A new approach for a simple electrochemical detection of PAT gene fragment is described. Poly(2,6-pyridinedicarboxylic acid) (PDC) modified glassy carbon electrode (GCE) was prepared by potential scan electropolymerization in an aqueous solution. Mg2 ions were incorporated by immer-sion of the modified electrode in 0.5 mol/L aqueous solution of MgCl2 to complete the preparation of a generic "activated" electrode ready for binding the probe DNA. The ssDNA was linked to the conduct-ing polymer by forming a bidentate complex between the carboxyl groups on the polymer and the phosphate groups of DNA via Mg2 . DNA immobilization and hybridization were characterized with dif-ferential pulse voltammetry (DPV) by using methylene blue (MB) as indicator and electrochemical im-pedance spectroscopy (EIS). The EIS was of higher sensitivity for DNA detection as compared with voltammetric methods in our strategy. The electron transfer resistance (Ret) of the electrode surface in EIS in [Fe(CN)6]3-/4- solution increased after the immobilization of the DNA probe on the Mg/PDC/GCE electrode. The hybridization of the DNA probe with complementary DNA (cDNA) made Ret increase further. The difference between the Ret at ssDNA/Mg/PDC/GCE and that at hybridization DNA modified electrode (dsDNA/Mg/PDC/GCE) was applied to determine the specific sequence related to the target PAT gene with the dynamic range comprised between 1.0 × 10-9 and 1.0 × 10_5 mol/L. A detection limit of 3.4 × 10-10 mol/L of oligonucleotides can be estimated.  相似文献

A novel electrochemical DNA biosensor based on zinc oxide (ZnO) nanoparticles and multi-walled carbon nanotubes (MWNTs) for DNA immobilization and enhanced hybridization detection is presented. The MWNTs/nano ZnO/chitosan composite film modified glassy carbon electrode (MWNTs/ZnO/CHIT/GCE) was fabricated and DNA probes were immobilized on the electrode surface. The hybridization events were monitored by differential pulse voltammetry (DPV) using methylene blue (MB) as an indicator. The sensor can effectively discriminate different DNA sequences related to PAT gene in the transgenic corn, with a detection limit of 2.8× 10^-12 mol/L of target sequence.  相似文献

A new type of ultrathin conducting polymer/carbon nanotubes (UCP/CNTs) nanomaterials with high catalytic surface area was proposed in this work. The electrode covered with an ultrathin layer of poly(azure A)/carbon nanotubes nanomaterials was fabricated by two steps: firstly, the single-wall carbon nanotubes (SCNTs) modified glassy carbon (GC) electrode was immersed in an azure A solution to assemble a thin azure A monomer film. Secondly, the assembled AA was electropolymerized in a phosphate buffer solution by cyclic voltammetry (CV). The electrocatalytic activity of the obtained ultrathin conducting poly(azure A)/carbon nanotubes (UCPAA/CNTs) nanomaterials was tested using sodium nitrite. The results showed thus prepared electrode exhibited excellent electrocatalytic behavior to the reduction of nitrite and facilitates the detection of nitrite at an applied reduction peak potential of 0.1 V. A linear range from 3.0 μM to 4.5 mM for the detection of sodium nitrite has been observed with fast response (within 3 s) and a detection limit of 1.0 μM based on a signal-to-noise ratio of 3.  相似文献

There is a high overvoltage in the oxidation of methanol in fuel cells,and so modified electrodes are used to decrease it.A modified electrode that used Ni(II) loaded analcime zeolite to catalyze the electrooxidation of methanol in alkaline solution was proposed.Analcime zeolite was synthesized by hydrothermal synthesis,and Ni(II) ions were incorporated into the analcime structure,which was then mixed with carbon paste to prepare modified electrode.The electrocatalytic oxidation of methanol on the surface of the modified electrode in alkaline solution was investigated by cyclic voltammetry and chronoamperometry.The effects of the scan rate of the potential,concentration of methanol,and amount of zeolite were investigated.The rate constant for the catalytic reaction of methanol was 6 × 103 cm3 mol-1 s-1 from measurements using chronoamperometry.The proposed electrode significantly improved the electron transfer rate and decreased the overpotential for methanol oxidation.  相似文献

The homogeneous electrocatalytic oxidation of hydrazine(HZ) has been studied by indigocarmine(IND) as a mediator at the surface of TiO2 nanoparticles modified carbon paste electrode(TNMCPE).Cyclic voltammetry was used to study the electrochemical behavior of IND at different scan rates.The voltammetric response of the modified electrode was linear against the concentration of HZ in the ranges of 3.0×l0-8-7.0×106 mol/L with differential pulse voltammetry method.The detection limit(3σ) was determined as 27.3 nmol/L.To evaluate the applicability of the proposed method to real samples,the modified CPE was applied to the determination of HZ in water samples.  相似文献

This work describes the promising activity of silver nanoparticles on the surface of a poly(2‐amino diphenylamine) modified carbon paste electrode(CPE) towards formaldehyde oxidation. Electro‐deposition of the conducting polymer film on the CPE was carried out using consecutive cyclic voltammetry in an aqueous solution of 2‐aminodiphenylamine and HCl. Nitrogen groups in the polymer backbone had a Ag ion accumulating effect, allowing Ag nanoparticles to be electrochemi‐cally deposited on the surface of the electrode. The electrochemical and morphological characteris‐tics of the modified electrode were investigated. The electro‐oxidation of formaldehyde on the sur‐face of electrode was studied using cyclic voltammetry and chronoamperometry in aqueous solu‐tion of 0.1 mol/L Na OH. The electro‐oxidation onset potential was found to be around-0.4 V, which is unique in the literature. The effect of different concentrations of formaldehyde on the electrocat‐alytic activity of the modified electrode was investigated. Finally, the diffusion coefficient of formal‐dehyde in alkaline media was calculated to be 0.47 × 10–6 cm2/s using chronoamperometry.  相似文献

Nafion solution as a binder,the Gr/CeO2 /Nafion/GCE modified electrode was prepared based on graphene (Gr)-nano cerium oxide (CeO2 )composite materials. The electrochemical behavior of Pb(II)on the modified electrode was investigated using differential pulse anodic stripping voltammetry (DPASV). Subsequently,a determination method for Pb(II)was established. The concentrations and ratio of composite materials,buffer solution pH,modification amount,enrichment potential and time were optimized. Under the optimal conditions ,the peak current of Gr/CeO2 /Nafion/GCE showed good linearity in the range of 0.24-48.31 μµmol/L of Pb(II),and the detection limit was 4.3 nmol/L. Interference experiments showed that the other common anions and cations in water samples caused little interference to Pb(II)detection. Furthermore,the interference of Cu(II)within 10 times is acceptable. The modified electrode was used to the detection of Pb(II) in four complex water samples,and the recovery rates of the spiked assays were from 89.5% to 107.7%. © 2023, Youke Publishing Co.,Ltd. A copper hexacyanoferrate modified ceramic carbon electrode (CuHCF/CCE) had been prepared by two-step sol-gel technique and characterized using electrochemical methods. The resulting modified electrode showed a pair of well-defined surface waves in the potential range of 0.40 to 1.0 V with the formal potential of 0.682 V (vs. SCE) in 0.050 mol·dm^-3 HOAc-NaOAc buffer containing 0.30 mol·dm^-3 KCl. The charge transfer coefficient (a) and charge transfer rate constant (ks) for the modified electrode were calculated. The electrocatalytic activity of this modified electrode to hydrazine was also investigated, and chronoamperometry was exploited to conveniently determine the diffusion coefficient (D) of hydrazine in solution and the catalytic rate constant (kcat). Finally, hydrazine was determined with amperometry using the resulting modified electrode. The calibration plot for hydrazine determination was linear in 3.0 × 10^-6--7.5 × 10^-4 mol·dm^-3 with the detection limit of 8.0 × 10^-7 molodm^-3. This modified electrode had some advantages over the modified film electrodes constructed by the conventional methods, such as renewable surface, good long-term stability, excellent catalytic activity and short response time to hydrazine.  相似文献

A novel electrochemical aptasensor based on a Cu2+-induced signal amplification strategy was constructed for the rapid, sensitive and specific detection of ochratoxin A (OTA). The OTA aptamer with poly (T) was hybridized with the captured DNA probe on the electrode surface. In the presence of Cu2+ and ascorbic acid, the end of poly(T) was used as a template to in situ grow copper nanoclusters (Cu NCs). In the absence of targeted OTA, the gold electrodes after decorating Cu NCs were immersed into an acidic environment to release Cu2+. After enriching Cu2+ at a potential of − 1.6 V, the strongest current value of copper was recorded by measuring differential pulse voltammetry (DPV). In the presence of OTA, the OTA aptamer was tightly bound to the target OTA. The OTA aptamer broke away from the electrode to reduce the growth of Cu NCs, resulting in lower DPV current response. This proposed method was employed to detect OTA with linear range from 0.1 to 50.0 ng/mL, and the detection limit was 41.2 pg/mL. A carbon paste electrode modified with 2‐((7‐(2,5-dihydrobenzylideneamino)heptylimino)methyl) benzene‐1,4‐diol(DHB) and carbon nanotubes were used to simultaneously determine the concen-trations of isoproterenol(IP), uric acid(UA), and folic acid(FA) in solution. First, cyclic voltammetry was used to investigate the redox properties of the modified electrode at various scan rates. Next, the mediated oxidation of IP at the modified electrode is described. At the optimum pH of 7.0, the oxidation of IP occurs at a potential about 90 mV less than that of an unmodified carbon paste elec-trode. Based on the results of differential pulse voltammetry(DPV), the oxidation of IP showed a dynamic range between 10 and 6000 μmol/L, and a detection limit of 1.24 μmol/L. Finally, DPV was used to simultaneously determine the concentrations of IP, UA, and FA in solution at the modified electrode.  相似文献

We present a new strategy for the label‐free electrochemical detection of DNA hybridization based on gold nanoparticles (AuNPs)/poly(neutral red) (PNR) modified electrode. Probe oligonucledotides with thiol groups at the 5‐end were covalently linked onto the surface of AuNPs/PNR modified electrode via S‐Au binding. The hybridization event was monitored by using differential pulse voltammetry (DPV) upon hybridization generates electrochemical changes at the PNR‐solution interface. A significant decrease in the peak current was observed upon hybridization of probe with complementary target ssDNA, whereas no obvious change was observed with noncomplementary target ssDNA. And the DNA sensor also showed a high selectivity for detecting one‐mismatched and three‐mismatched target ssDNA and a high sensitivity for detecting complementary target ssDNA, the detection limit is 4.2×10?12 M for complementary target ssDNA. In addition, the DNA biosensor showed an excellent reproducibility and stability under the DNA‐hybridization conditions.  相似文献

We report the new method for detection of DNA hybridization using enzymatic cleavage. The strategy is based on that S1 nuclease is able to specifically cleave only single strand DNA, but not double strand DNA. The capture probe DNA, thiolated single strand DNA labeled with electroactive ferrocene group, was immobilized on a gold electrode. After hybridization of target DNA of complementary and noncomplementary sequences, nonhybridized single strand DNA was cleaved using S1 nuclease. The difference of enzymatic cleavage on the modified gold electrode was characterized by cyclic voltammetry and differential pulse voltammetry. We successfully applied this method to the sequence‐selective discrimination between perfectly matched and mismatched target DNA including a single‐base mismatched target DNA. Our method does not require either hybridization indicators or other exogenous signaling molecules which most of the electrochemical hybridization detection systems require.  相似文献

In this paper, we present an impedance‐based DNA biosensor using thionine intercalation to amplify DNA hybridization signal. Beacon single‐stranded DNA (ssDNA) probe and mercaptoacetic acid were self‐assembled onto a Au electrode by forming Au? S bonds. These beacon ssDNAs were hybridized with the complementary sequences around the loop structure. Then thionine was intercalated into the double‐stranded DNA (dsDNA) immobilized on the Au electrode surface. Due to the neutralization of the negative charges of dsDNA by the intercalated thionine, the electronic transfer resistance (Ret) of the DNA modified Au electrode was significantly diminished. Herein, the decreased value of Ret resulted from the thionine intercalating into dsDNA was employed as the hybridization signal. SDS was used to reduce the unspecific adsorption between ssDNA and thionine. Several experimental conditions, including the surface coverage of ssDNA probe on Au electrode, the hybridization temperature and time were all optimized. Moreover, the hybridization reactions of the unstructured linear ssDNA probe and the structured beacon ssDNA probe with their complementary sequences were compared in this work. The sensitivity of the presented DNA biosensor highlighted that the intercalation of thionine into dsDNA was an efficient approach to amplify the hybridization signal using impedance detection technique. Additionally, in this DNA biosensing protocol, beacon ssDNA has a good ability to distinguish target DNA sequences. This results in a higher specificity than using traditional unstructured DNA probe.  相似文献

An electrochemical DNA sensing film was constructed based on the multilayers comprising of poly‐L ‐lysine (pLys) and Au‐carbon nanotube (Au‐CNT) hybrid. A precursor film of mercaptopropionic acid (MPA) was firstly self‐assembled on the Au electrode surface. pLys and Au‐CNT hybrid layer‐by‐layer assembly films were fabricated by alternately immersing the MPA‐modified electrode into the pLys solution and Au‐CNT hybrid solution. Cyclic voltammetry was used to monitor the consecutive growth of the multilayer films by utilizing [Fe(CN)6]3?/4? and [Co(phen)3]3+/2+ as the redox indicators. The outer layer of the multilayer film was the positively charged pLys, on which the DNA probe was easily linked due to the strong electrostatic affinity. The hybridization detection of DNA was accomplished by using methylene blue (MB) as the indicator, which possesses different affinities to dsDNA and ssDNA. Differential pulse voltammetry was employed to record the signal response of MB and determine the amount of the target DNA sequence. The established biosensor has high sensitivity, a relatively wide linear range from 1.0×10?10 mol/L to 1.0×10?6 mol/L and the ability to discriminate the fully complementary target DNA from single or double base‐mismatched DNA. The sequence‐specific DNA related to phosphinothricin acetyltransferase gene from the transgenically modified plants was successfully detected.  相似文献

We report a method for the detection of DNA hybridization in connection to lead sulfide (PbS) nanoparticle tags and electrochemical stripping measurement of the lead. A kind of lead sulfide nanoparticle with free carboxyl groups on its surface was synthesized in aqueous solution. The nanoparticle was used as a marker to label a sequence‐known oligonucleotide, which was then employed as a DNA probe for identifying a target ssDNA immobilized on a PPy modified electrode based on a specific hybridization reaction. The hybridization events were monitored by the oxidation dissolution of the lead sulfide anchored on the hybrids and the indirect determination of the lead ions by anodic stripping voltammetry (ASV). The detection limit is 0.3 pmol L?1 of target oligonucleotides. The PbS nanoparticle combining its easy conjugation to the DNA molecule with the highly sensitive stripping voltammetry detection of lead shows its promising application in the electrochemical DNA hybridization analysis assay.  相似文献

The polymer, poly(vinylferrocenium) (PVF+) modified electrode was developed as the first time herein for the improved electrochemical sensing of DNA based on the oxidation signals of polymer and guanine. The morphologies of polymer film and DNA immobilized polymer film were examined using scanning electron microscope (SEM). The electrochemical behavior of polymer modified electrode was investigated by using cyclic voltammetry (CV) and differential pulse voltammetry (DPV) in the absence/presence of DNA. Experimental parameters, such as the polymeric film thickness, the DNA immobilization time, the concentration of buffer solution, pH and DNA concentration were examined in order to obtain more sensitive and selective electrochemical signals. Abstract

Highly sensitive label-free techniques of DNA determination are particularly interesting in relation to the present development of an electrochemical hybridization biosensor for the detection of short DNA fragments specific to the human papilloma virus (HPV). Unlabeled DNA probes have been immobilized by spontaneous coadsorption of thiolated single-stranded oligonucleotides (HS-ssDNA) onto the sensing surface of a screen-printed gold electrode (SPGE). The covalently immobilized single-stranded DNA probe (HS-ssDNA) could selectively hybridize with its complementary DNA (cDNA) in solution to form double-stranded DNA (dsDNA) on the surface. DNA is treated with acid (e.g., 0.5 M chloridric acid), and the acid-released purine bases are directly determined by square wave voltammetry (SWV).

Variables of the probe-immobilization and hybridization steps are optimized to offer convenient quantitation of HPV DNA target, in connection with a short hybridization time. Peak currents were found to increase in the following order: hybrid-modified SPGE, 11-base mismatched modified SPGE, 18-base mismatched SPGE, and the probe modified SPGE. Control experiments with noncomplementary oligonucleotides were carried out to assess whether the suggested DNA sensor responds selectively to the target. The effect of the target DNA concentration on the hybridization signal was also studied. Under optimal conditions, this sensor has a good calibration range with HPV DNA sequence detection limit of 2 pg · ml?1 (S/N = 3).  相似文献

Gold electrodes were modified with submonolayers of mercaptoacetic acid (RSH) and further reacted with poly(amidoamine) (PAMAM) dendrimers (generation 4.0) to obtain thin films, on which DNA probe was later immobilized to afford a stable recognition layers. The characterization of the PAMAM/RSH‐modified electrode was investigated by cyclic voltammetry (CV) and electrochemical impedance measurement. Differential pulse voltammogram (DPV) measurement was used to monitor DNA hybridization with daunomycin (DNR) as indicator. Experiments carried out with these novel materials not only showed an improved DNA attachment quantity on the dendrimers‐modified electrodes compared to DNA sensors with oligonucleotides directly immobilized on Au electrodes, but also exhibited a high selectivity, sensitivity and stability for the measurement of DNA hybridization.  相似文献

A sensitive electrochemical method for the detection of DNA hybridization based on the probe labeled with multiwall carbon‐nanotubes (MWNTs) loaded with silver nanoparticles (Ag‐MWNTs) has been developed. MWNTs were electroless‐plated with a large number of silver nanoparticles to form Ag‐MWNTs. Probe single strand DNA (ss‐DNA) with a thiol group at the 3′‐terminal labeled with Ag‐MWNTs by self‐assembled monolayer (SAM) technique was employed as an electrochemical probe. Target ss‐DNA with a thiol group was immobilized on a gold electrode by SAM technique and then hybridized with the electrochemical probe. Binding events were monitored by differential pulse voltammetric (DPV) signal of silver nanoparticles. The signal difference permitted to distinguish the match of two perfectly complementary DNA strands from the near perfect match where just three base pairs were mismatched. There was a linear relation between the peak current at +120 mV (vs. SCE) and complementary target ss‐DNA concentration over the range from 3.1×10?14 to 1.0×10?11 mol/L with a detection limit of 10 fmol/L of complementary target ss‐DNA. The proposed method has been successfully applied to detection of the DNA sequence related to cystic fibrosis. This work demonstrated that the MWNTs loaded with silver nanoparticles offers a great promising approach for sensitive detection of DNA hybridization.  相似文献

Development of electrochemical DNA hybridization biosensors based on carbon paste electrode (CPE) and gold nanoparticle modified carbon paste electrode (NGMCPE) as transducers and ethyl green (EG) as a new electroactive label is described. Electrochemical impedance spectroscopy and cyclic voltammetry techniques were applied for the investigation and comparison of bare CPE and NGMCPE surfaces. Our voltammetric and spectroscopic studies showed gold nanoparticles are enable to facilitate electron transfer between the accumulated label on DNA probe modified electrode and electrode surface and enhance the electrical signals and lead to an improved detection limit. The immobilization of a 15‐mer single strand oligonucleotide probe on the working electrodes and hybridization event between the probe and its complementary sequence as a target were investigated by differential pulse voltammetry (DPV) responses of the EG accumulated on the electrodes. The effects of some experimental variables on the performance of the biosensors were investigated and optimum conditions were suggested. The selectivity of the biosensors was studied using some non‐complementary oligonucleotides. Finally the detection limits were calculated as 1.35×10?10 mol/L and 5.16×10?11 mol/L on the CPE and NEGCPE, respectively. In addition, the biosensors exhibited a good selectivity, reproducibility and stability for the determination of DNA sequences.  相似文献
