Authentic α-santolinenone ( = (+)-(4R)-1(7)-p-menthen-2-one; (+)- 1 ) is made available for the the first time in 30% overall yield from (+)-(4R)-p-menthene ((+)- 2 ) via the diastereoisomeric allylic alcohols (+)- 4a /(+)- 4b , which are oxidized to (+)- 1 with Ag2CO3/Celite. Yields are good, except for the last stage; indeed, only alcohol (+)- 4a , with equatorial OH-group, undergoes oxidation, and (+)- 1 is partly substracted via a hetero Diels-Alder dimerization giving a mixture of the diastereoisomeric dihydropyrans (+)- 5a /(+)- 5b . When Cr(VI) reagents ae used, (+)- 4a /(+)- 4b mainly give phellandral ( 6 ) and carvotanacetone ( 7 ), NnO2 reacts too sluggishly with (+)- 4a /(+)- 4b . A camphor pyrolyzate, previously thought to be 1 must be a different compound, probably 7 .  相似文献

Yeast reduction of rac-ethyl 2-methyl-6-oxocylohexanecarboxylate (rac- 1 ) yielded selectively (+)-ethyl 2-hydroxy-6-methylcyclohexane carboxylate (+)- 2 (Scheme 1) which has been alkylated with 5-iodo-2-methylbut-2-ene by (the dianion method to furnish the 4-methylbut-3-enyl derivat 3 (Scheme 3)). NaBH4 reduction of (+)- 1 led to three hydroxy-carboxylates (?)- 2 , (+)- 5 , and (?) -6 (Scheme 4). Allylation of the dianion of (+)- 5 afforded (+)- 7 .  相似文献

Novel chiral (S)-(+)-4?-(1-methylalkoxycarbonyl)biphenyl-4-yl 4-[ω-(2,2,3,3,4,4,4-hepta?uoro-butoxy)alkoxy]benzoates and 4-[ω-(2,2,3,3,4,4,4-hepta?uorobutoxy)alkoxy]-2-fluorobenzoates were prepared using different optically active alcohols: (S)-(+)-2-hexanol, (S)-(+)-2-heptanol, (S)-(+)-2-nonanol, (S)-(+)-2-decanol, (S)-(+)-2-undecanol and (S)-(+)-5-methylhexan-2-ol. Properties, such as the sequence of phases, transition temperatures and enthalpies, were tested by polarising optical microscopy and differential scanning calorimetry. Extremely low melting compounds were found (below 10°C) for derivatives of (S)-(+)-2-hexanol. The antiferroelectric smectic phase (SmCA*) with a direct transition from the antiferroelectric to isotropic phase (SmCA*-Iso) was observed, usually for propoxy-(CH2)3- spacing group. Initially, multicomponent mixtures with broad temperature ranges of antiferroelectric phase and direct SmCA*-Iso transition were formulated.  相似文献

Resolution of racemic cis-3-(2-aminophenylthio)-2-hydroxy-3-(4-methoxyphenyl) propionic acid ( 2 ) via the cinchonidine salt 3 , and brucine salt 4 , isolation of the calcium salts (+)- and (?)- 5 , as well as their cyclization to enantiomeric 1,5-benzothiazepines (+)- and (?)- 1 , are described. X-Ray single-crystal analysis reveals (2S, 3S) absolute configuration of (+)- 1 on the basis of tentative comparison of CD data with those for the 1,4-benzodiazepine derivative (+)- 8 of known absolute configuration.  相似文献

The l-azabicyclo[2.2.1]heptan-3-exo-ol ( 2 ) was resolved by fractional crystallisation of its hydrogen tartrate salts. The enantiomers (+)- and (?)- 2 were oxidised to the ketones (?)- 4 and (+)- 4 , respectively (Scheme). CD spectroscopy suggested that (?)- 4 possesses the (1R,4S)-configuration. This absolute configuration was confirmed by single-crystal X-ray diffraction of the derivative (+)-(1R,4R)-3-(1,3-dithian-2-ylidene)-1-azabicyclo [2.2.1]-heptane ((+)- 5 ).  相似文献

Mitsunobu displacement of (−)-(1S,4R,5S,6S)-4,5,6-tris{[(tert-butyl)dimethylsilyl]oxy}cyclohex-2-en-1-ol ((−)- 12 ; a (−)-conduritol-F derivative) with 4-ethyl-7-hydroxy-2H-1-benzopyran-2-one ( 16 ) provided a 5a-carba-β-D -pyranoside (+)- 17 that was converted into (+)-4-ethyl-7-[(1′R,4′R,5′S,6′R)-4′,5′,6′-trihydroxycyclohex-2′-en-1′-yloxy]-2H-1-benzopyran-2-one ((+)- 5 ) and (+)-4-ethyl-7-[(1′R,2′R,3′S,4′R)-2′,3′,4′-trihydroxycyclohexyloxy]-2H-1-benzopyran-2-one ((+)- 6 ). The 5a-carba-β-D -xyloside (+)- 6 was an orally active antithrombotic agent in the rat (venous Wessler's test), but less active than racemic carba-β-xylosides (±)- 5 and (±)- 6 . The 5a-carba-β-L -xyloside (−)- 6 was derived from the enantiomer (+)- 12 and found to be at least 4 times as active as (+)- 6 . (+)-4-Cyanophenyl 5-thio-β-L -xylopyranoside ((+)- 3 ) was synthesized from L -xylose and found to maintain ca. 50% of the antithrombotic activity of its D -enantiomer. Compounds (±)- 5 , (±)- 6 , and (−)- 6 are in vitro substrates for galactosyltransferase 1.  相似文献

The 6 R configuration of (+)-cis-γ-irone [(+)- 4 ] was established by chemical correlation with (—)-camphor. (+)-cis-γ-irone [(+)- 4 ] was converted into (+)-cis-α-irone [(+)- 1 ], (?)-trans-α-irone [(minus;;)- 2 ], and (+)-β-irone [(+)- 3 ], which therefore also have the 6 R configuration. The 2 S configurations of (+)-cis-α-irone [(+)- 1 ] and (+)-trans-α-irone [(+)- 2 ] were determined by comparison of their circular dichroism with that of R-α-ionone [(+)- 5 ]. The 2 S configuration of (+)-cis-γ-irone [(+)- 4 ] was established by chemical correlation with (+)-cis-α-irone [(+)- 1 ].  相似文献

Continuing studies of the global extracts from cultures of the marine deuteromycete Dendryphiella salina have led to the isolation of novel compounds that add to the scarce list of marine fungal metabolites. Besides (22E)-ergosta-4.6,8(14),22-tetraen-3-one which, though known from basidiomycetes, was unknown in the sea, they are an unusual glyceryl ester, i.e. glycer-1-yl dendryphiellale A (= (+)-(2R)-2,3-dihydroxyprop-l-yl (6S,2E,4E)6-methylocta-2,4-dienoate; (+)- 1 ), a trinor-eremophilane, i.e. dendryphiellin A1 ( = (+)-(3R*,4E,6E)-7-{[(1R*,2S*,7R*,8aR*)-1,2,6,7,8,8a-hexahydro-7-hydroxy-1,8a-dimethyl-6-oxonaphthalen-2yl]oxycarbonyl}-3-methylhepta-4,6-dienoic acid; (+)- 11 ), and two eremophilanes, i.e. dendryphiellin El ( = (+)-(1R*, 2S*, 7S*,8aR*)-1,2,6,7,8,8a-hexahydro-1,8a-dimethyl-7-(1-methylethenyl)-6-oxonaphthalen-2-yl(6S,2E,4E)-6-methyl-octa-2,4-dienoate; (+)- 13 ) and dendryphiellin E2 ( = (+)-(1R*, 2S*, 8aR*)-1,2,6,7,8,8a-hexahydro-7-isopropyl-idene-1,8a-dimethyl-6-oxonaphthalen-2-yl (6S,2E,4E)-6-Methylocta-2,4-dienoate; (+)- 14 ). Absolute configurations have been established for (+)- 1 via total synthesis and for the acid portion of (+)- 13 and (+)- 14 via transesterification in NaOMe/MeOH which gave in both cases melhyl dendryphiellate A ((+)- 16 ) of known configuration and the free alcoholic moiety of (+)- 14 , i.e. (+)- 17 .  相似文献

The stereospecific synthesis of the monoterpene alkaloids (?)-α-skytanthine ((?)- 2 ), (?)-N -demethyle-δ-sky-tanthine((?)- 7 ), and (+)-epidihydrotecomanine (+)- 4 was achieved from a common intermediate 22 , which in turn was obtained from (1R,4S,1′S)-2-(1′-phenylethyl)-2-azabicyclo[2.2.1]hept-5-ene (10) ,via a ketene aza-Claisen rearrangement. Epoxidation of (?)-(1R,2R,4R)-2-endo-cyano-7-oxabicyclo[2.2.1]hept-5-en-2-exo-yl acetate ((?)-5) followed by saponification afforded (+)-(1R,4R,5R,6R)-5,6-exo-epoxy-7-oxabicyclo[2.2.1]heptan-2-one ((+)-7). Reduction of (+)-7 with diisobutylaluminium hydride (DIBAH) gave (+)-1,3:2,5-dianhydroviburnitol ( = (+)-(1R,2R,3S,4R,6S)-4,7-dioxatricyclo[3.2.1.03,6]octan-2-ol; (+)-3). Hydride reductions of (±)-7 were less exo-face selective than reductions of bicyclo[2.2.1]heptan-2-one and its derivatives with NaBH4, AlH3, and LiAlH4 probably because of smaller steric hindrance to endo-face hydride attack when C(5) and C(6) of the bicyclo-[2.2.1]heptan-2-one are part of an exo oxirane ring.  相似文献

Synthesis of the optical isomers of (±)-methyl 6,7-dimethyl-3′,4′-dideoxynorlaudanosoline-1-carboxylate ((±)- 2 ) was accomplished by reaction of (±)- 2 with (+)-(R)-1-phenylethyl isocyanate, separation of the urea diastereoisomers (?)- 4A and (+)- 4B , and alcoholysis of the ureas in refluxing BuOH. Optically active isoquinoline-carboxylates 2A , B and hydantoins 8A , B isolated were characterized. The absolute configuration of the reaction products was established by X-ray analysis of the optically active hydantoin (+)- 8A . Hydrolysis of the methyl isoquinolinecarboxylates 2A , B with 48% HBr soln. at reflux afforded the desired optically active 3′,4′-dideoxynorlaudanosoline-1-carboxylic acids 1A , B required for enzyme-inhibition studies. Details of the X-ray diffraction analysis of (+)-methyl salsoline-1-carboxylate hydrobromide ((+)- 11A ·HBr) prepared earlier are included. CD spectra of (+)-(S)-methyl 6,7-dimethyl-3′,4′-dideoxynorlaudanosoline-1-carboxylate hydrobromide ((+)- 2A . HBr) and (?)-(R)-methyl salsoline-1-carboxylate hydrochloride ((?)- 11B ·HCl) confirmed the assignment of their (S)- and (R)-configurations, respectively.  相似文献

In four synthetic steps, (+)- and (–)-methyl 8-epinonactate ((+)- and (–)− 4 ) have been derived from (+)- and (–)-7-oxabicyclo[2.2.1]heptan-2-one ((+)- and (–)− 9 ), respectively. The (+)- and (–)-methyl nonactate ((+)- and (–)− 3 ) were obtained from (+)- and (–)− 4 , respectively, by Mitsunobu displacement reactions. Optical resolution of (±)− 9 via chromatographic separation of the corresponding N-methyl-S-alkyl-S-phenylsulfoximides 24 and 25 yielded the starting materials (+)- and (–)− 9 , respectively.  相似文献

The absolute configurations of acetylated bretonin A (= (+}-( R )-1-[(acetoxy)methyl]-2-{[(4E,6E,8E)-dodeca-4,6,8-trienyl]oxy}ethyl 4-acetoxybenzoate; (?)- 1b ) and isobretonin A (= (+)-(S)-3-{[(4E,6E,8E)-do-deca-4,6,8-trienyl]oxy}-2-hydroxypropyl 4-hydroxybenzoate; (+)-2), previously isolated from an undetermined sponge of the North Brittany sea, were established by comparison with synthetic (+)- lb and (+)- 2 , obtained from the condensation of commerical (?)-(R)-2,2-dimethyl-1,3-dioxolan-4-yl p-toluenesuifonate ((?)-(R)- 15 ) with a mixture of (4E,6E,8E)- ( 14e ) and (4E,6Z,8E)-dodeca-4,6,8-trien-1-ol ( 14z ). This also allowed confirming the structure and configuration of bretonin B (= (S)-2-{[(4E,6Z,8E)-dodeca-4,6,8-trienyl]oxy}-1-(hydroxy-methyl)ethyl 4-hydroxybenzoate; 3 ) which was also isolated from the same sponge, albeit in a too small amount for a complete study. As concerns the glyceryl ethers precursors of the bretonins, co-occurrence of the usual (S)-con-figuration (from 1a ) with the unusual (R)-configuration (from (+)- 2 )) poses intriguing biogenetic problems.  相似文献

The absolute configurations of the two algal pheromones multifidene 1 and viridiene 2 were determined as (+)-(3S, 4S)-3-[(Z)-1-butenyl]-4-vinylcyclopentene and (+)-(3R, 4S)-3-[(1Z)-1, 3-butadienyl]-4-vinylcyclopentene, respectively. The strategy involves enzyme-initiated asymmetric synthesis of the ring-saturated pheromone analogues (+)- 8a and (?)- 8b and their subsequent catalytic hydrogenation to the chiral cycloalkanes 9a and 9b , only the letter of which is also obtained from the two natural messengers (+)- 1 or (+)- 2 . Biological activity assays proved these enantiomers of 1 or 2 to be the characteristic pheromones for male gametes of the seaweeds Syringoderma, Cutleria multifida and Chorda tomentosa.  相似文献

Thermal Rearrangement of 2-Oxa-bicyclo [3.3.1]-non-7-en-3-ones; a Novel Lactone Rearrangement Lactone (+)- 2 was prepared by iodolactonisation and subsequent elimination in 51% yield from the known acid (+)- 1 (Scheme 1). Alkylation of (+)- 2 furnished (+)- 3a , (+)- 3b and (+)- 3c , respectively (Scheme 2). Heating of (+)- 3a in boiling DMF racemized the compound ((+)- 3a ? (?)- 4a ). Heating of (+)- 3b and (+)- 3c , respectively, equilibrated them with (?)- 4b and (?)- 4c , respectively. (+)-cis-Achillene ( 10 ) and (?)-trans-achillenol (7), two monoterpenes recently isolated [1] from the essential oil of Achilleafilipendulina, were synthesized, together with their stereoisomers (?)-(9) and (+)-(8), starting from (S)-(+)-2,6-trans-dimethylocta-1,3, 7-triene ( 1 ). The isomeric ß-hymen thereties ((?)- 3 and (+)-4), often quoted [2] [3] [4] but never isolated, were obtained as intermediates. The mode of synthesis chosen establishesis (R)-chirality for naturally occurring (?)-trans-achillenol (7) and (+)-cis-achillene ( 10 ) as well as for the purely synthetic 4, 7-diene derivatives described in this paper.  相似文献

Enantiospecific Synthesis of (+)-(2R)- and (?)-(2S)-6-Ethyl-3,4-dihydro-2-methyl-4-oxo-2H-pyran-5-carboxylic Acid The two enantiomers (?)-(2S)- and (+)-(2R)-6-ethyl-3,4-dihydro-2-methyl-4-oxo-2H-pyran-5-carboxylic acid ((S)- and (R)- 7 ) have been synthesized from (+)-(3S) and (?)-(3R)-3-hydroxybutanoates, respectively (Scheme 1). By reduction and decarboxylation, the tetrahydro-2H-pyranols (2R, 4R, 6S)- and (2S, 4S, 6R)- 13 , respectively, were obtained with an enantiomeric excess of ≥ 93%.  相似文献

Stereospecific Synthesis of (+)-(3R, 4R)-4-Methyl-3-heptanol, the Enantiomer of a Pheromone of the Smaller European Elm Bark Beetle (Scolytus multistriatus) Reduction of 2 with actively fermenting baker's yeast gave (?) -3. Stereospecific alkylation [3] of (?) -3 with propyl iodide furnished ethyl (+)-(2R, 3R)-2-propyl-3-hydroxypentanoate ((+) -4 , 58%) which was converted to the tetrahydropyranyl ether (?) -5 , then the alcohol 6 , the p-toluenesulfonate 7 and the thiophenyl ether 8 to give the title compound (+) -1. The latter consisted of 97% of the threo- and 3% of the erythro-isomer. The above synthesis also correlates the absolute configuration of (?)-(R) -3 with that of (+)-(R)-citronellic acid (see [2]).  相似文献
