This paper reports the influence of organic additives (alcohols, amino acids, sugars) on the micellization and cloud point (CP) of a phenothiazine drug, promethazine hydrochloride (PMT). The critical micelle concentration (CMC) of the drug, determined by surface tension measurements in the presence of a representative of each additive class (i.e., butanol, leucine, arabinose), are used to evaluate the maximum surface excess concentration (Γ max) and the minimum area per surfactant molecule (A min) at the air/water interface. Γ max increases and CMC/A min decreases with increasing concentration of the additives, which indicate mixed micelle formation. The intermicellar interaction coefficients in the mixed micelles (β m and β σ are also calculated, and their negative values imply attractive interactions. Effect of pH revealed CP decrease with increasing pH due to deprotonation of PMT molecules. Effect of amino acids depended upon their nature and polarity, whereas sugars caused a CP decreasing effect. Aliphatic alcohols as well as cycloalkanols and diols decreased the CP. In the presence of arabinose, increase in drug concentration resulted in the CP increase, while increase in pH showed an opposite trend. Results are interpreted on the basis of mixed micelle formation, hydrophobic interactions, and change in solvent structure.  相似文献

Herein we report the effect of additives (salts and organics) on the cloud point (CP) of nonionic surfactant Triton X‐114 (TX‐114) aqueous solutions. CP showed a concentration dependent variation in the absence of any added compound. Addition of quaternary ammonium (or phosphonium) bromides to 0.8 mM TX‐114 solutions increased the CP. It was found that long chain alcohols and amines decreased the CP of 0.8 mM TX‐114 +80 mM Bu4AmB aqueous system, while it either remained constant or increased in the presence of short chain additives. The effect of first group additives (long chain) can be explained by considering that these additives solubilize in interfacial region and assist in micellar growth. Short chain additives remain in aqueous phase and affect the micelle hydration by affecting the solvent. Pentylamine behaved differently than pentanol: pentylamine increased the CP (like short chain additives) while pentanol decreased the CP. In pentylamine, the hydrophilicity of NH2 group and its dissociation into NH3 + dominates over the hydrophobicity of its alkyl chain. Aliphatic hydrocarbons first decreased and then increased the CP. The overall behavior depended upon the chain length of the hydrocarbon. With decane, the CP decreasing region disappeared completely.  相似文献

The effect of sugar on plant metabolism, which is known to be similar to hormone-like signaling, was metabolomically studied using Melissa officinalis (lemon balm). The metabolite profiles of M. officinalis treated with sucrose were analyzed by gas chromatography-mass spectrometry (GC-MS) and principal component analysis (PCA). A total of 64 metabolites from various chemical classes including alcohols, amines, amino acids, fatty acids, inorganic acids, organic acids, phosphates, and sugars were identified by GC-MS. Three groups treated with different sucrose concentrations were clearly separated by PCA of their metabolite profiles, indicating changes in the levels of many metabolites depending on the sucrose concentration. Metabolite profiling revealed that treatment with a higher sucrose level caused an increase in the levels of metabolites such as sugars, sugar alcohols, and sugar phosphates, which are related to the glycolytic pathway of M. officinalis. Furthermore, proline and succinic acid, which are associated with the proline-linked pentose phosphate pathway, the shikimic acid pathway, and the biosynthesis of phenylpropanoids, also increased with increasing sucrose concentration. Therefore, these metabolic changes induced by sucrose ultimately led to the increased production of flavonoids such as caffeic acid via the biosynthetic pathway of phenylpropanoids. This study demonstrated that the abundance changes in some primary and secondary metabolites were somewhat interlocked with each other in response to sucrose.  相似文献

The cloud point (CP) studies on aqueous solutions of two ethylene oxide-propylene oxide triblock copolymers (EO)2.5(PO)31(EO)2.5 and (EO)13(PO)30(EO)13 with varying number of ethylene oxide (EO) units were carried out in the presence of series of additives, such as alkali, acids, ionic surfactants, alcohols, salts, and hydrotropes. The results of this study show that sodium hydroxide decreases the CP of the two copolymers. Acids increase the CP in the order hydrochloric acid > acetic acid > formic acid for both the triblock copolymers. Hydrotropes increase the CP, whereas salts decrease or increase the CP based on their salting-out/salting-in nature. Alcohols, which are polar organic additives, affect the CP of the two copolymers differently. The change in the CP of the triblock copolymers depends upon the structure and concentration of the additives and on the number of EO units of the two triblock copolymers.  相似文献

Surfactants/polymers are used extensively in drug delivery as drug carriers. We herein report the effect of surfactants and polymers on the cloud point (CP) of amphiphilic drug chlorpromazine hydrochloride. At fixed drug concentration (50 mM) and pH (6.7) these additives affect the CP in accordance to their nature and structure: anionic surfactants show an increase followed by a decrease, whereas cationic (conventional as well as gemini) and nonionic surfactants show continous increase. The behavior with polymers is dictated by the number of units present in a particular polymer. Increase in drug concentration and pH, in presence of fixed amounts of CTAB, increases and decreases the CP, respectively. Variation of CP with pH at various fixed gemini concentrations shows that gemini surfactants are better candidates for drug delivery.  相似文献

Cloud point (CP) phenomenon occurring in amphiphilic drug chlorpromazine hydrochloride (CPZ) solutions with and without salts is reported herein. The CP of a 50mM CPZ solution (prepared in 10mM sodium phosphate, SP, buffer) was found to decrease with increasing pH, both in the absence as well as presence (50mM) of added salts (NaCl, NaBr, LiBr, KBr, tetra-n-butylammonium bromide). Whereas, at a fixed concentration of NaCl, the CP increased with increasing CPZ concentration, addition of increasing amounts of salts (NaF, NaCl, NaBr, LiCl, KCl) to 50mM CPZ solution (at pH 6.7) caused continuous increase in CP. On the basis of these studies the binding-effect orders of counterions and co-ions have been deduced, respectively, as: Br(-)>Cl(-)>F(-) and Li(+)>Na(+)>K(+). The similar trend of increasing CP with addition of increasing amounts of quaternary bromides (tetramethylammonium bromide, TMeAB; tetraethylammonium bromide, TEtAB; tetra-n-propylammonium bromide, TPrAB; tetra-n-butylammonium bromide, TBuAB; tetra-n-pentylammonium bromide, TPeAB) to 50mM CPZ solutions (at pH 6.7) was found to be dependent upon the alkyl chain length of the particular salt. The overall behaviour has been discussed in terms of electrostatic interactions, micellar growth, and mixed micelle formation.  相似文献

Antidepressant drug imipramine hydrochloride (IMP) is amphiphilic which shows surfactant-like behavior in aqueous solutions. We have studied the effect of adding electrolytes and non-electrolytes on the micellar behavior of IMP by making cloud point (CP) and dye solubilization measurements. The CP of a 100mM IMP solution (prepared in 10mM sodium phosphate (SP) buffer) was found to decrease with increasing pH, both in the absence as well as presence of added salts. Increase in pH increased the visible absorbance of Sudan III dye solubilized in the drug micelles, implying micellar growth. Addition of increasing amounts of salts to 100mM IMP solutions (at pH 6.7) caused continuous increase in CP due to micellar growth. On the basis of these studies, the binding-effect orders of counter- and co-ions have been deduced, respectively, as: Br(-)>Cl(-)>F(-) and Li(+)相似文献

The effect of different kinds of additives (electrolytes, nonelectrolytes, hydrotropes, and surfactants) on the cloud point (CP) of low molecular weight and narrow dispersed poly(n-isopropylacrylamide) (PNIPAM) synthesized via reversible addition-fragmentation chain transfer (RAFT) controlled radical polymerization was examined. The CP showed a concentration dependent variation and it is greatly modified in the presence of additives. The size of the random polymer coil at 30°C obtained from dynamic light scattering (DLS) measurements is often influenced by the presence of additives. We have explained the effects of different additives on PNIPAM in terms of their interaction with polymer and resultant changes in the coil structure.  相似文献

The synergistic effect of two acidic amino acids, aspartic and glutamic acid, on the electrophoretic enantioseparation of four basic drugs was evaluated in the BGE containing a CD and at different pHs. Chlorpheniramine, hydroxyzine, propranolol and tramadol were used as the basic model drugs. However, no enantioseparations were achieved with a BGE containing sole amino acid, but the combined use of an acidic amino acid and a CD showed improved enantioseparations (synergistic effect) compared with the single CD system. The results demonstrated that at optimized pH, the electrostatic interactions of the anionic amino acids with the positively charged basic drugs could result in a decrease of the analyte migration velocity and it consequently improved the enantioseparation. The effective parameters such as the amino acid and chiral selector type and concentration, buffer pH, applied voltage, and capillary temperature were optimized. Favorable enantiomeric resolution and migration times of the model drugs were achieved with a 100 mM phosphate buffer solution (pH 3.0) containing 5.0 mM HP‐α‐CD/HP‐β‐CD and 20 mM aspartic acid with an 18 kV applied voltage at 25°C. 1H NMR experiments were also carried out in a mixture of an analyte and CD in the absence and presence of aspartic acid. The NMR results were consistent with the results obtained by CE which showed the synergistic effect of amino acid.  相似文献

Herein we report clouding phenomenon occurring in amphiphilic drug promethazine hydrochloride (PMT) in the presence of electrolytes. The CP of 50 mM drug solution, prepared in 10 mM sodium phosphate buffer, was found to decrease with increasing pH due to deprotonation of drug molecules at high pH. Addition of inorganic salts (KF, KCl and KBr) to drug solutions at fixed pH (6.7) and drug concentration (50 mM) caused an increase in CP. The results have been discussed on the desorption/adsorption of counterions to the headgroups. Cations also increased the CP by affecting the water structure with their effectiveness being in the order: Li+<Na+<K+<NH4 +. In the presence of NaCl, increase in drug concentration increased the CP while increase in pH showed an opposite trend.  相似文献

Dynamic light scattering, cloud point (CP), and surface tension studies have been carried out to examine the influence of six (including two acetates, two alkoxyethanols, and two sugars) nonelectrolyte additives on two silicone surfactants based on poly (dimethyl siloxane)—graft—polyethers in aqueous solutions. The results indicate that the presence of alkoxyethanols induced the oblate ellipsoidal to spherical micellar transition, while sugars increase the size of the ellipsoidal micelles. The effect of cosolvent or additives on critical micelle concentration (CMC) and CP is discussed on the basis of water structure making and breaking effect. The thermodynamic and surface active parameters were calculated from the surface tension isotherm curves.  相似文献

以针叶材杉木树皮和阔叶材桉木树皮为原料,利用X射线衍射 (XRD) 对两种树皮热解前后固体颗粒微晶结构的变化进行了比较,利用气质联用 (GC-MS)、凝胶渗透色谱 (GPC) 等手段对两种树皮生物油组分进行了对比分析。结果表明,两种树皮中的纤维结构及脂肪链结构在热解过程中发生了分解,基本被破坏。杉木树皮和桉木树皮生物油主要组分相似,含有酸类、酮类、酚类、醇类、醛类、糖类、酯类等类物质,但相对含量存在差异;桉木树皮相对杉木树皮热解生成了较多的酸类、酮类物质,而酚类、醇类、糖类物质相对较少。两种树皮生物油中酚类物质占有较大的比例,以苯酚和邻苯二酚为主。两种树皮生物油主要物质分子量集中在300~500 g/mol,但桉木树皮生物油中分子量在300~500 g/mol的相对含量 (48.18%) 相比杉木树皮 (61.14%) 较少。  相似文献

The separation of racemic derivatized amino acids (N-acetyl) into their enantiomers was achieved using capillary zone electrophoresis employing vancomycin as a chiral selector. Due to the strong absorption properties of the chiral selector at the low wavelengths used, the partial-filling countercurrent method was adopted in order to improve method sensitivity. In the separation system studied, the chiral selector filled only a part of the capillary and, due to the appropriate selection of the pH, was moving in the opposite direction of the analytes keeping the detector free from absorbing compounds. The effect of several experimental parameters on the enantioresolution of analytes was studied, e.g., vancomycin concentration (0-5 mM), pH of the background electrolyte (pH 4-7), capillary temperature (15-35 degrees C), and the presence of an organic modifier in the run buffer (methanol or ethanol or n-propanol). N-Acetyl glutamic acid, serine, cystine, tyrosine, and proline were all baseline-resolved into their enantiomers and the enantioresolution factor (R(s)) was increased by raising the vancomycin concentration. pH 4 allowed the baseline resolution of the five studied analytes in the presence of 2.5 mM of chiral selector and an increase in pH caused a decrease of R(s).  相似文献

The stability, storage, and distribution of certified reference materials which consist of liquid or frozen fruit juices is rather problematic. As a better alternative a stable orange juice powder has been obtained by freeze-drying to a moisture content below 0.4%. However, freeze-drying lemon juices resulted in unstable materials which collapsed into lumps after standing some weeks in brown bottles under argon. An additive was sought which can be used to obtain stable lemon juice powders without influence on important analytical parameters as amino acids, sugars, organic acids, minerals and flavanone glycosides. The additives pectin, hydroxy ethylcellulose, and hydroxypropyl methylcellulose were examined. Powders containing pectin quickly showed a loss of structure (collapse) and resulted in non-acceptable changes in amino acids, sugars or minerals compared to the original lemon juice. The addition of up to 3% hydroxypropyl methylcellulose seemed more promising, although some minor changes in the sugar and sodium concentration occurred.  相似文献

Micellar electrokinetic chromatography (MEKC) was used for the chiral separation of uncharged analytes (C- and N-protected amino acids). Sodium dodecyl sulfate (SDS) was the micelle forming agent, and different cyclodextrin (CD) derivatives were added as chiral selectors. Suitable conditions for the enantioseparation were found by variation of the separation conditions. The influence of addition of organic solvents like acetonitrile or methanol, and other chiral additives (camphor-10-sulfonic acid, malic acid) was examined. The addition of an organic modifier resulted in different effects on micelle formation, and thereby on the separation. The used chiral additives did not improve the selectivity. Furthermore, dependence of the electroosmotic flow (EOF), and the capacity factors on the concentration of CDs was investigated. Increasing the CD concentration, both the EOF to a smaller extent as well as the capacity factors decrease. Nevertheless, the enantioseparation is improved with a CD-concentration up to 30 mM. Higher CD-concentrations reduce the separation of the analytes.  相似文献
