A newly designed synthetic entry to the C1-C27 domain of okadaic acid has been developed. This incorporates substantial improvements in the preparations of the key okadaic acid building blocks representing the C3-C8, C9-C14, and C16-C27 portions. The synthesis of the C3-C8 lactone used (R)-glycidol as the origin of the C4 stereogenic center and featured a late-stage optional incorporation of the C7 hydroxyl group. The complementary C9-C14 fragment was synthesized in a concise route from (R)-3-tert-butyldimethylsilyloxy-2-methylpropanal and propargyl bromide. Assembly of the C3-C14 spiroketal-containing intermediate from the constitutent fragments revealed a dramatic effect of C7 functionalization upon spiroketalization efficiency. In contrast, both (9E)- and (9Z)-enones converged readily to the C8 spiroketal upon treatment with acid. Modifications to the central C16-C27 fragment of okadaic acid included the early replacement of benzylic protecting groups by more suitable functionalities to facilitate both the generation of the C15-C27 intermediate and the deprotection of the final products. These modular building blocks were deployed for the synthesis of the C1-C27 scaffold of 7-deoxyokadaic acid. This work demonstrates improvements in the formation of versatile okadaic acid intermediates, as well as a reordering of fragment couplings. This alternative order of coupling was designed to promote the late stage incorporation of nonnatural lipophilic extensions from the C27 terminus.  相似文献

A series of 2-(1-(arylimino)propyl)quinolin-8-olate half-titanocene dichlorides, Cp'TiCl(2)L (Cp' = η(5)-C(5)H(5) or η(5)-C(5)Me(5), L = 2-(1-(2,6-R(1)-4-R(2)-phenylimino)propyl)quinolin-8-olate), was synthesized via the stoichiometric reaction of Cp'TiCl(3) with the corresponding potassium 2-(1-(2,6-R(1)-4-R(2)-phenylimino)propyl)quinolin-8-olate salt. All titanium compounds were characterized by elemental analysis, (1)H NMR and (13)C NMR spectroscopy; the molecular structures of two representative compounds were determined by single crystal X-ray diffraction. On activation with methylaluminoxane (MAO), all half-titanocene compounds showed high activity in ethylene polymerization, and furthermore, performed with good to high activities in the co-polymerization of ethylene with either 1-hexene or 1-octene affording polyethylenes with high co-monomer incorporation. Less bulky ortho-substituents (R(1)) on the phenylimino groups were found to enhance the catalytic activities of their titanium compounds. In general, the titanium pro-catalysts containing η(5)-C(5)Me(5) (C7-C12) exhibited higher activities than did their analogues bearing η(5)-C(5)H(5) (C1-C6). Reaction of C70 with ten equivalents of silver(I) trifluoroacetate at 320-340 degrees C followed by fractional sublimation at 420-540 degrees C and HPLC processing led to the isolation of a single abundant isomer of C70(CF3)n for n = 2, 4, 6, and 10, and two abundant isomers of C70(CF3)8. These six compounds were characterized by using matrix-assisted laser desorption ionization (MALDI) mass spectrometry, 2D-COSY and/or 1D 19F NMR spectroscopy, and quantum-chemical calculations at the density functional theory (DFT) level. Some were also characterized by Raman spectroscopy. The addition patterns for the isolated compounds were unambiguously found to be C1-7,24-C70(CF3)2, C1-7,24,44,47-C70(CF3)4, C2-1,4,11,19,31,41-C70(CF3)6, Cs-1,4,11,19,31,41,51,64-C70(CF3)8, C2-1,4,11,19,31,41,51,60-C70(CF3)8, and C1-1,4,10,19,25,41,49,60,66,69-C70(CF3)10 (IUPAC numbering). Except for the last compound, which is identical to the recently reported, crystallographically characterized C70(CF3)10 derivative prepared by a different synthetic route, these compounds have not previously been shown to have the indicated addition patterns. The largest relative yield under an optimized set of reaction conditions was for the Cs isomer of C70(CF3)8 (ca. 30 mol % of the sublimed mixture of products based on HPLC integration). The results demonstrate that thermally stable C70(CF3)n isomers tend to have their CF3 groups arranged on isolated para-C6(CF3)2 hexagons and/or on a ribbon of edge-sharing meta- and/or para-C6(CF3)2 hexagons. For Cs- and C2-C70(CF3)8 and for C2-C70(CF3)6, the ribbons straddle the C70 equatorial belt; for C1-C70(CF3)4, the para-meta-para ribbon includes three polar hexagons; for C1-7,24-C70(CF3)2, the para-C6(CF3)2 hexagon includes one of the carbon atoms on a C70 polar pentagon. The 10.3-16.2 Hz 7JF,F NMR coupling constants for the end-of-ribbon CF3 groups, which are always para to their nearest-neighbor CF3 group, are consistent with through-space Fermi-contact interactions between the fluorine atoms of proximate, rapidly rotating CF3 groups.  相似文献

The design, chemical synthesis, and biological evaluation of a series of cyclopropyl and cyclobutyl epothilone analogues (3-12, Figure 1) are described. The synthetic strategies toward these epothilones involved a Nozaki-Hiyama-Kishi coupling to form the C15-C16 carbon-carbon bond, an aldol reaction to construct the C6-C7 carbon-carbon bond, and a Yamaguchi macrolactonization to complete the required skeletal framework. Biological studies with the synthesized compounds led to the identification of epothilone analogues 3, 4, 7, 8, 9, and 11 as potent tubulin polymerization promoters and cytotoxic agents with (12R,13S,15S)-cyclopropyl 5-methylpyridine epothilone A (11) as the most powerful compound whose potencies (e.g. IC(50) = 0.6 nM against the 1A9 ovarian carcinoma cell line) approach those of epothilone B. These investigations led to a number of important structure-activity relationships, including the conclusion that neither the epoxide nor the stereochemistry at C12 are essential, while the stereochemistry at both C13 and C15 are crucial for biological activity. These studies also confirmed the importance of both the cyclopropyl and 5-methylpyridine moieties in conferring potent and potentially clinically useful biological properties to the epothilone scaffold.  相似文献

Adding 1% of the metallic elements cerium, lanthanum, and yttrium to graphite rod electrodes resulted in different amounts of the hollow higher fullerenes (HHFs) C76-D2(1), C78-C2v(2), and C78-C2v(3) in carbon-arc fullerene-containing soots. The reaction of trifluoroiodomethane with these and other soluble HHFs at 520-550 degrees C produced 21 new C76,78,84,90(CF3)n derivatives (n = 6, 8, 10, 12, 14). The reaction with C76-D2(1) produced an abundant isomer of C2-(C76-D2(1))(CF3)10 plus smaller amounts of an isomer of C1-(C76-D2(1))(CF3)6, two isomers of C1-(C76-D2(1))(CF3)8, four isomers of C1-(C76-D2(1))(CF3)10, and one isomer of C2-(C76-D2(1))(CF3)12. The reaction with a mixture of C78-D3(1), C78-C2v(2), and C78-C2v(3) produced the previously reported isomer C1-(C78-C2v(3))(CF3)12 (characterized by X-ray crystallography in this work) and the following new compounds: C2-(C78-C2v(3))(CF3)8; C2-(C78-D3(1))(CF3)10 and C(s)-(C78-C2v(2))(CF3)10 (both characterized by X-ray crystallography in this work); C2-(C78-C2v(2))(CF3)10; and C1-C78(CF3)14 (cage isomer unknown). The reaction of a mixture of soluble higher fullerenes including C84 and C90 produced the new compounds C1-C84(CF3)10 (cage isomer unknown), C1-(C84-C2(11))(CF3)12 (X-ray structure reported recently), D2-(C84-D2(22))(CF3)12, C2-(C84-D2(22))(CF3)12, C1-C84(CF3)14 (cage isomer unknown), C1-(C90-C1(32))(CF3)12, and another isomer of C1-C90(CF3)12 (cage isomer unknown). All compounds were studied by mass spectrometry, (19)F NMR spectroscopy, and DFT calculations. An analysis of the addition patterns of these compounds and three other HHF(X) n compounds with bulky X groups has led to the discovery of the following addition-pattern principle for HHFs: In general, the most pyramidal cage C(sp(2)) atoms in the parent HHF, which form the most electron-rich and therefore the most reactive cage C-C bonds as far as 1,2-additions are concerned, are not the cage C atoms to which bulky substituents are added. Instead, ribbons of edge-sharing p-C6(X)2 hexagons, with X groups on less pyramidal cage C atoms, are formed, and the otherwise "most reactive" fullerene double bonds remain intact.  相似文献

The new tantalum(II) complex (eta (6)-C 7H 8)TaCl 2(PMe 3) 2 ( 1) was synthesized by the reduction of TaCl 5 with n-butyllithium in the presence of PMe 3 and cycloheptatriene. Compound 1 adopts a four-legged piano stool structure in which the tantalum center is bound to a eta (6)-cycloheptatriene ring in addition to two chlorides and two phosphine ligands in a transoid arrangement. Treatment of 1 with methyllithium results in a loss of the equivalents of HCl and formation of the eta (7)-cycloheptatrienyl complex (eta (7)-C 7H 7)TaCl(PMe 3) 2 ( 2), whereas treatment of 1 with sodium or sodium borohydride affords small amounts of the eta (5)-cycloheptadienyl complex (eta (5)-C 7H 9)TaCl 2(PMe 3) 2 ( 3). Compound 2 adopts a three-legged piano stool structure; the eta (7)-C 7H 7 ring is fully aromatic and planar. The molecular structure of 3 is similar to that of 1, except for the eta (5) binding mode of the seven-membered ring. Treatment of the previously described sandwich compound (C 5Me 5)Ta(C 7H 7) with allyl bromide affords the tantalum(V) product (C 5Me 5)Ta(C 7H 7)Br ( 4), which reacts with LiAlH 4 to give the tantalum(V) hydride (C 5Me 5)Ta(C 7H 7)H ( 5). Compound 4 also reacts with alkylating agents to generate the methyl, allyl, and cyclopropyl complexes (C 5Me 5)Ta(C 7H 7)Me ( 6), (C 5Me 5)Ta(C 7H 7)(eta (1)-CH 2CHCH 2) ( 7), and (C 5Me 5)Ta(C 7H 7)(c-C 3H 5) ( 8). Compounds 4- 8 all adopt bent sandwich structures in which the dihedral angle between the two carbocyclic rings is 34.9 degrees for the bromo compound 4, 26.6 degrees for the hydride 5, 33.1 degrees for the methyl compound 6, 34.2 degrees for the allyl compound 7, and 37.5 degrees for the cyclopropyl compound 8. A series of 2-benzoimidazol-8-ethoxyquinolines (L1-L6) and the nickel dihalides thereof (C1-C12) were synthesized and characterized by elemental analysis and infrared spectroscopy. The molecular structures of representative complexes were determined by single crystal X-ray diffraction which revealed a distorted pyramidal geometry at nickel. All complexes, when activated with Et(2)AlCl, showed high activities towards ethylene oligomerization and enhanced thermodynamic stability.  相似文献

The equilibrium molecular structure and conformation of 1,5-diazabicyclo[3.1.0]hexane (DABH) has been studied by the gas-phase electron-diffraction method at 20 degrees C and quantum-chemical calculations. Three possible conformations of DABH were considered: boat, chair, and twist. According to the experimental and theoretical results, DABH exists exclusively as a boat conformation of C s symmetry at the temperature of the experiment. The MP2 calculations predict the stable chair and twist conformations to be 3.8 and 49.5 kcal mol(-1) above the boat form, respectively. The most important semi-experimental geometrical parameters of DABH (r(e), A and angle)e), deg) are (N1-N5) = 1.506(13), (N1-C6) = 1.442(2), (N1-C2) = 1.469(4), (C2-C3) = 1.524(7), (C6-N1-C2) = 114.8(8), (N5-N1-C2) = 107.7(4), (N1-C2-C3) = 106.5(9), and (C2-C3-C4) = 104.0(10). The natural bond orbital (NBO) analysis has shown that the most important stabilization factor in the boat conformation is the n(N) --> sigma*(C-C) anomeric effect. The geometry calculations and NBO analysis have been performed also for the bicyclohexane molecule.  相似文献

The Deltadelta (deltaS-deltaR) values for the C-1 methyl 1H signals in the 1H-NMR spectroscopy of the bis-MTPA esters of four synthetic stereoisomers of alkane-6,8-diols, viz., bis-MTPA esters of (6S,8R)-C27- (1a) and C29- (3a) (Deltadelta = -0.05 ppm), (6R,8S)-C27- (2a) and C29- (4a) (Deltadelta = +0.05 ppm), (6S,8S)-C27- (5a) (Deltadelta = -0.01 ppm), and (6R,8R)-C27- (6a) (Deltadelta = +0.01 ppm) alkane-6,8-diols, made it possible to differentiate unequivocally among the four stereoisomers. This allowed the determination of the (6S,8R)-stereochemistry (Deltadelta = -0.05 ppm for the bis-MTPA esters) for the natural C27- and C29-alkane-6,8-diols isolated from the flowers of three Compositae plants, Carthamus tinctorius, Cynara cardanclus, and Taraxacum platycarpum.  相似文献

Reactions of the diphosphine 1,3-C6H4(CH2PH2)2 and fluorous alkenes H2C=CHR(fn)(R(fn)=(CF2)(n-1)CF3; n = 6, 8) at 75 degrees C in the presence of AIBN give the title ligands 1,3-C6H4(CH2P(CH2CH2R(fn))2)2(3-R(fn)) and byproducts 1,3-C6H4(CH3)(CH2P(CH2CH2R(fn))2)(4-R(fn)) in 1 : 3 to 1 : 5 ratios. Workups give -R(fn) in 4--17% yields. Similar results are obtained photochemically. Reaction of 1,3-C6H4(CH2Br)2 and HP(CH2CH2R(f8))2 (5) at 80 degrees C (neat, 1 : 2 mol ratio) gives instead of simple substitution the metacyclophane [1,3-C6H4(CH2P(CH2CH2R(f8))2 CH2-1,3-C(6)H(4)CH(2)P[lower bond 1 end](CH2CH2R(f8))2C[upper bond 1 end]H2](2+)2Br-, which upon treatment with LiAlH(4) yields 3-R(f8)(20%), 4-R(f8), and other products. Efforts to better access 3-R(f8), either by altering stoichiometry or using various combinations of the phosphine borane (H3B)PH(CH2CH2R(f8))2 and base, are unsuccessful. Reactions of 3-R(fn) with Pd(O2CCF3)2 and [IrCl(COE)2]2(COE=cyclooctene) give the palladium and iridium pincer complexes (2,6,1-C6H3(CH2 P(CH2CH2R(fn))(2)(2)Pd(O2CCF3)(10-R(fn); 80-90%) and (2,6,1-C6H3(CH2P(CH2CH2R(f8))2)2)Ir(Cl)(H)(11-R(f8); 29%), which exhibit CF3C6F(11)/toluene partition coefficients of >96 : <4. The crystal structure of 10-R(f8) shows CH2CH2R(f8) groups with all-anti conformations that extend in parallel above and below the palladium square plane to create fluorous lattice domains. NMR monitoring shows a precursor to 11-R(f8) that is believed to be a COE adduct.  相似文献

The crystal and molecular structure of 6b,7,8,8a-tetramethyl-6b,8a-dihydrocyclobut[a]acenaphthylene (I) and 1,2,2a,10b-tetramethyl-2a,10b-dihydrocyclobuta[l]phenanthrene (II) is established by XRD analysis. The C(6b)-C(8a) bond in I is lengthened to 1.603(3) ∢, and the C(2a)-C(10b) bond in II is prolonged to 1.589(6) ∢. The acenaphthene and dihydrophenanthrene frameworks are planar; the rms deviations of atoms from the plane are 0.011 and 0.032 ∢, respectively. The stoichiometric reaction of the salicylaldimine derivatives (L1-L12) with trimethylaluminium afforded the corresponding dimethylaluminium aldiminophenolates (C1-C12), which were fully characterized by NMR spectroscopy and elemental analysis. The molecular structures of the representative complexes C1, C6, and C8 were determined by the single-crystal X-ray diffraction, which revealed distorted tetrahedral geometry at aluminium. Activation of the dimethylaluminium aldiminophenolates for the ring-opening polymerization required one equivalent of BnOH. On the basis of the polymerization results for l-lactide, d-lactide or rac-lactide, higher efficiency was observed for the ROP of d-lactide, and the nature of the ligands present significantly affected the observed catalytic activities and the properties of the resultant polylactides.  相似文献

The C9 position of cinchona alkaloids functions as a molecular hinge, with internal rotations around the C8-C9 (τ(1)) and C9-C4' (τ(2)) bonds giving rise to four low energy conformers (1; anti-closed, anti-open, syn-closed, and syn-open). By substituting the C9 carbinol centre by a configurationally defined fluorine substituent, a fluorine-ammonium ion gauche effect (σ(C-H) → σ(C-F)*; F(δ-)???N(+)) encodes for two out of the four possible conformers (2). This constitutes a partial solution to the long-standing problem of governing internal rotations in cinchonium-based catalysts relying solely on a fluorine conformational effect.  相似文献
