Summary: We have successfully constructed a redox‐responsible hydrogel system by combination of β‐cyclodextrin (β‐CD), dodecyl‐modified poly(acrylic acid) [p(AA/C12)], and a redox‐responsive guest, ferrocenecarboxylic acid (FCA). In the reduced state of FCA, the ternary mixture exhibited a gel‐like behavior, whereas, in its oxidized state, the mixture exhibited a sol behavior.

Summary: β‐Cyclodextrin (β‐CD) pseudopolyrotaxanes containing poly(thiophene‐2,5‐diyl), PTh , or poly(3‐methylthiophene‐2,5‐diyl)s, P3MeTh s, as an axle were prepared. Structures of the pseudopolyrotaxanes and their inclusion behavior with β‐CD were investigated. The UV‐vis measurements revealed that inclusion of P3MeTh s by β‐CD depended on the flexibility of the main chain and their molecular weight.

This work is focused on the controlled drug release behavior of hyperbranched HPMA in the presence of β‐CD. Hence, three HPMA‐β‐CDs and a pure HPMA were synthesized by Michael addition polymerization. As a model drug, CLB (an anti‐cancer drug) was loaded into them via a solution method for in vitro release studies. The DSC results indicate that the CLB/polymer interactions are at the molecular level. Loading CLB into these polymers results in an evident increase in their glass transition temperatures, and ΔTg depends on the β‐CD content. The controlled‐release experiments show that the presence of β‐CD can appropriately slow the release of CLB from HPMA‐β‐CDs and adjust the ratio of CLB released in total drug loading.

Novel functional polymers utilizing specific host/guest interactions were designed by introducing α‐CD host molecules into poly(ε‐lysine) chains as side groups. An interesting phase separation was observed as a result of the inclusion complexation between the polymeric host and 3‐(trimethylsilyl)propionic acid as a model guest in aqueous media. This water‐soluble polymeric host would be useful for various applications, particularly drug delivery, due to its biodegradability, low toxicity, and unique functionality represented as a complexation‐induced phase separation.  相似文献

Summary: Host‐guest complexes of α‐cyclodextrin (α‐CD) and methylated β‐cyclodextrin (Me‐β‐CD) with diacrylates and dimethacrylates of butan‐1,4‐diol and hexan‐1,6‐diol at varying stoichiometries were studied. The complexes were analyzed by means of 1H NMR, two‐dimensional ROESY spectroscopy and Job's curves, which clearly revealed the discriminating influence of the two hosts towards complex formation. The corresponding polymers were obtained using a redox initiator system in water. Thermal analysis and IR measurements of the polymers provided evidence for the existence of a polyrotaxane architecture.

Two geometrically isomeric γ‐cyclodextrin derivatives with a thymolphthalein moiety in the secondary hydroxyl side were prepared as guest‐responsive color change indicators. The isomers exhibit a pH dependence of their absorption spectra and a remarkable change in the absorbance around 610 nm upon guest addition. One of the isomers formed complexes of 2 : 1, the other of 1 : 1 stoichiometry (host/guest).  相似文献

A new diphenylphosphane based on a β‐cyclodextrin skeleton that exhibits a dual solubility in water and in organic solvent was synthesised. Interestingly, a solvent‐dependent conformation change was evidenced by NMR spectroscopy studies; the self‐inclusion of a phenyl group of the phosphane moiety into cyclodextrin cavity observed in water disappeared in organic solvents due to a change in conformation. Hydrogenation or hydroformylation reactions performed in water and in organic solvents showed that this ligand was able to stabilise catalytically active rhodium species in solution. In the case of the hydroformylation reaction, it was demonstrated that regioselectivity was influenced by the solvent‐dependent conformation of the ligand.  相似文献

Molecular Recognition of α,α,α,β-ZnT(o-BocThr)APP (1) toward a series of imidazole derivatives and amino acid esters was investigated. Association constants were determined in chloroform by means of UV-Vis titration method. The association constants of 1 with imidazole derivatives are larger than those of 1 with amino acid esters. 1H NMR spectra were investigated to describe the binding mode of the recognition system, showing that all the protons of the guests were shifted to upfield. The circular dichroism spectra of 1-L-/D-ValOMe showed a split cotton effect in Soret region, while those of 1-L-/D-PheOMe showed no split cotton effect. Molecular modeling was performed to understand chiral recognition on a molecular level. Quantum chemical calculation was carried out based on the stable conformations of these recognition systems, which gave a reasonable explanation for the behavior of molecular recognition. The results indicated that the conformation of 1-D-ValOMe was more stable than that of 1-L-ValOMe.  相似文献

O‐Methacryloyl‐N‐(tert‐butoxycarbonyl)‐β‐hydroxyaspartic acid dimethyl ester was synthesized by methyl esterification of β‐hydroxyaspartic acid, followed by protection of the amino group with the tert‐butoxycarbonyl group and then the reaction of the hydroxyl group with methacryloyl chloride. The monomer efficiently underwent radical polymerization to afford the corresponding polymer with a number‐average molecular weight of 42,000 in good yields. The alkaline hydrolysis of the polymer occurred not only at the methyl ester but also at the ester moiety between the main and side chains of the polymer. The methyl ester‐free polymer gradually released β‐hydroxyaspartic acid moiety in a phosphate buffer solution with pH = 7.3 and 7.8. © 2002 Wiley Periodicals, Inc. 1H NMR spectroscopic titrations in competitive and non‐competitive media, as well as binding studies in two‐phase systems, such as phase transfer of sugars from aqueous into organic solvents and dissolution of solid carbohydrates in apolar media revealed both highly effective recognition of neutral carbohydrates and interesting binding preferences of an acyclic phenanthroline‐based receptor 1 . Compared to the previously described acyclic receptors, compound 1 displays significantly higher binding affinities, the rare capability to extract sugars from water into non‐polar organic solutions and α‐ versus β‐anomer binding preference in the recognition of glycosides, which differs from those observed for other receptor systems. X‐ray crystallographic investigations revealed the presence of water molecules in the binding pocket of 1 that are engaged in the formation of hydrogen‐bonding motifs similar to those suggested by molecular modelling for the sugar OH groups in the receptor–sugar complexes. The molecular modelling calculations, synthesis, crystal structure and binding properties of 1 are described and compared with those of the previously described receptors.  相似文献

A novel achiral monomer end‐capped with a phenyl‐[1,3,4]oxadiazolyl group and threaded through β‐cyclodextrin was synthesized to investigate the host‐guest interactions in the inclusion complex. 1H NMR studies revealed that one or two cyclodextrin molecules were threaded onto the synthesized achiral monomer, leading to the formation of a fibrous construction of self‐assembled inclusion complexes. The formation of a self‐assembled inclusion complex was identified using SEM and TEM. The highly ordered alignment of self‐assembled supramolecules was confirmed using polarized optical microscopy. We demonstrate an easy process for the fabrication of nano‐structured self‐assembled inclusion complexes in pyridine/ethanol (1 mL/10 mL) as well as the enhancement of photo‐induced fluorescence via monomers end‐capped with a phenyl‐[1,3,4]oxadiazolyl moiety threaded with β‐cyclodextrins. © 2010 Wiley Periodicals, Inc. Three different characterization methods—13C NMR spectroscopy, a terminal terpolymerization model, and a probability analysis based on the Poisson distribution—were used to determine the microstructure of random terpolymers. The methods were used to determine the amino acid sequence distribution of random terpolymers prepared from the polymerization of N‐carboxyanhydrides that contained L ‐leucine, β‐benzyl‐L ‐aspartate, and L ‐valine. Poly(L ‐leucine‐L ‐aspartic acid‐L ‐valine) [poly(LDV)] was designed as a target specific substrate for the α4β1 integrin that recognizes the tripeptide sequence leucine‐aspartic acid‐valine (LDV). The presence of the tripeptide sequence LDV within the polymer was determined to be eight LDV triad sequences on average in terpolymers of approximately 100 kDa. © 2006 Wiley Periodicals, Inc. Amphiphilic diblock copolymers with various block compositions were synthesized on poly(2‐ethyl‐2‐oxazoline) (PEtOz) as a hydrophilic block and poly(4‐methyl‐ε‐caprolactone) (PMCL) or poly(4‐phenyl‐ε‐caprolactone) (PBCL) as a hydrophobic block. These PEtOz‐b‐PMCL and PEtOz‐b‐PBCL copolymers consisting of soft domains of amorphous PEtOz and PM(B)CL had no melting endothermal peaks but displayed Tg. The lower critical solution temperature (LCST) values for the PEtOz‐b‐PMCL, and the PEtOz‐b‐PBCL aqueous solution were observed to shift to lower temperature than PEtOz homopolymers. Their aqueous solutions were characterized using fluorescence techniques and dynamic light scattering (DLS). The block copolymers formed micelles with critical micelle concentrations (CMCs) in the range 0.6–11.1 mg L?1 in an aqueous phase. As the length of the hydrophobic PMCL or PBCL blocks elongated, lower CMC values were generated. The mean diameters of the micelles were between 127 and 318 nm, with PDI in the range of 0.06–0.21, suggesting nearly monodisperse size distributions. The drug entrapment efficiency and drug‐loading content of micelles depend on block polymer compositions. In vitro cell viability assay showed that PEtOz‐b‐PMCL has low cytotoxicity. Doxorubicin hydrochloride (DOX)‐loaded micelles facilitated human cervical cancer (HeLa) cell uptake of DOX; uptake was completed within 2 h, and DOX was able to reach intracellular compartments and enter the nuclei by endocytosis. © 2013 Wiley Periodicals, Inc. The synthesis and characterisation of a novel isomeric family of closo‐carborane‐containing PtII complexes ((R/S)‐( 1 – 4 )?2 NO3) are reported. Related complexes ( 5 ?NO3 and 6 ?NO3) that contain the 7,8‐nido‐carborane cluster were obtained from the selective deboronation of the 1,2‐closo‐carborane analogues. The corresponding water‐soluble supramolecular 1:1 host–guest β‐cyclodextrin (β‐CD) adducts ((R/S)‐( 1 – 4 ) ? β‐CD?2 NO3) were also prepared and fully characterised. HR‐ESI‐MS experiments confirmed the presence of the host–guest adducts, and 2D‐1H{11B} ROESY NMR studies showed that the boron clusters enter the β‐CD from the side of the wider annulus. Isothermal titration calorimetry (ITC) experiments revealed enthalpically driven 1:1 and higher‐order supramolecular interactions between β‐CD and (R/S)‐( 1 – 4 )?2 NO3 in aqueous solution. A comparison of the predominate 1:1 binding mode established that the affinity of β‐CD for the guest molecule is mainly influenced by the pyridyl ring substitution pattern and chirality of the host, whilst the nature of the closo‐carborane isomer also plays some role, with the most favourable structural features for β‐CD binding being the presence of the 4‐pyridyl ring, 1,12‐closo‐carborane, and an S configuration. The results reported here represent the first comprehensive calorimetric study of the supramolecular interactions between closo‐carborane compounds and β‐CD, and it provides fascinating insights into the structural features influencing the thermodynamics of this phenomenon.  相似文献

Changing employment : Receptor 1 binds β‐N‐acetylglucosaminyl (β‐GlcNAc) up to 100 times more strongly than it does glucose. This synthetic lectin shows affinities similar to wheat germ agglutinin (WGA), a natural lectin used to bind GlcNAc. Remarkably, 1 is more selective than WGA. It favors especially the glycoside unit in glycopeptide 2 , a model of the serine‐O‐GlcNAc posttranslational protein modification.
