Well-defined poly(epsilon-caprolactone) (PCL)/poly(N,N-dimethylamino-2-ethyl methacrylate (PDMAEMA) diblock copolymers were synthesized, and their self-assembly was investigated as micelles both in aqueous solutions and in thin solid deposits. The synthetic approach combines controlled ring opening polymerization (ROP) of epsilon-caprolactone (CL) and atom transfer radical polymerization (ATRP) of N,N-dimethylamino-2-ethyl methacrylate (DMAEMA). Diblock copolymers were prepared by ROP of CL initiated by (Al(OiPr)3), followed by quantitative reaction of the PCL hydroxy end-groups with bromoisobutyryl bromide. The alpha-isopropyloxy omega-2-bromoisobutyrate poly(epsilon-caprolactone) (PCL-Br) obtained was used as a macroinitiator for the ATRP of DMAEMA. The molecular characterization of those diblock copolymers was performed by 1H NMR spectroscopy and gel permeation chromatography (GPC) analysis. The self-assembly of the copolymers into micellar aggregates in aqueous media was followed with dynamic light scattering (DLS), as a function of concentration and the pH. In parallel, the morphology of the solid deposits of those micelles was examined with atomic force microscopy (AFM).  相似文献

Biomimetic star‐shaped poly(ε‐caprolactone)‐b‐poly(gluconamidoethyl methacrylate) block copolymers (SPCL‐PGAMA) were synthesized from the atom transfer radical polymerization (ATRP) of unprotected GAMA glycomonomer using a tetra(2‐bromo‐2‐methylpropionyl)‐terminated star‐shaped poly(ε‐caprolactone) (SPCL‐Br) as a macroinitiator in NMP solution at room temperature. The block length of PGAMA glycopolymer within as‐synthesized SPCL‐PGAMA copolymers could be adjusted linearly by controlling the molar ratio of GAMA glycomonomer to SPCL‐Br macroinitiator, and the molecular weight distribution was reasonably narrow. The degree of crystallization of PCL block within copolymers decreased with the increasing block length ratio of outer PGAMA to inner PCL. Moreover, the self‐assembly properties of the SPCL‐PGAMA copolymers were investigated by NMR, UV‐vis, DLS, and TEM, respectively. The self‐assembled glucose‐installed aggregates changed from spherical micelles to worm‐like aggregates, then to vesicles with the decreasing weight fraction of hydrophilic PGAMA block. Furthermore, the biomolecular binding of SPCL‐PGAMA with Concanavalin A (Con A) was studied by means of UV‐vis, fluorescence spectroscopy, and DLS, which demonstrated that these SPCL‐PGAMA copolymers had specific recognition with Con A. Consequently, this will not only provide biomimetic star‐shaped SPCL‐PGAMA block copolymers for targeted drug delivery, but also improve the compatibility and drug release properties of PCL‐based biomaterials for hydrophilic peptide drugs. © 2007 Wiley Periodicals, Inc. An effective strategy was developed to fabricate the supramolecular hydrogels with high mechanical strength and adjustable thermosensitivity in aqueous systems, in which physical hydrogel precursors were first formed by the inclusion complexation of Pluronic F68/poly(epsilon-caprolactone) block copolymer end-capped with acryloyl groups with alpha-cyclodextrin (alpha-CD) and subsequently in situ UV photo-cross-linking was carried out. In this way, strong supramolecular hydrogels with elastic moduli greater than 100 000 Pa could be created, which is an order of magnitude higher than that previously achieved with related supramolecular hydrogels. Moreover, the stimuli-responsive property of these hydrogels could be tailored by changing the molar feed ratio of alpha-CD to the macromer. By X-ray diffraction and thermogravimetric analyses, the polypseudorotaxane structure of the inclusion complexes in as-obtained hydrogels was confirmed.  相似文献

Biodegradable and biomimetic SPCL-PLAMA biohybrids were synthesized via ATRP and characterized by FT-IR, (1)H NMR, GPC and DSC. Biohybrids with small PDI were obtained, and the block length of the PLAMA glycopolymer could be varied linearly by the varying the molar ratio of glycomonomer to macroinitiator. The outer PLAMA glycopolymer restrained the crystallization of inner PCL segments. The self-assembly properties of amphiphilic biohybrids were studied. Lactose-installed aggregates were fabricated in aqueous solution; they changed from spherical micelles to vesicles with increasing weight fraction of hydrophobic PCL. The SPCL-PLAMA biohybrids showed specific recognition for RCA(120) lectin.  相似文献

Well-defined amphiphilic seven-arm star triblock copolymers containing hydrophobic crystalline poly(ε-caprolactone)(PCL) blocks, hydrophobic non-crystalline poly(tert-butyl acrylate)(PtBA) blocks and hydrophilic poly(ethylene glycol)(PEG) blocks were precisely synthesized by a combination of ring-opening polymerization(ROP), atom transfer radical polymerization(ATRP) and “click” reaction. Such star copolymers could self-assemble into “core-shell-corona” micelles and “multi-layer” vesicles depending on the fraction of each block. Meanwhile, the selective hydrolysis of middle PtBA blocks into the poly(acrylic acid)(PAA) blocks allowed the star block copolymers to further change their morphologies of aqueous aggregates in response to pH values.  相似文献

Novel, unique amphiphilic pentablock terpolymers consisting of the highly hydrophobic polyisobutylene (PIB) mid-segment attached to the hydrophilic combshaped poly(poly(ethylene glycol) methacrylate) (PPEGMA) polymacromonomer chains, which are coupled to poly(methyl methacrylate) (PMMA) outer segments were synthesized by the combination of quasiliving carbocationic polymerization and atom transfer radical polymerization (ATRP). First, a bifunctional PIB macroinitiator was prepared by quasiliving carbocationic polymerization and subsequent quantitative chain end derivatizations. Quasiliving ATRP of PEGMAs with different molecular weights (Mn = 188, 300 and 475 g/mol) led to triblock copolymers which were further reacted with MMA under ATRP conditions to obtain PMMA-PPEGMA-PIB-PPEGMA-PMMA ABCBA-type pentablock copolymers. It was found that slow initiation takes place between the PIB macroinitiator and PEGMA macromonomers with higher molecular weights via ATRP. ATRP of MMA with the resulting block copolymers composed of PIB and PPEGMA chain segments led to the desired block copolymers with high initiating efficiency. Investigations of the resulting pentablock copolymers by DSC, SAXS and phase mode AFM revealed that nanophase separation occurs in these new macromolecular structures with average domain distances of 11-14 nm, and local lamellar self-assembly takes place in the pentablocks with PPEGMA polymacromonomer segments of PEGMAs with Mn of 118 g/mol and 300 g/mol, while disordered nanophases are observed in the block copolymer with PEGMA having molecular weight of 475 g/mol. These new amphiphilic block copolymers composed of biocompatible chain segments can find applications in a variety of advanced fields.  相似文献

Carbohydrate residues are found on the extracellular side of the cell membrane. They form a protective coating on the outer surface of the cell and are involved in intercellular recognition. Synthetic carbohydrate-based polymers, so-called glycopolymers, are emerging as important well-defined tools for investigating carbohydrate-based biological processes and for simulating various functions of carbohydrates. In this work, the surface of a polypropylene microporous membrane (PPMM) was modified with comb-like glycopolymer brushes by a combination of UV-induced graft polymerization and surface-initiated atom-transfer radical polymerization (ATRP). 2-Hydroxyethyl methacrylate (HEMA) was first grafted to the PPMM surface under UV irradiation in the presence of benzophenone and ferric chloride. ATRP initiator was then coupled to the hydroxyl groups of poly(HEMA) brushes. Surface-initiated ATRP of a glycomonomer, D-gluconamidoethyl methacrylate, was followed at ambient temperature in aqueous solvent. Water had a significant acceleration effect on the ATRP process; however, loss of control over the polymerization process was also observed. The addition of CuBr2 to the ATRP system largely increased the controllability at the cost of the polymerization rate. The grafting of HEMA, the coupling of ATRP initiator to the hydroxyl groups, and the surface-initiated ATRP were confirmed by Fourier transform infrared spectroscopy and X-ray photoelectron spectroscopy.  相似文献

ABA triblock copolymers were synthesized using two polymerization techniques, polycondensation, and atom transfer radical polymerization (ATRP). A telechelic polymer was synthesized via polycondensation, which was then functionalized into a difunctional ATRP initiator. Under ATRP conditions, outer blocks were polymerized to form the ABA triblock copolymer. Six types of samples were prepared based on a poly(ether ether ketone) or poly(arylene ether sulfone) center block with either poly(methyl methacrylate), poly(pentafluorostyrene), or poly(ionic liquid) outer blocks. As polycondensation results in polymers with broad molecular weight distribution (MWD), the center of these triblock copolymers are disperse, while the outside blocks have narrow MWD due to the control afforded from ATRP. © 2014 Wiley Periodicals, Inc. Novel biocompatible, biodegradable, four‐arm star, triblock copolymers containing a hydrophobic poly(ε‐caprolactone) (PCL) segment, a hydrophilic poly(oligo(ethylene oxide)475 methacrylate) (POEOMA475) segment and a thermoresponsive poly(di(ethylene oxide) methyl ether methacrylate) (PMEO2MA) segment were synthesized by a combination of controlled ring‐opening polymerization (ROP) and atom transfer radical polymerization (ATRP). First, a four‐arm PCL macroinitiator [(PCL‐Br)4] for ATRP was synthesized by the ROP of ε‐caprolactone (CL) catalyzed by stannous octoate in the presence of pentaerythritol as the tetrafunctional initiator followed by esterification with 2‐bromoisobutyryl bromide. Then, sequential ATRP of oligo(ethylene oxide) methacrylate (OEOMA475, Mn = 475) and di(ethylene oxide) methyl ether methacrylate) (MEO2MA) were carried out using the (PCL‐Br)4 tetrafunctional macroinitiator, in different sequence, resulting in preparation of (PCL‐b‐POEOMA475‐b‐PMEO2MA)4 and (PCL‐b‐PMEO2MA‐b‐POEOMA475)4 star triblock copolymers. These amphiphilic copolymers can self‐assemble into spherical micelles in aqueous solution at room temperature. The thermal responses of the polymeric micelles were investigated by dynamic light scattering and ultraviolet spectrometer. The properties of the two series of copolymers are quite different and depend on the sequence distribution of each block along the arms of the star. The (PCL‐b‐POEOMA475‐b‐PMEO2MA)4 star copolymer, with the thermoresponsive PMEO2MA segment on the periphery, can undergo reversible sol‐gel transitions between room temperature (22 °C) and human body temperature (37 °C). © 2011 Wiley Periodicals, Inc. Novel amphiphilic star‐block copolymers, star poly(caprolactone)‐block‐poly[(2‐dimethylamino)ethyl methacrylate] and poly(caprolactone)‐block‐poly(methacrylic acid), with hyperbranched poly(2‐hydroxyethyl methacrylate) (PHEMA–OH) as a core moiety were synthesized and characterized. The star‐block copolymers were prepared by a combination of ring‐opening polymerization and atom transfer radical polymerization (ATRP). First, hyperbranched PHEMA–OH with 18 hydroxyl end groups on average was used as an initiator for the ring‐opening polymerization of ε‐caprolactone to produce PHEMA–PCL star homopolymers [PHEMA = poly(2‐hydroxyethyl methacrylate); PCL = poly(caprolactone)]. Next, the hydroxyl end groups of PHEMA–PCL were converted to 2‐bromoesters, and this gave rise to macroinitiator PHEMA–PCL–Br for ATRP. Then, 2‐dimethylaminoethyl methacrylate or tert‐butyl methacrylate was polymerized from the macroinitiators, and this afforded the star‐block copolymers PHEMA–PCL–PDMA [PDMA = poly(2‐dimethylaminoethyl methacrylate)] and PHEMA–PCL–PtBMA [PtBMA = poly(tert‐butyl methacrylate)]. Characterization by gel permeation chromatography and nuclear magnetic resonance confirmed the expected molecular structure. The hydrolysis of tert‐butyl ester groups of the poly(tert‐butyl methacrylate) blocks gave the star‐block copolymer PHEMA–PCL–PMAA [PMAA = poly(methacrylic acid)]. 用酶促开环聚合与ATRP方法相结合,制备了聚甲基丙烯酸六氟丁酯-聚己内酯-聚乙二醇-聚己内酯-聚甲基丙烯酸六氟丁酯(PHFMA-b-PCL-b-PEG-b-PCL-b-PHFMA)五嵌段聚合物.首先用Novozym e 435作为催化剂合成了聚己内酯-聚乙二醇-聚己内酯三嵌段聚合物,然后通过端基官能化法合成了大分子引发剂,并用其引发甲基丙烯酸六氟丁酯(HFMA)的ATRP反应,合成了五嵌段聚合物.通过核磁和GPC证明了大分子引发剂和五嵌段共聚物的结构,五嵌段共聚物的GPC分析表明这种合成方法的可行.共聚物胶束的直径和大小通过动态光散射方法和原子力显微镜测试,五嵌段共聚物在水中的的自组装行为也被研究.结果证明胶束是球形,其平均直径为77 nm.聚合物在四氢呋喃中的浓度对聚合物的聚集形貌有很大的影响.  相似文献

A novel class of biomimetic glycopolymer–polypeptide triblock copolymers [poly(L ‐glutamate)–poly(2‐acryloyloxyethyllactoside)–poly(L ‐glutamate)] was synthesized by the sequential atom transfer radical polymerization of a protected lactose‐based glycomonomer and the ring‐opening polymerization of β‐benzyl‐L ‐glutamate N‐carboxyanhydride. Gel permeation chromatography and nuclear magnetic resonance analyses demonstrated that triblock copolymers with defined architectures, controlled molecular weights, and low polydispersities were successfully obtained. Fourier transform infrared spectroscopy of the triblock copolymers revealed that the α‐helix/β‐sheet ratio increased with the poly(benzyl‐L ‐glutamate) block length. Furthermore, the water‐soluble triblock copolymers self‐assembled into lactose‐installed polymeric aggregates; this was investigated with the hydrophobic dye solubilization method and ultraviolet–visible analysis. Notably, this kind of aggregate may be useful as an artificial polyvalent ligand in the investigation of carbohydrate–protein recognition and for the design of site‐specific drug‐delivery systems. © 2004 Wiley Periodicals, Inc. Libraries of siloxane-polyurethane coatings were designed, formulated, and screened using high-throughput experimentation. Four independent variables that were analyzed were the molecular weight of poly(dimethylsiloxane) (PDMS), presence or absence of poly(epsilon-caprolactone) (PCL) blocks attached to the PDMS backbone, the length of the PCL blocks, and the siloxane polymer level in the coating formulations. In addition to the siloxane libraries (3-aminopropyl-terminated PDMS and poly(epsilon-caprolactone)-poly(dimethylsiloxane)-poly(epsilon-caprolactone) (PCL-PDMS-PCL) triblock copolymers), the coating formulation included a trifunctional isocyanate crosslinker, trifunctional poly(epsilon-caprolactone) polyol, 2,4-pentanedione (pot-life extender), dibutyltin diacetate (catalyst), and a blend of solvents. The resulting coatings were analyzed for their surface energy and pseudobarnacle adhesion both before and after aging the coatings for 30 days in water. The water and methylene iodide contact angle averages increase with increasing molecular weight of PDMS. Coatings prepared from PCL-PDMS-PCL triblock copolymers have lower surface energies than coatings prepared from 3-aminopropyl-terminated PDMS; however, lower pseudobarnacle adhesion results were obtained for the coatings prepared from 3-aminopropyl-terminated PDMS than coatings prepared from PCL-PDMS-PCL triblock copolymers. The siloxane polymer level in the coating formulations does not have a significant effect on the surface energy of the coatings, but it resulted in higher pseudobarnacle adhesion.  相似文献

Amphiphilic diblock copolymer polycaprolactone‐block‐poly(glycidyl methacrylate) (PCL‐b‐PGMA) was synthesized via enzymatic ring‐opening polymerization (eROP) and atom transfer radical polymerization (ATRP). Methanol first initiated eROP of ?‐caprolactone (?‐CL) in the presence of biocatalyst Novozyme‐435 under anhydrous conditions. The resulting monohydroxyl‐terminated polycaprolactone (PCL–OH) was subsequently converted to a bromine‐ended macroinitiator (PCL–Br) for ATRP by esterification with α‐bromopropionyl bromide. PCL‐b‐PGMA diblock copolymers were synthesized in a subsequent ATRP of glycidyl methacrylate (GMA). A kinetic analysis of ATRP indicated a living/controlled radical process. The macromolecular structures were characterized for PCL–OH, PCL–Br, and the block copolymers by means of nuclear magnetic resonance, gel permeation chromatography, and infrared spectroscopy. Differential scanning calorimetry and wide‐angle X‐ray diffraction analyses indicated that the copolymer composition (?‐CL/GMA) had a great influence on the thermal properties. Atom transfer radical polymerization (ATRP) and ring‐opening polymerization (ROP) were combined to synthesize poly(?‐caprolactone‐co‐octadecyl methacrylate‐co‐dimethylaminoethyl methacrylate) copolymers possessing a triblock or random block structure. Various synthetic pathways (sequential or simultaneous approaches) were investigated for the synthesis of both copolymers. For the preparation of these copolymers, an initiator with dual functionality for ATRP/anionic ring‐opening polymerization, 2‐hydroxyethyl 2‐bromoisobutyrate, was used. Copolymers were prepared with good structural control and low polydispersities (weight‐average molecular weight/number‐average molecular weight < 1.2), but one limitation was identified: the dimethylaminoethyl methacrylate (DMAEMA) block had to be synthesized after the ?‐caprolactone block. ROP could not proceed in the presence of DMAEMA because the complexation of the amine groups in poly(dimethylaminoethyl methacrylate) deactivated tin(II) hexanoate, which was used as a catalyst for ROP. © 2005 Wiley Periodicals, Inc. A series of novel side‐chain liquid crystalline ABC triblock copolymers composed of poly(ethylene oxide) (PEO), polystyrene (PS), and poly[6‐(4‐methoxy‐4′‐oxy‐azobenzene) hexyl methacrylate] (PMMAZO) were synthesized by atom transfer radical polymerization (ATRP) using CuBr/1,1,4,7,7‐pentamethyldiethylenetriamine (PMDETA) as a catalyst system. First, the bromine‐terminated diblock copolymer poly(ethylene oxide)‐block‐polystyrene (PEO‐PS‐Br) was prepared by the ATRP of styrene initiated with the macro‐initiator PEO‐Br, which was obtained from the esterification of PEO and 2‐bromo‐2‐methylpropionyl bromide. An azobenzene‐containing block of PMMAZO with different molecular weights was then introduced into the diblock copolymer by a second ATRP to synthesize the novel side‐chain liquid crystalline ABC triblock copolymer poly(ethylene oxide)‐block‐polystyrene‐block‐poly[6‐(4‐methoxy‐4′‐oxy‐azobenzene) hexyl methacrylate] (PEO‐PS‐PMMAZO). These block copolymers were characterized using proton nuclear magnetic resonance (1H NMR) and gel permeation chromatograph (GPC). Their thermotropic phase behaviors were investigated using differential scanning calorimetry (DSC) and polarized optical microscope (POM). These triblock copolymers exhibited a smectic phase and a nematic phase over a relatively wide temperature range. The synthesis of ABA triblock copolymers of the type PDMAEMA-PCL-PDMAEMA was achieved by atom transfer radical polymerization (ATRP) of DMAEMA using difunctional polycaprolactone (PCL) as macroinitiator. First, ring-opening polymerization (ROP) of ε-caprolactone (ε-CL) was carried out in the presence of 1,2-diaminoethane/tin (II) octanoate. Dihydroxy PCL thus obtained was end-functionalized in a quantitative manner using 2-bromoisobutyryl bromide. The resulting Br-PCL-Br was used as macroinitiator in the ATRP of DMAEMA leading to triblock copolymers with PCL as the central block and PDMAEMA sequences of different lengths. NMR and SEC analyses confirmed the formation of ABA triblocks.  相似文献

Novel amphiphilic eight‐arm star triblock copolymers, star poly(ε‐caprolactone)‐block‐poly(acrylic acid)‐block‐poly(ε‐caprolactone)s (SPCL‐PAA‐PCL) with resorcinarene as core moiety were prepared by combination of ROP, ATRP, and “click” reaction strategy. First, the hydroxyl end groups of the predefined eight‐arm SPCLs synthesized by ROP were converted to 2‐bromoesters which permitted ATRP of tert‐butyl acrylate (tBA) to form star diblock copolymers: SPCL‐PtBA. Next, the bromide end groups of SPCL‐PtBA were quantitatively converted to terminal azides by NaN3, which were combined with presynthesized alkyne‐terminated poly(ε‐caprolactone) (A‐PCL) in the presence of Cu(I)/N,N,N,N,N″‐pentamethyldiethylenetriamine in DMF to give the star triblock copolymers: SPCL‐PtBA‐PCL. 1H NMR, FTIR, and SEC analyses confirmed the expected star triblock architecture. The hydrolysis of tert‐butyl ester groups of the poly(tert‐butyl acrylate) blocks gave the amphiphilic star triblock copolymers: SPCL‐PAA‐PCL. These amphiphilic star triblock copolymers could self‐assemble into spherical micelles in aqueous solution with the particle size ranging from 20 to 60 nm. A series of well‐defined amphiphilic triblock copolymers, poly(ethylene glycol)‐b‐poly(tert‐butyl acrylate)‐b‐poly(2‐hydroxyethyl methacrylate) (PEG‐b‐PtBA‐b‐PHEMA), were synthesized via successive atom transfer radical polymerization (ATRP). ATRP of tBA was first initiated by PEG‐Br macroinitiator using CuBr/N,N,N′,N″,N′″‐pentamethyldiethylenetriamine as catalytic system to give PEG‐b‐PtBA diblock copolymer. This copolymer was then used as macroinitiator to initiate ATRP of HEMA, which afforded the target triblock copolymer, PEG‐b‐PtBA‐b‐PHEMA. The critical micelle concentrations of obtained amphiphilic triblock copolymers were determined by fluorescence spectroscopy using N‐phenyl‐1‐naphthylamine as probe. The morphology and size of formed aggregates were investigated by transmission electron microscopy and dynamic light scattering, respectively. Finally, an acid‐sensitive PEG‐b‐PtBA‐b‐P(HEMA‐CAD) prodrug via cis‐aconityl linkage between doxorubicin and hydroxyls of triblock copolymers with a high drug loading content up to 38%, was prepared to preliminarily explore the application of triblock copolymer in drug delivery. © 2012 Wiley Periodicals, Inc. Inclusion complexes (ICs) between alpha-cyclodextrin (alpha-CD) and three kinds of biodegradable aliphatic polyesters with different sequence lengths of the monomeric repeating units poly(3-hydroxypropionate) [P(3HP)], poly(4-hydroxybutyrate) [P(4HB)] and poly(epsilon-caprolactone)(PCL) were prepared by mixing a solution of alpha-CD with that of the polymer, followed by stirring. The ICs were obtained as insoluble precipitates and characterized by FT-IR, WAXD and DSC. All measurements showed that the polymer chains of all three kinds of polyester were included into the alpha-CD cavity and formed ICs with different stoichiometries. WAXD patterns and thermal analysis indicated that these ICs possessed a channel structure and the crystallization of the polyester chains was suppressed upon inclusion into the alpha-CD cavity.  相似文献
