This Letter describes for the first time the synthesis of pyrido[2′,3′:4,5]furo[3,2-d]pyrimidines substituted by a primary or secondary amino group on position 4 of the pyrimidine ring. Application of microwave irradiation technology allowed fast and convenient procedures.  相似文献

In this work, we described an easy preparation of substituted 4-amino-5-cyano-1,3-thiazoles. These compounds have been used as starting materials to obtain two classes of compounds. New substituted [1,3]thiazolo[4,5-e]pyridines were synthesized in one step via Friedländer reaction. Diazotation of 4-amino-5-cyano-1,3-thiazoles afforded 4-chloro[1,3]thiazolo[4,5-d][1,2,3]triazines in one step. The later was substituted by a secondary amine to obtain substituted 4-amino[1,3]thiazolo[4,5-d][1,2,3]triazines.  相似文献

In this work, we described the synthesis of 3-isothiocyanatoselenophenes starting from 3-aminoselenophenes (using thiophosgene). Those compounds were then converted to the corresponding thioureas in the presence of ammonium hydroxide. Cyclization in 2-aminoselenolo[3,2-d]thiazoles was achieved using DDQ. 2-Aminothieno[3,2-d]thiazoles were obtained from 3-isothiocyanatothiophenes using the same pathway.  相似文献

A small library of hitherto unprepared pyrazolo[3,4-d]-4,5-dihydropyrimidin-6-ones was synthesized on a preparative scale. The synthesis starts with a substituted 5-aminopyrazole that reacts with an isocyanate to give the corresponding urea. The latter undergoes a chlorotrimethylsilane-promoted [5+1] cyclocondensation with an aldehyde yielding the title pyrazolo[3,4-d]-4,5-dihydropyrimidin-6-one. Both synthetic steps are high-yielding (74–94%). The intermediates and the target compounds were isolated by simple crystallization. Ketones with the exception of isatin do not react with the open-chain urea intermediates.  相似文献

N,N-Dimethyl-5-formylbarbituric acid 1 reacts with maleimide 2 and phenyl isocyanate/phenyl isothiocyanate 4 under microwave-assisted conditions in the solid phase to afford pyrano[2,3-d]pyrimidines 3 and oxazino[4,5-d]pyrimidines 5 in excellent yields. Under identical conditions, N,N-dimethyl-6-amino-5-formyluracil 6 reacts with 2 and 4 to give pyrido[2,3-d]pyrimidine derivative 7 and pyrimido[4,5-d]pyrimidines 8 in high yields.  相似文献

3-Alkyl-5-phenyl-3H-[1,2,3]triazolo[4,5-d]pyrimidin-7(6H)-ones were prepared by nitrosative cyclization of the appropriate 5,6-diamino-2-phenylpyrimidin-4(3H)-ones with nitrous acid and were subjected to regioselective alkylation with several alkylating agents in aprotic solvent at different temperature. Simultaneous 6-N- and 7-O-alkylation were observed and the regioselectivity varied remarkably with size and shape of the alkylating agents as well as with the reaction temperature. Similarly, N- and O-alkylation as well as selectivity was also observed in the case of 2-phenyl-9-propyl-9H-purin-6(1H)-one. Some of the synthesized compounds showed moderate antiviral and antitumor activities.  相似文献

The pyrazolo[3,4-d][1,2,3]triazin-4-ones 3 and imidazo[4,5-d][1,2,3]triazin-4-ones 4 are analogs structurally related to purines that have showed a wide and significant variety of biological activity. These compounds were synthesized by one-pot diazotization of 5-amino-1H-pyrazole-4-carbonitriles 1 and 5-amino-1H-imidazole-4-carbonitriles 2, respectively.  相似文献

7-Hydroxy-4-methyl-2H-chromen-2-one (2), 7-hydroxy-4,5-dimethyl-2H-chromen-2-one (15) and their some derivatives were synthesized for exploring selected biological screening. The compounds 9 and 13 had shown high degree of cytotoxic activity. Three compound 9, 10 and 13 showed high degree of bactericidal activity amongst the present series.  相似文献

1,3-Dipolar cycloadditions of diazoalkanes to pyridazin-3(2H)-ones 1–7 and pyridazin-3(2H)-thiones 8 and 9 are regioselective producing 3H -pyrazolo[3,4-d]pyridazin-4(5H)-ones 15–19, 27–29 and 34–38 as the major products. In some instances, the isomeric 3H-pyrazolo[3,4-d]pyridazin-7(6H)-ones, such as 20 and 23 were isolated as the minor products. From 3 and 6 the primary 3a,7a-dihydro cycloadducts 25 and 26 , and rearranged 1,2-dihydro intermediate 31 were isolated. From 10 and 1-diazoindane the isomeric exo- and endospiro products 39 and 40 were formed.  相似文献

Sharpless epoxidation of 4-amino-3-methyl-5-styrylisoxazoles 1 resulted in the formation of 3-methyl-5-aryl-4H-pyrrolo[2,3-d]-isoxazoles 4 in a one-step reaction. The reaction initially involves epoxide formation, followed by ring-opening and cyclization. Finally dehydration leads to the title compounds. The pyrrolo[2,3-d]-isoxazoles 4 were also synthesized via an alternative procedure.  相似文献

New aminomethyl derivatives of 3-methyl-4,7-dihydro-3H-imidazo[4,5-d][l,2,3]triazin-4-one were synthesized by Mannich reaction. These Mannich bases is also possible to prepare by reaction of 7(5)-hydroxymethyl-3-methyl-4,7(5)dihydro-3H-imidazo[4,5-d][l,2,3]triazin-4-one with amines. The compounds obtained exist in solutions as mixtures of N5 and N7 isomers, and the fraction of the latter grows with the polarity of the solvent.  相似文献

An efficient approach for the synthesis of 3-alkyl-8-arylamino-1H-imidazo[4,5-g]quinazolin-2(3H)-thiones and 3-alkyl-8-arylamino-1H-imidazo[4,5-g]quinazolin-2(3H)-ones on solid phase has been developed. The synthesis of a 5,7-dichloropyrido[4,3-d]pyrimidine scaffold is described. The chlorine at position 5 can selectively be displaced by different palladium-catalyzed cross-coupling reactions and nucleophilic aromatic substitutions. In the subsequent step, the chlorine at position 7 can be further derivatized. The described synthetic sequence allows for the construction of a diverse pyrido[4,3-d]pyrimidine library with structural variations at positions 5 and 7.  相似文献
