The composition of the fatty acids and phospholipids of 16 species of lichens collected in the basin of the river Volga has been studied. The main phospholipid was phosphatidylcholine the amount of which ranged in the various species from 33.3 to 85.5% of the total phospholipids. Other phospholipids were also found: phosphatidylethanolamine, phosphatidylserine, phosphatidylinositol, and phosphatidylglycerol. The composition of lipids from the aerial parts of two species of halophytes from the family Chenopodiaceae, Halostachys caspica C. A. Mey. and Halocharis hispida Bge. was determined. Neutral lipids (NL, 62.1 and 54.2%, respectively) dominated the total lipids (TL) of these plants. More than a third of the NL were esters of aliphatic alcohols and phytosterols (FAE). Fatty acids 16:0, 18:1, and 18:2 dominated the acids of FAE; 16:0, 18:1, and 18:3, the phospholipids. The principal fatty acids of glycolipids were unsaturated acids (68.3 and 75.1%) with linolenic acid dominating (44.9 and 43.5%). The content and composition by class and fatty acid of neutral (NL), glyco- (GL), and phospholipids (PL) in leaves ofEphedra equizetina Bunge (Ephedraceae) are determined. The acid composition of NL, GL, and PL includes saturated 12∶0–32∶0 acids and unsaturated 15∶1, 16∶1, 18∶1, 18∶2, and 18∶3 acids. Unsaponified components of the total lipids also contained biologically active substances such as α-tocopherol, carotenoids, high-molecular-weight fatty alcohols, triterpenes, and sterols. A liquid chromatographic–electrospray ionization–tandem mass spectrometric (LC–ESI–MS2) method has been developed for determination of the molecular species of phosphatidylethanolamine (PE) in four food matrices (soy, egg yolk, ox liver, and krill oil). The extraction and purification method consisted of a pressurized liquid extraction procedure for total lipid (TL) extraction, purification of phospholipids (PLs) by adsorption on a silica gel column, and separation of PL classes by semi-preparative normal-phase HPLC. Separation and identification of PE molecular species were performed by reversed-phase HPLC coupled with electrospray ionization tandem mass spectrometry (ESI–MS2). Methanol containing 5 mmol L−1 ammonium formate was used as the mobile phase. A variety of PE molecular species were detected in the four food matrices. (C16:0–C18:2)PE, (C18:2–C18:2)PE, and (C16:0–C18:1)PE were the major PE molecular species in soy. Egg yolk PE contained (C16:0–C18:1)PE, (C18:0–C18:1)PE, (C18:0–C18:2)PE, and (C16:0–C18:2)PE as the major molecular species. Ox liver PE was rich in the species (C18:0–C18:1)PE, (C18:0–C20:4)PE, and (C18:0–C18:2)PE. Finally, krill oil which was particularly rich in (C16:0(alkyl)–C22:6(acyl))plasmanylethanolamine (PakE), (C16:0–C22:6)PE, and (C16:0–C20:5)PE, seemed to be an interesting potential source for supplementation of food with eicosapentaenoic acid and docosahexaenoic acid.  相似文献

In the present work, the unsaturated fatty acid substituents of some phosphatidic acid, phosphatidylserine, phosphatidylinositol, and phosphatidylglycerol species were converted to their 1,2-di-hydroxy derivatives by OsO4. The subsequent electrospray ionization tandem low-energy mass spectrometry analysis of the deprotonated species allowed positional determination of the double bonds by the production of specific product-ions. The product-ions are formed by charge-remote and charge-proximate homolytic cleavages as well as charge-directed heterolytic cleavages and rearrangements. The commercial availability of pure species of the phospholipids in question was limited, and a number of species were therefore synthesized. The developed method was used to fully characterize the two isobaric phosphatidylglycerol species 16:0/16:1Δ9 and 16:0/16:1Δ10 extracted from the bacteria Methylococcus capsulatus. The presence of these fatty acids was supported by a gas-chromatography mass spectrometry investigation of the dimethyloxazoline derivatives of the species of the lipid extract. The present work demonstrates that a total structure characterization of acidic unsaturated phospholipids in isolate or in mixtures is accomplished by vicinal di-hydroxylation of olefinic sites and subsequent electrospray ionization mass spectrometry of the derivatized phospholipids.  相似文献

Methyl-branched fatty acids (MBFAs) are the dominant form of fatty acid found in many bacteria. They are also found at low levels in a range of foodstuffs, where their presence has been linked to bacterial sources. In this study we evaluated the potential of compound-specific isotope analysis to obtain insights into the stable carbon isotope ratios (δ13C values in ‰) of individual MBFAs and to compare them to the stable carbon isotope ratios of straight-chain fatty acids in food. Due to their low abundance in foodstuffs, the MBFAs were enriched prior to gas chromatography coupled to isotope ratio mass spectrometric (GC–IRMS) analysis. After transesterification, urea complexation was used to suppress the 16:0 and 18:0 methyl esters that were dominant in the samples. Following that, silver-ion high performance liquid chromatography was used to separate the saturated from the unsaturated fatty acids. The resulting solutions of saturated fatty acids obtained from suet, goat’s milk, butter, and human milk were studied by GC–IRMS. The δ13C values of fatty acids with 12–17 carbons ranged from −25.4‰ to −37.6‰. In all samples, MBFAs were most depleted in carbon-13, followed by the odd-chain fatty acids 15:0 and 17:0. 14:0 and 16:0 contained the highest proportions of carbon-13. The results from this study illustrate that MBFAs have distinctive δ13C values and must originate from other sources and/or from very different substrates. These measurements support the initial hypothesis that δ13C values can be used to attribute MBFAs to particular sources.  相似文献

The dynamics of the amount of total lipids, the ratio of the main classes of lipids, the amount of fatty acids, and the ratio of individual phospholipids in three epiphytic species of lichens from the Zhigulevsk reserve have been followed. It has been shown that all the classes of lipids of the lichens change to some degree or other in this period. The dynamics of the fatty acids has also confirmed a known rule: an increase in the amount of saturated and a decrease in the number of unsaturated fatty acids of the membranes with a rise in the temperature of the environment. The phospholipid and fatty-acid compositions of nine species of higher fungi have been investigated. In the various species studied the amount of total lipids ranged from 14 to 78 mg per gram dry weight. Phosphatidylcholine and phosphatidylethanolamine were found as the main classes of phospholipids. In addition to these, phosphatidylserine, phosphatidylinositol, phosphatidylglycerol, cardiolipin and lyso derivatives of phosphatidylcholine and phosphatidylethanolamine were also found. The composition and amounts of liposoluble substances in the flesh and peel of feijoa fruit has been investigated. About 30 groups of lipid substances have been identified. The main groups of lipids in the flesh were triacylglycerols, sterols, cerebrosides, ceramide phosphate inositol oligosides, sulfoquinovosyldiacylglycerols, phosphatidic acids, phosphotidylglycerols and phosphatidylcholines. In the peel, hydrocarbons, sterols, esters of fatty acids and lower alcohols, cerebrosides, digalactosyldiacylglycerols, ceramide oligosides, phosphatidylglycerols, and include predominated. The fatty acids of the flesh were found to include 15 representatives (C12:0–C28:0), and those of the flesh 11 representatives tives (C12:0–C18:3). M. V. Pigments and lipids from the aerial parts of six plant species from the genus Limonium (Plumbaginaceae, Limoniacea) were studied. Xanthophylls dominated among the carotinoids; chlorophyll, the chlorophylls, according to spectrophotometry. Unsaturated fatty acids, the content of which reached 80%, dominated in all samples according to GC. Qualitative and quantitative profiling of six different categories of urinary phospholipids (PLs) from patients with prostate cancer was performed to develop an analytical method for the discovery of candidate biomarkers by shotgun lipidomics method. Using nanoflow liquid chromatography–electrospray ionization–tandem mass spectrometry, we identified the molecular structures of a total of 70 PL molecules (21 phosphatidylcholines (PCs), 11 phosphatidylethanolamines (PEs), 17 phosphatidylserines (PSs), 11 phosphatidylinositols (PIs), seven phosphatidic acids, and three phosphatidylglycerols) from urine samples of healthy controls and prostate cancer patients by data-dependent collision-induced dissociation. Identified molecules were quantitatively examined by comparing the MS peak areas. From statistical analyses, one PC, one PE, six PSs, and two PIs among the PL species showed significant differences between controls and cancer patients (p < 0.05, Student’s t test), with concentration changes of more than threefold. Cluster analysis of both control and patient groups showed that 18:0/18:1-PS and 16:0/22:6-PS were 99% similar in upregulation and that the two PSs (18:1/18:0, 18:0/20:5) with two PIs (18:0/18:1 and 16:1/20:2) showed similar (>95%) downregulation. The total amount of each PL group was compared among prostate cancer patients according to the Gleason scale as larger or smaller than 6. It proposes that the current study can be utilized to sort out possible diagnostic biomarkers of prostate cancer.  相似文献

A sulfolipid, the structure of which was established by NMR spectroscopy, electrospray-ionization mass spectrometry (ESI-MS), and GC–MS, was isolated for the first time from the sea urchin Scaphechinus mirabilis. The sulfolipid was identified as a sulfoquinovosyldiacylglycerin (SQDG) and was the sum of related compounds. The fatty acids (FAs) of the SQDGs included saturated 14:0–24:0 FAs (95.8% of total FAs) and mono-unsaturated 20:1-24:1 FAs (4.2%). The principal FAs were saturated 14:0 (33.1%) and 16:0 (54.2%). ESI-MS/MS detected 15 molecular species of SQDGs, among which the contents of 14:0/14:0 and 16:0/16:0 were relatively high although 16:0/14:0 dominated.  相似文献

The compositions of the lipids and fatty acids of the seeds ofCynoglossum officinale, familyBoraginaceae have been established. The bulk of the lipids consisted of netural compounds (95.2%), while the amounts of glycolipids and phospholipids were 3.1 and 1.7%, respectively. A study of the feasibility of Fourier transform medium infrared spectroscopy (FT-midIR) for analytical determination of fatty acid profiles, including trans fatty acids, is presented. The training and validation sets—75% (102 samples) and 25% (36 samples) of the samples once the spectral outliers have been removed—to develop FT-midIR general equations, were built with samples from 140 commercial and home-made bakery products. The concentration of the analytes in the samples used for this study is within the typical range found in these kinds of products. Both sets were independent; thus, the validation set was only used for testing the equations. The criterion used for the selection of the validation set was samples with the highest number of neighbours and the most separation between them (H<0.6). Partial least squares regression and cross validation were used for multivariate calibration. The FT-midIR method does not require post-extraction manipulation and gives information about the fatty acid profile in two min. The 14:0, 16:0, 18:0, 18:1 and 18:2 fatty acids can be determined with excellent precision and other fatty acids with good precision according to the Shenk criteria, R 2≥0.90, SEP=1–1.5 SEL and R 2=0.70-0.89, SEP=2–3 SEL, respectively. The results obtained with the proposed method were compared with those provided by the conventional method based on GC-MS. At 95% significance level, the differences between the values obtained for the different fatty acids were within the experimental error. 应用自制硅胶板的薄层色谱法分离了红细胞膜上四种主要磷脂。将分离后的每种磷脂斑点硅胶涂层刮下,不经萃取,直接在无水甲醇-苯-乙酰氯溶液中进行转移甲基化,然后用毛细管相色谱分离测定其脂肪酸组成和含量。上述方法已成功地用于先天愚型病人和正常人红细胞膜上各种磷脂中脂肪酸轮廓分析,获得有意义的结果。  相似文献

The fractional composition of the polar lipids and also the fatty-acid composition of the main components of the phospholipids of the Chinese tallow tree have been studied. It has been established that in the seeds of the Chinese tallow tree there are 11 phospholipids, two glycolipids, and two phosphonolipids. The lipid composition of rat caecal mucosa, including the fatty acid composition of phospholipids and triacylglycerols, has been examined by capillary gas chromatography. Thirty-seven peaks were resolved, ranging in chain length from 12 to 24 carbon atoms. Preliminary identification of fatty acids by comparison with authentic standards was confirmed by gas chromatography-mass spectrometry using electron-impact ionization. The neutral and polar components were examined. Fatty acid methyl esters were quantified in absolute amounts with respect to the percentage of total phospholipid and triacylglycerols. The results show significantly higher levels of 16:0, 18:0, 18:1(n-9), 18:1(n-7), 18:2(n-6) and 20:4(n-6) in phospholipids, and higher levels of 16:0, 18:1(n-9) and 18:2(n-6) in triacylglycerols. On the other hand, analysis of caecal triacylglycerols revealed sn-glycerol-palmitate-oleate-palmitate, sn-glycerol-palmitate-linoleate-palmitate and sn-glycerol-palmitate-linoleate-oleate as major components.  相似文献

Archaeological samples are complex in composition since they generally comprise a mixture of materials submitted to deterioration factors largely dependent on the environmental conditions. Therefore, the integration of analytical tools such as TXRF, FT-IR and GC–MS can maximize the amount of information provided by the sample. Recently, two black rock art samples of camelid figures at Alero Hornillos 2, an archaeological site located near the town of Susques (Jujuy Province, Argentina), were investigated. TXRF, selected for inorganic information, showed the presence of manganese and iron among other elements, consistent with an iron and manganese oxide as the black pigment. Aiming at the detection of any residual organic compounds, the samples were extracted with a chloroform–methanol mixture and the extracts were analyzed by FT-IR, showing the presence of bands attributable to lipids. Analysis by GC–MS of the carboxylic acid methyl esters prepared from the sample extracts, indicated that the main organic constituents were saturated (C16:0 and C18:0) fatty acids in relative abundance characteristic of degraded animal fat. Lipids from the air-dried aerial part (AP) and seeds of Silybum marianum (L.) Gaerth. (Asteraceae) were studied. The class and fatty acid compositions of neutral lipids (AP, seeds) and glyco- and phospholipids (AP) were determined. Neutral lipids (NL) with a complicated set of lipophilic components, mainly triterpenols, sterols, and their esters predominated in the AP. The fatty acids of the AP were dominated by 16:0, 18:2 (glycolipids), and 18:3 (neutral lipids, phospholipids); of seed NL, by 18:2 and 18:1. The content and composition of flavolignans isolated from defatted seeds and the content of total protein in the meal were found.  相似文献
