A systematic comparison between the grafting‐to (convergent) and grafting‐from (divergent) synthetic routes leading to cyclic peptide–polymer conjugates is described. The reversible addition–fragmentation chain transfer (RAFT) process was used to control the polymerizations and the couplings between cyclic peptide and polymer or RAFT agent were performed using N‐hydroxysuccinimide (NHS) active ester ligation. The kinetics of polymerization and polymer conjugation to cyclic peptides were studied for both grafting‐to and grafting‐from synthetic routes, using N‐acryloyl morpholine as a model monomer. The cyclic peptide chain transfer agent was able to mediate polymerization as efficiently as a traditional RAFT agent, reaching high conversion in the same time scale while maintaining excellent control over the molecular weight distribution. The conjugation of polymers to cyclic peptides proceeded to high conversion, and the nature of the carbon at the α‐position to the NHS group was found to play a crucial role in the reaction kinetics. The study was extended to a wider range of monomers, including hydrophilic and temperature responsive acrylamides, hydrophilic and hydrophobic acrylates, and hydrophobic and pH responsive methacrylates. Both approaches lead to similar peptide–polymer conjugates in most cases, while some exceptions highlight the advantages of one or the other method, thereby demonstrating their complementarity. © 2015 Wiley Periodicals, Inc. The temperature‐responsive poly (N, N‐diethylacrylamide) (pDEAAm) with narrower molecular weight distribution was prepared by the atom transfer radical polymerization and characterized by 1HNMR and gel permeation chromatography. The temperature‐responsive “tadpole‐shaped” BSA–pDEAAm hybrids were fabricated via a free Cys‐34 residue of bovine serum albumin (BSA) site specifically binding to the end group disulfide bonds of pDEAAm and characterized by native‐polyacrylamide gel electrophoresis (Native‐PAGE) and matrix‐assisted laser desorption/ionization time of flight mass spectrometry. Their temperature‐responsive behaviors were measured by ultraviolet‐visible spectra (UV‐Vis). The lower critical solution temperature (LCST) of the pDEAAm was identified as 28°C, and the LCST of BSA–pDEAAm hybrids was identified as 31°C. The morphologies of BSA–pDEAAm hybrids self‐assembled in the aqueous solutions with two different temperatures at 25 °C and 40°C were investigated by transmission electron microscopy. Below the LCST of BSA–pDEAAm hybrids, the separate spherical nanoparticles were observed. In contrast, bundles and clusters were observed above the LCST of BSA–pDEAAm hybrids. The results suggested that the self‐assembly morphology of BSA–pDEAAm hybrids depended upon the pDEAAm block in BSA–pDEAAm hybrids, and the morphology transitions were effected by the LCST of BSA–pDEAAm hybrids. The dispersion reversible addition‐fragmentation chain transfer (RAFT) polymerization of 4‐vinylpyridine in toluene in the presence of the polystyrene dithiobenzoate (PS‐CTA) macro‐RAFT agent with different chain length is discussed. The RAFT polymerization undergoes an initial slow homogeneous polymerization and a subsequent fast heterogeneous one. The RAFT polymerization rate is dependent on the PS‐CTA chain length, and short PS‐CTA generally leads to fast RAFT polymerization. The dispersion RAFT polymerization induces the self‐assembly of the in situ synthesized polystyrene‐b‐poly(4‐vinylpyridine) block copolymer into highly concentrated block copolymer nano‐objects. The PS‐CTA chain length exerts great influence on the particle nucleation and the size and morphology of the block copolymer nano‐objects. It is found, short PS‐CTA leads to fast particle nucleation and tends to produce large‐sized vesicles or large‐compound micelles, and long PS‐CTA leads to formation of small‐sized nanospheres. Comparison between the polymerization‐induced self‐assembly and self‐assembly of block copolymer in the block‐selective solvent is made, and the great difference between the two methods is demonstrated. The present study is anticipated to be useful to reveal the chain extension and the particle growth of block copolymer during the RAFT polymerization under dispersion condition. © 2013 Wiley Periodicals, Inc. Three new ureidopyrimidinone(UPy)‐functionalized chain‐transfer agents (CTAs) have been synthesized for use in reversible addition‐fragmentation chain transfer (RAFT) polymerization. These UPy‐CTAs are able to polymerize a wide variety of vinyl monomers to yield UPy‐functionalized polymers, including homopolymers, block copolymers, and amphiphilic block copolymers. These polymers have been characterized via 1H and 13C NMR spectroscopy, gel permeation chromatography (GPC), UV/visible spectroscopy and differential scanning calorimetry (DSC) to demonstrate end‐group fidelity. © 2010 Wiley Periodicals, Inc. In this study, we designed and investigated pH‐responsive nanoparticles based on different ratios of monomers with primary, secondary or tertiary amino groups. For this purpose, copolymers of methyl methacrylate (MMA) with different compositions of amino methacrylates (2‐(dimethylamino)ethyl methacrylate (DMAEMA), 2‐(tert‐butylamino)ethyl methacrylate (tBAEMA) and 2‐aminoethyl methacrylate hydrochloride (AEMA·HCI)) were synthesized using the reversible addition‐fragmentation chain transfer (RAFT) polymerization process. The controlled nature of the radical polymerization was demonstrated by kinetic studies. All copolymers show low dispersities (?M < 1.2) with amino contents between 9 and 21 mol %. For the nanoparticle formation, nanoprecipitation with subsequent solvent evaporation was used. All suspensions were characterized by dynamic light scattering (DLS) and scanning electron microscopy (SEM). Different initial conditions of the formulations resulted in differently sized nanoparticles that have monomodal size distributions, relatively narrow polydispersity index (PDI) values and positive zeta potential values. The pH‐stability test results demonstrated that, depending on the structure and amount of the amino content, the obtained nanoparticles reveal a reversible pH‐response, such as dissolution at acidic pH values. The ability of the nanoparticles to encapsulate guest molecules was confirmed by pyrene fluorescence studies. This review summarizes recent advances in the design and synthesis of amino‐acid‐based block copolymers by reversible addition–fragmentation chain transfer (RAFT) polymerization of amino‐acid‐bearing monomers. We will mainly focus on stimuli‐responsive block copolymers, such as pH‐, thermo‐, and dual‐stimuli‐responsive block copolymers, and self‐assembled block copolymers, including amphiphilic and double‐hydrophilic block copolymers having tunable chiroptical properties. We will also highlight recent results in RAFT synthesis of amino‐acid‐based copolymers having various properties, such as catalytic and optoelectronic properties, cross‐linked block copolymer micelles, unimolecular micelles, and organic–inorganic hybrids.  相似文献

Dispersion RAFT polymerization of styrene in the alcohol/water mixture mediated with the brush macro‐RAFT agent of poly[poly(ethylene oxide) methyl ether vinylphenyl‐co‐styrene] trithiocarbonate [P(mPEGV‐co‐St)‐TTC] with similar molecular weight but different chemical composition is investigated. Well‐controlled RAFT polymerization including an initial slow homogeneous polymerization and a subsequent fast heterogeneous polymerization at almost complete monomer conversion is achieved. The molecular weight of the synthesized block copolymer increases linearly with the monomer conversion, and the polydispersity is relatively narrow (PDI < 1.3). The RAFT polymerization kinetics is dependent on the chemical composition in the brush macro‐RAFT agents, and those with high content of hydrophobic segment lead to fast RAFT polymerization. The growth of the block copolymer nano‐objects during the RAFT polymerization is explored, and various block copolymer nano‐objects such as nanospheres, worms, vesicles and large‐compound‐micelle‐like particles are prepared. The parameters such as the chemical composition in the brush macro‐RAFT agent, the chain length of the solvatophobic block, the concentration of the feeding monomer and the solvent character affecting the size and morphology of the block copolymer nano‐objects are investigated. © 2013 Wiley Periodicals, Inc. The combination of metals and polymers in hybrid materials is a research area of great current interest. A number of methods for controlling the positioning of metallic species within polymer matrices on the nanometer scale have been developed. This highlight focuses on the use of functional block copolymers for the localization of metal species, especially nanoparticles, on the nanometer scale through block copolymer phase segregation. A synthetic protocol for the preparation of hetero‐biofunctional protein–polymer conjugates is described. A chain transfer agent, S,S‐bis (α,α′‐dimethyl‐α″‐acetic acid) trithiocarbonate was functionalized with α,ω‐pyridyl disulfide (PDS) groups, Subsequently, one of the PDS groups was covalently attached to bovine serum albumin (BSA) at the specific free thiol group on the cysteine residue through a disulfide linkage. The second PDS group remained intact, as it was found to be inaccessible to further BSA functionalization. The BSA‐macro‐reversible addition‐fragmentation chain transfer (RAFT) agent was then used to prepare BSA‐polymer conjugates via in situ polymerization of oligo (ethyleneglycol) acrylate and N‐(2‐hydroxypropyl) methacrylamide using an ambient temperature initiator, 4,4′‐azobis [2,9‐imidazolin‐2‐ethyl)propane] dihydrochloride in an aqueous medium. Sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS‐PAGE) confirmed that the in situ polymerization occurred at the protein surface where the RAFT agent was attached and the molecular weights of the BSA–polymer conjugates were found to increase concomitantly with monomer conversion and polymerization time. After polymerization the remaining terminal PDS groups were then utilized to attach thiocholesterol and a flurophore, rhodamine B to the protein–polymer conjugates via disulfide coupling. UV–Vis and fluorescence analyses revealed that ~80% of the protein conjugates were found to retain integral PDS end groups for further attachment to free thiol‐tethered precursors. © 2010 Wiley Periodicals, Inc. Endowing unimolecular soft nanoobjects with biomimetic functions is attracting significant interest in the emerging field of single‐chain technology. Inspired by the compartmentalized structure and polymerase activity of metalloenzymes, copper‐containing compact nanoglobules have been designed, synthesized, and characterized endowed with metalloenzyme mimicking characteristics toward controlled synthesis of water‐soluble polymers and thermoresponsive hydrogels. When compared to metalloenzymes, artificial nanoobjects endowed with metalloenzyme mimicking characteristics offer increased stability against thermal changes and reduced degradability by hydrolytic enzymes.

A facile, general, and highly efficient one‐pot approach to obtain azobenzene (azo)‐containing molecularly imprinted polymer (MIP) nanoparticles with photoresponsive template binding and release properties in aqueous media is described, which involves the combined use of hydrophilic macromolecular chain transfer agent‐mediated reversible addition‐fragmentation chain transfer precipitation polymerization and easily available water‐insoluble azo functional monomers. The resulting azo‐containing MIPs were characterized with dynamic laser scattering (DLS), SEM, FTIR, static contact angle and water dispersion studies, and equilibrium binding experiments. They have proven to be nanoparticles (their diameters being around 104–397 nm, as determined by DLS in methanol) with surface‐grafted hydrophilic polymer brushes and exhibit excellent pure water‐compatible template binding properties. Moreover, obvious photoregulated template binding behaviors were observed for such azo‐containing MIP nanoparticles, which led to their largely accelerated template release in the aqueous media under the UV light irradiation. A series of environmentally sensitive ABA triblock copolymers with different block lengths were prepared by reversible addition‐fragmentation chain transfer (RAFT) polymerization from acrylic acid (AA) and N‐isopropylacrylamide (NIPAAm). The GPC and 1H NMR analyses demonstrated the narrow molecular weight distribution and precise chemical structure of the prepared P(AA‐b‐NIPAAm‐b‐AA) triblock copolymers owing to the controlled/living characteristics of RAFT polymerization. The lower critical solution temperature (LCST) of the triblock copolymers could be tailored by adjusting the length of PAA block and controlled by the pH value. Under heating, the triblock copolymers underwent self‐assemble in dilute aqueous solution and formed nanoparticles revealed via TEM images. Physically crosslinked nanogels induced by inter‐/intra‐hydrogen bonding or core‐shell micelle particles thus could be obtained by changing environmental conditions. With a well‐defined structure and stimuli‐responsive properties, the P(AA‐b‐NIPAAm‐b‐AA) copolymer is expected to be employed as a nanocarrier for biomedical applications in controlled‐drug delivery and targeting therapy. © 2015 Wiley Periodicals, Inc. Controlling the spatial distribution of catalytic sites in metallo‐folded single‐chain nanoparticles (SCNPs) is a first step toward the rational design of improved catalytic soft nano‐objects. Here an unexplored pathway is reported for tuning the internal structure of metallo‐folded SCNPs. Unlike the conventional SCNP synthesis in good solvent (protocol I), the proposed new route (protocol II) is based on the use of amphiphilic random copolymers and transfer, after SCNP formation, from selective to good (nonselective) solvent conditions. The size and morphology of the SCNPs obtained by the two protocols, and the corresponding spatial distribution of the catalytic sites, have been determined by combining results from size exclusion chromatography with triple detection, small‐angle X‐ray scattering and molecular dynamics (MD) simulations. Remarkably, the use of these protocols allows the tuning of the internal structure of the metallo‐folded SCNPs, as supported by MD simulations results. While the conventional protocol I yields a homogeneous distribution of the catalytic sites in the SCNP, these are arranged into clusters in the case of protocol II.

Nanostructures derived from amphiphilic DNA–polymer conjugates have emerged prominently due to their rich self‐assembly behavior; however, their synthesis is traditionally challenging. Here, we report a novel platform technology towards DNA–polymer nanostructures of various shapes by leveraging polymerization‐induced self‐assembly (PISA) for polymerization from single‐stranded DNA (ssDNA). A “grafting from” protocol for thermal RAFT polymerization from ssDNA under ambient conditions was developed and utilized for the synthesis of functional DNA–polymer conjugates and DNA–diblock conjugates derived from acrylates and acrylamides. Using this method, PISA was applied to manufacture isotropic and anisotropic DNA–polymer nanostructures by varying the chain length of the polymer block. The resulting nanostructures were further functionalized by hybridization with a dye‐labelled complementary ssDNA, thus establishing PISA as a powerful route towards intrinsically functional DNA–polymer nanostructures.  相似文献

A carboxylic acid based reversible additionfragmentation transfer (RAFT) agent is used to prepare gels composed of worm‐like diblock copolymers using two non‐ionic monomers, glycerol monomethacrylate (GMA) and 2‐hydroxypropyl methacrylate (HPMA). Ionization of the carboxylic acid end‐group on the PGMA stabilizer block induces a worm‐to‐sphere transition, which in turn causes immediate degelation. This morphological transition is fully reversible as determined by TEM and rheology studies and occurs because of a subtle change in the packing parameter for the copolymer chains. A control experiment where the methyl ester derivative of the RAFT agent is used to prepare the same diblock copolymer confirms that no pH‐responsive behavior occurs in this case. This end‐group ionization approach is important for the design of new pH‐responsive copolymer nano‐objects as, unlike polyacids or polybases, only a minimal amount of added base (or acid) is required to drive the morphological transition.  相似文献

Superhydrophobic surfaces (SHS) find versatile applications as coatings due to their very high water‐repellency, self‐cleaning, and anti‐icing properties. This investigation describes the preparation of a SHS from surfactant‐free hybrid fluoropolymer latex. In this case, reversible addition‐fragmentation chain transfer (RAFT) polymerization was adopted to prepare a copolymer of 4‐vinyl pyridine (4VP) and vinyl triethoxysilane (VTES), where the pyridine units were quaternized to make the copolymer soluble in water. The copolymer was further used as a macro‐RAFT agent to polymerize 2,2,2‐trifluoroethyl methacrylate (TFEMA) in a surfactant‐free emulsion via polymerization‐induced self‐assembly (PISA). The macro‐RAFT agent contained a small amount of VTES as co‐monomer which was utilized to graft silica nanoparticles (SNPs) onto the P(TFEMA) spheres. The film prepared using the nanocomposite latex exhibited a nano‐structured surface as observed by SEM and AFM analyses. Polymer–drug conjugates have attracted great interest as one category of various promising nanomedicines due to the advantages of high drug‐loading capacity, negligible burst release, and improved pharmacokinetics as compared with the small molecular weight drugs or the polymeric delivery systems with physically encapsulated drugs. Herein, a new type of oxidation‐responsive polymer–drug conjugates composed of a poly(ethylene glycol) (PEG) block and a hydrophobic polyacrylate block to which Naproxen is attached through a phenylboronic ester linker is reported. The amphiphilic block copolymers are synthesized through the reversible addition–fragmentation chain transfer polymerization of the Naproxen‐containing acrylic monomer using a PEG chain transfer agent. In neutral aqueous buffer, the conjugates formed nanoparticles with diameters of ≈150–300 nm depending on the length of the hydrophobic segment. The dynamic covalent bond of the phenylboronic ester is stabilized due to the hydrophobic microenvironment inside the nanoparticles. Upon exposure to H2O2, the phenylboronic ester is oxidized rapidly into the phenol derivative which underwent a 1,6‐elimination reaction, releasing the intact Naproxen. The rate of drug release is influenced by the concentration of H2O2 and the hydrophobic block length. This type of oxidation‐responsive polymer–drug conjugate is feasible for other drugs containing hydroxyl group or amino group.

Polymerization‐induced self‐assembly of block copolymer through dispersion RAFT polymerization has been demonstrated to be a valid method to prepare block copolymer nano‐objects. However, volatile solvents are generally involved in this preparation. Herein, the in situ synthesis of block copolymer nano‐objects of poly(ethylene glycol)‐block‐polystyrene (PEG‐b‐PS) in the ionic liquid of 1‐butyl‐3‐methylimidazolium hexafluorophosphate ([BMIN][PF6]) through the macro‐RAFT agent mediated dispersion polymerization is investigated. It is found that the dispersion RAFT polymerization of styrene in the ionic liquid of [BMIN][PF6] runs faster than that in the alcoholic solvent, and the dispersion RAFT polymerization in the ionic liquid affords good control over the molecular weight and the molecular weight distribution of the PEG‐b‐PS diblock copolymer. The morphology of the in situ synthesized PEG‐b‐PS diblock copolymer nano‐objects, e.g., nanospheres and vesicles, in the ionic liquid is dependent on the polymerization degree of the solvophobic block and the concentration of the fed monomer, which is somewhat similar to those in alcoholic solvent. A facile synthetic pathway to miktoarm star copolymers with multiple arms has been developed by combining reversible addition–fragmentation chain transfer (RAFT) arm‐first technique and aldehyde–aminooxy “click” coupling reaction. Star polystyrene (PS) with aldehyde functionalized core was initially prepared by RAFT arm‐first technique via crosslinking of the preformed linear macro‐RAFT agents using a newly designed aldehyde‐containing divinyl compound 6,6′‐(ethane‐1,2‐diylbis(oxy))bis(3‐vinylbenzaldehyde) (EVBA). It was then used as a multifunctional coupling agent for the subsequent formation of the second generation poly(ethylene glycol) (PEG) arms via the click coupling reaction between its aldehyde groups and aminooxy‐terminated PEGs.
