Ochratoxin A (OTA) is one of the most studied mycotoxins, with great public health and agroeconomic significance. The exposure of both humans and animals to this fungal toxin has been associated to food matrices since its discovery. However, according to recent reports, OTA may also represent a potential airborne hazard, in water-damaged buildings, or occupational contamination, in workplaces with high mould exposure, such as agricultural, farm and alimentary industries. Further, in addition to the conventional studied food matrices, worldwide consumed, there are increasing reports on different and less obvious sources of alimentary exposure. These include traditional and home-made food products, highly consumed by specific groups of populations that thus might be at risk. This paper aims to spotlight the current knowledge on these non-conventional OTA exposure sources.  相似文献

Contamination of food by mycotoxin occurs in minute/trace quantities. Nearly 92.5% of the cocoa samples present Ochratoxin A (OTA) levels at trace quantity. Hence, there is a necessity for a highly sensitive and selective device that can detect and quantify these organic toxins in various matrices such as cocoa beans. This work reports for the first time, a facile and label-free electrochemical impedimetric aptasensor for rapid detection and quantitation of OTA in cocoa beans. The developed aptasensor was constructed based on the diazonium-coupling reaction mechanism for the immobilization of anti-OTA-aptamer on screen printed carbon electrodes (SPCEs). The aptasensor exhibited a very good limit of detection (LOD) as low as 0.15 ng/mL, with added advantages of good selectivity and reproducibility. The increase in electron transfer resistance was linearly proportional to the OTA concentration in the range 0.15–2.5 ng/mL, with an acceptable recovery percentage (91–95%, RSD = 4.8%) obtained in cocoa samples. This work can facilitate a general model for the detection of OTA in cocoa beans based on the impedimetric aptasensor. The analysis can be performed onsite with pre-constructed and aptamer modified electrodes employing a portable EIS set up.  相似文献

Ochratoxin A (OTA) is a highly toxic mycotoxin and poses great threat to human health. Due to its serious toxicity and widespread contamination, great efforts have been made to evaluate its human exposure. This review focuses on the OTA occurrence and contamination level in nine plant and animal derived food commodities: cereal, wine, coffee, beer, cocoa, dried fruit, spice, meat, and milk. The occurrence and contamination level varied greatly in food commodities and were affected by many factors, including spices, geography, climate, and storage conditions. Therefore, risk monitoring must be routinely implemented to ensure minimal OTA intake and food safety.  相似文献

The aim of this study was to estimate the contamination of grain coffee, roasted coffee, instant coffee, and cocoa purchased in local markets with ochratoxin A (OTA) and its isomerization product 2′R-ochratoxin A (2′R-OTA), and to assess risk of dietary exposure to the mycotoxins. OTA and 2′R-OTA content was determined using the HPLC chromatography with immunoaffinity columns dedicated to OTA. OTA levels found in all the tested samples were below the maximum limits specified in the European Commission Regulation EC 1881/2006. Average OTA concentrations calculated for positive samples of grain coffee/roasted coffee/instant coffee/cocoa were 0.94/0.79/3.00/0.95 µg/kg, with the concentration ranges: 0.57–1.97/0.44–2.29/0.40–5.15/0.48–1.97 µg/kg, respectively. Average 2′R-OTA concentrations calculated for positive samples of roasted coffee/instant coffee were 0.90/1.48 µg/kg, with concentration ranges: 0.40–1.26/1.00–2.12 µg/kg, respectively. In turn, diastereomer was not found in any of the tested cocoa samples. Daily intake of both mycotoxins with coffee/cocoa would be below the TDI value even if the consumed coffee/cocoa were contaminated with OTA/2′R-OTA at the highest levels found in this study. Up to now only a few papers on both OTA and 2′R-OTA in roasted food products are available in the literature, and this is the first study in Poland.  相似文献

A polyclonal antibody against ochratoxin A (OTA) was produced from rabbits immunized with the OTA–BSA conjugate. A competitive direct enzyme-linked immunosorbent assay (cdELISA) and a membrane-base colloidal gold immunoassay in flow-through format were developed for the rapid detection of OTA in various food matrices. In the cdELISA, the concentration causing 50% inhibition was 0.07 ng mL−1, and the effects of different chemical conditions (ionic strength, pH value, and organic solvent) were studied. The sensitivity of the assay was higher than those previously reported. A simple, rapid, and efficient extraction method was developed and 74–110% recoveries of spiked samples were obtained. Fifty percent methanol extracts of some food samples such as barley, wheat, oat, corn, rice, and raisins could be analyzed directly by immunoassay after dilution in PBS; grape juice and beer samples could be analyzed directly after dilution with PBS; for coffee samples, a more complex method was used to remove the matrix effect effectively. Membrane-based colloidal gold immunoassays had a visual detection limit of 1.0 ng mL−1 for OTA with a detection time of less than 10 min. For the validation of the cdELISA and membrane-based colloidal gold immunoassay, samples were analyzed by high-performance liquid chromatography. The correlation between data obtained using the microwell assay and HPLC was good (R 2 = 0.984). The developed immunoassay methods are suitable for the rapid quantitative or qualitative determination of OTA in food samples.  相似文献

One hundred samples of commercial bread purchased from January to October (2006) from retail bakeshops in five different cities (Rabat, Témara, Salé, Casablanca and Meknès) in Morocco were surveyed for the presence of ochratoxin A (OTA) using liquid chromatography coupled to fluorescence detection. The identification of OTA in positive bread samples was confirmed by methyl ester derivatization. Analytical results showed that forty eight (48%) samples were positive with OTA greater than the limit of quantification (LOQ = 0.051 ng/g). Levels of OTA in positive samples ranged between 0.14 and 149 ng/g. The average contamination of bread samples with OTA was 13 ± 1.5 ng/g. The highest frequency of positive samples (61.5%) and the most contaminated sample (149 ng/g) were found in bread commercialized in the Casablanca area. Twenty six of the positive samples exceeded the maximum level of 3 ng/g set by European regulations for OTA in cereals and derivatives. Based in the results presented in this study, the estimated daily intake of OTA from bread was 126 ng/kg bw/day. The present paper is the first ever drafted on the natural occurrence of OTA in bread consumed in Morocco. Data on the daily intake of OTA by the Moroccan population are also estimated for the first time.  相似文献

This article describes a simple and homogeneous fluorescent aptasensor for the detection of ochratoxin A (OTA). With its high specificity and simplicity; RecJf exonuclease is used to cleave DNA strand of the FAM-aptamer/OTA complex and realize target recycling signal amplification. In order to avoid the loss of reaction system, magnetic beads (MBs) are added only once at the last experimental step. This proposed fluorescent aptasensor showed the higher sensitivity in the range of 0.1–100 ng/mL with LOD of 0.056 ng/mL, and the good selectivity against other interfering toxins. The feasibility of the prepared aptasensor was studied by detecting OTA in spiked liquor and cereal samples. The obtained average recoveries ranged from 92% to 115%. This study provides a promising application with convenience and rapidness in the aptasensor fabrication for food safety analysis.  相似文献

Wine and grape juices were identified as one of the most important sources of ochratoxin A (OTA), a mycotoxin with diverse toxic effects that naturally appears in food and foodstuffs all over the world.The aim of this study was to assess the OTA levels in Portuguese wines through the application of a simple and accurate method based on liquid chromatography (LC) with direct injection, followed by fluorescence detection (FD).Randomly selected wine samples were used to evaluate the performance of direct injection as efficient, fast, inexpensive and safe sample preparation method. The proposed method was successfully validated. The limit of quantification (LOQ) was 1.0 μg/L and OTA recoveries from wine samples, spiked at the three fortification levels, were higher than 85.4%, with RSDs lower than 9.6% for both red and white wines. The presence of OTA was confirmed by methyl ester derivatization followed by LC analysis.Data on OTA levels were obtained for 60 Portuguese red and white wine samples. OTA was found in 12 samples, nine (26%) red wine samples and three (12%) white wine samples. Only one red wine sample and one white wine sample presented a contamination level above the LOQ, with 1.23 and 2.4 μg/L, respectively. It should be pointed out that this white wine sample exceeded the EC maximum permitted level of 2.0 μg/L. The safe dose established as 120 ng/kg body weight/week was not exceeded by the weekly intake estimated for the samples contaminated above the LOQ.  相似文献

Abstract

Due to the growing public concern regarding food safety, reliable, nondemanding and robust analytical methods are needed for quantitative determination of toxic compounds in complex matrices. Sample preparation is frequently a crucial step in determination of ochratoxin A (OTA) in wine, and a simplified and automated procedure is described, using solid‐phase extraction coupled on‐line to high pressure liquid chromatography (HPLC) with fluorimetric detection (λex=333 nm, λem=460 nm). While the limit of quantitation is frequently better compared to off‐line procedures (30 ng/L), the decisive advantages of the new procedure are the absence of all sample manipulation during preconcentration and subsequent analysis, and consequentially no risk of analyte loss or sample contamination. Furthermore, using the standard addition method, matrix interferences can be avoided and the determination of extraction efficiency is unnecessary. These improvements have important consequences for the overall uncertainty of the analytical procedure. The developed method was applied for determination of OTA in 12 selected Slovenian wines. The typical relative standard deviation (RSD) was 10%. In none of the samples, did the OTA amount exceeded 2 µg/kg, the limit regulated by the EC.

The photo‐stability of the mycotoxin in solutions was examined. During irradiation of OTA solutions, its content was quickly reduced, while three fluorescent degradation products were detected. The degradation proceeds faster in water and 12% ethanolic solutions than in organic solvents or wine. Identification of the fluorescent degradation products was attempted using LC‐MS/MS with electrospray ionization.  相似文献

Ochratoxin A (OTA) is a carcinogenic fungal secondary metabolite which causes wide contamination in a variety of food stuffs and environments and has a high risk to human health. Developing a rapid and sensitive method for OTA detection is highly demanded in food safety, environment monitoring, and quality control. Here, we report a simple molecular aptamer beacon (MAB) sensor for rapid OTA detection. The anti-OTA aptamer has a fluorescein (FAM) labeled at the 5′ end and a black hole quencher (BHQ1) labeled at the 3′ end. The specific binding of OTA induced a conformational transition of the aptamer from a random coil to a duplex–quadruplex structure, which brought FAM and BHQ1 into spatial proximity causing fluorescence quenching. Under the optimized conditions, this aptamer sensor enabled OTA detection in a wide dynamic concentration range from 3.9 nM to 500 nM, and the detection limit was about 3.9 nM OTA. This method was selective for OTA detection and allowed to detect OTA spiked in diluted liquor and corn flour extraction samples, showing the capability for OTA analysis in practical applications.  相似文献

Mycotoxins are toxic secondary metabolites of fungal origin, the major mycotoxins of food concern are aflatoxins and ochratoxin A. Due to the wide range of matrices susceptible to mycotoxin contamination, the possible co-occurrence, and the very wide range of concentration, validated versatile multi-mycotoxin and multi-matrix methods are strongly requested. A reversed phase HPLC method for the simultaneous determination of aflatoxins and ochratoxin A in baby foods and paprika was set up. Three bulk samples were prepared according to commercial availability, one for paprika and for baby foods, two different bulks were set, a corn based and a multi-cereal based baby food. A single-laboratory validation was performed, for each investigated level ten analyses were performed, relative standard deviations of repeatability (RSDr) and recovery factors were calculated; RSDr values ranged from 2% to 10% for AFB1 and from 3% to 10% for OTA, while the recovery factors ranged from 86% to 96% for AFB1 and from 77% to 96% for OTA. The checked compliance of the RSDr and recovery with the values reported in the current EU Regulations confirmed the fitting for purpose of the method. Limit of detection and LoQ values of the method were respectively 0.002 and 0.020 μg/kg for AFB1 and 0.012 and 0.080 μg/kg for OTA in baby foods; and 0.002 and 0.200 μg/kg for AFB1 and 0.012 and 0.660 μg/kg for OTA in paprika. The current method represents a good example of the possibility of a multi-mycotoxin and/or a multi-matrix analysis depending on the laboratory research or official control purposes.  相似文献

Molecularly imprinted polymers (MIPs) displaying selective binding properties for the mycotoxin ochratoxin A (OTA) in polar/protic media were prepared. Crucial to the success of these efforts was the implementation of rationally designed OTA mimics as templates and a set of novel basic and neutral functional monomers, allowing the maximization of the template-functional monomer association via ion-pairing, hydrophobic and steric interactions. MIPs prepared with a 20:1:1:3 molar ratio of cross-linking agent, template mimic, basic functional monomer and hydrophobic auxiliary monomer produced polymers with superior recognition properties compared to materials generated with other stoichiometries. Chromatographic evaluation using the OTA mimics, OTA and a set of structurally closely related compounds as analytes revealed pronounced substrate selectivity of these MIPs in polar/protic media, the templates and OTA being bound with significantly higher affinities. Complementary substrate selectivities/affinities were observed in aprotic and apolar solvents. The possibility of solvent-dependent tuning of substrate selectivity/affinity and the high binding capacity recommend the developed MIPs as promising solid-phase extraction adsorbents for clean-up and pre-concentration of OTA from various biologically relevant matrices.  相似文献

Ochratoxin A (OTA) is a mycotoxin (potentially carcinogenic secondary metabolite derived from fungal contamination), produced by some Aspergillus and Penicillium strains. Although present and legislated in different food sources in the human diet, the regulation for wine intake is still under discussion. The Office International de la Vigne et du Vin (OIV) recommended maximum levels in wine of 2 μg l−1. Some reports refer to OTA contamination in wines up to 15 μg l−1 and a special incidence in red wines from the southern regions of Europe and the north of Africa, but the majority of the data available are below 1 μg l−1. When working at such low concentrations, the problem of the uncertainty of the results becomes decisive towards the implementation of legal limits. In order to assess the global uncertainty associated with OTA determination in wines and widen the data set and knowledge of the situation in Portugal, 340 wines were analysed (189 Port Wine, 85 Vinho Verde and 66 wines from other regions in the country) by a high performance liquid chromatography (HPLC)-fluorescence detection (FD) method using immunoaffinity columns for clean up. OTA was detected in 69 wines by the method used, but in concentrations below 0.5 μg l−1, except for two which showed levels up to a maximum of 2.1 μg l−1. However, the global uncertainty for OTA is close to 37% for concentrations above 0.5 μg l−1, and therefore, such value can be below or exceed the OIV limit. In the vicinity of the limit of detection, 0.084 μg l−1, the global uncertainty rises exponentially to a maximum of about 70%. This can be an obstacle when discussing safety intake limits. Ethanol and glucose content did not interfere in the clean up of OTA by immunoaffinity columns.  相似文献

Ochratoxin A (OTA) is a mycotoxin produced by fungi species belonging to the genera Aspergillus and Penicillium being isolated in alcoholic beverages. The aim of this work is developed and applied a procedure for the analysis of OTA in wines. An analytical method based on immunoaffinity column (IAC) for clean-up, liquid chromatography with fluorescence detection (LC-FD), and LC-FD after of OTA methylation was used to determine the occurrence of OTA in wines. Recoveries of this mycotoxin spiked to red wines at 0.5 ng/ml level were >90% with an average of relative standards deviations of 4%. Furthermore, 116 wine samples from designation of origin (DO) and three samples from food stores of Valencian Community (Spain) were examined for the occurrence of OTA being the levels of this mycotoxin ranged from <0.01 to 0.76 ng/ml. Finally, the estimated daily intake of OTA in this study was 0.15 ng/kg bw per day.  相似文献

To monitor the exposure of the Thai to pesticide residues in foods, the intake of pesticides based on the total diet approach has been studied in Thailand continually since 1989. Food items were chosen and their proportions of daily consumption were calculated from the national food consumption survey conducted by the Department of Health in 1986. Seventy-seven items of food including drinking water were classified into 12 groups according to their sources of contamination and the analytical methods used. Shopping lists and standard procedures for collecting samples and cooking were developed. Twelve food composites from each of the 4 regions were analyzed for nearly 100 pesticides. All analyses used multiresidue analytical methods, and the total daily dietary intakes of pesticide residues were estimated from the average amount of food consumed by Thais. Among 24 pesticides found in the 8 year study, DDT, dimethoate, methamidophos, and parathion methyl were found every year. However, dietary intakes of all pesticides were far below the established acceptable daily intake. In 1999, the study methodology was redesigned to accommodate the new national food consumption survey data.  相似文献

The detection of mycotoxins—toxic contaminants of fungal origin—is an important problem in the food and feed quality control. An immunochromatographic system was developed for the detection of ochratoxin A (OTA), which is one of the priority contaminants in grain. Monoclonal antibodies against OTA and their conjugates with colloidal gold nanoparticles were prepared. The detection is based on the competition of OTA in a sample and an OTA-protein conjugate immobilized on a test strip for the binding to anti-bodies on the colloidal particle surface. The method was tested in the analysis of plant extracts (maize and barley extracts). It was shown that OTA can be detected in a medium with a high content of an organic solvent (up to 35% of methanol). The disappearance of the line in the test zone is visually detected at OTA concentrations starting from 50 ng/mL. In the case of the video-digital detection of changes in the color intensity of the test zone, the limit of detection of OTA is 5 ng/mL. The duration of the assay is 10 min.  相似文献

Ochratoxin A (OTA) is a mycotoxin produced by Aspergillus and Penicillium that is widely found as a contaminant of food products. The toxin is a renal carcinogen in male rats, the cause of mycotoxicoses in pigs and has been associated with chronic human kidney diseases. Bioactivation has been implicated in OTA-mediated toxicity, although inconsistent results have been reported, due, in part, to the difficulty in detecting OTA metabolites in vivo. Liquid chromatography (LC) coupled with fluorescence detection (FLD) is the most widely used analytical detection method for OTA. Under acidic conditions the toxin generates blue fluorescence (465 nm) that is due to an excited state intramolecular proton transfer (ESIPT) process that generates an emissive keto tautomer. Disruption of this ESIPT process quenches fluorescence intensity and causes a blue shift in emission maxima. The aim of the present study was to determine the impact of the C5-chlorine atom, the lactone moiety and the amide bond on OTA fluorescence and derive optical parameters for OTA metabolites that have been detected in vitro. Our results highlight the limitations of LC/FLD for OTA metabolites that do not undergo ESIPT. For emissive derivatives, our absorption and emission data improves the sensitivity of LC/FLD (3-4-fold increase in the limit of detection (LOD)) for OTA analogues bearing a C5-OH group, such as the hydroquinone (OTHQ) metabolite and the glutathione conjugate of OTA (OTA-GSH). This increased sensitivity may facilitate the detection of OTA metabolites bearing a C5-OH group in biological fluids and enhance our understanding of OTA-mediated toxicity.  相似文献

A high-performance liquid chromatographic method (HPLC) with fluorescent detector is described for the determination of ochratoxin A (OTA). A mobile phase consisting of acetonitrile:water:acetic acid (99:99:2, v/v/v) was used for the resolution of the compound on a C(18) Hypersil column. The retention time for OTA and diflunisal which was used as an internal standard (IS) were 11.7 and 12.8 min, respectively. The method is selective, reliable, reproducable with relative standard deviation (RSD) of 1.70 and linear in the range of 2.5 x 10(-9)-1.5 x 10(-8) M OTA. The limit of detection (LOD) and limit of quantification (LOQ) were 2.5 x 10(-10) M corresponding to 0.1 ng mL(-1) and 8.2 x 10(-10) corresponding to 3.3 ng mL(-1), respectively. Recovery studies were 81.2 +/- 1.9 (SD). The method was applied for analysis of OTA in wheat, corn, red pepper, cheese and wine. The proposed method can be used for the routine analysis of OTA in food and animal feed.  相似文献
