In Vitro Binding of Furadan to Bovine Serum Albumin   总被引:1,自引:0,他引:1  
Under physiological conditions, the interaction between furadan (FRD) and bovine serum albumin (BSA) was investigated by spectroscopy including fluorescence emission, UV-visible absorption, scattering, circular dichroism (CD) spectra, synchronous and three-dimensional fluorescence spectra. The observed binding constant K b and the number of binding sites n were determined by the fluorescence quenching method. The distance r between donor (BSA) and acceptor (FRD) was obtained according to the Förster theory of non-radiation energy transfer. The enthalpy change (ΔH θ ), Gibbs energy change (ΔG θ ) and entropy change (ΔS θ ) at four different temperatures were calculated. The process of binding was proposed to be a spontaneous process since the ΔG θ values were negative. The positive ΔS θ and ΔH θ values indicated that the interaction of FRD and BSA was driven mainly by hydrophobic interactions. The addition of FRD to BSA solutions leads to enhancement in scattering intensity, exhibiting the formation of an aggregate in solution. CD spectra, synchronous and three-dimensional fluorescence spectra were used to measure the structural change of BSA molecules with FRD present.  相似文献

Fluorescence spectroscopy, Fourier transform infrared (FT‐IR) spectroscopy, circular dichroism (CD) and FT‐Raman spectroscopy were employed to analyze the binding of the asiatic acid (AA) to bovine serum albumin (BSA) under simulative physiological conditions. Fluorescence data revealed that the fluorescence quenching of BSA by AA was the result of the formation of BSA‐AA complex. The fluorescence quenching mechanism of BSA by AA was a static quenching procedure. According to the Van′t Hoff equation, the thermodynamic parameters enthalpy change (ΔH0) and entropy change (ΔS0) for the reaction were evaluated to be ?12.55 kJ·mol?1 and 67.08 kJ·mol?1, respectively, indicating that hydrophobic and electrostatic interactions played a major role in stabilizing the complex. The influence of AA on the conformation of BSA has also been analyzed on the basis of FT‐IR, CD and FT‐Raman spectra.  相似文献

The interaction between glyoxal-bis-(2-hydroxyanil) (GBH) and bovine serum albumin (BSA) was studied by spectroscopic methods including fluorescence spectroscopy, circular dichroism (CD) and UV–visible absorption spectra. The mechanism for quenching the fluorescence of BSA by GBH is discussed. The number of binding sites n and observed binding constant K b were measured by the fluorescence quenching method. The thermodynamic parameters ΔH θ , ΔG θ , and ΔS θ were calculated at different temperatures and the results indicate that hydrogen bonding and van der Waals forces played major roles in the reaction. The distance r between the donor (BSA) and acceptor (GBH) molecules was obtained according to Förster’s theory of non-radiation energy transfer. Synchronous fluorescence and three-dimensional fluorescence spectra were used to investigate the structural change of BSA molecules that occur upon addition of GBH, and these results indicate that the secondary structure of BSA molecules is changed by the presence of GBH.  相似文献

The interaction between 2,4-dichlorophenol (DCP) and bovine serum albumin (BSA) was investigated by fluorescence spectroscopy combined with UV-vis absorption and circular dichroism (CD) spectroscopy under simulative physiological conditions. The experiment results show that the fluorescence intensity of BSA is dramatically decreased owing to the formation of a DCP–BSA complex. The corresponding effective quenching constants (K a) between DCP and BSA at four different temperatures (292, 298, 304 and 310 K) were determined to be 10.08×104, 9.082×104, 8.177×104, and 7.260×104 L?mol?1, respectively. The thermodynamics parameters enthalpy change (ΔH) and entropy change (ΔS) were calculated to be ?13.64 kJ?mol?1 and 49.08 J?mol?1?K?1, which suggested that hydrophobic interaction was the predominant intermolecular force. Site marker competitive experiments indicated that the binding of DCP to BSA primarily takes place in subdomain IIA. The binding distance (r) between DCP and the tryptophan residue of BSA ias 4.09 nm according to Förster’s theory of non-radioactive energy transfer. The conformational investigation demonstrated that the presence of DCP decreased the α-helical content of BSA and induced a slight unfolding of the polypeptides of protein, which confirmed the occurrence some micro environmental and conformational changes of BSA molecules.  相似文献

采用荧光光谱、紫外光谱、CD光谱法研究了K2Cr2O7与牛血清白蛋白(BSA)的相互作用。实验结果表明,铬(Ⅵ)使BSA的紫外吸收降低,峰位红移,表明铬(Ⅵ)与BSA发生较强的相互作用;铬(Ⅵ)酸根离子与BSA形成基态复合物导致BSA内源荧光猝灭,猝灭机理主要为静态猝灭。测定了不同温度下该反应的热力学参数,ΔGθ0,ΔHθ和ΔSθ分别为-12.60kJ/mol和56.60J/(mol.k),表明上述作用过程是一个熵增加、自由能降低的自发分子间作用过程,铬(Ⅵ)酸根离子与BSA之间以静电作用力为主;非辐射能量转移机理确定了铬(Ⅵ)与牛血清白蛋白中色氨酸残基之间的距离r=2.85nm;同步荧光和CD光谱研究表明,铬(Ⅵ)使BSA的二级结构发生改变,α-螺旋含量降低,色氨酸残基所处微环境的极性减小。  相似文献

The interaction between N‐(4‐ethoxyphenyl)‐N′‐(4‐antipyrinyl)thiourea (EPAT) and bovine serum albumin (BSA) was studied by fluorescence spectroscopy in combination with UV absorption spectroscopy. The intrinsic fluorescence of bovine serum albumin was quenched by EPAT through a static quenching procedure. The binding constants of EPAT with BSA were estimated according to the fluorescence quenching results at different temperatures. The thermodynamic parameters: enthalpy change (ΔH) and entropy change (ΔS) were calculated to be ?10.69 kJ/mol and 42.64 J·mol?1·K?1 according to thermodynamic equations, respectively, and indicating that the binding force was suggested to be mainly a hydrophobic force. The effect of common ions on the binding constant was also investigated. A new fluorescence spectroscopy assay of the proteins was presented in this paper. The determination results of the proteins in bovine serum by means of this method were very close to those obtained using Coomassie Brilliant Blue G‐250 colorimetry.  相似文献

The mechanism and conformational changes of farrerol binding to bovine serum albumin (BSA) were studied by spectroscopic methods including fluorescence quenching technique, UV–vis absorption, circular dichroism (CD) spectroscopy and Fourier transform infrared (FT-IR) spectroscopy under simulative physiological conditions. The results of fluorescence titration revealed that farrerol could strongly quench the intrinsic fluorescence of BSA through a static quenching procedure. The thermodynamic parameters enthalpy change and entropy change for the binding were calculated to be −29.92 kJ mol−1 and 5.06 J mol−1 K−1 according to the van’t Hoff equation, which suggested that the both hydrophobic interactions and hydrogen bonds play major role in the binding of farrerol to BSA. The binding distance r deduced from the efficiency of energy transfer was 3.11 nm for farrerol–BSA system. The displacement experiments of site markers and the results of fluorescence anisotropy showed that warfarin and farrerol shared a common binding site I corresponding to the subdomain IIA of BSA. Furthermore, the studies of synchronous fluorescence, CD and FT-IR spectroscopy showed that the binding of farrerol to BSA induced conformational changes in BSA.  相似文献

The mechanism of the interaction between bovine serum albumin (BSA) and [Pt(phen) (histidine)]+ complex was studied employing ultraviolet (UV) absorption, circular dichroism (CD), FT-IR, differential pulse voltammetry (DPV), and fluorescence spectral methods. Fluorescence data showed that the intrinsic fluorescence of BSA was strongly quenched by Pt(II) complex in terms of an untypical static quenching process. The corresponding number of binding sites (n) and binding constant (K b) of BSA and complex at 283, 298, and 310 K were calculated to be 0.61?×?106, 19?×?106, and 42?×?106 M?1, respectively. The results showed that the increasing temperature improves the stability of the complex–BSA system, which results in a higher binding constant and the number of binding sites of the complex–BSA system. The positive ΔH and positive ΔS indicated that hydrophobic forces might play a major role in the binding between complex and BSA. Based on Forster’s theory of non-radiation energy transfer, the binding distance (r) between the donor (BSA) and acceptor (Pt(II) complex) was evaluated. The results of CD, UV–vis, DPV, and FT-IR spectroscopy showed that the binding of Pt(II) complex to BSA induced conformational changes in BSA  相似文献

The interaction of trans-resveratrol and BSA was investigated by means of fluorescence quenching, resonance light scattering, ultraviolet spectroscopy and Fourier Transform Infrared Spectroscopy. Binding of trans-resveratrol to BSA quenches the BSA fluorescence and both static and dynamic quenching occur with complex formation. The apparent binding constants of trans-resveratrol and BSA at 20, 30 and 40?°C are 1.95×106, 1.70×106 and 1.65×106 L?mol?1, respectively. The binding site values are (1.25±0.02). According to the Förster theory of non-radiation energy transfer, the binding distances between trans-resveratrol and BSA are 3.47, 3.73 and 3.99 nm at 20, 30 and 40?°C, respectively. The negative enthalpy change and positive entropy change indicated that the interaction of trans-resveratrol and BSA was driven mainly by electrostatic forces. The process of binding was spontaneous whereby the Gibbs energy change was negative.  相似文献

在模拟生理条件下,采用荧光光谱法、圆二色光谱法和红外光谱法研究了花椒油素(XT)与牛血清白蛋白(BSA)的相互作用。结果表明花椒油素与牛血清白蛋白之间发生动态和静态联合猝灭,二者间的的猝灭常数(K)在286, 298和310 K分别为3.31 × 105, 到2.03 × 105 和 0.94 × 105 L∙mol-1. 热力学参数表明, 花椒油素与牛血清白蛋白间以疏水作用力为主。圆二色光谱和红外光谱法表明加入花椒油素后,牛血清白蛋白的二级结构发生了变化,其中α-螺旋减少了3.9%。另外,我们还研究了共存离子对两者结合的影响。  相似文献

在模拟动物体生理条件和不同温度下,用荧光光谱和紫外-可见吸收光谱法研究了溴百里酚蓝(BTB)与牛血清白蛋白(BSA)结合反应的光谱行为。用Stern-Volmer和Lineweaver-Burk方程分别处理试验数据,发现BSA与BTB发生反应生成了新的复合物,属于静态荧光猝灭。求出了反应时复合物的形成常数KLB(2.792×105L.mol-1)、热力学参数(ΔHθ=(20.24 kJ.mol-1,ΔSθ=37.22J.K-1,ΔGθ=(31.25kJ.mol-1)与结合位点数(1.1578)。根据F rster偶极-偶极非辐射能量转移理论计算出结合位置距离212位色氨酸残基2.60nm,证明二者主要靠静电作用力结合。同时用同步荧光光谱和三维荧光光谱法探讨了BTB对BSA构象的影响,表明BTB使色氨酸残基所处微环境的极性减弱、疏水作用增强,为阐明BTB的染色机理、毒理效应和生物学效应提供重要信息。  相似文献

The binding interaction of a Schiff base compound containing a 1,2,4‐triazole ring [4‐(4‐chlorobenzyl‐ideneamino)‐5‐methyl‐1,2,4‐triazole‐3‐thiol, CTT] with bovine serum albumin (BSA) was studied by spectroscopy methods including fluorescence and circular dichroism spectrum under simulative physiological conditions. Fluorescence investigation revealed that the fluorescence quenching of BSA was induced by the formation of a relative stable CTT‐BSA complex. The corresponding binding constants (Ka) between CTT and BSA at three different temperatures were calculated according to the modified Stern‐Volmer equation. The enthalpy change (ΔH) and entropy change (ΔS) were calculated to be −15.78 kJ·mol−1 and 49.23 J·mol−1·K−1, respectively, which suggested that hydrophobic forces and hydrogen bond played major roles in stabilizing the CTT‐BSA complex. Site marker competitive experiments indicated that the binding of CTT to BSA primarily took place in sub‐domain IIIA (site II) of BSA. The binding distance (r) between CTT and the tryptophan residue of BSA was obtained to be 4.3 nm based on F?rster theory of non‐radioactive energy transfer. The conformational investigation revealed that the presence of CTT decreased the α‐helix content of BSA (from 58.62% to 54.66%) and induced the slight unfolding of the polypeptides of protein, which confirmed some micro‐environmental and conformational changes of BSA molecules.  相似文献

The interaction between juglone at the concentration range of 10–110 µM and bovine serum albumin (BSA) or human serum albumin (HSA) at the constant concentration of 11 µM was investigated by fluorescence and UV absorption spectroscopy under physiological-like condition. Performing the experiments at different temperatures showed that the fluorescence intensity of BSA/HSA was decreased in the presence of juglone by a static quenching mechanism due to the formation of the juglone–protein complex. The binding constant for the interaction was in the order of 103 M?1, and the number of binding sites for juglone on serum albumins was determined to be equal to one. The thermodynamic parameters including enthalpy (ΔH), entropy (ΔS) and Gibb’s free energy (ΔG) changes were obtained by using the van’t Hoff equation. These results indicated that van der Waals force and hydrogen bonding were the main intermolecular forces stabilizing the complex in a spontaneous association reaction. Moreover, the interaction of BSA/HSA with juglone was verified by UV absorption spectra and molecular docking. The results of synchronous fluorescence, UV–visible and CD spectra demonstrated that the binding of juglone with BSA/HSA induces minimum conformational changes in the structure of albumins. The increased binding affinity of juglone to albumin observed in the presence of site markers (digoxin and ibuprofen) excludes IIA and IIIA sites as the binding site of juglone. This is partially in agreement with the results of molecular docking studies which suggests sub-domain IA of albumin as the binding site.  相似文献

The interaction between colloidal AgTiO2 nanoparticles and bovine serum albumin (BSA) was studied by using absorption, steady state, time resolved and synchronous fluorescence spectroscopy measurements. Absorption spectroscopy proved the formation of a ground state BSA?AgTiO2 complex. Upon excitation of BSA, colloidal AgTiO2 nanoparticles effectively quenched the intrinsic fluorescence of BSA. The number of binding sites (n = 1.06) and apparent binding constant (K = 3.71 × 105 M−1) were calculated by the fluorescence quenching method. The effects of buflomedil (BFM), a novel cerebrovascular medicine, on serum albumin and the feasibility of ??-cyclodextrin (??-CD) acting as a controlled releaser for BFM were investigated by molecular spectroscopy methods, including fluorescence emission, UV?Cvisible absorption, Fourier transform infrared (FT-IR), synchronous and three-dimensional fluorescence spectroscopies. The interaction of BFM with bovine serum albumin (BSA) was analyzed by fluorescence quenching and it was found that BFM had reacted with BSA in the ground state and had affected the secondary structure of BSA. The observed binding constant K b, the number of binding sites n, the distance r between donor (BSA) and acceptor (BFM), the enthalpy change (??H ?? ), Gibbs energy change (??G ?? ) and entropy change (??S ?? ) at different temperatures were calculated. The inclusion reaction between ??-CD and BFM was explored by the Lineweaver?CBurk equation. The inclusion constants and the thermodynamic parameters were determined. The mechanism of the inclusion reaction was discussed and the controlled release characteristics of ??-CD to BFM were considered at the molecule level.  相似文献

The binding of kaempferol‐3,7‐αL‐rhamnopyranoside (KRR) with bovine serum albumin (BSA) was investigated by different spectroscopic methods under simulative physiological conditions. Analysis of ?uorescence quenching data of BSA by KRR at different temperatures using Stern‐Volmer methods revealed the formation of a ground state KRR‐BSA complex with moderate binding constant of the order 104 L·mol?1. The existence of some metal ions could weaken the binding of KRR on BSA. The changes in the van't Hoff enthalpy (ΔH0) and entropy (ΔS0) of the interaction were estimated to be ?26.53 kJ·mol?1 and 3.33 J·mol?1·K?1 and both hydrophobic and electrostatic forces contributed to stabilizing the BSA‐KRR complex. According to the F?ster theory of non‐radiation energy transfer, the distance r between the donor (BSA) and the acceptor (KRR) was obtained (r=2.83 nm). Site marker competitive experiments showed that KRR could bind to Site I of BSA. In addition, synchronous fluorescence, UV‐Vis absorption and circular dichroism (CD) results indicated that the KRR binding could cause conformational changes of BSA.  相似文献

The interaction between bovine serum albumin (BSA) and pegylated puerarin (Pur) in aqueous solution was investigated by UV-Vis spectroscopy, fluorescence spectroscopy and circular dichroism spectra (CD), as well as dynamic light scattering (DLS). The fluorescence of BSA was strongly quenched by the binding of pegylated Pur to BSA. The binding constants and the number of binding sites of mPEG(5000)-Pur with BSA were 2.67±0.12 and 1.37±0.05 folds larger after pegylating, which were calculated from the data obtained from fluorescence quenching experiments. The enthalpy change (ΔH) and entropy change (ΔS) were calculated to be 4.09 kJ mol(-1) and 20.01 J mol(-1) K(-1), respectively, according to Van't Hoff equation, indicating that the hydrophobic force plays a main role in the binding interaction between pegylated Pur and BSA. In addition, the negative sign for Gibbs free energy change (ΔG) implies that the interaction process is spontaneous. Moreover, the results of synchronous fluorescence and CD spectra demonstrated that the microenvironment and the secondary conformation of BSA were changed. Comparing with Pur, all our data collected indicated that pegylated Pur interacted with BSA in the same way as that of Pur, but docked into the hydrophobic pocket of BSA with more accessibility and stronger binding force. DLS measurements showed monomethoxy polyethylene glycol (mPEG) have an effect on BSA conformation, and revealed that changes in BSA size might be due to increases in binding constant and the absolute values of ΔG after Pur pegylation.  相似文献

The binding of isothipendyl hydrochloride (IPH) to bovine serum albumin (BSA) was investigated by fluorescence spectroscopy combined with UV-visible absorption and circular dichroism (CD) techniques under simulative physiological conditions for the first time. The quenching mechanism of fluorescence BSA by IPH was discussed. The binding parameters have been evaluated by fluorescence quenching method. The thermodynamic parameters, ΔH°, ΔS° and ΔG° calculated at different temperatures indicated that the hydrophobic force played a major role in the interaction of IPH to BSA. The distance, r between donor (BSA) and acceptor (IPH) was obtained according to the Förster's theory of non-radiation energy transfer and was found to be 2.21 nm. Experimental results showed that the α-helicity of BSA decreased from 66.4% (in free BSA) to 39.1% (in bound BSA). The effect of common ions on the binding constant was also investigated.  相似文献

Cyromazine (CMZ) had been believed to be one of the safest pesticides and widely used for many years until its carcinogenesis was revealed recently. In this work, the interaction between cyromazine and human serum albumin (HSA) was systematically investigated by multiple spectroscopic methods and molecular docking techniques using warfarin and flufenamic acid as probes. The results demonstrated the fluorescence of HSA had been quenched by CMZ through static mechanism, with new non-covalent complexes formed at ground state. Fluorescence probe experiments indicated CMZ bound to Sudlow’s site I in subdomain IIA of HSA, having no competition with site marker in site II. The number of binding sites, equilibrium constants and thermodynamic parameters were calculated by monitoring the binding equilibriums at different temperatures. The positive enthalpy change (ΔH θ) and entropy change (ΔS θ) implied the binding was mainly conducted by hydrophobic interactions. The binding was an endothermic, spontaneous (ΔG θ?<?0) and entropy-driven process which made the energy distribution of the system more evenly. The force of interaction and the conformation of binding pocket were displayed by molecular simulation and discussed at molecular level. Circular dichroism (CD) spectra indicated distorted α-helix content of HSA decreased while other fine secondary structure increased when CMZ was added.  相似文献

The interaction between dioxopromethazine hydrochloride (DPZ) and human serum albumin (HSA), in vitro, was investigated by means of fluorescence, absorption and circular dichroism (CD) spectroscopy. The results obtained from analysis of the fluorescence spectra indicate that DPZ has a strong ability to quench the intrinsic fluorescence of HSA through a static quenching mechanism. The HSA?CDPZ binding distance was determined to be less than 7?nm, suggesting that energy transfer from HSA to DPZ may occur. The thermodynamic parameters of the interaction between DPZ and HSA were measured according to the van??t Hoff equation. The negative enthalpy change (??H 0) and positive entropy change (??S 0) values indicate that hydrophobic and electrostatic interactions play major roles in the binding between DPZ and HSA. The binding process was a favorable process in which the Gibbs energy change (??G 0) is negative. The results of displacement experiments suggested that DPZ is located in site I of HSA within subdomain IIA. In addition, the results of absorption, synchronous fluorescence and CD spectra show that binding of DPZ with HSA can induce conformational changes in HSA.  相似文献
