The radiosensitivity of codling moth, Cydia pomonella (L.), eggs in different stages of development was studied. Eggs ranging in age from 1–24 to 97–120 h were exposed, at 24 h intervals, to gamma radiation doses ranging from 10 to 350 Gy. The effects of gamma radiation on egg hatch, pupation and adult emergence was examined. Results showed that the radiosensitivity of codling moth eggs decreased with increasing age. Egg hatch in 1–24 h old eggs was significantly affected at 20 Gy dose and at 60 Gy dose, egg hatch decreased to about 1%. At the age of 25–48 h, however, egg hatch at 60 Gy dose was about 10%, and egg sensitivity to gamma irradiation decreased significantly in the 49–72 h age group; 60 Gy dose had no significant effect on egg hatch. Eggs irradiated few hours before hatch (at the blackhead stage), were the most resistant ones; 100 Gy had no significant effect on egg hatch and at 350 Gy dose over 56% of the eggs hatched. When adult emergence was used as a criterion for measuring effectiveness, however, the effect of gamma radiation was very sever. A dose of 60 Gy completely prevented adult emergence and at 100 Gy dose all resulted larvae died before pupation.  相似文献

As insects increase in radiotolerance as they develop and usually several developmental stages of the pest may be present in the fresh shipped commodity, it is important to know the radiation susceptibility of the stages of the target insect before the establishment of ionizing radiation quarantine treatments. This study was performed to determine the radiotolerance of eggs of the oriental fruit moth, Grapholita molesta (Busck) (Lepidoptera: Tortricidae), to gamma radiation. This species is considered as one of the most serious worldwide pests for temperate fruits, especially peaches. Eggs (12 h old) were exposed to 0 (control), 25, 35, 50, 75, 100, 125 and 150 Gy of gamma radiation. Surviving larvae were allowed to feed on an artificial diet. Three days after irradiation, it was verified that larvae’s cephalic capsules were significantly affected by gamma radiation, and the estimated mean LD90 and LD99 were 66.3 Gy and 125.8 Gy, respectively. Oriental fruit moth eggs revealed to be quite radiosensitive and very low doses as 50 Gy were sufficient to disrupt G. molesta embryogenesis. At 25 Gy, only male adults originated from the surviving larvae and, after mating with untreated fertile females, shown to be sterile.  相似文献

Although considered the most complete food and nutritionally shown to be part of a healthy diet, the egg is the source of many eating disorders, especially for infants. Irradiation has been used in studies not only as a means of microbiological control, but also on its structural action in the substances molecules and has been used to reduce the allergenic effects. The aim of this study was to evaluate the sensory effects of Co60 gamma radiation on proteins, enabling the acceptability of allergy food for genetically intolerant people. Eggs commercial fresh and freeze-dried and subjected to gamma irradiation by Co60 source at doses 0 (control), 10 kGy; 20 kGy and 30 kGy and rates of doses of 19.4 kGy/h and 31.8 kGy/h. Acceptability test was used by the hedonic scale, since it is necessary to know the “affective status” of consumers for the product, implying a preference, i.e. the most preferred samples are the most accepted and vice versa. The samples were presented as the habit of consumption (cooked) to a group of 41 adults panelists of both gender, aged from 21 to 40 years, and served under complete block design balanced with respect to the order of presentation. The evaluated attributes was flavor, appearance and overall acceptability. In general, for boiled eggs and freeze-dried, it was observed that the control sample was the most acceptable, followed by the sample irradiated with 10 kGy in both dose rates. In addition, panelists presented in testimony that they found interesting changes due to irradiation; also said they would not buy the product because of the marked change in appearance and smell, which at one point he ended up in disgust and detract from sales of the product, but they would buy irradiated with 10 kGy in both dose rate and dose of 20 kGy at a dose rate of 19.4 kGy/h.  相似文献

Samples of egg shell powder were annealed in air in the temperature range 298–1173 K. Samples were analyzed by X-ray diffraction (XRD), Fourier transform infrared (FTIR) spectroscopy, thermogravimetric analysis and electron spin resonance (ESR). ESR measurements were made on annealed egg shells before and after gamma irradiation at 4 kGy. Annealing egg shells produced a range of organic radical with g-values from g=2.005 to 2.002. The thermal dependence of this signal may provide valuable information about the extent of the thermal treatment that the sample was subjected to. Samples irradiated from 3Gy to 10 kGy were analyzed by ESR in order to obtain the response curve and to show ESR signal behavior.  相似文献

The yield increase of secondary metabolite production was examined in plant cell cultures with the use of relatively low to high doses gamma irradiation. Suspension culture of Lithospermum erythrorhizon cells was irradiated to 2, 16, and 32 Gy. The gamma irradiation significantly stimulated the shikonin biosynthesis of the cells and increased the total shikonin yields (intracellular+extracellular shikonin yields) by 400% at 16 Gy and by only 240% and 180% at 2 and 32 Gy, respectively. One of the key enzymes for the shikonin biosynthesis of cells, p-hydroxylbenzoic acid (PHB) geranyltransferase, was found to be stimulated by the gamma-radiation treatments. The activity of PHB geranyltransferase was increased at 2 and 16 Gy with a negligible change at 32 Gy. In contrast, the activity of PHB glucosyltransferase was slightly changed at all doses of gamma radiation compared with the control cells. Therefore, the increase in PHB geranyltransferase activity leads to the accumulation of secondary metabolites such as a shikonin, which may contribute to plant defense against the stresses induced by gamma irradiation.  相似文献

Wound debridement is essential for the removal of necrotic or nonviable tissue from the wound surface to create an environment conducive to healing. Nonsurgical enzymatic debridement is an attractive method due to its effectiveness and ease of use. Papain is a proteolytic enzyme derived from the fruit of Carica papaya and is capable of breaking down a variety of necrotic tissue substrates. The present study was focused on the use of gamma radiation for sterilization of papain dressing with wound debriding activity. Membranes with papain were prepared using 0.5% chitin in lithium chloride/dimethylacetamide solvent and sterilized by gamma radiation. Fluid absorption capacity of chitin–papain membranes without glycerol was 14.30±6.57% in 6 h. Incorporation of glycerol resulted in significant (p<0.001) increase in the absorption capacity. Moisture vapour transmission rate of the membranes was 4285.77±455.61 g/m2/24 h at 24 h. Gamma irradiation at 25 kGy was found suitable for sterilization of the dressings. Infrared (IR) spectral scanning has shown that papain was stable on gamma irradiation at 25–35 kGy. The irradiated chitin–papain membranes were impermeable to different bacterial strains and also exhibited strong bactericidal action against both Gram-positive and Gram-negative bacteria. The fluid handling characteristics and the antimicrobial properties of chitin–papain membranes sterilized by gamma radiation were found suitable for use as wound dressing with debriding activity.  相似文献

The ethanolic extracts of red beet (Beta vulgaris L.) hairy root were used to investigate the removal of color and improvement of biological activity for enhanced industrial applications. The extracts were exposed to gamma rays ranging from 2.5 to 30 kGy. The red beet hairy root is composed of two major red-colorants, betanin and isobetanin. Gamma ray radiation at 5 kGy remarkably reduced the levels of the major colorants by 94% and the reddish color was eliminated by doses greater than 10 kGy. Color removal was likely due to the gamma ray radiolysis of ethanol. Although details on the mechanism responsible for the decay of the chromophore have not been entirely determined, our results suggest that the free radicals that are produced during this process are capable of destroying the chromophore group in isobetanin, thus bleaching the substrate solution. In spite of the degradation of the major colorants, the biological activities of constituents of the extract such as DPPH radical scavenging and tyrosinase inhibition were negligibly affected by the gamma ray radiation up to 20 kGy. The antioxidant activity was 92.7% in control samples and 90.0–92.0% in irradiated samples (2.5–20 kGy), and a slight decrease to 87.5% was observed for gamma ray radiation at 30 kGy. In addition, tyrosinase inhibition activity has also the same pattern; the activity is slightly increased from 50.7% of control to 49.1–52.8% of irradiated samples (2.5–20 kGy) with a 46.8% at 30 kGy.  相似文献

The aim of this paper is to investigate structural changes of linear low density polyethylene (LLDPE) modified by ionizing radiation (gamma rays) in different atmospheres. The gamma radiation process for modification of commercial polymers is a widely applied technique to promote new physical–chemical and mechanical properties. Gamma irradiation originates free radicals which can induce chain scission or recombination, providing its annihilation, branching or crosslinking. This polymer was irradiated with gamma source of 60Co at doses of 5, 10, 20, 50 or 100 kGy at a dose rate of 5 kGy/h. The changes in molecular structure of LLDPE, after gamma irradiations were evaluated using thermogravimetric analyzer (TGA) and tensile machine and oscillatory rheology. The results showed the variations of the properties depending on the dose at each atmosphere.  相似文献

In this study, radiation stability of poly(bisphenol-a-epichlorohydrin) (PBEH) was studied via gamma irradiations at two different dose rates of 1540 and 82.8 Gy/h, in order to understand the possible use of PBEH in radioactive waste management as a solidifying agent. The total dose of irradiation was up to 2150 kGy. Degradation nature was tested by studying the changes in mechanical and thermal properties with the change of dose rate and total dose of irradiation. Tensile strength at yield was increased first then decreased when irradiated, while toughness decreased. The half value dose (HVD) of elongation was found as 29 kGy at dose rate of 1540 Gy/h. The non-irradiated PBEH was transparent, and the color changed to yellow with irradiation. Mechanical tests showed that PBEH has high radiation stability although there were some structural changes. It was seen that PBEH is a candidate polymer to be used in the immobilization of radioactive waste regarding radiation stability.  相似文献

The study was conducted to compare the radiation types of a gamma ray and an electron beam for the inhibition and reduction of a food allergy. OVA (2 mg/ml) were irradiated at 3, 5, 7 and 10 kGy. Patterns detected by the SDS-PAGE and an immunoblot showed that the intact OVA band disappeared and that it was dependant upon the radiation doses regardless of the radiation types. Binding abilities of the irradiated OVA against the monoclonal IgG and the egg allergic patients’ IgE decreased due to a conformational change of the epitope, but differences from using the two different radiation types were not observed. The results indicate that both the radiation types can be used for an inhibition and a reduction of a food allergy regardless of the radiation types.  相似文献

Thin films of isotactic polypropylene (iPP) are of great economical importance and their production is quite challenging due to the need of very fast uniaxial or biaxial expansion. During the expansion, critical problems usually arise, like structure disruption, shear thinning, causing material, energy and time losses. This work aims to study the surface morphology and compare the thermal, mechanical properties of PP films irradiated by gamma ray in an acetylene atmosphere after uniaxial expansion. PP films were made by compression molding at 190 °C with cooling in water at room temperature and irradiated by gamma ray, at (5, 12.5 and 20 kGy) under acetylene atmosphere. After irradiation the samples were submitted to thermal treatment at 90 °C for 1 h and then stretched out at 170 °C using an Instron machine. The surface of PP films, pristine and modified, (i.e., irradiated), was studied using optical microscopy (OM) and scanning electron microscopy (SEM). The changes in morphology, crystallinity and tensile parameters, like yield stress, rupture stress and elongation strain of the PP with irradiation dose were investigated. The results showed some evidences of gel formation due to crosslinking and/or long chain branching induced by radiation.  相似文献

In the present study, the effects of ionizing radiation of Cs-137 and Co-60 from 4.95 to 743.14 Gy and from 40 Gy to 300 kGy, respectively, on some bovine hemoglobin characteristics were studied. Such an effect was evaluated using electron paramagnetic resonance (EPR) spectroscopy, and infra-red (IR) spectroscopy. Bovine hemoglobin EPR spectra were recorded and analyzed before and after irradiation and changes were explained in detail. IR spectra of unirradiated and irradiated Bovine hemoglobin were recorded and analyzed also. It was found that ionizing radiation may lead to the increase of free radicals production, the decrease in α-helices contents, which reflects the degradation of hemoglobin molecular structure, or at least its incomplete performance. Results also show that the combined application of EPR and FTIR spectroscopy is a powerful tool for determining structural modification of bovine hemoglobin samples exposed to gamma irradiation.  相似文献

PP1084 protein was exposed to gamma irradiation ranging from 5 to 500 kGy. Native PAGE showed minor structural changes in PP1084 at 5 kGy, and major structural changes at >15 kGy. Size-exclusion chromatography (SEC) showed the formation of a new shoulder peak when the protein was irradiated with 15 and 30 kGy, and a double peak appeared at 100 kGy. The results of PAGE and SEC imply that PP1084 protein is degraded by gamma irradiation, with simultaneous oligomerization. PP1084 chaperone activity reached the highest level at 30 kGy of gamma irradiation, and then, decreased in a dose-dependent manner with increasing gamma irradiation. However, the peroxidase activity significantly decreased following exposure to all intensities of gamma irradiation. The improvement of chaperone activity using gamma irradiation might be promoted by the oligomeric structures containing covalently cross-linked amino acids. Consequently, PP1084 modification using gamma irradiation could elevate chaperone activity by about 3–4 folds compared to the non-irradiated protein.  相似文献

Chitosan (1 wt%, in 2% aqueous acetic acid solution) and starch (1 wt%, in deionised water) were dissolved and mixed in different proportions (20–80 wt% chitosan) then films were prepared by casting. Tensile strength and elongation at break of the 50% chitosan containing starch-based films were found to be 47 MPa and 16%, respectively. It was revealed that with the increase of chitosan in starch, the values of TS improved significantly. Monomer, 2-butane diol-diacrylate (BDDA) was added into the film forming solutions (50% starch-based), then casted films. The BDDA containing films were irradiated under gamma radiation (5–25 kGy) and it was found that strength of the films improved significantly. On the other hand, synthetic petroleum-based polymeric films (polycaprolactone, polyethylene and polypropylene) were prepared by compression moulding. Mechanical and barrier properties of the films were evaluated. The gamma irradiated (25 kGy) films showed higher strength and better barrier properties.  相似文献

Concentration of p-nonylphenols (NPs) in water at 1 μmol dm−3 was decreased exponentially with absorbed dose when NPs were irradiated by 60Co γ-rays. Two products having molecular weight of 236, presumably OH adducts of NPs, were detected by LC–MS analyses. The elimination of estrogen activity of aqueous NPs solution including such irradiation products at 5000 Gy (J kg−1) was confirmed by the yeast two-hybrid assay. These results should expand the application of ionizing radiation to the treatment of NPs.  相似文献

Gamma-ray irradiation is a very useful tool to improve the physicochemical properties of various biodegradable polymers without the use of a heating and crosslinking agent. The purpose of this study was to investigate the degradation behavior of poly (l-lactide-co-glycolide) (PLGA) depending on the applied gamma-ray irradiation doses. PLGA films prepared through a solvent casting method were irradiated with gamma radiation at various irradiation doses. The irradiation was performed using 60Co gamma-ray doses of 25–500 kGy at a dose rate of 10 kGy/h.The degradation of irradiated films was observed through the main chain scission. Exposure to gamma radiation dropped the average molecular weight (Mn and Mw), and weakened the mechanical strength. Thermograms of irradiated film show various changes of thermal properties in accordance with gamma-ray irradiation doses. Gamma-ray irradiation changes the morphology of the surface, and improves the wettability. In conclusion, gamma-ray irradiation will be a useful tool to control the rate of hydrolytic degradation of these PLGA films.  相似文献

Fresh-cut Iceberg lettuce packaged in modified atmosphere packages and spinach in perforated film bags were irradiated with gamma rays at doses of 0, 1, 2, 3, and 4 kGy. After irradiation, the samples were stored for 14 days at 4 °C. O2 levels in the packages of fresh-cut Iceberg lettuce decreased and CO2 levels increased with increasing radiation dose, suggesting that irradiation increased respiration rates of lettuce. Tissue browning of irradiated cut lettuce was less severe than that of non-irradiated, probably due to the lower O2 levels in the packages. However, samples irradiated at 3 and 4 kGy had lower maximum force and more severe sogginess than the non-irradiated control. In addition, ascorbic acid content of irradiated lettuce was 22–40% lower than the non-irradiated samples after 14 days of storage. The visual appearance of spinach was not affected by irradiation even at a dose of 4 kGy. Consumer acceptance suggested that more people would dislike and would not buy spinach that was treated at 3 and 4 kGy as compared to the non-irradiated sample. Overall, irradiation at doses of 1 and 2 kGy may be employed to enhance microbial safety of fresh-cut Iceberg lettuce and spinach while maintaining quality.  相似文献

The efficacy of gamma irradiation as a method of decontamination for food and herbal materials is well established. In the present study, Glycyrrhiza glabra roots were irradiated at doses 5, 10, 15, 20 and 25 kGy in a cobalt-60 irradiator. The irradiated and un-irradiated control samples were evaluated for phenolic contents, antimicrobial activities and DPPH scavenging properties. The result of the present study showed that radiation treatment up to 20 kGy does not affect the antifungal and antibacterial activity of the plant. While sample irradiated at 25 kGy does showed changes in the antibacterial activity against some selected pathogens. No significant differences in the phenolic contents were observed for control and samples irradiated at 5, 10 and 15 kGy radiation doses. However, phenolic contents increased in samples treated with 20 and 25 kGy doses. The DPPH scavenging activity significantly (p<0.05) increased in all irradiated samples of the plant.  相似文献

The commercially packed samples of raisins, dried figs and dried apricots were irradiated using doses in the range of 0.5–1.0 kGy for disinfestation and 0.5–5.0 kGy for sensory analysis with the dose rate ranging from 1.44 to 1.92 kGy/h. Pests on dried fruits were evaluated after 0, 1, 2 and 3 months of storage for irradiated dried figs and 1, 3, 6 and 12 months of storage for raisins and dried apricots. Sensory analysis of dried figs, dried apricots and raisins were carried out after 0, 1, 3, 6 and 12 months of storage. The results indicated that radiation processing at low doses, (∼1.0 kGy) is an effective post-harvest treatment and quarantine control for these products with no adverse effects on sensory (marketing) attributes.  相似文献

Poly(carbonate urethane) (PCU), is a valuable commercial engineering polymer. In order to understand the possible use of PCU in radioactive waste management as a solidifying agent or as a disposal container, radiation stability of the PCU is studied by Co-60 gamma irradiations at two different dose rates of 1540 and 82.8 Gy/h. The total dose of irradiation was up to 6.24 MGy. Degradation nature was tested by studying the changes in mechanical and thermal properties with rate and total dose of irradiation. Ultimate tensile strength and toughness first increased and then decreased with the irradiation dose. Half value dose (HVD) for elongation was 4010 kGy and for tensile strength 6010 kGy at the dose rate of 1540 Gy/h. The non-irradiated PCU transparent color changed to yellow and then brown with increased irradiation dose. The FTIR spectral analysis showed a random scission of polymer with irradiation. From the experimental observation, it was shown that PCU can be used for embedding radioactive waste for about 300 years.  相似文献
