The alkylation of unactivated β‐methylene C(sp3)? H bonds of α‐amino acid substrates with a broad range of alkyl iodides using Pd(OAc)2 as the catalyst is described. The addition of NaOCN and 4‐Cl‐C6H4SO2NH2 was found to be crucial for the success of this transformation. The reaction is compatible with a diverse array of functional groups and proceeds with high diastereoselectivity. Furthermore, various β,β‐hetero‐dialkyl‐ and β‐alkyl‐β‐aryl‐α‐amino acids were prepared by sequential C(sp3)? H functionalization of an alanine‐derived substrate, thus providing a versatile strategy for the stereoselective synthesis of unnatural β‐disubstituted α‐amino acids.  相似文献

A palladium‐catalyzed arylation of unactivated γmethylene C(sp3)?H and remote δ‐C?H bonds by using an oxazoline‐carboxylate directing group has been developed. Arylation occurs with a broad substrate scope and high tolerance of functional groups (i.e., halogen, nitro, cyano, ether, trifluoromethyl, amine, and ester). The oxazoline‐type auxiliary can be removed under acidic conditions.  相似文献

Polyheterocycles are found in many natural products and are useful moieties in functional materials and drug design. As part of a program towards the synthesis of Stemona alkaloids, a novel palladium(II)‐catalyzed C? H activation strategy for the construction of such systems has been developed. Starting from simple 1,3‐dienyl‐substituted heterocycles, a large range of polycyclic systems containing pyrrole, indole, furan and thiophene moieties can be synthesized in a single step.  相似文献

The commonly used para‐nitrobenzenesulfonyl (nosyl) protecting group is employed to direct the C? H activation of amines for the first time. An enantioselective ortho‐C? H cross‐coupling between nosyl‐protected diarylmethylamines and arylboronic acid pinacol esters has been achieved utilizing chiral mono‐N‐protected amino acid (MPAA) ligands as a promoter.  相似文献

The N‐tosylcarboxamide group can direct the room‐temperature palladium‐catalyzed C?H alkoxylation and halogenation of substituted arenes in a simple and mild procedure. The room‐temperature stoichiometric cyclopalladation of N‐tosylbenzamide was first studied, and the ability of the palladacycle to react with oxidants to form C?X and C?O bonds under mild conditions was demonstrated. The reaction conditions were then adapted to promote room‐temperature ortho‐alkoxylations and ortho‐halogenations of N‐tosylbenzamides using palladium as catalyst. The scope and limitation of both alkoxylations and halogenations was studied and the subsequent functional transformation of the N‐tosylcarboxamide group through nucleophilic additions was evaluated. This methodology offers a simple and mild route to diversely functionalized arenes.  相似文献

β‐Lactams are very important structural motifs because of their broad biological activities as well as their propensity to engage in ring‐opening reactions. Transition‐metal‐catalyzed C? H functionalizations have emerged as strategy enabling yet uncommon highly efficient disconnections. In contrast to the significant progress of Pd0‐catalyzed C? H functionalization for aryl–aryl couplings, related reactions involving the formation of saturated C(sp3)? C(sp3) bonds are elusive. Reported here is an asymmetric C? H functionalization approach to β‐lactams using readily accessible chloroacetamide substrates. Important aspects of this transformation are challenging C(sp3)? C(sp3) and strain‐building reductive eliminations to for the four‐membered ring. In general, the β‐lactams are formed in excellent yields and enantioselectivities using a bulky taddol phosphoramidite ligand in combination with adamantyl carboxylic acid as cocatalyst.  相似文献

Synthesis of heteroatom‐containing ladder‐type π‐conjugated molecules was successfully achieved via a palladium‐catalyzed intramolecular oxidative C?H/C?H cross‐coupling reaction. This reaction provides a variety of π‐conjugated molecules bearing heteroatoms, such as nitrogen, oxygen, phosphorus, and sulfur atoms, and a carbonyl group. The π‐conjugated molecules were synthesized efficiently, even in gram scale, and larger π‐conjugated molecules were also obtained by a double C?H/C?H cross‐coupling reaction and successive oxidative cycloaromatization.  相似文献

An improved and practical procedure for the stereoselective synthesis of anti‐β‐hydroxy‐α‐amino acids (anti‐βhAAs), by palladium‐catalyzed sequential C(sp3)?H functionalization directed by 8‐aminoquinoline auxiliary, is described. followed by a previously established monoarylation and/or alkylation of the β‐methyl C(sp3)?H of alanine derivative, β‐acetoxylation of both alkylic and benzylic methylene C(sp3)?H bonds affords various anti‐β‐hydroxy‐α‐amino acid derivatives. As an example, the synthesis of β‐mercapto‐α‐amino acids, which are highly important to the extension of native chemical ligation chemistry beyond cysteine, is described. The synthetic potential of this protocol is further demonstrated by the synthesis of diverse β‐branched α‐amino acids. The observed diastereoselectivities are strongly influenced by electronic effects of aromatic AAs and steric effects of the linear side‐chain AAs, which could be explained by the competition of intramolecular C?OAc bond reductive elimination from PdIV intermediates vs. intermolecular attack by an external nucleophile (AcO?) in an SN2‐type process.  相似文献

A rhodium(III)‐catalyzed carboxylic acid directed decarboxylative C? H/C? H cross‐coupling of carboxylic acids with thiophenes has been developed. With a slight adjustment of the reaction conditions based on the nature of the substrates, aryl carboxylic acids with a variety of substituents could serve as suitable coupling partners, and a broad variety of functional groups were tolerated. This method provides straightforward access to biaryl scaffolds with diverse substitution patterns, many of which have conventionally been synthesized through lengthy synthetic sequences. An illustrative example is the one‐step gram‐scale synthesis of a biologically active 3,5‐substituted 2‐arylthiophene by way of the current method.  相似文献

The nickel‐catalyzed α‐arylation of ketones with readily available phenol derivatives (esters and carbamates) provides access to useful α‐arylketones. For this transformation, 3,4‐bis(dicyclohexylphosphino)thiophene (dcypt) was identified as a new, enabling, air‐stable ligand for this transformation. The intermediate of an assumed C? O oxidative addition was isolated and characterized by X‐ray crystal‐structure analysis.  相似文献

The regiodivergent palladium‐catalyzed C? H arylation of pyrazolo[1,5‐a]pyrimidine has been achieved, wherein the switch in regioselectivity between positions C3 and C7 is under complete catalyst control. A phosphine‐containing palladium catalyst promotes the direct arylation at the most acidic position (C7), whereas a phosphine‐free catalyst targets the most electron‐rich position (C3).  相似文献

An efficient and facile method for synthesis of 2‐arylpyrroles through Rh(III)‐catalyzed direct C? H arylation with pyrrole derivatives and aryl boronic acids has been developed. This reaction could proceed under mild reaction conditions and afford a series of 2‐arylated products in good to excellent yields. The gram‐scale experiment has been conducted to demonstrate the synthetic potential of this methodology.  相似文献

C?H arylation of polyfluoroarenes and heteroarenes with aryl sulfides proceeds smoothly with the aid of a palladium–N‐heterocyclic carbene catalyst. A bulky zinc amide, TMPZnCl ? LiCl, plays a key role as an effective base to generate the corresponding arylzinc species in situ. This arylation protocol is practically much easier to perform than our previous method, which necessitates preparation of the arylzinc reagents in advance from the corresponding aryl halides. Aryl sulfides that are prepared through sulfur‐specific reactions, such as SNAr sulfanylation and extended Pummerer reactions, undergo this direct arylation, offering interesting transformations that are otherwise difficult to achieve with conventional halogen‐based organic synthesis.  相似文献

An intermolecular C(sp3)? H amination using a Pd0/PAr3 catalyst was developed. The reaction begins with oxidative addition of R2N? OBz to a Pd0/PAr3 catalyst and subsequent cleavage of a C(sp3)? H bond by the generated Pd? NR2 intermediate. The catalytic cycle proceeds without the need for external oxidants in a similar manner to the extensively studied palladium(0)‐catalyzed C? H arylation reactions. The electron‐deficient triarylphosphine ligand is crucial for this C(sp3)? H amination reaction to occur.  相似文献

Two new rhodium‐catalyzed oxidative couplings between sulfoximine derivatives and alkenes by regioselective C?H activation, affording ortho‐olefinated (Heck‐type) products, are reported. A synthetic application of the ortho‐alkenylated products into the corresponding cyclic derivatives has been demonstrated, and a mechanistic rational for the rhodium catalysis is presented.  相似文献

A palladium‐catalyzed enantioselective C? H arylation of N‐(o‐bromoaryl)‐diarylphosphinic amides is described for the synthesis of phosphorus compounds bearing a P‐stereogenic center. The method provides good enantioselectivities and high yields. The products were readily transformed into P‐chiral biphenyl monophosphine ligands.  相似文献

A novel protocol for the Pd‐catalyzed ortho‐arylation of aryl phosphinamide with boronic acid is reported. By using phosphinamide as a new directing group, the reaction proceeds efficiently under mild conditions at 40 °C. Mechanistic studies reveal that the reaction proceeds via a PdII to Pd0 cycle. The phosphinamide group is also shown to be an effective orienting group for direct C?H amination.  相似文献

Versatile ruthenium(II) complexes allow for site‐selective C? H oxygenations with weakly‐coordinating aldehydes. The challenging C? H functionalizations proceed with high chemoselectivity by rate‐determining C? H metalation. The first organocatalytic enantioselective C? H alkenylation and arylation reactions of N‐carbamoyl tetrahydropyridines and tetrahydro‐β‐carbolines (THCs) are described. The metal‐free processes represent an efficient and straightforward approach to a variety of structurally and electronically diverse α‐substituted tetrahydropyridines and THCs in good yields with excellent regio‐ and enantioselectivities. Preliminary control experiments provide important insights into the reaction mechanism.  相似文献
