Summary Guaran, a neutral polysaccharide, has been used as a buffer modifier to improve the separation of basic proteins and drugs. Migration reproductibility, peak shape and efficiency were improved when 0.1% guaran was added to the buffer. The concentration of guaran, ionic strength, and pH of buffer solution were optimized to obtain the optimum separation of proteins. Possible separation efficiencies of 700,000 plates per meter were obtained for test proteins. The relative standard deviation (% RSD) of the migration time of all test proteins was less than 0.5%. Improved separation of β-blockers was also observed when guaran was added to the buffer.  相似文献

A method using capillary zone electrophoresis has been developed for separation of a series of isomeric benzene polycarboxylic acids. The separation was found to depend largely on the geometry of the acids and their degree of ionization as well as on the buffer pH and composition. The presence of additives to the buffer such as cyclodextrin and cationic surfactants was found necessary for complete resolution of all the isomers.  相似文献

A wide number of chiral selectors have been employed in CE and among them macrocyclic antibiotics exhibited excellent enantioselective properties toward plenteous racemic drugs. Different from macrocyclic antibiotics, the use of lincomycin antibiotics as chiral selectors has not been reported previously. In this study clindamycin phosphate belonging to the group of lincomycin antibiotics is first used as a novel chiral selector for the enantiomeric separations of several racemic basic drugs, which possess high separability, consisting of nefopam, citalopram, tryptophan, chlorphenamine and propranolol. Other basic drugs giving partial enantioseparation include tryptophan methyl ester, metoprolol and atenolol. Clindamycin phosphate possesses advantages such as high solubility and low viscosity in the water and very weak UV absorption. In the course of this work we observed that both migration time and enantioseparation were influenced by several parameters such as pH of the BGE, clindamycin concentration, capillary temperature, applied voltage and organic modifier. The optimum pH that was in the neutral or weak basic region but varied among drugs, a low capillary temperature and a clindamycin concentration of 60 or 80 mM are recommended as the optimum conditions for chiral separation of these drugs. Moreover, comparison of the influences of the studied parameters was further investigated by means of Statistical Product and Service Solutions in this paper.  相似文献

Carbon nanoparticles obtained from the flame of an oil lamp were examined by means of capillary electrophoresis. The influence of buffer composition on the separation of the mixture of negatively charged carbon nanoparticles was studied by varying buffer selection, pH, and concentration. The electrophoretic pattern was affected by both the co- and counter-ion in the buffer solution, influencing selectivity and peak shape. The capillary electrophoretic separations at different pH revealed species with large electrophoretic mobilities under a wide range of pH. The mobility of selected species in the mixture of nanoparticles showed a strong dependence upon the solution ionic strength. The mobility of these nanoparticles as a function of ionic strength was compared to classical electrokinetic theory, suggesting that under the experimental conditions utilized, the species are small, highly charged particles with appreciable zeta potentials, even at low pH.  相似文献

A new method involving headspace single-drop microextraction (SDME) and capillary electrophoresis (CE) is developed for the preconcentration and determination of ammonia (as dissolved NH3 and ammonium ion). An aqueous microdrop (5 μL) containing 1 mmol/L H3PO4 and 0.5 mmol/L KH2PO4 (as internal standard) was used as the acceptor phase. Common experimental parameters (sample and acceptor phase pH, extraction temperature, extraction time) affecting the extraction efficiency were investigated. Proposed SDME-CE method provided about 14-fold enrichment in about 20 min. The calibration curve was linear for concentrations of NH4+ in the range from 5 to 100 μmol/L (R2 = 0.996). The LOD (S / N = 3) was estimated to be 1.5 μmol/L of NH4+. Such detection sensitivity is high enough for ammonia determination in common environmental and biological samples. Finally, headspace SDME was applied to determine ammonia in human blood, seawater and milk samples with spiked recoveries in the range of 96-107%.  相似文献

The combination of capillaries with different internal diameters was used to accelerate the separation of enantiomers in capillary electrophoresis. Separation of R ,S‐1,1′‐binaphthalene‐2,2′‐diyl hydrogen phosphate using isopropyl derivative of cyclofructan 6 was studied as a model system. The best separation conditions included 500 mM sodium borate pH 9.5 with 60 mM concentration of the chiral selector. Separation lasted approx. 1.5 min using the combination of 50 and 100 μm id capillaries of 9.7 cm and 22.9 cm, respectively. It allowed approx. 12‐fold acceleration in comparison to the traditional long‐end separation mainly due to the higher electroosmotic flow generated in the connected capillaries.  相似文献

A CE protocol was developed for the identification and separation of inorganic polysulfides simultaneously with other inorganic sulfur-bearing species coexisting in aqueous hydrosulfide/sulfur solutions. The electrophoretic separation of thiosulfate, sulfate, hydrosulfide, sulfite, tetrathionate, and polysulfides was achieved at pH values between 8.2 and 12.2. The peaks attributed to the polysulfide species were strongly sensitive to pH. CE analysis of hydrosulfide/sulfur solutions at different pH values permitted possible identification of two forms of polysulfides: S4(2-) and S3(2-). Upon exposure to air at ambient temperature, thiosulfate was the main oxidation product of hydrosulfide/sulfur solutions mainly in the first 60 min, when hydrosulfide was rapidly consumed. Analysis of the oxidation reaction products provided retrospectively tentative evidence that the peaks separated and identified as tri- and tetrasulfide may be ascribed to polysulfides.  相似文献

The electro‐osmotic flow, a significant factor in capillary electrophoretic separations, is very sensitive to small changes in structure and surface roughness of the inner surface of fused silica capillary. Besides a number of negative effects, the electro‐osmotic flow can also have a positive effect on the separation. An example could be fused silica capillaries with homogenous surface roughness along their entire separation length as produced by etching with supercritical water. Different strains of methicillin‐resistant and methicillin‐susceptible Staphylococcus aureus were separated on that type of capillaries. In the present study, fused‐silica capillaries with a gradient of surface roughness were prepared and their basic behavior was studied in capillary zone electrophoresis with UV‐visible detection. First the influence of the electro‐osmotic flow on the peak shape of a marker of electro‐osmotic flow, thiourea, has been discussed. An antifungal agent, hydrophobic amphotericin B, and a protein marker, albumin, have been used as model analytes. A significant narrowing of the detected zones of the examined analytes was achieved in supercritical‐water‐treated capillaries as compared to the electrophoretic separation in smooth capillaries. Minimum detectable amounts of 5 ng/mL amphotericin B and 5 μg/mL albumin were reached with this method.  相似文献

A rapid, simple, and practical method for the determination of four of the most used thyreostatic drugs (methimazole, 2‐thiouracil, 6‐methyl‐2‐thiouracil, and 6‐propyl‐2‐thiouracil) using CE coupled to electrochemiluminescence detection has been established, based on the electrochemiluminescence enhancement of tris(2,2‐bipyridyl)ruthenium(II) with these analytes. Parameters that affect separation and detection were optimized. Under the optimum experimental conditions, the four analytes could be well separated within 11 min at the separation voltage of 16 kV in a running solution containing 20 mM phosphate buffer (pH 9.0) and 1.0 × 10?4 M Ru(bpy)32+, with a solution of 20 mM phosphate buffer (pH 12.0) containing 1.0 × 10?4 M Ru(bpy)32+ in the electrochemiluminescence detection cell. The detection limits for methimazole, 6‐methyl‐2‐thiouracil, 6‐propyl‐2‐thiouracil, and 2‐thiouracil were 0.1, 0.05, 0.05, and 0.01 μM, respectively. The proposed method was applied to analyze these drugs in spiked animal feed samples. The recoveries were 88.2~99.0 and 86.4~98.7% for the intraday and interday analyses, respectively. The RSDs were 2.7~4.8 and 1.8~5.0% for the intraday and interday analyses, respectively. The results demonstrate that the proposed method has promising applications in the detection of thyreostatic drugs in animal feeds.  相似文献

Here we describe a capillary electrophoretic method for the separation of double-stranded oligonucleotides (ds-ODNs) ranging from 16-20 bp with 2 bp resolution using a low concentration of poly(ethylpyrrolidine methacrylate-co-methyl methacrylate) (PEPyM-co-PMMA) copolymer physically adsorbed to a capillary surface. Contrary to traditional DNA separations, we show that the ds-ODN with the highest molecular size eluted first and propose that this phenomena is due to a screening effect by the PEPyM-co-PMMA coating on the smaller ds-ODNs negative charge during elution. Key to the performance of this separation was a sample preparation time of less than 1 h and analysis time of 40 min. Repeatability of intraday migration time for the mixtures was typically < 1% relative standard deviation (n = 3). In addition, we demonstrate that the coating has an acceptable capillary lifetime of over 70 injections.  相似文献

The migration behaviour of isoquinoline, quinoline, and methyl derivatives of quinoline in different capillary electrophoretic modes has been systematically investigated. Optimised separation conditions were established by varying the key parameters (solvent, pH, temperature, surfactant concentration, core phase) for aqueous and non‐aqueous capillary zone electrophoresis (NACE), micellar electrokinetic chromatography (MEKC) with anionic or non‐ionic micelles (SDS, Brij 35), and microemulsion electrokinetic chromatography (MEEKC) with charged or uncharged microemulsion droplets. A separation of all quinolines could be achieved by MEEKC with charged droplets, by MEKC or by formamide‐based NACE. Comparing the separations with respect to separation selectivity, substantial changes in migration order could be observed between the different techniques. Regarding separation efficiency, the number of theoretical plates and limits of detection (LOD) have been compared. The best LODs were achieved using SDS as surfactant in MEKC, followed by MEEKC.  相似文献

A capillary electrophoretic method for resolving selected imidazolium ionic liquid cations is reported. The method, in which citric buffer is used as the running electrolyte, is simple and reproducible. The separation of a standard mixture is in linear accordance with the relative molecular mass (M(r)) of solutes regardless of the type of substitution (alkyl or aryl). The theoretical prediction of compounds as yet not analyzed is therefore possible; however, cations with identical molecular masses are inseparable with this method. Nevertheless, the method's quantitative analytical performance was excellent. The paper also discusses the applicability of a method for tracking the photodegradation kinetics of an exemplary ionic liquid.  相似文献

Nowadays, macrocyclic antibiotics are presenting an increasing number of enantioseparation applications. The macrocyclic antibiotics used as chiral selectors in capillary electrophoresis (CE) include the ansamycins and the glycopeptides. The macrolides, another important class of macrocyclic antibiotics, have been reported as a new type of chiral selectors recently. In this study, clarithromycin lactobionate (CL), belonging to the group of macrolide antibiotics, was first investigated for its potential as a novel chiral selector in CE for enantiomeric separation of several basic drugs. As observed, CL allowed excellent separation of the enantiomers of metoprolol, atenolol, propranolol, bisoprolol, esmolol, ritodrine, and amlodipine, as well as partial enantioresolution of labetalol and nefopam. In addition, CL possesses advantages such as high solubility and low viscosity in the solvent and very weak UV absorption. In the course of this study, it was found that both migration times and enantioseparation of the basic drugs were influenced by several experimental parameters, e.g. selector concentration, the composition and pH of the BGE, the type and concentration of organic modifier, and applied voltage. Thus, the effects of these factors were systematically investigated, and satisfactory enantioseparations of the studied drugs were achieved at the buffer pH range of 7.3–7.5 using 12.5 mM borax buffer with 50% v/v methanol, 60 mM CL, and 20 kV applied voltage. Moreover, comparison of the influences of the studied parameters was further investigated by means of Statistical Product and Service Solutions (SPSS) in this article.  相似文献

A novel peptide containing antimicrobial sequence and gelatinase cleavage sites was designed for Staphylococcus aureus detection. Since Staphylococcus aureus could secrete gelatinase, the fluorescein labeled peptide GKRWWKWWRRPLGVRGC could be recognized and cleaved. The obtained products were able to be analyzed by capillary electrophoresis with fluorescence detection. To explore the effect of Staphylococcus aureus concentration on enzyme digestion ability of peptide, Staphylococcus aureus with different concentrations were incubated with the peptide. Results indicated that capillary electrophoretic method was efficient for determining Staphylococcus aureus content. Compared with traditional approaches for Staphylococcus aureus detection, capillary electrophoresis possessed higher efficiency, enhanced sensitivity, and low sample consumption. Moreover, the proposed peptide also presented desirable antimicrobial activity. It suggested that the novel antimicrobial peptide used in this research opens a new path of detecting Staphylococcus aureus by capillary electrophoretic method.  相似文献

A capillary electrophoretic (CE) method was developed for the separation of diastereoisomers of a new human immunodeficiency virus (HIV) protease inhibitor TMC114. In total 16 isomers of this drug have been synthesized (eight pairs of enantiomers). We succeeded in the separation of the eight diastereoisomers, but no enantiomers could be separated. Because of the high similarity and water-insolubility of these isomers, the separation is a real challenge. Different CE modes were tried out: capillary zone electrophoresis (CZE), nonaqueous capillary electrophoresis (NACE), micellar electrokinetic capillary chromatography (MEKC), and microemulsion electrokinetic capillary chromatography (MEEKC). Only MEEKC offered resolution of these compounds.  相似文献

Summary Capillary electrophoresis has been used to separate inorganic (Hg2+) and organic (methyl-, ethyl-, and phenylmercury) mercury compounds as their cysteine complexes. The optimized electrophoretic separation was performed in fused-silica capillary tubing at 25 kV with 25mm sodium borate buffer (pH 9.3). Identification and quantification of the mercury species at mg L−1 levels was achieved by use of UV detection at 200 nm. The relative standard deviation (n=10) ranged from 0.38 to 0.51% for migration times and from 0.43 to 2.94% for corrected peak areas. Good recovery (>90%) was obtained for all four mercury species in surface waters, and for inorganic mercury and methylmercury in five- to tenfold diluted biofluids (urine, saliva, and cerebrospinal fluid). TheLOQ values obtained were too high to be useful for determination of mercury species in real samples. Capillary gel electrophoresis (CGE) has been used for protein separation for more than two decades. Due to the technology advancement, current CGE methods are becoming more and more robust and reliable for protein analysis, and some of the methods have been routinely used for the analysis of protein-based pharmaceuticals and quality controls. In light of this progress, we survey 147 papers related to CGE separations of proteins and present an overview of this technology. We first introduce briefly the early development of CGE. We then review the methodology, in which we specifically describe the matrices, coatings, and detection strategies used in CGE. CGE using microfabricated channels and incorporation of CGE with two-dimensional protein separations are also discussed in this section. We finally present a few representative applications of CGE for separating proteins in real-world samples.  相似文献
