New microporous particles with large pore size (mean pore diameter of 820 nm) are successfully prepared from a mixture of cellulose and konjac glucomannan (RC-KGM3) in 1.5 M NaOH-0.65 M thiourea aqueous solution by coagulating with 5 weight percentage (wt%) CaCl(2), and then 2 wt% HCl aqueous solution. A preparative size-exclusion chromatographic (SEC) column packed with the gel particles is used for the fractionation of a dextran in water. The exclusion limit and fractionation range of the stationary phase are molecular masses of 125 yen 104 g/mol and 5.6 yen 104 to 125 yen 104 g/mol, respectively. The dextran [dextran 50, weight-average molecular mass (M(w)) = 40.1 yen 104 g/mol, polydispersity index (d) = 3.5] is fractionated by the preparative SEC column to obtain six fractions, and four of them are refractionated twice by the same preparative SEC column. The refractionated samples F-3-3 and F-4-3 are characterized by analytical SEC combined with laser light scattering and light scattering to obtain M(w) of 91.8 and 61.9 yen 104 g/mol, as well as d of 1.3 and 1.4, respectively. The results indicate that the fractions having narrow molecular mass distribution are satisfactorily prepared with the SEC column. The described SEC column can be successfully used to fractionate polymers in aqueous solution.  相似文献

A lignosulfonate sample was fractionated according to the solubility in ethanol-water. The fractions were analysed by aqueous size exclusion chromatography (SEC) combined with in-line multi-angle laser light scattering (MALLS), and by static MALLS. Satisfactory SEC results were obtained with aqueous phosphate buffer containing DMSO and SDS. The refractive index increment (dn/dc) varied from 0.186 to 0.205 ml/g, depending on Mw and the degree of sulfonation. The second viral coefficient (A2) was 7 x 10(-3) ml mol/g2. The weight-average molecular weight (Mw) of the fractions varied from 4600 up to 398 000 g/mol. and the polydispersity (Mw/Mn) varied between 1.3 and 3.5.  相似文献

A series of short block length methoxy poly(ethylene glycol)-block-poly(caprolactone) diblock copolymers was synthesized and characterized in order to assess the potential of these copolymers as a micellar drug-delivery system. Varying the caprolactone:MePEG weight ratio in the reaction mixture allowed the synthesis of diblock copolymers with a MePEG molecular weight of 750 g/mol and PCL block lengths of 2, 5 or 10 repeat units. Phase diagrams of aqueous solutions of the copolymers were constructed which displayed characteristic cloud points and Krafft points. As the degree of polymerization of PCL increased, critical micelle concentration (CMC) values decreased from 6.97 x 10(-1) to 3.38 x 10(-3) g/l, partition equilibrium coefficients (Kv) increased from 1.09 x 10(4) to 22.2 x 10(4),and hydrodynamic diameters increased from 12.2 to 19.5 nm. The micelle morphology was determined to be spherical by transmission electron microscopy.  相似文献

Microporous regenerated cellulose gel particles were prepared by mixing cellulose cuoxam with silk fibroin aspore former, and the mean pore size and pore volume of the pallicles were 525 nm and 7.27 mL g~(-1), respectively. Apreparative size-exclusion chromatography (SEC) column (550 mm×20 mm) packed with the cellulose gel particles wasused for the fractionation of two polysaccharides Dextran 07 (M_w = 7.14×10~4, d= 1.7) and Dextran 50(M_w = 50.5×10~4,d = 3.8) in water phase. The fractionation range of the stationary phase covered M_w from 3×10~3 to 1.1×10~6. The dailythroughput was 2.9 g for Dextran 07 (D07) and 4.3 g for Dextran 50 (D50) with a flow-rate of 1.5 mL min~(-1). The fractionsobtained by using the SEC were analyzed by an analytical SEC combined with laser light scattering (LLS), and thepolydispersity indices of fractions for Dextran 07 and Dextran 50 were determined to be 1.34-1.57 and 1.53-3.36,respectively. The preparative SEC is a simple, rapid, and suitable means not only for the fractionation of polysaccharides inwater but also for other polymers in organic solvents.  相似文献

Pseudo-first-order rate constants (k(obs)) for alkaline hydrolysis of N-benzylphthalimide (1) show a nonlinear decrease with the increase in [C(m)E(n)]T (total concentration of Brij 58, m = 16, n = 20 and Brij 56, m = 16, n = 10) at constant [CH(3)CN] and [NaOH]. These nonionic micellar effects, within the certain typical reaction conditions, have been explained in terms of the pseudophase micellar (PM) model. The values of micellar binding constants (KS) of 1 are 1.04 x 10(3) M(-1) (at 1.0 x 10(-3) M NaOH) and 1.08 x 10(3) M(-1) (at 2.0 x 10(-3) M NaOH) for C(16)E(20) as well as 600 M(-1) (at 7.6 x 10(-4) M NaOH) and 670 M(-1) (at 1.0 x 10(-3) M NaOH) for C(16)E(10) micelles. The pseudo-first-order rate constants (kM) for hydrolysis of 1 in C(16)E(20) micellar pseudophase are approximately 90-fold smaller than those (kW) in water phase. The values of kM for hydrolysis of 1 in C(16)E(10) micelles are almost zero. Kinetic coupled with UV spectral data reveals significant irreversible nonionic micellar binding of 1 molecules in the micellar environment of nearly zero hydroxide ion concentration at >or=0.14 M C(16)E(20) and 1.0 x 10(-3) M NaOH while such observations could not be detected at or=3 x 10(-3) M C(16)E(10) and 7.6 x 10(-4) M NaOH, while the rate of hydrolysis of 1 is completely ceased at >or=0.05 M C(16)E(10) and 7.6 x 10(-4) M NaOH. The rate of hydrolysis of 1 at 5.0 x 10(-2) and 8.8 x 10(-2) M C(16)E(10) and 1.0 x 10(-3) M NaOH reveals the formation of presumably phthalic anhydride, whereas such observation was not observed in the C(16)E(20) micellar system under similar experimental conditions.  相似文献

Regenerated cellulose microspheres (RCM) with different diameters were prepared from cellulose solution using 7 wt% NaOH/12 wt% urea aqueous solvent pre-cooled to −12 °C by the sol–gel transition method via a “green” process. By varying the hydrophile–lipophile balance, the amount of the surfactants, the proportion of the water to the oil phase and the stirring speed, the mean diameter of the cellulose microsphere with nano-scale pore size could be controlled easily from 5 μm to 1 mm. The structure and physicochemical properties of the microspheres were characterized by FT-IR spectroscopy, scanning electron microscopy, X-ray diffraction, mercury intrusion-porosimetry and particle size analyzer. The RCM microspheres exhibited spherical shape with the cellulose II structure. A preparative size-exclusion chromatography (SEC) column packed with the cellulose microspheres was used for the fractionation of a polyethylene oxide (PEO) in water, which indicated high efficiency for the fractionations and a large daily throughput of 4 g. Moreover, they had good adsorption capacity to dye particles through physical interaction. The cellulose microspheres would have potential applications in the fields of purification, separation and fractionation of polymers as chromatography packing and adsorbent both at laboratory and industrial scale.  相似文献

The apparent second-order rate constant (k OH) for hydroxide-ion-catalyzed conversion of 1 to N-(2'-methoxyphenyl)phthalamate (4) is approximately 10(3)-fold larger than k OH for alkaline hydrolysis of N-morpholinobenzamide (2). These results are explained in terms of the reaction scheme 1 --> k(1obs) 3 --> k(2obs) 4 where 3 represents N-(2'-methoxyphenyl)phthalimide and the values of k(2obs)/k(1obs) vary from 6.0 x 10(2) to 17 x 10(2) within [NaOH] range of 5.0 x 10(-3) to 2.0 M. Pseudo-first-order rate constants (k(obs)) for alkaline hydrolysis of 1 decrease from 21.7 x 10(-3) to 15.6 x 10(-3) s(-1) with an increase in ionic strength (by NaCl) from 0.5 to 2.5 M at 0.5 M NaOH and 35 degrees C. The values of k obs, obtained for alkaline hydrolysis of 2 within [NaOH] range 1.0 x 10(-2) to 2.0 M at 35 degrees C, follow the relationship k(obs) = kOH[HO(-)] + kOH'[HO (-)] (2) with least-squares calculated values of kOH and kOH' as (6.38 +/- 0.15) x 10(-5) and (4.59 +/- 0.09) x 10(-5) M (-2) s(-1), respectively. A few kinetic runs for aqueous cleavage of 1, N'-morpholino-N-(2'-methoxyphenyl)-5-nitrophthalamide (5) and N'-morpholino-N-(2'-methoxyphenyl)-4-nitrophthalamide (6) at 35 degrees C and 0.05 M NaOH as well as 0.05 M NaOD reveal the solvent deuterium kinetic isotope effect (= k(obs) (H 2) (O)/ k(obs) (D 2 ) (O)) as 1.6 for 1, 1.9 for 5, and 1.8 for 6. Product characterization study on the cleavage of 5, 6, and N-(2'-methoxyphenyl)-4-nitrophthalimide (7) at 0.5 M NaOD in D2O solvent shows the imide-intermediate mechanism as the exclusive mechanism.  相似文献

A potentiometric and/or visual method for the determination of sulphamethoxazole (SULF) in pure form in the range of 5.3 x 10(-5) to 5.0 x 10(-4) mol/L is proposed. To enhance the solubility, the determination has been carried out in a micellar medium formed by an aqueous 5.0 x 10(-2) mol/L N-hexadecyl-N,N,N-trimethyl ammonium bromide (CTAB) solution which increases the dissociation constant K(alpha) of SULF about tenfold. The titration is performed with NaOH (2.5 x 10(-3) or 5.0 x 10(-2) mol/L) and the end-point is determined by the second derivative graph. The results agree with those obtained by the official method of the USP XXIII. The method is simple, accurate, economical and can successfully replace the more complicated, more expensive and time-consuming existent procedures which are carried out in a non-aqueous medium.  相似文献

An aqueous deoxygenated dilute PVME solution (cP = 0.5 mass %) was irradiated with gamma-rays at different radiation doses (D = 0.2-10 kGy). At these concentrations and doses contracted molecules are formed but no macroscopic networks. For the analysis of the structural changes, the irradiated samples were analyzed by size exclusion chromatography (SEC) with a triple detector system, static light scattering, and viscometry. SEC measurements necessitate for the application of the universal calibration principle the knowledge of the Kuhn-Mark-Houwink (KMH) parameters. To obtain these parameters commercially available poly(vinyl methyl ether) was fractionated and analyzed by the same means. We found at 30 degrees C a KMH relationship to [eta] = 0.0226 (mL/g) x Mw(0.67) in THF. We observed an increase in molar masses without any significant structural changing for D < 0.5 kGy. With increasing radiation dose, the intramolecular cross-linking reaction becomes more and more important. The results of viscosity measurements show a slight increase in contraction for 1.0 kGy. For irradiation dose higher than D > 1.5 kGy, a strong contraction occurs. For D > 5.0 kGy, the favored intramolecular reactions lead to the formation of microgels.  相似文献

Six water-soluble polysaccharide-protein complexes coded as GM1, GM2, GM3, GM4, GM5 and GM6 wereisolated from the mycelium of Ganoderma tsugae by extracting with 0.2 mol/L phosphate buffer solution at 25, 40 and80℃, water at 120℃, 0.5 mol/L aqueous NaOH solution at 25 and 65℃, consecutively. Their chemical components wereanalyzed by using IR, GC, HPLC and ~(13)C-NMR, and some new results were obtained. The four samples GM1, GM2, GM3and GM4 are heteropolysaccharide-prote in complexes, in which, α- (1→3) linked D-glucose is the major monosaccharidewhile galactose, mannose and ribose are the secondary ones. GM5 and GM6 are β-(1→3)-D-glucan-protein complexes. Theprotein content increased from 32% to 69% with the progress of isolation. Weight-average molecu1ar mass M_w and theintrinsic viscosity [η] of the GM samples in 0.5 mol/L aqueous NaCl solution at 25℃ were measured systematically by laserlight scartering (LLS), size exclusion chromatography (SEC) combined with LLS, and viscometry. The M_w of GM1 to GM6are 35.5, 46.8, 58.9, 41.6, 3.3 and 22.0×10~4, respectively. The conformation and molecular mass of the two fractions of sample GM5 were characterized satisfactorily by SEC-LLS without further fractionation.  相似文献

Poly[n-hexyl-(S)-3-methylpentylsilane] aggregates confined in microcapsules to keep the aggregation number and ranging in average polymer mass m(p) in a microcapsule from 2 x 10(-)(16) to 2 x 10(-)(14) g were studied by circular dichroism measurements in ethanol (a nonsolvent) and tetrahydrofuran (an associative solvent at low temperature) at various temperatures. The weight-average molecular weight M(w) and the polydispersity index (the ratio of M(w) to the number-average molecular weight) of the polysilane sample were 6.6 x 10(4) and 1.07, respectively, and the average number of polymer molecules in each capsule was estimated to be 1.9 x 10(3) for m(p) = 2 x 10(-)(16) g and 1.7 x 10(5) for m(p) = 2 x 10(-)(14) g. The size of each aggregate did not affect the optical activity because the circular dichroism thus obtained was proportional to m(p) under the same conditions in the investigated m(p) range; on the other hand, the peak height of the circular dichroism in tetrahydrofuran had a significant hysteresis between 0 and 25 degrees C. Moreover, the circular dichroism appreciably reflected the prepared method, that is, the temperature and solvent; in other words, the aggregates memorized the initial conditions in their stacking structures.  相似文献

Capillary zone electrophoresis was employed for the determination of clozapine using an end-column amperometric detection at a carbon fiber array microdisk electrode with simplified capillary/electrode alignment. The optimum conditions of separation and detection are: Britton-Robinson buffer, pH 2.0 (1.3 x 10(-2) mol/L total concentration of acids, 3.2 x 10(-3) mol/L NaOH), 15 kV for separation voltage, 5 kV and 10 s for injection voltage and injection time, respectively. The limit of detection is 4.2 x 10(-7) mol/L or 1.2 fmole (signal to noise, S/N = 2). The relative standard deviation is 1.4% for the migration time and 2.5% for the electrophoretic peak current. The method was applied to the determination of clozapine in human blood. The recovery of the method is between 94-104%.  相似文献

Poly(epsilon-caprolactone) (PCL) macromers (M(n) = 1.7-3.8 kDa) which contain one Z-protected -NH2 group per chain were synthesized by ring-opening polymerization of epsilon-caprolactone in the presence of Sn(oct)2 using as initiator a diamine prepared by condensation of N-Boc-1,6-hexanediamine and N(alpha)-Boc-N(epsilon)-Z-L-Lysine. The coupling of these macromers with -COCl end-capped poly(oxyethylene) (PEO), M(n) = 1.0 kDa, afforded amphiphilic multiblock poly(ether ester)s (PEEs) which have, along the chain, regularly spaced pendant protected amino groups. Deprotection, accomplished without chain degradation, yielded -NH2 groups available for further reactions. The molecular structure of macromers and PEEs was investigated by 1H NMR and SEC. DSC and WAXS analyses showed that macromers and copolymers were semicrystalline and their T(m) increased with increase in the molecular weight of PCL segments. The inherent viscosity values (0.25-0.30 dL x g(-1)), together with SEC analysis results, indicated moderate polymerization degrees.  相似文献

The evidences of spontaneous oxidation of Mn(II) by the dissolved oxygen in azide buffer medium, which is dependent on the N (-)(3)HN (3) concentration, suggested a formation of stable Mn(III) complexes due to marked colour changes. Spectrophotometric studies combined with coulometric generation of Mn(III), in presence of large excess of Mn(II), showed a maximum absorbance peak at 432 nm. The molar absorptivity increases with azide concentration (0.44-3.9 mol 1(-1)) from 3100 to 6300 mol(-1) 1 cm(-1), showing a stepwise complex formation. Potential measurements of the Mn(III) Mn(II) system in several azide aqueous buffers solutions: 1.0 x 10(-2) mol 1(-1) HN(3), (0.50-2.0 mol 1(-1)) N(-)(3) and 5.0 x 10(-2) mol 1(-1) Mn(II) and constant ionic strength 2.0 mol 1(-1), kept with sodium perchlorate, leads to the conditional potential, E(0')x, in several azide concentrations at 25.0 +/- 0.1 degrees C. Considering the overall formation constants of Mn(II) N (-)(3), from former studies, and the potential, E(0')s = 1.063 V versus SCE, for Mn(III) Mn(II) system in non-complexing media, it was possible to calculate the Fronaeus function, F(0)(L), and the following overall formation constants: beta(1) = 1.2 x 10(5) M(-1), beta(2) = 6.0 x 10(8) M(-2), beta(3) = (2.4 +/- 0.7) x 10(11) M(-3), beta(4) = (1.5 +/- 0.5) x 10(11) M(-4) and beta(5) = (9.6 +/- 0.8) x 10(11) M(-5) for the Mn(III) N (-)(3) complexes. These data give important support to understand the importance of Mn(II) and Mn(III) synergistic effect on the analytical method of S(IV) determination based on the Co(II) autoxidation.  相似文献

The sorption of Hg(II) in the presence of sodium thiocyanate solution onto polyurethane (PUR) foam, an excellent sorbent, has been investigated in detail. Maximum sorption of Hg(II) is achieved from 0.1 M hydrochloric acid solution containing 7.5x10(-2) M sodium thiocyanate in 5 min. The sorption data followed both Freundlich and Langmuir adsorption isotherms. The Freundlich constants 1/n and sorption capacity, C(m), are evaluated to be 0.44+/-0.02 and (3.86+/-0.89)x10(-3) mol g(-1). The saturation capacity and adsorption constant derived from Langmuir isotherm are (6.88+/-0.28)x10(-5) mol g(-1) and (5.6+/-0.37)x10(4) dm(3) mol(-1) respectively. The mean free energy (E) of Hg(II)-SCN sorption onto PUR foam computed from D-R isotherm is 12.4+/-0.3 kJ mol(-1) indicating ion-exchange type mechanism of chemisorption. The variation of sorption with temperature yields thermodynamic parameters of DeltaH=-30.7+/-1.2 kJ mol(-1), DeltaS=-70.1+/-4.1 J mol(-1) K(-1) and DeltaG=-9.86+/-0.77 kJ mol(-1) at 298 K. The negative value of enthalpy and free energy reflects the exothermic and spontaneous nature of sorption. On the basis of the sorption data, sorption mechanism has been proposed.  相似文献

A novel cellulose solvent, 1.5 M NaOH/0.65 M thiourea aqueous solution, was used to dissolve cotton linters having a molecular weight of 10.1 × 104 to prepare cellulose solution. Regenerated cellulose (RC) films were obtained from the cellulose solution by coagulating with sulfuric acid (H2SO4) aqueous solution with a concentration from 2 to 30 wt %. Solubility of cellulose, structure, and mechanical properties of the RC films were examined by infrared spectroscopy, X‐ray diffraction, scanning electron microscopy, 13C NMR, and tensile tests. 13C NMR analysis indicated that the novel solvent of cellulose is a nonderivative aqueous solution system. The presence of thiourea enhanced significantly the solubility of cellulose in NaOH aqueous solution and reduced the formation of cellulose gel; as a result, thiourea prevented the association between cellulose molecules, leading to the solvation of cellulose. The RC film obtained by coagulating with 5 wt % H2SO4 aqueous solution for 5 min exhibited higher mechanical properties than that with other H2SO4 concentrations and a homogenous porous structure with a mean pore size of 186 nm for free surface in the wet state. The RC film plasticized with 10% glycerin for 5 min had a tensile strength of 107 MPa and breaking elongation of 10%, and about 1% glycerin in the RC film plays an important role in the enhancement of the mechanical properties. © 2002 Wiley Periodicals, Inc. The polysaccharide having weight-average molecular weight M_w=1. 09×10~5, isolated from the sap of lac trees (Vietnam), was separated into 12 fractions by aqueous-phase preparative gel permeation chromatography. The molecular weights and molecular weight distributions of the fractions were measured in aqueous 0.08M KCl/0.01 M NaAc and 0.4M KCl/0.05M NaAc at pH =7. 6 by light scattering, viscometry and gel permeation chromatography. The Mark-Houwink equation in aqueous 0.08M KCl/0.01M NaAc at 30℃was found to be [η]= 2.28×10~(-2) M_w~(0.52) (cm~3/g), which indicated the polysaccharide chain in the aqueous solution to be a spherical random coil.  相似文献

Two samples of cellulose (molecular weight 2.97 × 105 and 1.25 × 105) were transformed into carbanilates (CTC) which were then fractionated by the elution method at a constant composition of the acetone-water elution mixture with the column temperature gradually increasing from ?30°C to 30°C, and by the GPC method in acetone and tetrahydrofuran. Tetrahydrofuran appeared to be a more suitable solvent. The molecular weights of fractions obtained by the elution fractionation were determined by the light-scattering method in tetrahydrofuran. The width of fractions was determined by the GPC method (average M w/M n = 1.37); the [η] values and the Mark-Houwink constants (K = 5.3 × 10-3, a = 0.84) for tetrahydrofuran at 25°C were determined. The calibration curve for the GP method was constructed by means of the fractions thus obtained; it was demonstrated that the universal calibration curve according to Benoit can also be used. It was demonstrated that the molecular weight distribution of cellulose can be conveniently determined by conversion into CTC followed either by the elution fractionation (for preparative purposes) or by fractionation by the GPC method (for analytical purposes).  相似文献

A new post-chemiluminescence (PCL) reaction was observed when piroxicam was injected into the reaction mixture after the CL reaction of N-bromosuccinimide (NBS) and luminol. A possible reaction mechanism was proposed based on the studies of the CL kinetic characteristics, the CL spectra, fluorescence spectra and other experiments. A new flow injection CL method for the determination of piroxicam was established based on the PCL reaction. The relative standard deviation (RSD) for the determination of piroxicam was 1.2% (n = 11, c = 2.0 x 10(-6) g/ml). The CL intensity responded linearly to the concentration of piroxicam in the range 1.0 x 10(-7) - 1.0 x 10(-5) g/ml (r = 0.9991). The detection limit was 4.0 x 10(-8) g/ml. The method had been applied to the determination of piroxicam in tablets with satisfactory results.  相似文献

Critical concentrations of α-(1→3)-D-glucan L-FV-Ⅱ from Lentinus edodes were studied by viscometry andfluorescence probe techniques. The dependence of the reduced viscosity on concentration of the glucan in 0.5 mol/L NaOHaqueous solutions with or without urea showed two turning points corresponding to the dynamic contact concentration c_s andthe overlap concentration c~* of the polymer. The values of c_s and c~* were found to be 1×10~(-3) g cm~(-3) and 1.1×10~(-2) g cm~(-3),respectively, for L-FV-Ⅱ in 0.5 mol/L NaOH aqueous solutions. The two critical concentrations of L-FV-Ⅱ in 0.5 mol/LNaOH aqueous solutions were also found to be 1.2×10~(-3) g cm~(-3) fbr c_s and 9.2×10~(-3) g cm~(-3) for c~* from the concentrationdependence of phenanthrene fluorescence intensities. The overlap concentration c~* of L-FV-Ⅱ in 0.5 mol/L NaOH aqueoussolutions was lower than that of polystyrene with same molecular weight in benzene, owing to the fact that polysaccharidetends to undergo aggregation caused by intermolecular hydrogen bonding. A normal viscosity behavior of L-FV-Ⅱ in 0.5 mol/L urea/0.5 mol/L NaOH aqueous solutions can still be observed in an extremely low concentration range at 25℃.  相似文献
