应用高解析离子淌度质谱(HDMS)与纳升级超高效液相色谱(UPLC)联用, 寻找冠心病不稳定性心绞痛血瘀证血浆差异蛋白, 探索冠心病不稳定性心绞痛血瘀证的蛋白质组学特点. 采用美国Agilent公司多克隆抗体亲和柱去除冠心病不稳定性心绞痛血瘀证患者和健康人血浆中6种丰度最高的蛋白后, 进行无标记定量蛋白质组(Label free proteome)分析. 结果表明, 本方法的离子强度变异系数小于5%, 保留时间变异系数小于3%, 具有良好的重现性. 动态范围约104数量级. 总蛋白数3843种, 差异倍数大于1.5倍的差异蛋白数25种, 上调蛋白数13种(包括心绞痛血瘀证患者独有蛋白3种), 下调蛋白数12种. ACTA1, ITIH3和 LBP仅在冠心病不稳定性心绞痛血瘀证患者血浆中表达, Haptoglobin, SAA, CP, C6, MYH11, APOH和ANXA6在冠心病不稳定性心绞痛血瘀证患者中高表达, 而HBB, HBA, HBE, HBD, HBG, HRG, IGHG, GSN和TF在冠心病不稳定性心绞痛血瘀证患者中低表达. 表达上调的差异蛋白根据功能可分为: (1) 急性时相反应蛋白; (2) 补体蛋白; (3) 细胞骨架蛋白; (4) 凝血相关蛋白. 表达下调的差异蛋白根据功能可分为: (1) 载脂蛋白; (2) 运输蛋白; (3) 抗凝血相关蛋白; (4) 免疫球蛋白; (5) 细胞骨架调控蛋白. 以上结果提示, 冠心病不稳定性心绞痛血瘀证可能属于一种炎症反应; 冠心病不稳定性心绞痛血瘀证患者可能同时存在心肌损伤、凝血因子异常、脂代谢紊乱与氧运输障碍; 这些方面相互影响, 互为因果. 这些差异蛋白可为研究或发现抗心绞痛药物作用的新靶标提供线索. 本研究结果表明, 非标记定量蛋白质组学(Label free proteomics)是疾病及证候生物标志物研究的一种有效手段.  相似文献

采用双向凝胶电泳和生物质谱技术, 对12对胰腺癌组织和癌旁组织样品、3个胰腺良性疾病样品、3个正常胰腺组织样品的蛋白质进行了分离和鉴定, 获得了重复性较好的双向凝胶电泳图谱; 鉴定了胰腺癌和癌旁组织的差异表达蛋白质, 发现了30个差异表达蛋白质; 应用MALDI-TOF-MS/MS技术对差异表达蛋白质进行鉴定, 共有24个蛋白质得到鉴定, 其中15个蛋白质在胰腺癌组织中表达上调, 9个蛋白质表达下调. 这些蛋白质与胰腺癌的发生相关, 可能成为胰腺癌的分子标志物和药物治疗的靶蛋白.  相似文献

以6月及12月龄SAMP 8及同龄SAMR 1为研究对象, 应用双向凝胶电泳法, 分析比较了快速老化模型小鼠(Senescence accelerated mice, SAM)的快速老化亚系SAMP 8及抗快速老化亚系SAMR 1血清蛋白表达的差异. 与同龄SAMR 1比较, 6月龄SAMP 8血清中有15个蛋白点表达显著上调, 3个蛋白点表达显著下调, 7个蛋白点只在SAMP 8中有表达; 12月龄SAMP 8血清中有9个蛋白点表达显著上调, 7个蛋白点表达显著下调, 12个蛋白点只在SAMP 8中有表达. 应用质谱进行肽质量指纹图谱分析和数据库检索共鉴定了19种蛋白质. 其中6个蛋白只在6月龄SAMP 8中表达, 4个蛋白只在12月龄SAMP 8中表达. 此外, 在6月龄及12月龄SAMP 8血清差异蛋白中, 存在9个共同的差异蛋白, 按照功能可分为4类: (1) 免疫相关蛋白; (2) 老化相关蛋白; (3) 糖代谢及神经元凋亡相关蛋白; (4) 其它蛋白. 上述研究结果显示, SAMP 8和SAMR 1血清蛋白表达存在明显差异, 其中一些差异蛋白可能是SAMP 8老化进程中相关病理生理变化的重要原因.  相似文献

以急性心梗大鼠为研究对象, 应用双向凝胶电泳法(2-DE)分析比较了维拉帕米作用下急性心梗大鼠心肌蛋白表达的差异, 从蛋白质水平探讨了维拉帕米心肌保护作用的发生机制. 结果表明, 与假手术组及模型组相比, 维拉帕米给药组心肌组织中有8个蛋白点表达显著上调, 7个蛋白点表达显著下调. 采用质谱(MALDI-TOF-MS)分析结合数据库检索, 共鉴定了其中的15种蛋白质, 可按功能分为如下4类: (1) 能量代谢及线粒体功能相关蛋白; (2) 氧化应激相关蛋白; (3) 细胞骨架蛋白; (4) 其它蛋白. 研究结果表明, 维拉帕米的心肌保护作用与恢复心肌损伤过程中的能量供应及对抗氧化应激等作用有关.  相似文献

采用MTT法考察了La3 在2×10-4~4×10-7 mol·L-1浓度范围内对体外培养的人正常肝细胞株7701的生长和增殖的影响及其对浓度的依赖关系.结果表明,低浓度的La3 对细胞有明显的促增殖作用,而高浓度的La3 对细胞的生长表现抑制作用.为探讨促增殖的细胞是否正常,利用双向凝胶电泳技术,对La3 诱导增殖的细胞和对照组正常细胞进行蛋白质分离和分析,发现两组细胞蛋白质的表达存在着明显的差异,表示在稀土促进增殖下的细胞可能并非正常,并讨论了La3 促进细胞增殖的可能机制及其过程.  相似文献

RP-HPLC分离三七粉提取液,并鉴定含有Rb1、Rg1、Re、R1等皂甙成分。以蓝斑背肛海兔(Notarcusleachii cirrosus Stimpson,NLCS)为分析模型,三七粉提取液为诱导剂,选用蛋白质组技术研究NLCS神经连索诱导前后所表达的差异蛋白质。通过优化双向凝胶电泳分离NLCS神经连索全蛋白质组技术,获得496个蛋白质斑点。采用肽指纹图谱技术和数据库检索比对法,初步鉴定了NLCS受三七粉提取液诱导前后,其神经连索表达13个差异蛋白质,其中较高的匹配率蛋白质为肌动蛋白、3-羟酯酰辅酶A脱氢酶、ATP结合转运子和甲基转移酶12。选用LOC tree软件对13个差异蛋白质进行亚细胞定位,认为它们在保护神经系统中发挥重要的调节作用。  相似文献

Radiation causes severe constraint on numerous pathological functions of cells, such as cell growth, nuclear genetic material expression and cell functions. In this study, we performed proteomic profiling of a nuclear fraction of Jurkat T lymphocyte cells under radiation along different time course by means of 2DE and MALDI TOF-MS. We found 24 protein spots whose expression had changed after radiation, including relevant proteins, genetic material proteins, metabolism proteins, molecular chaperon and nuclear membrane proteins. Based on the above it is concluded that the combination of fluorescence labeled 2D-PAGE and MALDI-TOF MS is more precisely and effectively to elucidate the protein changes in Jurkat T lymphocyte cells after irradiation.  相似文献

The response criteria for complete remission (CR) in acute myeloid leukemia (AML) are currently based on morphology and blood cell counts. However, these criteria are insufficient to establish a diagnosis in cases with poor quality bone marrow (BM) samples demonstrating a loss of cellular morphology. We investigated whether the sera of patients contained biomarkers that indicate disease response status. First, we performed multidimensional liquid chromatography-differential gel electrophoresis (MDLC-DIGE) to generate protein profiles of two pooled, paired serum samples from patients who had achieved CR; one collected at diagnosis (PreCR) and the other collected after chemotherapy (CR). Then, with the biomarker candidates found, ELISA was carried out for individual PreCR and CR samples, and for other verification sets including nonremission (NR) patients and normal samples. We selected two proteins, complement factor H (CFH) and apolipoprotein H (ApoH), with dye (Cy) ratios showing greater than 2.0-fold differences between the pooled samples. ELISA showed that CFH and ApoH are useful for distinguishing between the recovered (CR and normal) and nonrecovered (PreCR, PreNR, and NR) states in AML (p <0.001). We successfully applied a protein profiling technology of MDLC-DIGE and LC-MS/MS to discover two biomarkers for CR which needs further validation for a clinical setting.  相似文献

Examining the toxic effects of heavy metals on protein expression can be useful for gaining insight into the biomolecular mechanisms of toxicity and for identifying potential candidate metal-specific protein markers of exposure and response. In this article, we present the state of the art of proteomics in metal-toxicity-related studies. We consider different methods used for sample preparation that depend on the nature of the sample (plants, microorganisms and animals). We also describe different proteomic strategies, both gel-based and gel-free technologies, including two-dimensional gel electrophoresis (2-DE) and multi-dimensional protein-identification technology (MudPIT). We critically review the advantages and the disadvantages of such techniques and discuss the main studies carried out so far. Abstract

A wireless magnetoelastic glucose biosensor in blood plasma is described, based on using a mass sensitive magnetoelastic sensor as transducer. The glucose biosensor was fabricated by coating the ribbon‐like, magnetoelastic sensor with a pH sensitive polymer and a biolayer of glucose oxidase (GOx) and catalase. The pH response polymer swells or shrinks, thereby changing sensor mass loading, respectively, in response to increase or decrease of pH values. The GOx–catalyzed oxidation of the glucose in blood plasma produces gluconic acid, resulting in the pH sensitive polymer shrinking, which in turn decreases the sensor mass loading. The results show that the proposed magnetoelastic glucose biosensor can be successfully applied to determine the concentration of glucose in blood plasma. At glucose concentration range of 2.5–20.0 mmol/l, the biosensor responses are reversible and linear, with a detection limit of 1.2 mmol/l. Since no physical connections between the sensor and the monitoring instruments are required, this proposed biosensor can potentially be applied to in vivo and in situ measurement of glucose concentration in physiological fluids.  相似文献

目的探讨综合护理干预对妊娠高血压综合征患者血压及妊娠结局的影响。方法选取2012年6月至2015年2月来江西省新余市中医院接受治疗的88例PIH患者作为研究对象。按照入院顺序抽签后随机分为实验组和对照组,对照组患者进行常规护理。实验组患者在对照组护理的基础上,进行综合护理干预。观察患者血压、分娩方式结局、胎儿结局。结果实验组护理后收缩压和舒张压均低于对照组和护理前(P0.05)。实验组早产、产后出血、胎盘早剥、剖宫产总发生率均低于对照组(P0.05)。实验组胎儿窘迫、新生儿窒息总发生率均低于对照组(P0.05)。结论综合护理干预能够降低妊娠高血压综合征患者的血压,改善妊娠结局。  相似文献
