谢图谱的PCA分析结果表明不同除蛋白试剂和衍生化试剂对代谢物谱存在不同的影响,其中衍生化试剂的影响大于除蛋白试剂的影响。信息量计算显示MSTFA组图谱的信息量(约8.9)大于BSTFA组的信息量(约8.7)。对血清样本定性共得到44种代谢物,MSTFA组血清样本特有代谢物5种,BSTFA血清样本特有代谢物6种。对定性结果进行分析后建议:当需要研究极性氨基酸和短链脂肪酸时,采用BSTFA作衍生化试剂较合适;而当需要研究非极性氨基酸和长链脂肪酸时,采用MSTFA作衍生化试剂较合适。  相似文献

茶叶中的化学成分是构成其风味特征的物质基础。本研究建立了基于气相色谱-质谱联用和液相色谱-质谱联用的非靶向代谢组学方法。完成预处理及分析条件优化后,使用标准样品考察方法的线性、回收率及重复性,结果表明方法整体稳定且结果可靠。将该方法用于绿茶、乌龙茶和红茶中化学成分的分析。通过超声辅助溶剂提取及气相色谱-质谱联用分析共获得1812个特征峰,而使用加热溶剂提取及液相色谱-质谱联用分析可获得2608个特征峰。结合保留规律及质谱数据库,共定性173种化合物(109种随后经标准样品验证),其中只有9种化合物在上述两类分析中被同时检出,表明方法互补性良好。3类茶叶数据的偏最小二乘判别分析结果表明,三者间存在显著差异。结合模型的变量重要因子(VIP)与非参数检验共筛选出90种化合物,其中包含儿茶素、氨基酸、糖、有机酸和黄酮苷类等众多与茶叶滋味密切相关的化学成分。  相似文献

Fermented bean products are used worldwide; most of the products are made using only a few kinds of beans. However, the metabolite changes and contents in the beans generally used during fermentation are unrevealed. Therefore, we selected four different beans (soybean, Glycine max, GM; wild soybean, Glycine soja, GS; common bean, Phaseolus vulgaris, PV; and hyacinth bean, Lablab purpureus, LP) that are the most widely consumed and fermented with Aspergillus oryzae. Then, metabolome and multivariate statistical analysis were performed to figure out metabolite changes during fermentation. In the four beans, carbohydrates were decreased, but amino acids and fatty acids were increased in the four beans as they fermented. The relative amounts of amino acids were relatively abundant in fermented PV and LP as compared to other beans. In contrast, isoflavone aglycones (e.g., daidzein, glycitein, and genistein) and DDMP-conjugated soyasaponins (e.g., soyasaponins βa and γg) were increased in GM and GS during fermentation. Notably, these metabolite changes were more significant in GS than GM. In addition, the increase of antioxidant activity in fermented GS was significant compared to other beans. We expect our research provides a basis to extend choice for bean fermentation for consumers and food producers.  相似文献

Solid-phase microextraction (SPME) was coupled to gas chromatography mass spectrometry (GC-MS) and a method optimized to quantitatively and qualitatively measure a large array of volatile metabolites in alfalfa glandular trichomes isolated from stems, trichome-free stems, and leaves as part of a non-targeted metabolomics approach. Major SPME extraction parameters optimized included SPME fiber composition, extraction temperature, and extraction time. The optimized SPME method provided the most chemically diverse coverage of alfalfa volatile and semi-volatile metabolites using a DVB/CAR/PDMS fiber, extraction temperature of 60 °C, and an extraction time of 20 min. Alfalfa SPME-GC-MS profiles were processed using automated peak deconvolution and identification (AMDIS) and quantitative data extraction software (MET-IDEA). A total of 87 trichome, 59 stem, and 99 leaf volatile metabolites were detected after background subtraction which removed contaminants present in ambient air and associated with the fibers and NaOH/EDTA buffer solution containing CaCl2. Thirty-seven volatile metabolites were detected in all samples, while 15 volatile metabolites were uniquely detected only in glandular trichomes, 9 only in stems, and 33 specifically in leaves as tissue specific volatile metabolites. Hierarchical cluster analysis (HCA) and principal component analysis (PCA) of glandular trichomes, stems, and leaves showed that the volatile metabolic profiles obtained from the optimized SPME-GC-MS method clearly differentiated the three tissues (glandular trichomes, stems, and leaves), and the biochemical basis for this differentiation is discussed. Although optimized using plant tissues, the method can be applied to other types of samples including fruits and other foods.  相似文献

Mass spectrometry (MS) driven metabolomics is a frequently used tool in various areas of life sciences; however, the analysis of polar metabolites is less commonly included. In general, metabolomic analyses lead to the detection of the total amount of all covered metabolites. This is currently a major limitation with respect to metabolites showing high turnover rates, but no changes in their concentration. Such metabolites and pathways could be crucial metabolic nodes (e.g., potential drug targets in cancer metabolism). A stable-isotope tracing capillary electrophoresis–mass spectrometry (CE-MS) metabolomic approach was developed to cover both polar metabolites and isotopologues in a non-targeted way. An in-house developed software enables high throughput processing of complex multidimensional data. The practicability is demonstrated analyzing [U-13C]-glucose exposed prostate cancer and non-cancer cells. This CE-MS-driven analytical strategy complements polar metabolite profiles through isotopologue labeling patterns, thereby improving not only the metabolomic coverage, but also the understanding of metabolism.  相似文献

采用气相色谱-串联四极杆飞行时间质谱(GC/Q-TOF MS)建立了稻米的代谢组学分析方法,并将其应用于产地溯源。利用D-最优设计对提取溶剂进行优化,通过偏最小二乘分析法考察了不同溶剂对代谢物提取效率的影响。考察了衍生试剂种类、衍生温度及时间对代谢物检测的影响,最终确定分析条件为:甲醇-水(1∶1)为提取溶剂,N-甲基-N-三甲基硅基三氟乙酰胺-三甲基氯硅烷(MSTFA-TMCS,99∶1)于60℃条件下衍生1 h。选取10种代谢物考察仪器精密度、方法重现性和衍生产物稳定性,相对标准偏差(RSD)均小于9%,证明了方法的可靠性。结合Mass Profiler Professional(MPP)软件,以主成分分析(PCA)法区分黑龙江省4个地区的29个绥粳4号稻米样本,分类效果显著。对显著变化(P0.05,倍率变化≥2)的差异性代谢物进行鉴定,确定了11种特征标记物。结果表明,利用GC/Q-TOF MS建立代谢组学分析方法,结合主成分分析鉴别稻米产地具有可行性,为大米市场的规范和资源开发利用提供了依据。  相似文献

While coffee beans have been studied for many years, researchers are showing a growing interest in coffee leaves and by-products, but little information is currently available on coffee species other than Coffea arabica and Coffea canephora. The aim of this work was to perform a targeted and untargeted metabolomics study on Coffea arabica, Coffea canephora and Coffea anthonyi. The application of the recent high-resolution mass spectrometry-based metabolomics tools allowed us to gain a clear overview of the main differences among the coffee species. The results showed that the leaves and fruits of Coffea anthonyi had a different metabolite profile when compared to the two other species. In Coffea anthonyi, caffeine levels were found in lower concentrations while caffeoylquinic acid and mangiferin-related compounds were found in higher concentrations. A large number of specialized metabolites can be found in Coffea anthonyi tissues, making this species a valid candidate for innovative healthcare products made with coffee extracts.  相似文献

Morus alba L. is used in traditional Chinese medicine for its anti-diabetic activity; however, the part of the hypoglycemic activity and related active metabolites are still not fully clarified. In this study, the metabolites in the M. alba roots, leaves, twigs, and fruits extracts (70% ethanol extracts) were systematically identified, and their hypoglycemic activity was evaluated by the high-fat diet/streptozotocin-induced 2 diabetes mellitus (T2D) mouse model. A total of 60 high-level compounds, including 16 polyphenols, 43 flavonoids, and one quinic acid, were identified by high-performance liquid chromatography/quadrupole time-of-flight mass spectrometry (HPLC-Q-TOF-MS) combined with the fragmentation pathways of standards and the self-established database. Among them, 23 metabolites were reported for the first time from this plant. In contrast to the extracts of M. alba leaves and fruits, the extracts of roots and twigs displayed significant hypoglycemic activity The glycemia was significantly reduced from 32.08 ± 1.27 to 20.88 ± 1.82 mmol/L and from 33.32 ± 1.98 to 24.74 ± 1.02 mmol/L, respectively, after 4 weeks of treatment with roots and twigs extracts. Compound 46 (morusin), which is a high-level component identified from the extracts of M. alba roots, also displayed significant activity in decreasing the blood glucose level of T2D mice reduced from 31.45 ± 1.23 to 23.45 ± 2.13 mmol/L. In addition, the extracts of roots and twigs displayed significant activity in reducing postprandial glycemia. This work marks the first comparison of the metabolites and hypoglycemic activity of M. alba roots, leaves, twigs, and fruits extracts, and provides a foundation for further development of M. alba extracts as anti-diabetic drugs.  相似文献

This study reports differential expression of endoglucanase (EG) and β-glucosidase (βG) isoforms of Aspergillus terreus. Expression of multiple isoforms was observed, in presence of different carbon sources and culture conditions, by activity staining of poly acrylamide gel electrophoresis gels. Maximal expression of four EG isoforms was observed in presence of rice straw (28 U/g DW substrate) and corn cobs (1.147 U/ml) under solid substrate and shake flask culture, respectively. Furthermore, the sequential induction of EG isoforms was found to be associated with the presence of distinct metabolites (monosaccharides/oligosaccharides) i.e., xylose (X), G1, G3 and G4 as well as putative positional isomers (G1/G2, G2/G3) in the culture extracts sampled at different time intervals, indicating specific role of these metabolites in the sequential expression of multiple EGs. Addition of fructose and cellobiose to corn cobs containing medium during shake flask culture resulted in up-regulation of EG activity, whereas addition of mannitol, ethanol and glycerol selectively repressed the expression of three EG isoforms (Ia, Ic and Id). The observed regulation profile of βG isoforms was distinct when compared to EG isoforms, and addition of glucose, fructose, sucrose, cellobiose, mannitol and glycerol resulted in down-regulation of one or more of the four βG isoforms.  相似文献
