The cis-cxo- amino acid c with norbornene skeleton was converted into 2-aryvl-cis-cxo-1,3-oxazin-4-ones 5a-d. These compounds, similarly to the diendo isomers 1a-d studied by us earlier, undergo retrodiene decomposition under mild conditions to give 2-aryl-62-l,3-oxazin-6-ones (2a-d) in 50-60% yield. The ratio of the decomposition rate constants of the tricyclic diendo and diexo-1,3-oxazin-4-ones, measured in toluene solution, is about 2.  相似文献

The 1H and 13C nmr spectra of exo, exo-9-oxobicyclo[3.3.1]nonane-2,4-dicarboxylic acid, 3, in DMSO-d6 or alkaline D2O, clearly show that it exists in a boat-chair-conformation with equatorial carboxyl groups, thus being the first case of boat-chair preference of a bicyclo[3.3.1] nonan-9-one due to the presence of exo,exo-2,4- substituents.  相似文献

Reaction of cis,cis,cis,cis-[9]annulene anion (1) with electrophiles (2a-d) leads to substituted cis,cis,cis,cis-1,3,5,7-cyclononatetraenes (3a-d) which are precursors for the preparation of 10- and 10,10′-donor-substituted nonafulvenes (e.g. 7b′e′). The influence of solvent and temperature on the 1H-nmr spectra of the nonafulvenes 7b′-h′ has been investigated.  相似文献

The synthesis of two enantiomeric pairs of pyrimidoisoindoles 9a, 9b and 10a, 10b is reported. During a domino ring-closure reaction, followed by cycloreversion, the chirality of diendo-(?)-(1R,2S,3R,4S)-3-aminobicyclo[2.2.1]hept-5-ene-2-carboxamide [(?)-1] was successfully transfered to heterocycles (+)-9a, (+)-10a, (?)-9b, (?)-10b and (?)-10c.  相似文献

Summary Direct reversed-phase high-performance liquid chromatographic methods were developed for the separation and identification of the enantiomers of mono- and bicyclic racemic β-amino acids:cis- andtrans-2-aminocyclopentane-1-carboxylic acids,cis- andtrans-2-aminocyclohexane-1-carboxylic acids,cis- andtrans-2-amino-4-cyclohexene-1-carboxylic acids,diendo- anddiexo-3-aminobicyclo[2.2.1]heptane-2-carboxylic acids anddiendo- anddiexo-3-amino-5-bicyclo[2.2.1]heptene-2-carboxylic acids. Enantioseparation was carried out by the application of a chiral stationary phase, Crownpak CR(+). The conditions of separation were optimized by changing the temperature, the flow rate and the pH of the mobile phase. By the reaction of anthranilic hydrazide 1 with cis-2-(p-methylbenzoyl)-1-cyclohexanecarboxylic acid 2a or diendo-3-(p-methylbenzoyl)bicyclo[2.2.1]heptane-2-carboxylic acid 2b , fused tetra- and pentacyclic ring systems 3a, b were prepared, trans-2-Amino-1-cyclohexanecar-bohydrazide 4b was reacted with 3-(p-chlorobenzoyl)propionic acid 5 to yield the pyridazino[6,1-b]quinazolinone 6 . From the reaction of cis-2-amino-1-cyclohexanecarbohydrazide 4a with 2a , three isomeric partially saturated 8H-phthalazino[1,2-b]quinazolin-8-ones 7a-c were formed. The reaction of diexo-2-aminobicyclo[2.2.1]heptane-3-carbohydrazide 4c and 2a furnished the pentacyclic derivatives 8 and 9 containing a 3-aryl-4,5-dihydropyridazine or 3-arylhexahydropyridazine ring C with cis annelated C/D rings. The formation of 8 and 9 involving different ring systems can be rationalized by two reaction pathways: (i) in the bislactam 9 the carboxyl group acylates the hydrazide, while (ii) in 8 it forms a pyridazine ring with the cyclic amino group by cyclocondensation. The structures of the products were elucidated by 1H and 13C nmr methods, including DEPT, DNOE and 2D-HSC measurements.  相似文献

(1R,5S)-2S-Deuteriobicyclo[3.2.1]octan-3-one (1) and (1R,5S)-2R-Deuteriobicyclo[3.2.1]octan-3-one (2), prepared by diazomethane ring enlargement of (1S,4R)-2(exo)-deuteriobicyclo-[2.2.1] heptan-2-one and (1S,4R)-2(endo)-deuteriobicyclo[2.2.1]heptan-2-one respectively, both gave (?) n-π* circular dichroism (CD) Cotton effects, Δεmax294 = ?0.05 and Δεmax294=?0.1, respectively, in hydrocarbon solvent. (1S,5R)-2S-Deuterio-6,6-diaethylbicyclo[3.1.1] heptan-3-one (3) and (1S,5R)-2R-deuterio-6,6-dimethylbicyclo[3.1.1] heptan-3-one (4), prepared from (-) myrtenal, both exhibited extraordinary vibrational fine structure for the n-π* CD transitions observed in hydrocarbon solvent and oppositely?signed CEs, Δεmax282=?0.01 and Δεmax279=+0.01 respectively in CF3CH2OH solvent.  相似文献

The rate of the thermal rearrangement of (S) 2 chloromethyl-1-ethylpyrrolidine [(S)-1a] to (R)-3-chloro-1-ethylpiperidine [(R) 2a] has been examined at three temperatures in benzene by PMR and polarimetry. The rearrangement was shown to be completely stereospecific and to obey a simple first order rate law. The calculated Ea ΔH3 and ΔS3 were 22 ± 2 kcalmole (25°), 21 ± 2.5 kcalmole (25°) and - 10 ± 2 e.u. (0°K) respectively. The effect of solvents having differing dielectric constants was also studied. A transition state 9'a and an ion pair intermediate 3a are suggested for the rearrangement. The stereochemical course of the reactions of (S)-1a, (R)-2a and (S)-2a with hydroxide and methoxide ions have been shown to be 100% stereospecific with an uncertainty of about 1%. The absolute configurations of all optically active reactants and products [(S)- and (R)-4a, (S)-4b (R)- and (S)-5a, (R)-5b, (S,S')-6a, (S,R')-7a and (R,R')-8a] were established by chemical correlations with known compounds or by ORD and chemical inference. The ring opening of both the primary and secondary aziridinium ion positions of 1-azonia-1-ethylbicyclo [3.1.0]hexane [(S)-3a] by nucleophiles proceeds entirely by SN2 processes. The conversion of (R)-1-ethyl-3-hydroxypiperidine [(R)-5a] to (S)-2a. HCl with thionyl chloride in chloroform proceeds by inversion with 4.8% racemization, whereas the thermal rearrangement of (S)-1a to (R)-2a occurs with complete retention of absolute configuration.  相似文献

The synthesis of 7-(β-D-ribofuranosyl)-4-amino-5H-pyrrolo[3,2-d]pyrimidine (9-deazaadenosine) 9 is described. It involves base-catalyzed cyclization of N-carboethoxyenamine 4 to give β- and α-ribosylated 3-amino-2-cyanopyrroles 6 and 7, respectively, followed by a one-step conversion to the desired pyrrolo[3,2-d]pyrimidine system.  相似文献

The efficacy of a new acid-catalyzed intramolecular C-alkylation has been demonstrated by the synthesis of 1-methyl-4-p-methoxyphenylbicyclo [2.2.2] octan-2-one (5) and 4-p-methoxyphenylbicyclo [2.2.2] octan-2-one (6) from easily accessible starting materials. The carbinol 20, derived from 5, undergoes facile rearrangement leading to 1-p-methoxyphenyl-4-methyl bicyclo [3.2.1] oct-3-ene (22), which has been transformed to endo-1-p-methoxyphenyl-4-methylbicyclo [3.2.1] octan-3-one (25).  相似文献

3-(t-Butoxycarbonyl)-2-(trifluoromethyl)imidazo[1,2-a]-pyridine, prepared from trifluoroacetonitrile and pyridinium t-butoxycarbonylmethylide, reacts smoothly with trifluoroacetic acid to provide 2-(trifluoromethyl)imidazo[1,2-a]pyridine-3-carboxylic acid, which gives 2-(trifluoromethyl)imidazo[1,2-a]-pyridine when heated. 3-Cyano-2-(trifluoromethyl)imidazo[1,2-a]pyridine can be obtained via treatment of trifluoroacetonitrile with pyridinium cyanomethylide, which is sufficiently reactive to effect nucleophilic displacement of fluorine from pentafluoropyridine under mild conditions [→pyridinium cyano(tetrafluoro-4-pyridyl)methylide].  相似文献

Oxidative decarboxylation of [n.2.2]propellane carboxylic acids (1a-c) with lead tetraacetate gave the bicyclic acetates (2b, c) having a bridgehead double bond and/or the tricyclic acetates (3a, b) in good yields. Epoxidation of (?)-(1R,2R,4R)-2-endo-cyano-7-oxabicyclo[2.2.1]hept-5-en-2-exo-yl acetate ((?)-5) followed by saponification afforded (+)-(1R,4R,5R,6R)-5,6-exo-epoxy-7-oxabicyclo[2.2.1]heptan-2-one ((+)-7). Reduction of (+)-7 with diisobutylaluminium hydride (DIBAH) gave (+)-1,3:2,5-dianhydroviburnitol ( = (+)-(1R,2R,3S,4R,6S)-4,7-dioxatricyclo[3.2.1.03,6]octan-2-ol; (+)-3). Hydride reductions of (±)-7 were less exo-face selective than reductions of bicyclo[2.2.1]heptan-2-one and its derivatives with NaBH4, AlH3, and LiAlH4 probably because of smaller steric hindrance to endo-face hydride attack when C(5) and C(6) of the bicyclo-[2.2.1]heptan-2-one are part of an exo oxirane ring.  相似文献

Reaction of 7 with phenacyl bromides gave 1,3-thiazolo[3,2-b]pyrazolo [2,3-d]-1,2,4-triazepin-4-iums 10. Compound 3a on treatment with phenacyl bromides gave 1-(4'-aryl-thiazol-2'-yl)-2-pyrazoline derivatives 6. Compound 3b reacts with acyl chlorides to give pyrazolo[1,5-c]pyrimidine derivatives 14, and with aromatic aldehydes giving the 2-(Δ2'1 pyrazolin-1-yl)-5-aryl-1,3,4-Δ2-thiadiazolines 12 which were easily converted to the corresponding 2-(Δ2-pyrazolin-1-yl)-5-aryl-1,3,4-thiadiazoles 13.  相似文献

6, 7-Dimethoxy-2H-l,3-benzothiazine derivatives (1, 8) react with substituted acetyl chlorides to give angularly condensed β -lactams (3a-d, 10, 11). The cis compound 11 was epimerised to the trans derivative 12. From the interaction of 2-phenyl-6,7-dimethoxy-4H-1,3-benzothiazine (7) and α -chloro-phenylacetyl chloride two stereoisomeric β -lactam derivatives (9a, b) were isolated, whereas in the other cases studied the reactions leading to β -lactams proved to be stereospecific. Analogous reactions of 4-methyl-6,7-dimethoxy-2H-l,3-benzothiazine (5) furnished the enamides 6a,c, d. Structures of the new compounds and configurations of the diastereomers were elucidated by IR and NMR spectroscopy.  相似文献
