A simple and reliable method for the preparation of 2,2-dimethyl-5-(trifluoroacetyl)-1,3-dioxane-4,6-dione and 2,2-dimethyl-6-(trifluoromethyl)-4H-1,3-dioxin-4-one on a multigram scale was developed. These (trifluoroacetyl)ketene precursors were used in the hetero-Diels-Alder reaction with dialkylcyanamides and 1-ethoxyprop-1-yne, as well as in some reactions with nucleophiles.  相似文献

The stereoselective properties of modified thiourea organocatalysts were tested in the Friedel–Crafts alkylation of indole with 5-arylidene-2,2-dimethyl-1,3-dioxane-4,6-diones, which produces chiral 5-((1H-indol-3-yl)(aryl)methyl)-2,2-dimethyl-1,3-dioxane-4,6-diones. Based on a tentative reaction mechanism for ((S)-N-benzyl-2-(3-(3,5-bis(trifluoromethyl)phenyl)thioureido)-N,3,3-trimethylbutanamide organocatalysts, modifications were applied in four selected regions. Systematic structure-stereoselectivity relationship study allowed designing the best efficient organocatalyst for the investigated Friedel–Crafts alkylation of indole with 5-arylidene-2,2-dimethyl-1,3-dioxane-4,6-diones.  相似文献

By reaction of 4-substituted 5-amino-6-mercaptopyrimidines with 5-bromo-2,2-dimethyl-4,6-dioxo-1,3-dioxane, we have obtained 4′-substituted 5-(5-amino-6-pyrimidylthio)-2,2-dimethyl-4,6-dioxo-1,3-dioxanes. We have studied diazotization of these compounds by isoamyl nitrite. In the case of 4′-methoxy- and 4′-dimethylamino-substituted derivatives, we have obtained derivatives of novel heterocyclic systems: pyrimido[5,4-e][1,3,4]thiadiazine and pyrimido[5,4-e][1,3,4]thiadiazine-7-spiro-5′-1,3-dioxane, and in the case of the 4′-isopropylamino-substituted derivative we obtained 4-isopropyl-7-(2,2-dimethyl-4,6-dioxo-1,3-dioxan-5-ylidene)-1,2,3-triazolo[5,4-d]pyrimidin-7-ylidene.__________Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 4, pp. 613–623, April, 2005.  相似文献

Depending on the conditions of carrying out the reaction of dimethylformamide diethylacetal with the Meldrum's acid, either 2,2-dimethyl-4,6-dioxo-5-(N,N-dimethyl-aminomethylene)-1,3-dioxane or N,N,N1N1-tetramethylformamidinium salt of 2,2-dimethyl-4,6-dioxo-5-(2,2-dimethyl-4,6-dioxo-1,3-dioxan-5-yl)methylene-1,3-di-oxane are formed. We have obtained previously unknown 1-methyl- and 1,3-dimethyl-5-diethylaminothiocarbonylthiobarbituric acids by reaction of sodium diethyldithiocarbamate with 5-phenyliodonium betaines of 1-methyl- and 1,3-dimethylbarbituric acids. Cyclization of these compounds upon heating in conc. H2SO4 gives methyl-substituted 5,7-dioxo(4H,6H)-1,3-dithiolo[4,5-d]pyrimidine-2-diethylimmonium hydrosulfates; the derivative of 1-methylbarbituric acid forms a mixture of 4-methyl- and 6-methyl-substituted compounds (21). We isolated perchlorates of 4-methyl- and 4,6-dimethyl-substituted derivatives in pure form. By treatment of the immonium salt with sodium sulfide or selenide, we obtained 4-methyl- and 4,6-dimethyl-5,7-dioxo(4H,6H)-1,3-dithiolo[4,5-d]pyrimidine-2-thiones and 4-methyl-, 6-methyl-, and 4,6-dimethyl-2-selenones. We characterized the isomeric 4- and 6-methyl-substituted selenones by electronic absorption spectra and ionization constants (7.65 and 4.0). The differences in the pK values and in the electronic absorption spectra makes it possible to distinguish the substitution site in N-mono-substituted derivatives of 5,7-dioxo(4H,6H)-1,3-dithiolo[4,5-d]pyrimidine.Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 11, pp. 1526–1533, November, 1993.  相似文献

6-(2-Iodoethyl)-2,2-dimethyl-4H-1,3-dioxin has been prepared in five steps from ethyl acetoacetate. A variety of enolates were then alkylated with this iodide. The resulting 6-alkyl-4H-1,3-dioxins were either subjected to further transformations and/or heated (or subjected to a Lewis acid) to effect facile retrocycloaddition reactions. The resulting enones were found to smoothly participate in conjugate addition or olefin metathesis reactions to provide various carbocyclic ring systems. Collectively, these examples document the synthetic equivalency of dioxin 2 with iodoethyl vinyl ketone and, moreover, delineate a strategy for accomplishing the sequential reactions with nucleophiles at the β′, followed by the β electrophilic sites.  相似文献

The reaction of sodium diphenylamide with 2,2-dimethyl-4,5-bis(tosyloxymethyl)-1,3-dioxolane gave (+)-(4S,5S)-2,2-dimethyl-4,5-bis(diphenylaminomethyl)-1,3-dioxolane, which was brought into complex formation with cobalt chloride. Treatment of 2,2-dimethyl-4,5-bis(tosyloxymethyl)-1,3-dioxolane with sodium N-methylanilide resulted in cleavage of the SÄO bond in the p-toluenesulfonate moiety with formation of N-methyl-N-phenyl-p-toluenesulfonamide and 4,5-bis(hydroxymethyl)-2,2-dimethyl-1,3-dioxolane disodium salt. Diethyl (4R,5R)-2,2-dimethyl-1,3-dioxolane-4,5-dicarboxylate reacted with methylamine to give the corresponding dicarboxamide which was reduced with lithium aluminum hydride to (4S,5S)-2,2-dimethyl-4,5-bis(methylaminomethyl)-1,3-dioxolane having chiral carbon and nitrogen atoms.  相似文献

Six optical isomers of 2-(4-diphenylmethyl-1-piperazinyl)ethyl 5-(4,6-dimethyl-2-oxo-1,3,2-dioxaphosphorinan-2-yl)-1,4-dihydro-2, 6-dimethyl-4-(3-nitrophenyl)-3-pyridinecarboxylate dihydrochloride (NIP-101, 1.2HCl.2H2O), a potent calcium antagonist, were successfully prepared by using optically active (2R,4R)-(-)- and (2S,4S)-(+)-2,4-pentanediols, and cis-2,4-pentanediol and optically active (S)-(+)-2-methoxy-2-phenylethanol. Their proton nuclear magnetic resonance investigations demonstrate that the 1,3,2-dioxaphosphorinane group is conformationally constrained around the C-P bond. Calcium-antagonistic and hypotensive activities of the optical isomers were examined and found to depend mainly on the absolute configuration at a stereogenic center in the 1,4-dihydropyridine ring rather than the configuration of the 1,3,2-dioxaphosphorinane moiety.  相似文献

A variety of 2,2-dimethyl-6-(2-oxoalkyl)-1,3-dioxin-4-ones 5a-l and the corresponding 6-substituted 4-hydroxy-2-pyrones 3a-l were prepared in high yields under mild reaction conditions by the reaction of 2,2,6-trimethyl-1,3-dioxin-4-one 4 with 1-acylbenzotriazoles 9 in the presence of LDA followed by thermal cyclization of 5a-l to 3a-l. Synthesis of novel 6-(1-benzoylalkyl)-2,2-dimethyl-1,3-dioxin-4-ones 12a-c was achieved by alkylation of dioxinone 5a and their subsequent cyclization gave 5-alkyl-4-hydroxy-2-pyrones 13a-c.  相似文献

N,N-Dimethylazidochloromethyleniminium chloride (azidophosgeniminium chloride) ( 1 ) reacts by diazo group transfer with 1,3-dimethyl-4-aryl-(and alkyl)aminouracils 4 to give, under mild “one pot” reaction conditions, a very good yield of 3-aryl-(and 3-alkyl)-4,6-dimethyl-5,7-dioxo-1,2,3-triazolo[4,5-d]pyrimidines (8-azatheophyllines) 8 . The asymmetric transfer hydrogenation of a series of 2,2-dimethyl-6-(2-oxoalkyl)-1,3-dioxin-4-ones and 2,2-dimethyl-6-(2-oxoaryl)-1,3-dioxin-4-ones was achieved in high enantiomeric excess using a Ru(II) catalyst. A theoretical study is employed to describe the orbital interactions involved in the conformers' stability, the energies for the stereoelectronic interactions, and the corresponding effects of these interactions on the molecular structure (bond lengths) for cis- and trans-4,6-disubstituted-2,2-dimethyl-1,3-dioxanes. For cis-4,6-disubstituted-2,2-dimethyl-1,3-dioxanes, two LPO --> sigma*C(2)-Me(8) interactions are extremely important and the energies involved in these interactions are in the range 6.81-7.58 kcal mol(-1) for the LP(O)(1) --> sigma*C(2)-Me(8) and 7.58-7.71 kcal mol(-1) for the LP(O)(3) --> sigma*C(2)-Me(8) interaction. These two LP(O) --> sigma*C(2)-Me(8) interactions cause an upfield shift, indicating an increased shielding (increased electron density) of the ketal carbon C(2) as well as the axial Me(8) group in the chair conformation. These LP(O) --> sigma*C(2)-Me(8) hyperconjugative anomeric type interactions can explain the 13C NMR chemical shifts at 19 ppm for the axial methyl group "Me(8)" and 98.5 ppm for the ketal carbon "C(2)". The observed results for the trans derivatives showed that for compounds 2a-c (R = -CN, -C[triple bond]CH, and -CHO, respectively) the chair conformation is predominant, whereas for 2d,f-h [-CH3, -Ph, -C6H4(p-NO2), -C6H4(p-OCH3), respectively] the twist-boat is the most stable compound and for 2e [-C(CH3)3] is the only form.  相似文献

The synthesis of the first completely characterized transition-metal complex containing a sulfur-bound 4,6-dimethyldibenzothiophene (4,6-Me(2)DBT) ligand, [CpRu(CO)(2)(eta(1)(S)-4,6-Me(2)DBT)]BF(4) (1) (Cp = eta(5)-C(5)Me(5)), is reported. X-ray studies of 1 and its 4-methyldibenzothiophene and dibenzothiophene analogues, [CpRu(CO)(2)(eta(1)(S)-4-MeDBT)]BF(4) (2) and [CpRu(CO)(2)(eta(1)(S)-DBT)]BF(4) (3), show that the Ru-S bond distances increase in the order, 3 < 2 < 1. Equilibrium studies on the series of [CpRu(CO)(2)(eta(1)(S)-DBTh)](+) compounds, where DBTh = DBT, 4-MeDBT, 4,6-Me(2)DBT, and 2,8-Me(2)DBT, show that the relative binding strengths of the dibenzothiophene ligands increase in the order 4,6-Me(2)DBT (1) < 4-MeDBT (20.2(1)) < DBT (62.7(6)) < 2,8-Me(2)DBT (223(3)). These results are the first to quantify the steric effect of 4- and 6-methyl groups on the sulfur-coordinating ability of dibenzothiophenes to transition-metal centers. They are also consistent with the proposal that 4- and 6-methyl groups reduce the coordination of dibenzothiophenes to active metal sites on hydrodesulfurization catalysts, which could account for the slow rate of 4-MeDBT and 4,6-Me(2)DBT hydrodesulfurization in petroleum feedstocks.  相似文献
