正A new ionic liquid-based high-performance liquid chromatography stationary phase is reported.A derivative of N-methyl pyrrolidinium tetrafluoroborate was covalently immobilized on the surface of silica particles to prepare silica-based N-methyl pyrrolidinium tetrafluoroborate(SilprMP BF4)stationary phase.The obtained ionic liquid-modified silica was evaluated and confirmed by elemental analysis,infrared spectroscopy,and thermogravimetric analysis.A column was packed with the modified particles.The retention behavior of aromatic compounds,alkyl benzenes,and acidic and basic compounds on the SilprMP BF4 stationary phase was studied under reversed-phase liquid chromatography conditions.The effect of the eluent pH on the separation of the acidic and basic compounds was also studied.The new stationary phase involves multiple retention mechanisms,such as electrostatic,hydrophobic,ion-dipole,and anion-exchange interactions,which might lead to multipurpose separation media.  相似文献

键合型多糖手性固定相因具有化学稳定性高和溶剂耐受性好的特点而受到研究者的极大关注。采用施陶丁格(Staudinger)反应将6-叠氮-6-脱氧纤维素-3,5-二氯苯基氨基甲酸酯键合到氨丙基硅胶上得到一种新的键合型手性固定相(ImCel),研究了其手性分离性能,并探讨了非常规流动相(如氯仿、四氢呋喃等)的影响。结果表明,在20对手性化合物中,17对在合适的流动相下得到基线分离。ImCel在正相条件下的分离性能优于反相条件,且在含氯仿的流动相中仍对手性化合物表现出良好的分离能力。在分离一系列芴甲氧羰基(fmoc)-氨基酸衍生物时,ImCel与键合6-叠氮-6-脱氧纤维素-3,5-二甲基苯基氨基甲酸酯的手性固定相表现出互补性,出现了固定相改变引起的对映体洗脱反转现象。本研究丰富了键合型多糖手性固定相的种类和合成方法,为开发新的键合型手性固定相提供了参考。  相似文献

通过十八烷基胺的氨基与二醛微晶纤维素的醛基共价键合,制备了基于二醛微晶纤维素(DMCC)官能化C18的新型反相/亲水色谱固定相(C18-DMCC/SiO2),该色谱固定相被用于反相色谱(RPLC)和亲水相互作用色谱(HILIC)模式。C18-DMCC/SiO2色谱柱展现了良好的疏水选择性和芳香选择性,在反相色谱模式下可分离烷基苯和多环芳烃(PAHs)。苯胺类、酚类和糖苷类等极性化合物被用于评估该色谱柱在反相色谱模式下的极性选择性,商品C18柱作对照柱,色谱评价结果令人满意。核酸碱基被用于评估C18-DMCC/SiO2色谱柱的亲水色谱性能。通过考察有机溶剂含量对分析物保留的影响,发现该新型色谱固定相具有反相/亲水色谱的典型特征。  相似文献

采用纤维素-三(4-甲基苯甲酸酯)(CTMB)手性固定相,利用反相色谱法研究了氟比洛芬、普拉洛芬、布洛芬、萘普生、洛索洛芬5种芳基丙酸类手性药物的色谱拆分行为。考察了流动相组成、酸碱添加剂及柱温对上述5种药物对映体分离的影响,并通过热力学研究及对映体结构分析对CTMB固定相的手性拆分机理进行了探讨。结果表明,除萘普生采用乙腈-0.1%(v/v)甲酸溶液外,以甲醇-0.1%(v/v)甲酸水溶液为流动相可使普拉洛芬、洛索洛芬、氟比洛芬和布洛芬的对映体间的分离度均大于1.5,CTMB固定相对这5种芳基丙酸类药物的手性拆分能力依次为普拉洛芬>洛索洛芬>氟比洛芬>布洛芬>萘普生。在各自的优化色谱条件下,将方法应用于上述5种药物制剂的含量测定,结果令人满意。  相似文献

在纤维素-三(3,5-二甲基苯基氨基甲酸酯)(CDMPC)手性固定相上,分别采用正相、反相及极性有机相色谱模式对氟环唑外消旋体进行了拆分,并考察了流动相组成在手性识别中对手性分离的影响。氟环唑在Chiralcel OD-H手性色谱柱(填充CDMPC手性固定相)上采用反相色谱模式,以甲醇-水(体积比为80∶20)为流动相,获得了最佳的拆分,其两对对映异构体的分离度Rs分别为1.64和6.50。  相似文献

Determination of acetaminophen and its main impurities: 4-nitrophenol, 4'-chloroacetanilide, as well as 4-aminophenol and its degradation products, p-benzoquinone and hydroquinone has been developed and validated by a new high-performance liquid chromatography method. Chromatographic separation has been obtained on a Hypersil Duet C18/SCX column, using gradient elution, with a mixture of phosphate buffer (pH = 4.88) and methanol as a mobile phase. Analysis time did not exceed 14.5 min and good resolutions, peak shapes and asymmetries have resulted. The linearity of the method has been tested in the range of 5.0-60 μg/mL for acetaminophen and 0.5-6 μg/mL for the other compounds. The limits of detection and quantification have been also established to be lower than 0.1 μg/mL and 0.5 μg/mL, respectively. The method has been successfully applied for the analysis of commercial acetaminophen preparations.  相似文献

在反相以及正相争件下,利用自制的涂敷型纤维素-三(3,5-二甲基苯基氨基甲酸酯)手性固定相直接拆分了外消旋雷诺嗪,并考察了不同流动相对手性拆分的影响,特别是醇类物质对拆分影响。结果表明,醇的立体结构、极性对雷诺嗪的手性拆分均有影响。实验结果显示无论在正相条件下还是在反相条件下,涂敷型纤维素-三(3,5-二甲基苯基氨基甲酸酯)手性固定相均可以很好的拆分外消旋体雷诺嗪。  相似文献

Summary Amylose tris(phenylcarbamate) (ATPC) coated on a small particle silica gel was prepared. This ATPC chiral stationary phase (ATPC-CSP) was found to be useful for the enantiomeric separation of some novel chiral tetrahedrane-type clusters. Moreover, the influence of mobile phase modifier and of the structure of chiral tetrahedrane-type clusters on the chiral separation and retention were investigated. The results suggest that not only the structure and concentration of alcohol in mobile phase, but also the subtle structural differences in racemates can have a pronounced effect on enantiomeric separation and retention.  相似文献

A direct chiral LC‐UV method was optimized for the determination of salbutamol (SAL) β2‐agonist in environmental water. Two commercially available columns were evaluated: teicoplanin Chirobiotic‐T? (150 × 2.1 mm i.d., 5 µm) and vancomycin Chirobiotic‐V? (150 × 2.1 mm i.d., 5 µm). Finally, teicoplanin chiral stationary phase was selected for SAL enantiomer resolution. In order to preserve its integrity and maintain the column performance for longer time, the use of additives such as triethylamine (TEA) in the mobile phase was avoided. Experimental design was applied to simultaneously evaluate the influence of several parameters involved in enantiomer separation and to establish the conditions for acceptable resolution and performance in short analysis time. Optimum mobile phase was methanol–20 mM ammonium acetate buffer at pH 4.5 (98:2, v/v). A solid‐phase extraction procedure for sample pre‐concentration and clean‐up allowed the determination of chiral SAL residues in natural water samples spiked at low concentrations in the range 1.0–20 ng mL?1. Reproducible recoveries, between 77 and 98%, were obtained and matrix effect was negligible. Injection of sample solutions at low elution strength permitted the SAL enantioresolution in the natural water complex matrix with satisfactory sensitivity and precision. Summary High pressure liquid chromatographic methods for the determination of diphenylmethane-4,4-diisocyanate (MDI) and toluene diisocyanate (TDI) in chemical products are described. The MDI- and TDI monomers were determined as their urea derivative formed by the reaction with 9-(methyl aminomethyl)-anthracene. Using these methods MDI- and TDI monomer concentrations have been determined in 55 chemical products: sealing waxes, insulating- and adhesive foam, hardener, primer, adhesives and surface coatings. The recovery of both MDI and TDI monomers from various types of chemical product was found to be 92–97%, and the relative standard deviations of the methods was <5% for all types of products.  相似文献

高效液相色谱(HPLC)被广泛认为是分离制备光学纯单一对映体的最有效方法。在高效液相色谱手性拆分中,手性固定相(CSP)的性能直接影响到色谱柱的手性分离能力。在众多手性固定相中,键合型手性固定相具有溶剂耐受性好,分离模式灵活等优点,已经发展成为一类重要的手性固定相。本文通过两步化学反应合成了新型的光学活性丙烯酰胺衍生物--(S)-1-丙烯酰-2-(N-苯基甲酰胺基)吡咯烷((S)-APACP),采用核磁共振氢谱表征了(S)-APACP的化学结构;通过3步化学反应制备了键合型聚丙烯酰胺衍生物手性固定相,采用热重分析法表征了聚合物的键合量,采用HPLC评价了键合型手性固定相的识别能力,分析了影响其手性识别能力的因素。研究结果表明,APACP聚合物成功地键合到硅胶表面制备了具有良好溶剂耐受性的键合型手性固定相,其聚合物键合量为10.2%~11.8%,该键合型手性固定相对若干种对映体显示了较好的手性识别能力。  相似文献

以纤维素三-(3,5-二甲基苯基氨基甲酸酯)为手性固定相(Lux Cellulose-1),建立了在正相色谱条件下直接分离盐酸川丁特罗对映体的高效液相色谱法。考察了乙醇、异丙醇等有机改性剂,三氟乙酸、二乙胺等流动相添加剂和柱温对对映体分离的影响。结果显示,酸性和碱性添加剂对对映体分离的影响最为显著: 添加二乙胺时两对映体无分离趋势;添加三氟乙酸时对映体保留强,且分离趋势明显;而同时添加三氟乙酸和二乙胺则两对映体分离显著改善,分离度可达4.0。优化后的色谱条件: 色谱柱为Lux Cellulose-1手性柱(250 mm×4.6 mm, 5 μm),流动相为正庚烷-乙醇-三氟乙酸-二乙胺(88:12:0.3:0.05, v/v/v/v),流速为1.0 mL/min,紫外检测波长为246 nm,柱温为25 ℃。该方法简便,快速,可用于左旋盐酸川丁特罗原料中右旋异构体杂质的检查。  相似文献

采用直链淀粉手性固定相高效液相色谱法在正相条件下直接拆分了比索洛尔对映异构体。分别以异丙醇、乙醇为有机改性剂,考察了流动相的组成与配比、流速及柱温等因素对比索洛尔对映体分离的影响。确定了比索洛尔对映体的最佳拆分条件:流动相正己烷-乙醇-二乙胺(体积比为88∶12∶0.1),流速0.6 mL/min,检测波长270 nm,柱温20 ℃。该方法可快捷、简便地拆分比索洛尔对映体。  相似文献

Summary The high-performance liquid chromatographic resolution of the enantiomers of tebuconazole, a new anti-fungal agent with one chiral center, and the enantiomers of some impurities in technical tebuconazole, has been studied on a chiral stationary phase prepared by coating aminopropylated silica gel with celluloseris(3,5-dimethylphenylcarbamate). The effects of solute structure and the amount of the organic mobile-phase modifier, 2-propanol, on retention and resolution were studied. Under optimum conditions excellent enantiomer separations were achieved for tebuconazole and its impurities. As far as we are aware this is the only liquid chromatographic system enabling discrimination of the enantiomers of all of the racemates discussed in this paper.  相似文献

使用Chiralpak IC(纤维素-三(3,5-二氯苯基氨基甲酸酯)共价键合硅胶)手性柱,建立了采用手性固定相高效液相色谱拆分6种 α -芳基萘满酮类衍生物对映体的方法。考察了流动相中有机改性剂的种类和比例、柱温和流速对对映体分离的影响。结果显示6种化合物在异丙醇为改性剂的条件下均可获得较高的对映体分离度。热力学研究表明6种化合物对映体的手性拆分过程均受焓驱动影响,且低温有利于对映体分离。最终推荐分离化合物Ⅰ对映体的流动相是正己烷-异丙醇(90:10,v/v);分离化合物Ⅱ、Ⅲ、Ⅳ对映体的流动相是正己烷-异丙醇(99:1,v/v);分离化合物Ⅴ对映体的流动相是正己烷-异丙醇(85:15,v/v);分离化合物Ⅵ对映体的流动相是正己烷-异丙醇(80:20,v/v)。柱温为25℃,流速为1.0 mL/min。6种化合物对映体均可在Chiralpak IC手性固定相上得到完全分离,证明该色谱柱对6种化合物具有较高的对映体选择性。  相似文献

目前环糊精(CD)手性固定相(CSP)的研究大多集中于对CD或桥联臂进行功能衍生引入更多作用位点以提升手性拆分能力,鲜有能够反映天然CD本征识别能力的CSP的研究报道,该文通过"巯基-烯"点击化学反应合成了结构明确可控的单(6-巯基-6-去氧)-β -环糊精手性固定相(CSP1),其最大限度地保留了天然CD的本征结构,且桥联臂无识别作用位点,固体核磁共振(13 C SSNMR)和红外光谱(FTIR)的表征结果证明了CSP1的成功制备,元素分析结果表明,与双键功能化硅胶相比,CSP1的C、H、N的百分含量均得到了提高,计算得出CSP1的表面CD固载量为0.82 μmol/m2。采用高效液相色谱反相模式对50多种手性对映体包括异(口恶)唑啉、手性交酯、手性酮、黄烷酮以及丹磺酰氨基酸等进行了手性拆分,充分考察了天然CD的本征手性识别能力,结果表明CD的本征识别能力比较有利于异(口恶)唑啉类样品中含有两个疏水苯环基团Ph-Ph类样品的分离,对于其他几类样品仅利于部分样品的分离。同时与前期制备的功能三唑桥联CD-CSP及咪唑嗡桥联CD-CSP在同一色谱条件下进行了结果比对,结果证明样品的分离过程除了与手性介质的结构有关外,还与样品分子的结构有很大关系,对桥联臂进行功能改性可提升对部分对映体的选择性,但同时会小幅损失CD的本征手性识别能力。对于环糊精本征识别能力易于分离的样品,在设计手性介质时,其桥联臂不需要任何官能团,这为CD固定相结构的设计提供了有益参考。  相似文献

The enantioseparation of seven novel chiral transition metal tetrahedral clusters has been achieved for the first time on cellulose derivatized with tris(3,5-dimethylphenylcarbamate) (CDMPC) as chiral stationary phase (CSP) and hexane containing different alcohols as modifiers as mobile phases. The effect of mobile-phase composition on enantioselectivity was studied, and the effect of structural variation of the solutes on their enantioseparation was also investigated. It was found that both the metal in the tetrahedral core and the ligand coordinated to the atom in the tetrahedral core had significant effects on the chromatographic behavior of the analytes.  相似文献

使用亲水性的C16硅胶反相色谱柱分离了苯胺、间甲苯胺、4-氯苯胺、2,4-二氯苯胺、3,4-二氯苯胺、2,4,6-三氯苯胺、对溴苯胺、2-萘胺及N-硝基-2-萘胺等9种芳香胺类化合物,对影响分离的几种主要条件进行了优化。结果表明以乙腈-水为流动相,使用梯度淋洗程序,这9种芳香胺类化合物可以在大约40min内得到理想分离,分离后的芳香胺类化合物可在紫外检测器上进行检测。该方法对苯胺、间甲苯胺、4-氯苯胺、2,4-二氯苯胺、3,4-二氯苯胺、2,4,6-三氯苯胺、对溴苯胺、2-萘胺及N-硝基-2-萘胺的检出限分别为15、173、0、40、1.21、7、30、30、0.3μg/L。该方法可应用于环境水样中芳香胺类化合物的测定。  相似文献

We report the synthesis and enantioseparation characteristics of two novel covalently immobilized deoxycholic acid derivatives as chiral stationary phases for high‐performance liquid chromatography. In the structure of the first stationary phase, the 3‐position of deoxycholic acid is substituted with a 3,5‐dinitrophenylcarbamoyl group and the second one has an additional calix[4]arene attached to the carboxylic group of the deoxycholic acid. The chromatographic performance of the stationary phases was evaluated with enantioseparation of N‐(3,5‐dinitrobenzoyl)‐dl ‐leucine, N‐(3,5‐dinitrobenzoyl)‐dl ‐valine, omeprazole, diclofop‐methyl, dl ‐mandelic acid and (RS)‐pregabalin. Comparison of the performance characteristics of the prepared chiral stationary phases provided evidence for the active involvement of the calix[4]arene unit in the chiral recognition process. Both stationary phases are chemically bonded to the silica and can be used in both normal‐phase and reversed‐phase modes.  相似文献
