Aβ is a major component of the senile plaques characteristic of Alzheimer disease (AD) and sulfated GAGs such as chondroitin sulfates (CS) have been found in all types of amyloidosis. In this paper, a biochromatographic approach was developed to measure for the first time changes in enthalpy, heat capacity change and copper effect for the binding of Aβ to CS in a wide temperature range. For this, CS was immobilized on a chromatographic support. It was established that this novel CS column was stable during an extended period of time. The thermodynamic data showed that Aβ–CS binding, for low temperature (<10 °C), is enthalpically unfavourable and being dominated by a positive entropy change. This result suggested that dehydration at the binding interface and charge–charge interactions contribute to the Aβ–CS complex formation and a large heat capacity change, ΔC p = ?2.32 kJ mol?1 K?1, was determined. Above 10 °C, the thermodynamic data ΔH and ΔS became negative due to van der Waals interactions and hydrogen bonding which are engaged at the complex interface confirming strong Aβ–CS hydrogen bond networks. Also, for a copper concentration in the range 20–160 μM, it was shown that an increase of the Cu2+ concentration in the medium led an increase of this association classically attributed to salt effect (i.e. hydrophobic bonds) and to ion pair formations between the Cu2+ cation and Aβ to bind to chondroitin sulfate and could thus improve the Aβ aggregation by copper.  相似文献

Structurally complex diazo-containing scaffolds are formed by conjugate addition to vinyl diazonium salts. The electrophile, a little studied α-diazonium-α,β-unsaturated carbonyl compound, is formed at low temperature under mild conditions by treating β-hydroxy-α-diazo carbonyls with Sc(OTf)3. Conjugate addition occurs selectively at the 3-position of indole to give α-diazo-β-indole carbonyls, and enoxy silanes react to give 2-diazo-1,4-dicarbonyl products. These reactions result in the formation of tertiary and quaternary centers, and give products that would be otherwise difficult to form. Importantly, the diazo functional group is retained within the molecule for future manipulation. Treating an α-diazo ester indole addition product with Rh2(OAc)4 caused a rearrangement to occur to give a 2-(1H-indol-3-yl)-2-enoate. In the case of diazo ketone compounds, this shift occurred spontaneously on prolonged exposure to the Lewis acidic reaction conditions.  相似文献

Phosphorylation of proteins by kinases plays an important role in regulating cellular processes including melanin production in the skin cells. Protein kinase C β (PKCβ) is known to be involved in phosphorylating tyrosinase, the key enzyme of melanin production, regulating the skin pigmentation process. In melanogenesis, PKCβ activates the tyrosinase by phosphorylation of its two serine residues. In this study, phosphorylation activity by PKCβ was monitored on a protein chip for the screening of depigmenting agents. As a tyrosinase mimic, 11 or 30 amino acids of the C-terminal of tyrosinase was fused with maltose-binding protein (MBP). After immobilizing the MBP-fused PKCβ substrate peptide on epoxy-treated slide surface, PKCβ reaction mix was applied over the immobilized MBP-fused PKCβ substrate peptide. Phosphorylation was detected with anti-phosphoSer/Thr antibodies, followed by fluorescence-labeled second antibodies. Phosphorylation of MBP-30aa was observed on a protein chip, and this phosphorylation was inhibited by the PKC inhibitor (GF109203X). These results indicate the potential of PKCβ protein chip as a high-throughput screening tool in the screening of depigmenting agents.  相似文献

Tetrakis(alloxycarbonyl)porphyrin and its β-octafluoro-substituted derivatives were synthesized via Lindsey method and transformed to their zinc complexes.Single crystal X-ray structures of corresponding Zn(Ⅱ) porphyrins revealed that β-octafluorination will give more compactness of porphyrin moieties in the crystal structure owing to the hydrogen bonding interactions involving β-fluorine atoms.An unusual six-coordinated Zn(Ⅱ) was found via intramolecular coordination of oxygen atom of meso-substituents with central Zn(Ⅱ).  相似文献

The fatal neurological disorder Alzheimer's disease has been linked to soluble neurotoxic oligomers of amyloid-β (Aβ) peptides. Herein we demonstrate that Cu(1+) ligated within Aβ(42) oligomers (Aβ sequence: DAEFRHDSGYEVHHQKLVFFAEDVGSNKGAIIGLMVGGVVIA) possesses a highly dioxygen sensitive tetrahedral coordination geometry. In this paper, the interaction between squarylium cyanine and porphyrin in chloroform is investigated by absorption and fluorescence spectroscopy. Emphasis has been put on the mechanism of intermolecular energy transfer. The overlap integral J between the absorption spectrum of squarylium cyanine and the fluorescence spectrum of porphyrin was calculated, which reveals that the singlet-singlet energy transfer may occur from porphyrin to squarylium cyanine in solution. In comparison of the observed rate constant [kqII=6.1 ×1013 (mol/L)-1·s-1] for fluorescence quenching of porphyrin by squarylium cyanine with the diffusion rate constant in chloroform [kdif=1.1×1010 (mol/L)-1·s-1] and the rate of energy transfer [ket≤6.7×104 (mol/L)-1·s-1 in the experimentally dilute solutions] estimated from Forster formula, the possibility of energy transfer by electron exchange or/and coulombic mechanism could be excluded. The electronic structure of [Fe(TMP)F(2)], which is formally a one-electron oxidation equivalent above [Fe(III)(TMP)F(2)](-), has been examined in solution by (1)H NMR, UV-Vis, and M?ssbauer spectroscopy. In CD(2)Cl(2)-CD(3)OD solution at 193 K, the pyrrole-H and m-H signals appeared at 128.2 and 116.7 ppm, respectively. The UV-Vis spectrum showed broad absorption bands at 560-680 nm. The M?ssbauer spectrum taken in frozen toluene-methanol solution exhibited a very broad single line from which the IS and QS values were determined by computer simulation to be 0.50 and 0.14 mm s(-1), respectively. On the basis of these results, it was concluded that the one-electron oxidized product of [Fe(TMP)F(2)](-) should be formulated as the iron(III) radical cation [Fe(III)(TMP˙)F(2)], not as iron(IV) porphyrin [Fe(IV)(TMP)F(2)] as previously suggested.  相似文献

In view of the property of porphyrin's accumulation selectively in tumor,the ftorafur was modified by binding a porphyrin block to improve its tumor targeting and reduce its side effects.These novel porphyrin derivatives and metal compounds were synthesized under mild conditions with satisfactory yield,and the constructions of all these new compounds were characterized by UV,IR,MS, ~1H NMR spectra and elementary analysis.Their anticancer activities were evaluated by MTT assay;the results indicated that the anticancer activities of compounds 4a-c were twice as high as that of ftorafur.  相似文献

Coordination of a cyclometallated Pt(II) complex (1) to an amyloid-β peptide was probed by NMR and ESI-MS. Furthermore, EPR showed that binding of 1 to the Cu(II)-amyloid-β species resulted in a reshuffling of the Cu(II) coordination sphere, which was absent or lower for the sister non cyclometallated Pt(II) complexes.  相似文献

To search for more wider absorption and higher charge carriers mobilities materials of polymer solar cell, a series of soluble alternating polyfluorene copolymers were synthesized by palladium-catalyzed Suzuki coupling reaction. Their structures were determined by 1H NMR, IR and UV-vis. And their UV-vis absorption spectra indicated that they had strong absorption over 600 nm spectral range and nearly cover 400-700 nm visible region. The band gaps of copolymers calculated according to cyclic voltammetry (CV) were between 1.96 and 2.03 eV. Polymer:TiO2 bulk-heterojunction films were made from mixtures of polymer and titanium isopropoxide, a precursor for TiO2, via hydrolysis in air overnight. The photoluminescence at 380-800 nm of the blend film of PT-TPP20 (5 mg/mL):Ti(OC3H7)4 (80 μL/mL) (20% volume fraction) was significantly quenched in the 50% Ti(OC3H7)4 blend film, indicating that rapid and efficient separation of photoinduced electron-hole pairs.  相似文献
