The SELective INverse detection of carbon–proton CORrelation pulse sequence that yields a 1D spectrum of a proton directly bonded to a selected carbon resonance has been converted into a proton and carbon double‐selective variant that provides a 1H spectrum of a selected proton that is long‐range coupled to a specific carbon resonance. The resulting 1D proton multiplet exhibits a pure absorptive in‐phase lineshape for precise measurement of specific long‐range proton–carbon coupling constants in small organic molecules at natural abundance. A spin state‐selective Heteronuclear Single‐Quantum Multiple‐Bond Connectivities (HSQMBC‐COSY) experiment is proposed to measure the sign and the magnitude of long‐range proton‐carbon coupling constants (nJ(CH); n > 1) either for protonated or for non‐protonated carbons in small molecules. The simple substitution of the selective 180° 1H pulse in the original selHSQMBC pulse scheme by a hard one allows the simultaneous evolution of both proton‐proton and proton‐carbon coupling constants during the refocusing period and enables a final COSY transfer between coupled protons. The successful implementation of the IPAP principle leads to separate mixed‐phase α/β cross‐peaks from which nJ(CH) values can be easily measured by analyzing their relative frequency displacements in the detected dimension. A new pulse sequence for determining scalar long range C,H spin couplings in small organic molecules is proposed. It is based on the combination of a selective J‐resolved spectrum with a selective HMBC and displays the long‐range spin couplings of one chosen carbon atom in the indirect dimension. Four‐ and five‐bond heteronuclear J‐couplings between the hydrogen H‐8 and carbons C‐6 and C‐2 in a series of 7‐ and 9‐benzyl substituted purine derivaties with variuous substituents in positions 2 and 6 were studied by coupled 13 C NMR and H,C‐HMBC experiments and by DFT calculations. We have found that for some of the derivatives, the five‐bond coupling H8‐C2 is higher than the four‐bond H8‐C6 coupling, which is also evidenced by a stronger crosspeak in the HMBC. This finding contradicts the generally accepted opinion that only strong three‐bond crosspeaks and one weak four‐bond H8‐C6 crosspeak can be observed in the HMBC spectra of purine derivatives. The misinterpretation of HMBC spectra may lead to an incorrect determination of the purine derivatives' structure. Copyright © 2012 John Wiley & Sons, Ltd.  相似文献

An efficient pulse sequence for measuring long‐range C? H coupling constants (JC? H) named selective J‐resolved HMBC has been developed by replacing a 1H 180° pulse with a selective 1H 180° pulse and the HMBC pulse scheme with the constant time (CT) HMBC employed in the J‐resolved HMBC pulse sequence that we reported previously. The novel pulse sequence providing only long‐range JC? H cross peaks for easy and accurate analysis enables to overcome disadvantages of the previous HMBC‐based pulse sequences (J‐resolved HMBC‐1) along with maintaining high sensitivity. The efficiency of measuring long‐range JC? H using the proposed pulse sequence has been demonstrated in applications to the complicated natural products, portmicin and monazomycin. This work reports on the comprehensive calculation of the NMR one‐bond spin–spin coupling constants (SSCCs) involving carbon and tellurium, 1J(125Te,13C), in four representative compounds: Te(CH3)2, Te(CF3)2, Te(C?CH)2, and tellurophene. A high‐level computational treatment of 1J(125Te,13C) included calculations at the SOPPA level taking into account relativistic effects evaluated at the 4‐component RPA and DFT levels of theory, vibrational corrections, and solvent effects. The consistency of different computational approaches including the level of theory of the geometry optimization of tellurium‐containing compounds, basis sets, and methods used for obtainig spin–spin coupling values have also been discussed in view of reproducing the experimental values of the tellurium–carbon SSCCs. Relativistic corrections were found to play a major role in the calculation of 1J(125Te,13C) reaching as much as almost 50% of the total value of 1J(125Te,13C) while relativistic geometrical effects are of minor importance. The vibrational and solvent corrections account for accordingly about 3–6% and 0–4% of the total value. It is shown that taking into account relativistic corrections, vibrational corrections and solvent effects at the DFT level essentially improves the agreement of the non‐relativistic theoretical SOPPA results with experiment. © 2016 Wiley Periodicals, Inc.  相似文献

Experimental measurements and second‐order polarization propagator approach (SOPPA) calculations of 77Se? 1H spin–spin coupling constants together with theoretical energy‐based conformational analysis in the series of 2‐substituted selenophenes have been carried out. A new basis set optimized for the calculation of 77Se? 1H spin–spin coupling constants has been introduced by extending the aug‐cc‐pVTZ‐J basis for selenium. Most of the spin–spin coupling constants under study, especially vicinal 77Se? 1H couplings, demonstrated a remarkable stereochemical behavior with respect to the internal rotation of the substituent in the 2‐position of the selenophene ring, which is of major importance in the stereochemical studies of the related organoselenium compounds. Natural products often possess various spin systems consisting of a methine group directly bonded to a methyl group (e.g. –CHa–CHb(CH3)–CHc–). The methine proton Hb splits into a broadened multiplet by coupling with several vicinal protons, rendering analysis difficult of nJC–H with respect to Hb in the J‐resolved HMBC‐1. In purpose of the reliable and easy measurements of nJC–H and nJH–H in the aforesaid spin system, we have developed a new technique, named BASHD‐J‐resolved‐HMBC. This method incorporates band selective homo decoupled pulse and J‐scaling pulse into HMBC. In this method, high resolution cross peaks can be observed along the F1 axis by J‐scaling pulse, and band selective homo decoupled pulse simplified multiplet signals. The experimental spin–spin coupling constants (SSCCs) for 1,3‐ and 1,4‐difluorobenzene have been determined anew, and found to be consistent with previously determined values. SSCCs for 1,2‐, 1,3‐, and 1,4‐difluorobenzene have been analyzed by comparing them with the coupling constants computed using the second‐order polarization propagator approximation (SOPPA) and the equation‐of‐motion coupled cluster singles and doubles method (EOM‐CCSD). Eighty experimental values have been analyzed using SOPPA calculations, and a subset of 40 values using both SOPPA and EOM‐CCSD approaches. One‐bond coupling constants 1J(C? C) and 1J(C? F) are better described by EOM‐CCSD, whereas one‐bond 1J(C? H) values are better described by SOPPA. An empirical equation is presented which allows for the prediction of unknown coupling constants from computed SOPPA values. 2‐Bromocyclohexanone is a model compound in which a 4JH2, H6 coupling constant is observed, whereas the corresponding 4JH2, H4 is absent. The observed long‐range coupling is not only a result of the known W‐type coupling, in the axial conformation, but also because of the less usual diaxial spin–spin coupling in the equatorial conformer. The carbonyl group plays a determining role in describing the coupling pathway, as concluded by natural bond orbital (NBO) analysis; although the and interactions in the axial conformer contribute for transmitting the spin information associated with the W‐type coupling, the strong and hyperconjugations in the equatorial conformer define an enhanced coupling pathway for 4JH2, H6, despite the inhibition of this coupling because of interaction and the large carbonyl angle. These findings provide the experimental evidence that orbital interactions contribute for the conformational isomerism of 2‐bromocyclohexanone. Copyright © 2008 John Wiley & Sons, Ltd.  相似文献

Benchmark calculations of geminal and vicinal 29Si–1H spin–spin coupling constants across double bond in three reference alkenylsilanes have been carried out at both DFT and SOPPA levels in comparison with experiment. At the former, four density functionals, B3LYP, B3PW91, PBE0 and KT3, were tested in combination with five representative basis sets. At the latter, three main SOPPA‐based methods, SOPPA, SOPPA(CC2) and SOPPA(CCSD), were examined in combination with the same series of basis sets. On the whole, the wavefunction methods showed much better results as compared to DFT, with the most efficient combination of SOPPA/cc‐pVTZ‐su2 characterized by a mean absolute error of only 0.4 Hz calculated for a set of nine coupling constants in three compounds with a sample span of around 40 Hz. The computational study of the one‐bond 29Si–13C spin–spin coupling constants has been performed at the second‐order polarization propagator approximation (SOPPA) level in the series of 60 diverse silanes with a special focus on the main factors affecting the accuracy of the calculation including the level of theory, the quality of the basis set, and the contribution of solvent and relativistic effects. Among three SOPPA‐based methods, SOPPA(MP2), SOPPA(CC2), and SOPPA(CCSD), the best result was achieved with SOPPA(CCSD) when used in combination with Sauer's basis set aug‐cc‐pVTZ‐J characterized by the mean absolute error of calculated coupling constants against the experiment of ca 2 Hz in the range of ca 200 Hz. The SOPPA(CCSD)/aug‐cc‐pVTZ‐J method is recommended as the most accurate and effective computational scheme for the calculation of 1J(Si,C). The slightly less accurate but essentially more economical SOPPA(MP2)/aug‐cc‐pVTZ‐J and/or SOPPA(CC2)/aug‐cc‐pVTZ‐J methods are recommended for larger molecular systems. It was shown that solvent and relativistic corrections do not play a major role in the computation of the total values of 1J(Si,C); however, taking them into account noticeably improves agreement with the experiment. Conformational study of five derivatives of 2‐(pyrazol‐4‐yl)‐1,3‐diselenane together with related 1,2‐diselenolane in respect to the stereochemical trends of geminal and vicinal 77Se‐1H spin‐spin coupling constants has been carried out by means of high‐level theoretical calculations in combination with experiment. The marked dihedral angle dependences for both types of couplings accounted for the lone pair effect in the case of geminal coupling constants and the Karplus‐type relationship for vicinal couplings have been established, which is of major importance for the stereochemical analysis of saturated selenium containing heterocycles. The main photophysical properties of a series of recently synthetized 1,2‐ and 1,3‐squaraines, including absorption electronic spectra, singlet‐triplet energy gaps, and spin‐orbit matrix elements, have been investigated by means of density functional theory (DFT) and time‐dependent DFT approaches. A benchmark of three exchange‐correlation functionals has been performed in six different solvent environments. The investigated 1,2 squaraines have been found to possess two excited triplet states (T1 and T2) that lie below the energy of the excited singlet one (S1). The radiationless intersystem spin crossing efficiency is thus enhanced in both the studied systems and both the transitions could contribute to the excited singlet oxygen production. Moreover, they have a singlet‐triplet energy gap higher than that required to generate the cytotoxic singlet oxygen species. According to our data, these compounds could be used in photodynamic therapy applications that do not require high tissue penetration. © 2014 Wiley Periodicals, Inc.  相似文献

In 1975 a large number of coupling constants were measured in 2‐fluorobenzamide labeled with 15N. Some of them were assigned to couplings through intramolecular N? H···F hydrogen bonds (HBs). These couplings change dramatically when CDCl3 is replaced by DMSO‐d6. In this theoretical paper we provide density functional theory (DFT) calculations that justify the existence of a weak HB in the absence of solvent, while solvents that act as HB acceptors break down the intramolecular hydrogen bond (IMHB) of 2‐fluorobenzamide. A 3D 1H–13C–1H refocused INEPT transfer experiment is proposed in which the initial coherence transfer of 1H longitudinal to 13C transverse magnetization is tuned to the long‐range 1H, 13C couplings while the reverse INEPT component transfers the magnetization to the directly bonded 1H. Integration of a constant time 1H evolution period into the long‐range coherence transfer interval provides absorption mode signals for each dimension. A 13C purge component at the beginning of the sequence selects for 12C‐bound 1H magnetization that is then transferred to a 13C‐bound hydrogen, thus strongly suppressing the diagonal signals. This experiment is expected to be of particular value for situations in which resonance overlap in the 13C dimension renders 2D long‐range heteronuclear correlation data ambiguous. In combination with a diagonal‐suppressed 3D 1H–13C–1H TOCSY‐HSQC experiment, complete assignment of the ring resonances of the Lewis‐b hexasaccharide was obtained on a 4.2 mM sample using a conventional 500 MHz probe (0.1% ethylbenzene signal‐to‐noise ratio of 600), suggesting its applicability to sub‐millimolar samples using cryoprobe technology. Good performance of segmented contracted basis sets XZP, where X = D, T, Q and 5, for obtaining H2O, H2, HF, F2 and F2O nuclear isotropic shielding constants in the BHandH Kohn–Sham basis set limit was shown. The results of two‐ and three‐parameter complete basis set limit extrapolation schemes were compared with experimental results, earlier literature data and benchmark ab initio results. Similar convergence patterns of shieldings obtained from calculations using general purpose XZP basis sets and from polarization‐consistent basis sets pcS‐n and pcJ‐n, where n = 0, 1, 2, 3 and 4, designed to accurately predict magnetic properties were observed. On the contrary, the SSCCs were more sensitive to the XZP basis set size and generally less accurate than those estimated using pcJ‐n basis set family. The BHandH density functional markedly outperforms B3LYP method in predicting heavy atom shieldings and SSCCs values in the studied systems. Copyright © 2009 John Wiley & Sons, Ltd.  相似文献

This work describes the synthesis of novel 1,2,3‐triazole‐4‐linked (2E,6E)‐2‐benzylidene‐6‐(4‐nitrobenzylidene)cyclo‐hexanones starting from cyclohexanone. 1‐(Cyclohex‐1‐en‐1‐yl)piperidine, the enamine from cyclohexanone and piperidine, reacted with 4‐nitrobenzaldehyde to obtain 2‐(4‐nitrobenzylidene)cyclohexanone. Condensation of the latter compound with (prop‐2‐yn‐1‐yloxy)benzaldehyde derivatives under acidic conditions gave (4‐nitrobenzylidene)‐[(prop‐2‐yn‐1‐yloxy)‐benzylidene]cyclohexanones. Finally, ‘click reaction’ of these derivatives and various organic azides led to the title compounds. All compounds were examined by MTT assay for cytotoxic activity in one human breast cancer cell line, MDA‐MB‐231.  相似文献

Substituted 2‐(benzylamino)‐2H‐1,4‐benzoxazin‐3(4H)‐ones are unstable under alkaline and acidic conditions, undergoing opening of the benzoxazinone ring. 2‐Bromo‐2H‐1,4‐benzoxazin‐3(4H)‐ones show similar degradation under alkaline conditions, while replacement of Br at C(2) to give 2‐hydroxy‐2H‐1,4‐benzoxazin‐3(4H)‐ones was observed only under mild alkaline conditions. Mechanisms of ring opening and degradation to 2‐aminophenol derivatives are proposed.  相似文献
