The adsorption of bovine serum albumin (BSA) onto polystyrene latexes bearing various amounts of sugar moieties has been investigated as a function of pH and ionic strength and the results were compared to those for bare polystyrene latexes having negative surface charges. The functionalized latexes were produced by seeded copolymerization of (0.3 μm) liposaccharidic monomer onto polystyrene particles obtained by soap-free emulsion polymerization of styrene using potassium persulfate as initiator. At first, the electrophoretic mobility behavior of the various latexes was examined as a function of pH: a significant decrease was observed in the case of saccharide-containing latex particles compared to the bare particles. The adsorption of BSA onto these latexes exhibited a reduced amount of adsorbed BSA for those latex particles bearing saccharide groups. This adsorbed amount depends on the yield of saccharidic monomer incorporated onto the surfaces of the latex particles. 相似文献

Studies on Interaction between Gatifloxacin and Bovine Serum Albumin by Spectroscopy 总被引:1,自引:0,他引:1
The interaction of gatifloxacin (HGA) with bovine serum albumin (BSA) at 15 and 37 ℃ has been investigated by fluorescence quenching spectroscopy in aqueous solution. The bimolecular quenching rate constant was determined by Stem-Volmer curves and the values were Kq=9.28× 10^12 L·mol^-1·s^-1 (15 ℃) and Kq=8.51 ×10^12 L·mol^-1·s^-1 (37 ~C). The results showed that the fluorescence quenching mechanism of BSA by HGA was a static quenching procedure. The thermodynamic parameters indicated that electrostatic forces played major role in the interaction of BSA with HGA. Studies on the relationship between the concentration of HGA and the fluorescence intensity of BSA showed that BSA and HGA bound at the molar ratio 1 : 1 and the equilibrium constant K0 was 6.80 ×10^4 L·mol^-1. The binding distances between BSA and HGA and the energy transfer efficiency were obtained based on the Ftrster's theory. 相似文献

应用光谱法研究了水溶液体系中吖啶橙与牛血清白蛋白分子间的相互结合反应,其结合常数为k=4.8×10^3mol/L,结合位点数为n=0.28。并依据Forster非辐射能量转移机制,探讨了吖啶橙与牛血清白蛋白相互结合时,其给体-受体间距离(R0=1.5nm)和能量转移效率(E=0.32)。证实了吖啶橙与牛血清白蛋白的相互结合作用为单一的静态猝灭过程,且阐明了其猝灭机制是通过能量转移产生的。 相似文献

利用微分脉冲伏安法,研究了日落黄与牛血清白蛋白之间的相互作用.试验结果表明,日落黄与牛血清白蛋白之间形成近似1:1的复合物,从而造成日落黄峰电流的降低,两者之间的结合主要依靠静电力,且结合常数随温度升高而降低.研究有利于从分子水平上了解日落黄和牛血清白蛋白之间的结合作用,为更进一步了解日落黄的生物代谢机理提供一定的理论参考. 相似文献

运用荧光光谱(FS)、紫外光谱(UV)和超滤(UF)法, 研究了阿魏酸、毛蕊异黄酮及芒柄花素三种抗氧化小分子物质与牛血清白蛋白(BSA)的结合反应. 以Lineweaver-Burk双倒数方程和能量传递原理分别计算了三者与BSA反应的结合常数(KLB)和结合距离(r)以及由热力学参数的计算判断了三种小分子物质与BSA结合的作用力类型; 以Scatchard方程计算了三种小分子与BSA的结合常数(Ka)和结合位点数(n); 并对阿魏酸与毛蕊异黄酮及芒柄花素的竞争结合反应进行了研究. 实验结果表明: 这三种物质能与BSA结合形成非共价复合物, 导致BSA内源性荧光的静态猝灭; 它们与BSA的结合能力随着极性基团的增多、分子体积的增大而减弱; 静电作用和疏水作用可能是它们与BSA结合的主要作用力; 阿魏酸与芒柄花素或毛蕊异黄酮会产生与BSA的竞争结合效应. 相似文献

在模拟生理条件下,采用荧光光谱法、圆二色光谱法和红外光谱法研究了花椒油素(XT)与牛血清白蛋白(BSA)的相互作用。结果表明花椒油素与牛血清白蛋白之间发生动态和静态联合猝灭,二者间的的猝灭常数(K)在286, 298和310 K分别为3.31 × 105, 到2.03 × 105 和 0.94 × 105 L∙mol-1. 热力学参数表明, 花椒油素与牛血清白蛋白间以疏水作用力为主。圆二色光谱和红外光谱法表明加入花椒油素后,牛血清白蛋白的二级结构发生了变化,其中α-螺旋减少了3.9%。另外,我们还研究了共存离子对两者结合的影响。 相似文献

The interaction between CdTe quantum dots (QDs) and bovine serum albumin (BSA) was systematically investigated by fluorescence, UV‐vis absorption and circular dichroism (CD) spectroscopy under physiological conditions. The experimental results showed that the fluorescence of BSA could be quenched by CdTe QDs with a static quenching mechanism, indicating that CdTe QDs could react with BSA. The quenching constants according to the modified Stern‐Volmer equation were obtained as 1.710×106, 1.291×106 and 1.010×106 L·mol?1 at 298, 304, and 310 K, respectively. ΔH, ΔS and ΔG for CdTe QDs‐BSA system were calculated to be ?33.68 kJ·mol?1, 6.254 J·mol?1·K?1 and ?35.54 kJ·mol?1 (298 K), respectively, showing that electrostatic interaction in the system played a major role. According to F?rster theory, the distance between Trp‐214 in BSA and CdTe QDs was given as 2.18 nm. The UV‐vis, synchronous fluorescence and CD spectra confirmed further that the conformations of BSA after addition of CdTe QDs have been changed. 相似文献

应用荧光光谱研究了岩白菜素与牛血清白蛋白(BSA)分子间的相互作用.结果表明,岩白菜素对BSA内源荧光的猝灭机制属于形成化合物所引起的静态猝灭,猝灭常数Ksv为1.905×104L.mol-1;岩白菜素与BSA反应的结合常数为2.083×104,结合位点数为1.由热力学参数确定了岩白菜素与牛血清白蛋白的结合作用主要为静电作用.实验还发现随着岩白菜素的加入,BSA的猝灭值与岩白菜素浓度在1.5×10-5~1.5×10-4mol.L-1的范围内呈良好的线性关系,检出限2.0×10-6mol.L-1,可用于岩白菜素的测定. 相似文献

利用Dip-pen纳米刻蚀技术(简称DPN技术)在云母基底上构建出形状、尺寸可控的牛血清白蛋白(BSA)纳米结构.考察了针尖接触基底时间及针尖下行距离对构建的牛血清白蛋白纳米结构的影响.较长的针尖-基底接触时间及较深的下行距离可以沉积更多的牛血清白蛋白分子,构建牛血清白蛋白纳米结构的形状除了与墨水分子的本身性能有关,还与墨水-基底的相互作用有关.这些形状及尺寸可控的蛋白质纳米结构可以作为模板,进行金属、半导体等其它材料的组装,有望用于制造光电纳米器件及生物纳米器件. 相似文献
