The vinylogous Mukaiyama aldol reaction (VMAR) is a powerful tool of polyketide synthesis, which constructs a large size structure by simultaneous introduction of the stereogenic center(s) and the α,β-unsaturated carbonyl group. A variety of stereocontrolled VMARs have been developed and applied to natural product synthesis. N-Tosyl-2-1,3,2-oxazaborolidin-5-one 1 was used to probe the role of the Lewis basicity of aldehydes utilized in Mukaiyama aldol reactions promoted by oxazaborolidin-5-ones. Eight aldehydes were each allowed to undergo a Mukaiyama aldol reaction with ketene silyl acetal derived from methyl isobutanoate. Lewis basicity of the aldehydes was determined computationally. The best results were obtained with isobutyl aldehyde and 3,4,5-trimethoxy benzaldehyde (91% and 85% ee). The best fit of ee as a function of Lewis basicity gave a correlation of R2=0.79 [when two ortho-substituted aldehydes were excluded] suggesting that Lewis basicity of OCHO can play a role in reactions of aldehydes promoted by 1.  相似文献

The Stille coupling between a common oxazole vinyl iodide and stereodefined stannyl-diene units is described as the cornerstone of a unified synthetic route to the inthomycin family of bioactive Streptomyces metabolites. This procedure has been utilised to prepare (+)-inthomycin B and (+)-inthomycin C for the first time; in these examples the stereogenic centre was introduced using the Kiyooka ketene acetal/amino acid-derived oxazaborolidinone variant of the Mukaiyama aldol reaction. In addition, a convenient preparation of rac-inthomycin A is described based on the same strategy.  相似文献

We have developed asymmetric Mukaiyama aldol reactions of silicon enolates with aldehydes catalyzed by chiral FeII and BiIII complexes. Although previous reactions often required relatively harsh conditions, such as strictly anhydrous conditions, very low temperatures (?78 °C), etc., the reactions reported herein proceeded in the presence of water at 0 °C. To find appropriate chiral water‐compatible Lewis acids for the Mukaiyama aldol reaction, many Lewis acids were screened in combination with chiral bipyridine L1 , which had previously been found to be a suitable chiral ligand in aqueous media. Three types of chiral catalysts that consisted of a FeII or BiIII metal salt, a chiral ligand ( L1 ), and an additive have been discovered and a wide variety of substrates (silicon enolates and aldehydes) reacted to afford the desired aldol products in high yields with high diastereo‐ and enantioselectivities through an appropriate selection of one of the three catalytic systems. Mechanistic studies elucidated the coordination environments around the FeII and BiIII centers and the effect of additives on the chiral catalysis. Notably, both Brønsted acids and bases worked as efficient additives in the FeII‐catalyzed reactions. The assumed catalytic cycle and transition states indicated important roles of water in these efficient asymmetric Mukaiyama aldol reactions in aqueous media with the broadly applicable and versatile catalytic systems.  相似文献

N-Heterocyclic carbenes were shown to be highly effective catalysts to promote Mukaiyama aldol reactions. In the presence of only 0.5 mol % of N-heterocyclic carbene (5), various aldehydes and 2,2,2-trifluoroacetophenone underwent Mukaiyama aldol reactions in THF with trimethylsilyl ketene acetal (2) at 23 degrees C as well as with trimethylsilyl enol ether (7) at 0 degrees C to afford aldol adducts in good yields. The Mukaiyama aldol type condensation of t-butyldimethylsilyloxypyrrole 1b with methyl 2-formylbenzoate furnished the aldol adduct 9 with high yield and complete stereoselectivity. An erythro (anti) configuration was established (X-ray) in sharp contrast with the reaction of 1b with aliphatic aldehydes. The Mukaiyama aldol reaction of 2-aryl-1-cyclohexene-1-carboxaldehydes with phenyl trimethylsilyl ketene acetal unexpectedly resulted in the formation of 9-substituted-1,2,3,4-tetrahydrofluorene derivatives via a novel intramolecular titanium-promoted deoxygenative cyclization. By successive treatment of the corresponding allyl alcohols with n-butyllithium and titanium tetrachloride, the cyclization products were obtained in good yields.  相似文献

A highly stereocontrolled total synthesis of the 18-membered macrolide (+)-concanamycin F, a potent inhibitor of vacuolar ATPases, is described that proceeds in 5.8% yield over 26 steps. The three key fragments, C1-C13 vinyl iodide, C14-C22 vinyl stannane and C23-C28 aldehyde, were efficiently constructed using asymmetric boron-mediated aldol reactions of appropriate chiral ketone building blocks. The nature of the silyl protection of the C7/C9 hydroxyls proved to be critical for achieving macrocyclisation, with TES ethers being superior to a cyclic silylene derivative. Following a Liebeskind-Stille cross-coupling reaction between the C1-C13 vinyl iodide and C14-C22 vinyl stannane fragments to assemble the (12E,14E)-diene, a modified Yamaguchi macrolactonisation delivered the requisite 18-membered macrocyclic core. This advanced intermediate was also obtained by an alternative sequence using an esterification step to connect the C1-C13 and C14-C22 fragments followed by a Pd-catalysed intramolecular Stille reaction to install the (12E,14E)-diene. Conversion of the resulting macrocyclic intermediate into a methyl ketone then enabled a highly diastereoselective Mukaiyama aldol coupling of the derived silyl enol ether with the C13-C28 aldehyde fragment to install the fully elaborated side chain, whereby subsequent global deprotection of the resulting β-hydroxyketone under suitable conditions (TASF followed by p-TsOH) afforded (+)-concanamycin F.  相似文献

New phosphinooxazoline (PHOX) η5-indenyl complexes of iron were synthesized and applied as catalysts in the Mukaiyama aldol reaction. Reaction of three different PHOX ligands with [Fe(η5-Ind)I(CO)2] afforded the iodide salts of three complexes of the general formula [Fe(η5-Ind)(CO)(PHOX)]+ in 73-81% isolated yields. The molecular structure of one of the new complexes was determined, revealing a pseudo octahedral coordination geometry about the iron center. The iron complexes are catalytically active in the Mukaiyama aldol reaction between aldehydes and 1-(tert-butyldimethylsilyloxy)-1-methoxyethene to give the corresponding aldol adducts (3 mol % catalyst, 15 min, room temperature, 48-83% isolated yields). A previously synthesized iron complex of the general formula [Fe(η5-Cp)(CO)(PHOX)]+ was found to be catalytically active in the title reaction as well, but needed three hours at room temperature to convert the starting materials to the products.  相似文献

Novel C1-symmetric bis(oxazoline) ligands with a secondary binding sidearm were prepared in enantiomerically pure form in good yields, in only four steps starting from commercially available reagents. These new chiral ligands were tested in the enantioselective Mukaiyama aldol condensation between the trimethylsilyl keteneacetal of methyl isobutyrate and a non-chelating substrate such as benzaldehyde to afford the product in up to 55% ee.  相似文献

The design of novel nucleoside triphosphate (NTP) analogues bearing an all-carbon quaternary center at C2′ or C3′ is described. The construction of this all-carbon stereogenic center involves the use of an intramoleculer photoredox-catalyzed reaction. The nucleoside analogues (NA) hydroxyl functional group at C2′ was generated by diastereoselective epoxidation. In addition, highly enantioselective and diastereoselective Mukaiyama aldol reactions, diastereoselective N-glycosylations and regioselective triphosphorylation reactions were employed to synthesize the novel NTPs. Chiral N-sulfonylated α-amino acid monomer (5) derived from (S)-tryptophan was copolymerized with styrene and divinylbenzene under radical polymerization conditions to give a polymer-supported N-sulfonyl-(S)-tryptophan (6). Treatment of the polymer-supported chiral ligand with 3,5-bis(trifluoromethyl)phenyl boron dichloride afforded a polymeric Lewis acid catalyst (16) effective for asymmetric Mukaiyama aldol reaction of silyl enol ethers and aldehydes. Various aldehydes were allowed to react with silyl enol ethers in the presence of the polymeric chiral Lewis acid to give the corresponding aldol adducts in high yield with high levels of enantioselectivity.  相似文献
