Summary Microemulsion Electrokinetic Chromatography (MEEKC) has previously been reported to be useful for the separation of a range of hydrophobic solutes. The previous reports had tended to show relatively long separation times. In this report the separation and operation conditions were optimized and a method was successfully employed in a range of novel applications. High efficiencies, rapid separations and high repeatability were demonstrated. The applications developed included a range of both neutral and charged compounds. Both water-soluble and water-insoluble compounds were also well separated. The novel applications developed included analysis of basic drugs, various aromatic acids, a range of neutral aromatics, profiling of ink components, direct injection of urine for profiling purposes and the analysis of a range of amino acids and their derivatives. The method could also be used for hydrophobicity measurements of the resolved solutes based upon their migration times. This report clearly shows that MEEKC can be widely applied to a range of compounds and is especially useful for complex mixtures containing solutes with varying charge and hydrophobicity.  相似文献

A rapid screening assay for the determination of octanol-water partition coefficients (log P(OW)) of pharmaceuticals was developed by using pressure-assisted microemulsion electrokinetic chromatography (MEEKC). The microemulsion system contains 50 mM sodium dodecyl sulfate, 0.87 M l-butanol, 82 mM heptane, and 50 mM borate-phosphate (2:3) at pH 10. Ten standard compounds with known log P(OW) values from -0.26 to 4.88 were used for constructing the calibration curve of log P(OW) against the MEEKC retention factor, log k. The log P(OW) values of the compounds were calculated based on the log k values measured by MEEKC and the slope and intercept of the calibration curve. For 13 literature and 32 Roche compounds, about 90% of the log P(OW) values measured by MEEKC are within 0.5 log units of the values from the literature and potentiometric titration. The throughput is about 2 samples/h using +20 kV voltage plus 5 mbar air pressure for separation. This MEEKC method is applicable for log P(OW) screening of weakly basic, weakly acidic, and neutral pharmaceuticals with log P(OW) = 0-5 and pKa < or = 10.  相似文献

In this study, microemulsion electrokinetic chromatography (MEEKC) and micellar electrokinetic chromatography (MEKC) were compared for their abilities to separate and detect thirteen phenolic compounds (syringic acid, p-coumaric acid, vanillic acid, caffeic acid, gallic acid, 3,4-dihydroxybenzoic acid, 4-hydroxybenzoic acid, (+)-catechin, (-)-epigallocatechin, (-)-epicatechin gallate, (-)-epigallocatechin gallate, (-)-epicatechin, and (-)-gallocatechin), and two other ingredients (caffeine and theophylline) in teas and grapes. Separation of phenolic compounds was improved by changing the SDS concentration for MEEKC, but the SDS concentration rarely affected the resolution for MEKC. Organic modifier (acetonitrile or methanol) was found to markedly influence the resolution and selectivity for both MEEKC and MEKC systems. In addition, a higher voltage and a higher column temperature improved the separation efficiency without any noticeable reduction in resolution for MEEKC whereas they caused a poor resolution for the MEKC system. Although separations with baseline resolution were achieved by the optimized MEEKC and MEKC methods, the separation selectivity resulting from the proposed MEEKC method was completely different from that of MEKC.  相似文献

Microemulsion electrokinetic chromatography (MEEKC) and micellar electrokinetic chromatograpy (MEKC) were compared for their abilities to separate and detect ten similar benzophenones, which are commonly used as UV filters in various plastic and cosmetic products. Sodium dodecyl sulfate (SDS) concentration and column temperature rarely affected separation resolution for MEEKC, but separation of benzophenones could be improved by changing the SDS concentration and column temperature for MEKC. Buffer pH and ethanol (organic modifier) were found to markedly influence the separation selectivity for both MEEKC and MEKC systems. In addition, a higher electric voltage improved the separation efficiency without a noticeable reduction in separation resolution for MEEKC, whereas it caused a poor separation resolution for the MEKC system.  相似文献

In this study, separation and determination of nine preservatives ranging from hydrophilic to hydrophobic properties, which are commonly used as additives in various pharmaceutical and cosmetic products, by micellar electrokinetic chromatograpy (MEKC) and microemulsion electrokinetic chromatography (MEEKC) were compared. The effect of temperature, buffer pH, and concentration of surfactant on separation were examined. In MEKC, the separation resolution of preservatives improved markedly by changing the sodium dodecyl sulfate concentration. Temperature and pH of running buffers were used mainly to shorten the magnitude of separation time. However, in order to detect all preservatives in a single run in a MEEKC system, a microemulsion of higher pH was needed. The separation resolution was improved dramatically by changing temperature, and a higher concentration of SDS was necessary for maintaining a stable microemulsion solution, therefore the separation of the nine preservatives in MEEKC took longer than in MEKC. An optimum MEKC method for separation of the nine preservatives was obtained within 9.0 min with a running buffer of pH 9.0 containing 20 mM SDS at 25 degrees C. A separation with baseline resolution was also obtained within 16 min using a microemulsion of pH 9.5 which composed of SDS, 1-butanol, and octane, and a shorter capillary column at 34 degrees C. Finally, the developed MEKC and MEEKC methods determined successfully preservatives in various cosmetic and pharmaceutical products.  相似文献

Microemulsions are used in a similar way as micellar solutions are used for separations in capillary electrophoresis. Within the last approximately six years, a number of papers have appeared in the literature in which the separation characteristics of microemulsion electrokinetic chromatography (MEEKC) have been investigated. The total number of papers dealing with MEEKC is now close to 100. One of the major fields of application for MEEKC has been the characterization of the lipophilicity of chemical substances but the technique has also been applied for the analysis of substances within the fields of natural products, pharmaceuticals, vitamins, peptides, proteins, and nucleic acid bases as well as nucleosides. An overview of the applications is given together with the microemulsions used for each application.  相似文献

Compared to MEKC, the presence of a water-immiscible oil phase in the microemulsion droplets of microemulsion EKC (MEEKC) gives rise to some special properties, such as enhanced solubilization capacity and enlarged migration window, which could allow for the improved separation of various hydrophobic and hydrophilic compounds, with reduced sample pretreatment steps, unique selectivities and/or higher efficiencies. Typically, stable and optically clear oil-in-water microemulsions containing a surfactant (SDS), oil (octane or heptane), and cosurfactant (1-butanol) in phosphate buffer are employed as separation media in conventional MEEKC. However, in recent years, the applicability of reverse MEEKC (water-in-oil microemulsions) has also been demonstrated, such as for the enhanced separation of highly hydrophobic substances. Also, during the past few years, the development and application of MEEKC for the separation of chiral molecules has been expanded, based on the use of enantioselective microemulsions that contained a chiral surfactant or chiral alcohol. On the other hand, the application of MEEKC for the characterization of the lipophilicity of chemical substances remains an active and important area of research, such as the use of multiplex MEEKC for the high-throughput determination of partition coefficients (log P values) of pharmaceutical compounds. In this review, recent applications of MEEKC (covering the period from 2003 to 2005) are reported. Emphases are placed on the discussion of MEEKC in the separation of chiral molecules and highly hydrophobic substances, as well as in the determination of partition coefficients, followed by a survey of recent applications of MEEKC in the analysis of pharmaceuticals, cosmetics and health-care products, biological and environmental compounds, plant materials, and foods.  相似文献

A multiplexed capillary electrophoresis (CE) system equipped with 96 channels was evaluated for high-throughput screening in drug discovery by microemulsion electrokinetic chromatography (MEEKC). Method transfer from a single channel to a multichannel CE system is described. Loss of efficiency and reduced migration times could be elucidated to the poor efficacy in Joule heat dissipation by forced air cooling in the multiarray system compared to liquid cooling in the single channel instrument. On the other hand, only 48 channels could actually be used because of the maximum total current of 3 mA. Precision data remained below 8% and 9% for migration times and peak areas, respectively. Some UV-detector cross-talk interference between neighboring capillary channels was noted. Impurities at 0.5% compared to the main peak (100%) could be detected with the multiplexed system which is 10 times lower compared to the single capillary system. Higher efficiency and improved figures of merit (absolute sensitivity and no cross-talk interferences) were obtained by using an array of only 24 capillaries.  相似文献

In this study, on-line sample concentration methods, which coupled field-amplified sample injection and sweeping technology with MEEKC, were used to detect and analyze eight common penicillin antibiotics (nafcillin, dicloxacillin, ampicillin, oxacillin, penicillin V, cloxacillin, penicillin G, and amoxicillin). During the optimization of field-amplified sample injection-sweeping MEEKC, the composition of sample matrix and the length of acidic plug were found to be the predominant influences for penicillin stacking. Both zwitterionic ampicillin and amoxicillin could only be stacked through cation-selective-exhaustive-injection sweeping, whereas the other six penicillin compounds were found to be concentrated by anion-selective-exhaustive-injection sweeping. Hence, in order to simultaneously concentrate the eight penicillins in a single-run sweeping step, a combination of successive anion- and cation-selective injections was used. When compared with previous CE-UV methods, the proposed on-line concentration MEEKC method provided better detection sensitivity and faster separation for these penicillins either in single ion-selective injection or in successive anion-/cation-selective injection where the LODs were in the range of 0.2-2.8 microg/L and 0.5-5.8 microg/L, respectively.  相似文献

采用微乳电动毛细管色谱法(MEEKC)建立不同来源掌叶大黄的指纹图谱,为其质量控制提供依据。使用未涂层石英毛细管柱(75μm×65 cm,有效长度55 cm),采用的缓冲液为w(SDS)∶w(正丁醇)∶w(正辛烷)∶w(10 mmol/L硼砂溶液)=3.3∶6.6∶0.8∶89.3并通过添加10%的乙腈组成的O/W型微乳体系,分离电压18 kV,检测波长280 nm。结果通过聚类分析和相似度分析处理,14个掌叶大黄样品初步分为三类:道地药材,市售一般品和次品。本方法可用于掌叶大黄质量评价的有效手段,为MEEKC在中药指纹图谱研究中开辟了新的应用途径。  相似文献

Microemulsion electrokinetic chromatography (MEEKC) is an electrodriven separation technique. Separations are achieved using microemulsions which are nanometre-sized oil droplets suspended in aqueous buffer. The surface tension between the oil and water components is reduced by covered the oil droplet with an anionic surfactant such as sodium dodecyl sulphate and a co-surfactant such as a short-chain alcohol. This review summarises the various microemulsion types and compositions that have been used in MEEKC. The effects of key operating variables such as pH and temperature are also described. The application areas of MEEKC are also described in some detail. MEEKC has been applied to a wide range of water-soluble and insoluble both charged and neutral compounds. Examples are described which include analysis of derivatised sugars, proteins, pesticides and a wide range of pharmaceuticals. At present there are only a limited number of publications describing the use of MEEKC but it is anticipated that this number will increase rapidly in the near future as more awareness of the separation possibilities that MEEKC presents increases.  相似文献

In this work, the influences of ionic liquid (IL) as a modifier on microemulsion microstructure and separation performance in MEEKC were investigated. Experimental results showed that synergetic effect between IL 1‐butyl‐3‐methylimidazolium tetrafluoro‐borate (BmimBF4) and surfactant SDS gave a decreased CMC. With increment of IL in microemulsion, negative ζ potential of the microdroplets reduced gradually. The influence of IL on the dimensions of microdroplet was complicated. At BmimBF4 less than 8 mM, IL made microemulsion droplet smaller in size. While at BmimBF4 more than 10 mM, the size increased and reached to a maximum value at 12 mM, where the microdroplets were larger than that without IL. After that, the micreodroplet size decreased again. Relative fluorescence intensity of the first vibration band of pyrene to the third one (I1/I3) enhanced as IL was added to microemulsion, which indicated that this addition increased environmental polarity in the inner core of microdroplets. Prednisone, hydrocortisone, prednisolone, hydrocortisone acetate, cortisone acetate, prednisolone acetate, and triamcinolone acetonide were analyzed with MEEKC modified with IL to evaluate the separation performance. Cortisone acetate and prednisolone acetate could not be separated at all in typical microemulsion. The seven analytes could be separated by the addition of 10 mM BmimBF4 into the microemulsion system. The method has been used for analysis of corticosteroids in cosmetic samples with simple extraction; the recoveries for seven analytes were between 86 and 114%. This method provides accuracy, reproducibility, pretreatment simplicity, and could be applied to the quality control of cosmetics.  相似文献

A microemulsion electrokinetic chromatography (MEEKC) method has been developed and validated for the determination of 4-hydroxybenzoates and their impurities. These materials are commonly known as parabens and are widely used as preservatives in foods, cosmetics and pharmaceuticals. The method was shown to be selective and quantitative for the methyl, ethyl, propyl and butyl esters of 4-hydroxybenzoic acid. An internal standard, 4-hydroxyacetophenone, was employed to improve injection precision and detector linearity. In addition, 4-hydroxybenzoic acid, the major degradent, could also be monitored at the 0.1% (m/m) level. The method was successfully validated for assay and detection of the impurities in 4-hydroxybenzoic acid methyl ester and 4-hydroxybenzoic acid propyl ester samples and for the determination of 4-hydroxybenzoic acid methyl ester in a liquid pharmaceutical formulation. The determination of paraben content by MEEKC in a liquid sample was consistent with HPLC analysis. This work is the first reported validated MEEKC method and shows that the methodology can be successfully implemented into routine quality control testing.  相似文献

Electrokinetic chromatography is a new type of analytical separation method which belongs to the group of high performance capillary electrophoretic techniques but whose separation principle is based on that of chromatography. The solute distributes itself between a carrier and the surrounding medium. The carrier, which corresponds to the stationary phase in conventional chromatography, can be transported by electrophoresis with a different velocity from the surrounding medium. The separation is achieved by the differential solute distribution and the differential migration of the carrier. The charged molecules or charged molecular aggregates are employed as the carrier. Various kinds of carriers are available for electrokinetic chromatography along with different partition mechanisms. Among them, micellar electrokinetic chromatography, which employs an ionic micelle as a carrier, has become the most popular method because of its unique and attractive characteristics as well as the separating capability of electrically neutral or nonionic solutes in comparison with capillary zone electrophoresis. The present paper describes the principle, separation characteristics and its application to the analysis of pharmaceuticals.  相似文献

Retention index (I), rather than retention factor (k), was found to be a more reasonable parameter for comparison of the relative affinity of disubstituted benzenes in MEEKC and MEKC, due to independent of I with the SDS surfactant concentration. MEKC and MEEKC may give similar or different I values, depending on types of moieties. With known I and Kow for alkylbenzenes as references in MEKC and MEEKC, the values of Kow for disubstituted benzenes can be estimated from the observed I values, where Kow is the octanol–water distribution constant. In addition, a group additive approach can be used to predict I for disubstituted benzenes with different moieties from the average observed I for the disubstituted benzenes with same moieties. However, electronic effects and/or intramolecular interaction may result in the different observed I from prediction.  相似文献

A cosurfactant such as a short-chain alcohol is generally used to stabilize the microemulsion. This review summarizes the various microemulsion types and compositions that have been used in MEEKC. The effects of key-operating variables such as surfactant type and concentration, cosurfactant type and concentration, buffer pH and type, oil type and concentration, use of organic solvent and cyclodextrin additions, and temperature are described. A microemulsion electrokinetic chromatography (MEEKC) method was developed to analyze and detect eight food colorants (tartrazine, fast green FCF, brilliant blue FCF, allura red AC, indigo carmine, sunset yellow FCF, new coccine, and carminic acid), which are commonly used as food additives in various food products. The effects of sodium dodecyl sulfate (SDS) surfactant, organic modifier, cosurfactant, and oil were examined in order to optimize the separation. The amount of organic modifier (acetonitrile) and SDS surfactant were determined as apparent influences on the separation resolution while the type of oil and cosurfactant rarely affected the separation selectivity of the eight colorants. A highly efficient MEEKC separation method, where the eight colorants were separated with baseline resolution within 14 min, was achieved by using a microemulsion solution of pH 2.0 containing 3.31% SDS, 0.81% octane, 6.61% 1-butanol, and 10% acetonitrile. This optimal MEEKC method has a higher separation efficiency and similar detection limit when compared to conventional capillary electrophoresis (CE) method. Furthermore, a sample pretreatment is rarely needed when this MEEKC technique is used to analyze colorants in food products, whereas a suitable sample pretreatment (for example solid-phase extraction) has to be employed prior to CE separation in order to eliminate matrix interferences resulting from the constituents of the food sample.  相似文献

Separations of human urine by microemulsion electrokinetic chromatography (MEEKC) and micellar electrokinetic capillary chromatography (MEKC) with respect to resolution, migration times and efficiencies were optimized and compared. The optimised MEEKC and MEKC methods were simple and fast, both of which are excellent characteristics for the complex separations required in clinical and biomedical studies. However, resolution in MEKC was significantly greater than in MEEKC although migration times were 30% faster for the optimised MEEKC method. In addition, a faster analysis method (short-end injection) specifically for routine screening purposes was also investigated. With both MEEKC and MEKC modes, this provided short separations (less than 4 min for urine) with no major compromise in resolution. In conclusion, we found that MEEKC offered no real advantage over MEKC for urine analysis.  相似文献
