Two aromatic rings of a phlorizin molecule form inclusion complexes with -CD and -CD. Induced circular dichroism spectra of these complexes have been measured to estimate the orientation of the two aromatic rings in the hydrophobic space of CDs. Apparent complex formation constants have been also estimated for each complex. It is concluded that phlorizin forms a stronger inclusion complex with -CD than with -CD.  相似文献

We have developed a circular-dichroism thermal lens microscope for UV wavelengths (UV-CD-TLM), for the first time, to realize sensitive chiral analysis on a microchip. Quasi-continuous-wave phase modulation of a pulsed UV laser was used to generate left-circularly polarized light and right-circularly polarized light and to detect the generated TL signal amplitude and phase with a lock-in amplifier. The amplitude and phase were used to determine the concentration and chirality, respectively, of a sample. The basic principle of UV-CD-TLM for chiral analysis on a microchip was verified by measuring aqueous solutions of optically active camphorsulfonic acids (CSA). Lower limits of detection (LOD) were calculated at S/= 2 and were 8.7 × 10−4 mol L−1A = 5.2 × 10−6 Abs.) for (+)-CSA and 8.4 × 10−4 mol L−1A = 5.0 × 10−6 Abs.) for (−)-CSA. In terms of number of molecules, LODs for UV-CD-TLM were calculated to be 8.7 fmol and 8.4 fmol, respectively. This is at least three orders of magnitude lower than previously obtained. 探索蛋白质结构稳定性及其淀粉样纤维化的环境条件具有重要意义.本文采用了荧光光谱法研究溶液中的金属盐对鸡卵清溶菌酶内源荧光和淀粉样纤维化的影响.结果表明,金属盐能够增加溶菌酶的热稳定性,减小淬灭剂对内源荧光的作用,对溶菌酶的高级结构具有一定的稳定作用.另一方面,在长时间热胁迫的情况下,金属盐可促进溶菌酶分子的聚集而纤维化.金属盐的这种双重作用分别与其阳离子和阴离子的性质有关.  相似文献

The inclusion behaviors of three native or modified CDs including p-CD,2-hydroxypropyl-β-CD(2-Hp-β-CD) and 2,6-dimethyl-β-CD(Me-β-CD) toward 5-amino-6-methyl-2-benzimidazolone(AMBI) were comparatively investigated by NMR and fluorescence titration in combination with IR spectra,X-ray diffractometry and scanning electron microphotographs.The experimental results jointly demonstrated that the phenyl ring of AMBI entered into the cavity of the CDs and located close to the narrow rims accompanied by the formation of the 1:1 inclusion complex with large stability constant in aqueous solution.The introduction of the hydroxypropyl unit to the host improved the solubility,ultimately effecting an obvious promoting in the fluorescence intensity and the stability constant  相似文献

The interaction between lysozyme (LYSO) and puerarin has been studied at three temperatures (294, 302 and 310K) through/using fluorescence spectroscopy and circular dichroism (CD). The LYSO fluorescence was quenched by the binding of puerarin to LYSO. The binding constants and the number of binding sites can be calculated from the data obtained from fluorescence quenching experiments. According to the van't Hoff equation, the standard enthalpy change (DeltaH degrees ) and standard entropy change (DeltaS degrees ) for the reaction were calculated to be 17.47kJ/mol and 163.5J/molK. It indicated that the hydrophobic interactions play a main role in the binding of puerarin to LYSO. In addition, the distance between puerarin (acceptor) and tryptophan residues of LYSO (donor) was estimated to be 1.47nm on the basis of fluorescence energy transfer. The changes of LYSO secondary structure in the presence of puerarin were observed from CD spectroscopy.  相似文献

A sensitive and validated liquid chromatography with mass spectrometry method was developed for the enantioseparation of the racemic mixture of pomalidomide, a novel, second‐generation immunomodulatory drug, using β‐cyclodextrin‐bonded stationary phases. Four cyclodextrin columns (β‐, hydroxypropyl‐β‐, carboxymethyl‐β‐, and sulfobutyl‐β‐cyclodextrin) were screened and the effects of eluent composition, flow rate, temperature, and organic modifier on enantioseparation were studied. Optimized parameters, offering baseline separation (resolution = 2.70 ± 0.02) were the following: β‐cyclodextrin stationary phase, thermostatted at 15°C, and mobile phase consisting of methanol/0.1% acetic acid 10:90 v/v, delivered with 0.8 mL/min flow rate. For the optimized parameter at multiple reaction monitoring mode 274.1–201.0 transition with 20 eV collision energy and 100 V fragmentor voltage the limit of detection and limit of quantitation were 0.75 and 2.00 ng/mL, respectively. Since enantiopure standards were not available, elution order was determined upon comparison of the circular dichroism signals of the separated pomalidomide enantiomers with that of enantiopure thalidomide. The mechanisms underlying the chiral discrimination between the enantiomers were also investigated. Pomalidomide‐β‐cyclodextrin inclusion complex was characterized using nuclear magnetic resonance spectroscopy and molecular modeling. The perturbation of the three-site interaction of phenolphthalein on complexation with different cyclodextrins (CDs) has been investigated as a function of the type and degree of substitution of the CD. The UV visible spectra are practically identical, while the circular dichroism spectra change dramatically both in intensities and signs. Parallel to this, the stability constants are influenced not only by the size of the cavity but also by the substitution of its rims, particularly by that of the primary hydroxyls. The two phenomena can be related to allow conclusions concerning the mode of inclusion.  相似文献

Peptides and proteins have become very promising drug candidates in recent decades due to their unique properties. However, the application of these drugs has been limited by their high enzymatic susceptibility, low membrane permeability and poor bioavailability when administered orally. Considerable efforts have been made to design and develop drug delivery systems that could transport peptides and proteins to targeted area. Although it is of great importance to determine the composition after loading a drug to the carrier, the ability to do so is significantly limited by current analytical methods. In this letter, five important proteins, α1-antitrypsin, hemoglobin human, human serum albumin, human transferrin and r-globulin were chemically conjugated to two model drug carriers, namely carbon dots and polymer O-(2-carboxyethyl) polyethylene glycol. A simple yet convenient method based on circular dichroism spectroscopy was developed to determine the compositions of the various protein-carrier conjugates.  相似文献

The Induced Circular Dichroism of the complex formed by inclusion of Licochalcone-A (a powerful antimalarian of natural origin) and β-cyclodextrin was measured and calculated. The association constant was determined by titration experiments of ICD and fluorescence. The lowest energy conformations of the complex were obtained through docking procedures and their Circular Dichroism was evaluated within the Tinoco framework. The geometry of the complex was found to be consistent with the main experimental features.  相似文献

Microporous polyethylene (PE) membranes were coated on both sides with a polypyrrole (PPy) overlayer. PPy was deposited also on the walls of pores. Thermodeformation measurements indicate that the hard framework produced by rigid-chain PPy controls the mechanical properties of composite membranes. FTIR spectroscopy was used to detect oxidative processes in both polymer components above 200 °C. Elemental analysis showed that the nitrogen content remains at high level, even after the treatment of membranes in air at 300 °C, indicating that the decomposition involved mainly PE while PPy was much more stable. The changes in surface relief after annealing were assessed with electron microscopy. No oxidative processes were identified in the FTIR spectra of composites after annealing in vacuum, even after treatment at 300 °C.  相似文献

Multivariate curve resolution with alternating least squares (MCR-ALS) is applied for the first time to the simultaneous analysis of electrochemical and spectroscopic data. Then, a data analysis is done with augmented matrices constituted by Differential Pulse Polarography and Circular Dichroism data submatrices. The use of proper, and different for each submatrix, constrains in the iterative ALS optimization allows to obtain chemically meaningful results constituted by a common matrix containing the concentration profiles, and two matrices with the pure electrochemical and spectroscopic signals. MCR-ALS is applied to the study of the complexation of Cd by Cys-Gly, a glutathione-fragment of great interest for understanding metal-phytochelatins complexation.  相似文献

The interactions of calf thymus deoxyribonucleic acid (ct-DNA) with two antitumour drugs (5-fluorouracil and tegafur) in aqueous buffer solution (pH 7.40) have been investigated using nano-watt-scale isothermal titration calorimetry (ITC), circular dichroism (CD), ultraviolet absorption (UV) and fluorescence spectroscopy. Thermodynamic parameters, i.e., binding proportions and constants, standard changes of enthalpy (ΔH°), Gibbs free energy (ΔG°) and entropy (ΔS°) have been derived from the calorimetric data. The binding ratios of 5-fluorouracil and tegafur with base pairs in ct-DNA are 1:3 and 1:4, respectively. The thermodynamic parameters have been discussed according to the influence of drugs on molecular structure of the DNA shown spectrogram. The results indicate that molecule of 5-fluorouracil or tegafur can intercalate itself into the intra-molecular space formed by DNA double helix and cause some changes in the secondary structure of DNA molecule.  相似文献

The conformation of amphiphilic lipopolysaccharides (LPS) influences the behavior of free and cell-bound LPS in aqueous environments, including their adhesion to surfaces. Conformational changes in Pseudomonas aeruginosa serotype 10 LPS aggregates resulting from changes in solution pH (3, 6, and 9), ionic strength [I] 1, 10, and 100 mmol L−1, and electrolyte composition (NaCl and CaCl2) were investigated via attenuated total reflectance (ATR) Fourier transform infrared (FTIR) spectroscopy. ATR-FTIR data indicate that LPS forms more stable aggregates in NaCl relative to CaCl2 solutions. Time- and cation-dependent changes in ATR-FTIR data suggest that LPS aggregates are perturbed by Ca2+ complexation at lipid A phosphoryl groups, which leads to reorientation of the lipid A at the surface of a ZnSe ATR internal reflection element (IRE). Polarized ATR-FTIR investigations reveal orientation of LPS dipoles approximately perpendicular to the IRE plane for both Na- and Ca-LPS. The results indicate that changes in solution chemistry strongly impact the conformation, intermolecular and interfacial behavior of LPS in aqueous systems.  相似文献

A photo-induced de-oxygenation process of myoglobin (Mb) in aqueous solution was investigated by use of fluorescence spectroscopy. The spectra are characterized by the fluorescence intensity declining gradually after each scan,and the decay of fluorescence intensity being significant in each scan,which is assigned to the release of oxygen from the opening of the heme-pockets induced by illumination. More illumination will cause more release of oxygen; if the temperature of an Mb solution is increased when it is illuminated,the rate of de-oxygenation will be higher. It was found that ligand-oxygen in Fe-porphyrin could be removed from Mb by nitrogen. This indicates that the interac-tion between oxy-Mb and other different gases can be tested by the method of fluorescence spectros-copy. In addition,fluorescence spectroscopy can be employed to probe the energy transfer between Fe-porphyrin and tryptophan or tyrosine in Mb molecules.  相似文献

Circular dichroism spectral and fluorescence decay methods have been employed to determine the conformations of mono[6-(p-tolylseleno)-6-deoxy]-p-CD(1), mono(6-anilino-6-deoxy) β -CD (2) and mono[6-(L-tryptophan)-6-deoxy]-β-CD (3) in phosphate buffer solution (pH 7.2, 0.1 mol dm-3) at 298.15 K. The results indicate that compounds 2 and 3 formed self-inclusion complexes in aqueous buffer solution, while the substituent of compound 1 was not included into cyclodextrin cavity at all. Furthermore, the complex stability constant (logKs) and Gibbs free energy change (-ΔG° ) of these three cylcodextrin derivatives with several cycloalkanols have been determined by circular dichroism spectral titration in phosphate buffer solution at 298.15 K. It is found that the location of the substituent affects the stability of host-guest complex in aqueous solution.  相似文献

The diffusional behavior of amphiphilic poly(2-oxazoline) diblock copolymers in aqueous solution is studied using photon correlation spectroscopy (PCS) and fluorescence correlation spectroscopy (FCS). The polymers were synthesized by living cationic polymerization and were fluorescence-labeled with tetramethyl rhodamine isothiocyanate either at the end of the hydrophilic or the hydrophobic block. Temperature-resolved PCS showed that, at room temperature, large metastable aggregates are present along with unimers and micelles. An annealing above ~40 °C resulted in stable equilibrium micellar solutions. By means of FCS, the hydrodynamic radii of the unimers and the micelles were measured simultaneously in a broad concentration range, and the critical micelle concentration could be determined. The hydrodynamic radii of micelles formed by amphiphilic poly(2-alkyl-2-oxazoline) diblock copolymers in aqueous solution determined using fluorescence correlation spectroscopy (FCS) depend on the nature of the fluorescent tracer used. We have compared the values of the hydrodynamic radii of the unimers and the micelles as well as the critical micelle concentrations (CMC), using as tracers (1) the identical diblock copolymers being fluorescence-labeled at the hydrophilic or the hydrophobic block terminus [Bonné et al. Colloid Polym Sci (2004) 282:833–843], and (2) a low molar mass fluorescence dye, rhodamine 6G. Whereas similar values for the CMC were found for both probes, the hydrodynamic radius of micelles is significantly underestimated using a free dye as a tracer in FCS, especially near the CMC. We attribute this discrepancy to the fast exchange of the dye between micelles and solution.  相似文献

Severe thermal pretreatment of gold wire electrodes in an inert gas atmosphere resulted in the appearance of dramatic premonolayer oxidation responses, which in some instances commenced at 0.25 V (RHE), for the resulting active gold electrodes in aqueous acid media. Similar behaviour was reported earlier for platinum and gold activated by cathodic pretreatment in acid solution; these active noble metal surfaces are evidently more susceptible to oxidation than bulk copper. Such behaviour was attributed to the effect of surface disorder; many of the metal atoms at the interface are assumed to be in a very active, metastable state possessing quite low lattice stabilization energy. Premonolayer oxidation responses are again correlated with electrocatalytic behaviour and the existence of unusual chemisorption behaviour for molecules reacting at highly active states of metals is outlined. The ability of beta-cyclodextrin (beta-CD), sulfurbutylether-beta-CD (SBE-beta-CD) and hydroxypropyl-beta-CD (HP-beta-CD) to break the aggregate of the meso-Tetrakis (4-sulfonatophenyl) porphyrin (TPPS4) and to form 2:1 inclusion complexes has been studied by adsorption and fluorescence spectroscopy. The formation constants are calculated, respectively by fluoremetry, from which the inclusion capacity of different CDs is compared and the inclusion mechanism of charged-beta-CD (SBE-beta-CD) is quite different from that of parent beta-CD. At lower pH, the complexation between HP-beta-CD and H2TPPS(2+)4 (the form of the diprotonated TPPS4) hampers the continuous protonation of the pyrrole nitrogen of TPPS4 and the hydrophobic cavity may prefer to bind an apolar neutral porphyrin molecule. 1HNMR data support the inclusion conformation of the porphyrin-cyclodextrin supramolecular system, indicating the interaction of meso-phenyl groups of TPPS4 with the cavity of CDs. For this host-guest inclusion model, cyclodextrin, being regarded as the protein component, which acts as a carrier enveloping the active site of heme prosthetic group within its hydrophobic environment, provides a protective sheath for porphyrin, creating artificial analogues of heme-containing proteins. However, the TPPS4, encapsulated within this saccharide-coated barrier, its physico-chemical, photophysical and photochemical properties changed strongly.  相似文献

The binding of nevadensin to human serum albumin (HSA) in aqueous solution was investigated for the first time by molecular spectroscopy and modeling at pH 7.4. Spectrophotometric observations are rationalized in terms of a static quenching process and binding constant (KaKb) and the number of binding sites (n ≈ 1) were evaluated by fluorescence quenching methods. Thermodynamic data showed that nevadensin was included in the hydrophobic cavity of HSA mainly via hydrophobic interactions. The value of 3.09 nm for the distance r between the donor (HSA) and acceptor (nevadensin) was derived from the fluorescence resonance energy transfer. Spectrophotometric techniques were also applied to investigate the structural information of HSA molecules on the binding of nevadensin and the results showed that the binding of nevadensin to HSA did not change significantly molecular conformation of HSA in our experimental conditions. Furthermore, the study of molecular modeling also indicated that nevadensin could strongly bind to the site I (subdomain IIA) of HSA mainly by a hydrophobic interaction and there are hydrogen bond interactions between nevadensin and the residues Arg-218, Arg-222, Lys-195, and Asp-451. As compared to the other flavonoids, the flavonoids containing methoxy groups which are in aromatic rings can bind to HSA with higher affinity.  相似文献
