2-Iodoaniline derivatives were used as bifunctional substrates in palladium-catalysed carbonylation. Depending on the substituents, two types of compounds were synthesised: having methyl or hydrogen in 4-position 2-aryl-benzo[d][1,3]oxazin-4-one derivatives have been formed, chloro, bromo, cyano or nitro groups in the same position resulted in the formation of dibenzo[b,f][1,5]-diazocine-6,12-dione derivatives. In the presence of various primary and secondary amines (tert-butylamine, amino acid methyl esters) as N-nucleophiles 2-ketocarboxamides were obtained as major products in aminocarbonylation reaction with double carbon monoxide insertion.  相似文献

Thermocatalytic decomposition of diazo esters in the presence of 3-aryl-2H-azirines gives rise to azirinium ylides. The latter preferentially transform via isomerization into 2-azabuta-1,3-diene derivatives or, with excess diazo compound, via reaction with the Rh-carbenoid to form 3,4-dihydro-2H-pyrrole derivatives. In contrast, ylides generated from 2-monosubstituted or 2,2-disubstituted 3-phenyl-2H-azirines transform exclusively via isomerization into the corresponding 2-azabuta-1,3-dienes in high yields.  相似文献

Azulene analog of tryptanthrin, azuleno[1′,2′:4,5]pyrrolo[2,1-b]quinazoline-6,14-dione, was successfully prepared by the condensation reaction of azuleno[2,1-b]pyrrole-2,3-dione with isatoic anhydride in the presence of sodium hydride or diisopropylethylamine (DIPEA). Its 2-halo derivatives were also obtained in high yields by the condensation reaction with 5-haloisatoic anhydrides in the presence of DIPEA. Reactivity toward electrophilic reagents was revealed by halogenation with N-bromosuccinimide (NBS) or N-iodosuccinimide (NIS) to afford 12-halo derivatives in high yields. Among the halo derivatives, 2-iodo and 12-iodo derivatives were reactive enough to afford phenylethynyl derivatives under Pd-catalyzed Sonogashira cross-coupling conditions. Within the phenylethynyl derivatives, only 12-phenylethynyl derivative was transformed into its 1,1,4,4-tetracyanobutadiene (TCBD) derivative by the reaction with TCNE. Amphoteric redox properties of the novel azulene analogs of tryptanthrin were characterized by spectroscopic and voltammetric analyses.  相似文献

Hydrazones derived from substituted benzophenones and fluorenone reacted with 4-aryl-2-hydroxy-4-oxobut-2-enoic and 2-hydroxy-5,5-dimethyl-4-oxohex-2-enoic acids to give the corresponding 2-(2-ylidenehydrazino) derivatives which may be used as initial compounds for the synthesis of 3-hydrazono-3H-furan-2-ones. The obtained but-2-enoic and hex-2-enoic acid derivatives in solution may exist as Z- and E-isomeric enehydrazine tautomers or hydrazone tautomers with syn or anti orientation of substituents with respect to the double C=N bond.  相似文献

Sulfonation of the N-Boc derivatives of 1,2-aminoalcohols, such as ephedrine, pseudoephedrine, norephedrine, norpseudoephedrine, thiomicamine, and chloramphenicol yields a mixture of the corresponding oxazolidinones with inversion (erythro derivatives) and/or retention of configuration (threo derivatives)at C5. We suggest a competition between two mechanisms: an intramolecular SN2 process initiated by attack of the carbonyl oxygen of the Boc group to the benzylic carbon and the other one through a double SN2 process. In the erythro derivatives the first mechanism is predominant, while in the threo derivatives both mechanisms have similar energy. This hypothesis is supported by theoretical calculations and additional experimental assays.  相似文献

Cyclotrimerization of β- or α-ethynyl-3,5-di-O-toluoyl-2-deoxy-d-ribofuranose with α,ω-diynes proceeded smoothly under Rh-catalysis to afford the corresponding β- or α-benzene C-nucleoside derivatives. Analogous co-cyclotrimerization of α- or β-propynyl- and -phenylethynyl-3,5-di-O-toluoyl-2-deoxy-d-ribofuranose with α,ω-diynes gave the corresponding arene derivatives only under microwave irradiation in the presence of a Rh-catalyst in moderate yields. Attempted homocyclotrimerization of β- or α-ethynyl-3,5-di-O-toluoyl-2-deoxy-d-ribofuranose under Rh-catalysis led only to enynes while the use of Ru-catalyst gave the desired 1,2,4- and 1,3,5-tri-(2-deoxyribofuranose-1-yl)benzene.  相似文献

The deprotonation of 1-phenyl-3-phospholene 1-oxide, 1-sulfide or 1-borane with 1 or 2 equiv of LDA, followed by quenching with electrophiles gave a range of 2-mono- or 2,5-disubstituted phospholene derivatives in good yield. Only trans substitution in relation to the P-Ph group was observed. Treatment of lithiated phospholene intermediates with 1,3-dihaloalkanes afforded annulated 2-phenyl-2-phosphabicyclo[3.3.0]oct-3-ene derivatives. The annulation reactions occurred with high regio- and stereoselectivity and led to the exclusive formation of the exo-Ph-P substituted products.  相似文献

2-Bromomethyl- and 2-iodomethyl-2,3-dihydrothiazolo[3,2-a]pyrimidin-5-ones are prepared via the reaction of 3-allyl-2-thiouracil derivatives with bromine or iodine chloride, respectively. The 6-bromo and 6-nitro derivatives are synthesized by an electrophilic substitution at C-6 of the thiazolopyrimidine system. As a result, novel 2,3-dihydrothiazolo[3,2-a]pyrimidin-5-one derivatives are obtained. Hydrogen halide elimination from the 2-halomethyl-2,3-dihydrothiazolo[3,2-a]pyrimidin-5-ones is also reported.  相似文献

New 1-cycloalkenyl-1-diazenes have been obtained in good yields from cyclic β-ketoesters and hydrazine derivatives. They furnished new cycloalkyl[d][1,3]thiazolines with thioamides or new spirocycloalkyl-thiazolinones with thioureas. Moreover they gave, with imidazolidine-2-thione and tetrahydropyrimidine-2-thione, new and interesting spiro[cycloalkyl-1,2′-imidazo[2,1-b][1,3]thiazole] or spiro[cycloalkyl-1,2′-[1,3]thiazolo[3,2-a]pyrimidine] derivatives, respectively. Cycloalkyl[d][1,3]thiazolines were useful for the further preparation of unknown thia-triaza-tricyclo derivatives. Novel hexahydro-1,3-benzothiazoles have been achieved by reaction of N,N′-dialkylthioureas on N-1-phenyl-2-(1-cyclohexenyl)-1-diazene-1-carboxyamide. The acidic hydrolysis of spirocycloalkyl-thiazolinones produced 2-imino-5-(ω-carboxyalkyl)-4-thiazolidinones.  相似文献

1,2-Dibromoarenes were coupled with aniline derivatives to yield N,N-diaryl-o-phenylenediamines in moderate to good yield using a palladium/phosphine or palladium/carbene catalyst system. Under similar conditions, 1,2,4,5-tetrabromobenzene was coupled with aniline derivatives to produce the corresponding tetrasubstituted derivatives which are oxidized on workup to yield azophenines. The sequential reaction of two different anilines with 1-chloro-2-iodobenzene afforded mixed N,N-diaryl-o-phenylenediamines.  相似文献

The Pd-catalyzed cross-coupling reactions of B-I bond in m- and p-carboranes and cobalt bis(1,2-dicarbollide) anion with organomagnesium and organozinc compounds were studied. Carboranyl derivatives of furan, thiophene, indole, pyridine and quinoline were synthesized. 2-Pyridylethynyl and 3-quinolylethynyl derivatives of p-carborane were prepared by Pd-catalyzed cross-coupling reactions using corresponding alkynes or their magnesium derivatives.  相似文献

Novel l-arabinose, l-ribose, 2-deoxy-l-ribose and 2-fluoro-2-deoxy-l-arabinose derivatives were synthesized from readily available l-arabinose. Different synthetic routes to methyl 3,5-di-O-acylated-l-arabino(ribo)furanosides as valued intermediates for the preparation of C-2 functionalized l-pentoses were investigated via regioselective transformations of 1,2-di-O-acetyl-3,5-di-O-pivaloyl-l-arabinofuranose, and selective acylation of methyl l-arabinofuranoside with 4-chlorobenzoyl or pivaloyl chloride. Short three-five-step syntheses of methyl 2-deoxy-α-l-ribofuranoside, its 3,5-di-O-acyl derivatives, valuable precursors for preparation of antiviral 2′-deoxy-l-nucleosides, were accomplished via simple and efficient reduction of methyl 2-O-mesyl-l-arabinofuranoside with L-Selectride or tandem reaction involving a complex hydride 2-deoxygenation/acylation of intermediate 2-deoxysugar. A new synthesis of 2′-deoxy-2′-fluoro-β-l-arabinofuranosyl thymine (l-FMAU) was performed using a mild fluorination of protected l-ribofuranoside and a novel sequence of conversions for the preparation of 2-deoxy-2-fluoro-l-arabinofuranoside derivatives.  相似文献

In trifluoromethanesulfonic acid, 2-nitromethylene-1-(ω-phenylalkyl)imidazolidine or 2-nitromethylene-1-(ω-phenylalkyl)hexahydropyrimidine derivatives undergo an intramolecular cyclization to afford (E)-hydroxyiminohydroazaazoniabenzocycloalkenes, in their trifluoromethanesulfonate salt form. The reaction probably occurres via the formation of an electrophilic transient hydroxynitrilium ion (or O-protonated nitrile oxide). Reactions of 2-ethoxymethylidene 1,3-dicarbonyl compounds with pyridine-2,6- and pyrimidine-4,5-diamines gave the corresponding 2-hetarylaminomethylidene derivatives. Depending on the initial reactant structure, the formation of mono-and/or bis-condensation products is possible. Hetarylaminomethylidene derivatives showed a moderate tuberculostatic effect. 2-Pyridinylaminomethylidene 3-oxo esters were used as ligands to obtain coordination compounds with d- and f-metal ions.  相似文献

A series of oxadiazole pyridine derivatives were synthesized by using 2-chloro-6-hydrazinoisonicotinic acid hydrazide as starting material. Treatment of the hydrazide with carbon disulfide to afford the oxadiazole derivative, which was treated with 5-methyl-2-furancarbaldehyde, formic acid, acetic acid/acetic anhydride, or phthalic anhydride to yield the corresponding pyridinodiazoles and on imide. Condensation of the hydrazide with p-fluorobenzaldehyde in ethanol or acetic acid in the presence of sodium acetate afforded hydrazone and oxadiazole derivatives, which were acetylated and cyclized with acetic anhydride to N-acetyloxadiazole derivatives. The hydrazone was treated with acetic acid in the presence of sodium acetate, or bromine water/sodium acetate to give on oxadiazole, while it was cyclized with chloroacetyl chloride in the presence of TEA to oxoazetidinaminoisonicotinamide. Finally, condensation of the hydrazide with acid anhydrides in refluxing glacial acetic acid afforded the corresponding bisimide derivatives. The pharmacological screening showed that many of these obtained compounds have good antianexiety activity comparable to diazepam® as positive control.  相似文献

Chiral, non-racemic 1,2-disubstituted ferrocenes have been prepared from monosubstituted ferrocene derivatives by amine-mediated ortho-directed reactions and subsequent partial reductive removal of the stereogenic ortho-directing group. It was found that the ortho-directing amino group of 2-substituted derivatives of N,N-dimethylaminoethyl-ferrocene and similar compounds can, after quaternisation with methyl iodide, be reductively removed with sodium borohydride to give 2-substituted methyl- or ethylferrocenes. In most cases the substituents I, Br, COOEt, P(O)Ph2 and CN tolerate the reaction conditions used. In addition, a few examples are reported that show how the use of LiTMP allows 2-bromo- and especially 2-cyano-substituted derivatives to be further ortho-lithiated and reacted to give 1,2,3-trisubstituted ferrocenes.  相似文献

2-Quinolone analogs are powerful inhibitors of farnesyl transferase, and are a novel class of anticancer drugs. The present study focused on continual efforts to elucidate the anticancer activity of synthesized 2-quinolone derivatives without N-methyl or 3-aryl substitution. Three derivatives namely JST, JST2 and JST13 were synthesized in our lab and screened for in vitro and in vivo anticancer activities. Significant cytotoxicity was observed in MCF-7 cells treated with JST2 and JST13. Both the derivatives’ treatment showed damage to the DNA. In vivo studies for JST2 and JST13 were performed at two doses 100 and 200 mg/kg using Ehrlich ascites carcinoma (liquid) and Dalton lymphoma ascites (solid) models. Both derivatives showed a significant reduction in the tumor progression by increasing the mean life span and by improving the haematological profile and antioxidant status of the liver in a liquid tumor model. More prominent effect was observed in a solid tumor model by reduction in solid tumor weight and tumor volume.  相似文献

New pathways of reaction between 1-methylthiourea or 1-methylurea and benzil bring about new derivatives of (2S*,3aR*,6aS*)-perhydro-3aH-[1,3]dioxolo[4,5-d]imidazole and racemic (4S*,5R*)-4-alkoxy-5-hydroxy-1-methyl-4,5-diphenylimidazolidine-2-thiones. Some of the obtained urea-and thiourea derivatives were characterized by X-ray diffraction, which showed their supramolecular organization governed by the directionality of hydrogen bonds at the acceptor side C=O or C=S groups.  相似文献

Continuing investigations in the field of the synthesis of condensed heterocyclic systems containing pyrrole and indole fragments ¦1–3¦, we have condensed of 2-aminopyrrole derivatives with various 1, 3-dicarbonyl compounds. The reaction took place at the boil in solutions in pyridine or acetic acid, and also without a solvent at 150–160° C, forming in a single stage derivatives of pyrrolo[1, 2-a]pyrimidine (I–III) and pyrrolo[2, 1-b]-tetrahydroquinazoline (IV–VI). The condensation of 2-aminopyrrole with acetoacetic and substituted acetoacetic esters led to 4-oxo derivatives of pyrrolo[1, 2-β]pyrimidine (VII and VIII).  相似文献
