The chiral isoquinuclidine derivative, 2-azabicyclo[2.2.2]octane ring system, endo-(7R)-3 was obtained in good yield with excellent diastereoselectivity (up to 92% de) by Diels–Alder reaction of 1-(phenoxycarbonyl)-1,2-dihydropyridine 1 with N-acryloyl-(4S)-4-benzyloxazolidin-2-one (4S)-2 using titanium-(2R,3R)-TADDOLate 4 as a chiral Lewis acid catalyst in toluene at 0 °C. On the other hand, endo-(7S)-3 was obtained in good yield with excellent diastereoselectivity (up to 97% de) by Diels–Alder reaction of 1 with (4R)-2 using Cu(OTf)2/(4S,4′S)-bis(oxazoline) catalyst 8 as a chiral Lewis acid catalyst in dichloromethane at 0 °C. In these reactions, the choice of solvent and the combination of titanium-(2R,3R)-TADDOLate 4 {or Cu(II)/(4S,4′S)-bis(oxazoline) 8} and dienophile (4S)-2 {or (4R)-2} are very important. The stereochemistry of endo-(7R)-3 has been established to be (1R,4S,7R) and the reaction mechanism is proposed.  相似文献

The reaction of 2-lithiophenyldiphenylphosphine with phosphorus trichloride afforded the new unsymmetric phosphine, dichloro(2-diphenylphosphinophenyl)phosphine (4). Condensation of 4 with (a) (2R,3R)-dimethyl tartrate or (b) (S)-binaphthol in the presence of triethylamine gave new chiral phosphine-phosphonite ligands, (2R,3R)-[2-(2′-(diphenylphosphino)phenyl)-4,5-bis(carbomethoxy)-1,3,2-dioxaphospholane] ((2R,3R)-5) and (S)-[2-(diphenylphosphino)benzene][1,1′-binaphthalen-2,2′-diyl]phosphonite] ((S)-6). The analogous reaction of 4 with (1R,2S)-ephedrine using N-methylmorpholine as the base, gave [2-(2′-(diphenylphosphino)phenyl)-3,4-dimethyl-5-phenyl-1,3,2-oxazaphospholidine] (7) as a 95:5 mixture of diastereoisomers.  相似文献

In the present study, the synthesis and liquid phase extraction properties towards some amino acid methylesters and amino alcohols of Schiff base and amino alcohol substituted calix[4]arene are reported. The Schiff base substituted calix[4]arene 5 has been synthesized via condensation reaction involving 5,17-diformyl-11,23-di-tert-butyl-25,27-di[3-(4-formylphenoxy)propoxy]-26,28 dihydroxycalix[4]arene 4 and (R)-(?)-2-phenylglycine methyl ester in CHCl3:MeOH. To give the amino alcohol substituted calix[4]arene 6, the synthesized chiral compound 5 was reduced by LiAlH4. The new chiral Schiff base and amino alcohol derivatives of calix[4]arene have been characterized by a combination of FT-IR, 1H NMR, 13C NMR, FAB-MS and elemental analysis. Also, the extraction behaviors of 5 and 6 towards some selected amino acid methylesters and amino alcohols have been studied by liquid–liquid extraction.  相似文献

The synthesis of chiral diamide derivatives of calix[4]arene from the reaction of p-tert-butylcalix[4]arene diester 1a and calix[4]arene diester 1b with various amino alcohols were reported. The 1H and 13C NMR, data showed that the compounds synthesized exist in the cone conformation. A stereoselective synthesis of 1a {N-[(1R)-2-hydroxy-1-methoxy-methyl ethyl]-(4E,7S)-7-methoxy-4-eicosenamide} has been accomplished in 10 steps from 1-tetradecanol for the first time in 28% overall yield. The key steps involved the coupling reaction of a chiral alkyne with a protected bromide in the presence of t-BuLi, as well as the amidation reaction of (4E,7S)-7-methoxyeicos-4-enoic acid with (R)-methoxyamino alcohol. Acetylation of 1a finished the preparation of 1b {N-[(1S)-2-acetyloxy-1-methoxy-methyl ethyl]-(4E,7S)-7-methoxy-4-eicosenamide}. Their 1′-epi-isomers have also been synthesized with a similar strategy.  相似文献

Herein the synthesis and recognition abilities towards amino acids and amino alcohols of new d-/l-phenylalaninol substituted p-tert-butylcalix[6]arenas are reported. These compounds, 6 and 7 have been synthesized via nucleophilic substitution reactions involving 5,11,17,23,29,35-tert-butyl-37,38-dimethoxy-39,40,41,42-(p-tosylethoxy)calix[6]arene 5 with d-/l-phenylalaninol in dry THF. The extraction properties of 6 and 7 towards some selected amino acid methylesters and amino alcohols have been studied by liquid–liquid extraction. These results show that chiral calix[6]arene derivatives exhibit a good affinity towards all amino acids and amino alcohols.  相似文献

The synthesis of some chiral bis-(aminol)ethers are described. Reaction of a solution of the resorcin[4]arene derived from propanal with N,N-bis(methoxymethyl)-N-(S)-(−)-α-methylbenzylamine in toluene at 85 °C initially afforded a 1:1 mixture of two diastereoisomeric tetrakis(benzoxazines). Further, heating of this mixture under reflux in ethanol for 24 h afforded the crystalline (αS),(S)-diastereoisomer in 77% yield. N,N-bis(ethoxymethyl)-N-(S)-(−)-α-methylbenzylamine and N,N-bis(ethoxymethyl)-N-(R)-(+)-α-methylbenzylamine were reacted with β keto esters to afford a 1:1 mixture of the diastereoisomeric double Mannich adducts. Two of the double Mannich adducts were converted into tricyclic ABE analogues of the alkaloid methyllycaconitine 1.  相似文献

A chiral benzodiazepine derivative 1 was synthesized starting from o-nitrobenzoyl chloride and methyl l-prolinate hydrochloride. Diastereomeric (1R,2R,1′S)-(+)-2-[N-methyl-N-(α-phenylethyl)amino]cyclohexanol 3a and (1S,2S,1′S)-(+)-2-[N-methyl-N-(α-phenylethyl)amino]cyclohexanol 3b were synthesized starting from (S)-α-phenylethylamine and cyclohexene oxide via ring-opening, diastereomer separation and N-methylation. (S,S)-octahydrodipyrrolo[1,2-a:1′,2′-d]pyrazin 5 was synthesized from methyl l-prolinate. Chiral tertiary amines 1, 3a, 3b and 5 almost cannot catalyze the Baylis-Hillman reaction between o-nitrobenzaldehyde and methyl vinyl ketone (MVK). However, they functioned as efficient catalysts for this reaction in the presence of l-proline. The corresponding adducts were obtained in good yields with enantioselectivity of 83% ee, 81% ee, 51% ee and 66% ee, respectively.  相似文献

Racemic 2,2′-bis[di(p-tolyl)stibano]-1,1′-binaphthyl (BINASb) (±)-2 has been prepared from 2,2′-dibromo-1,1′-binaphthyl 1 via 2,2′-dilithio-1,1′-binaphthyl intermediate, and has been resolved via the separation of a mixture of the diastereomeric Pd complexes 4A and 4B, derived from the reaction of (±)-2 with di-μ-chlorobis{(S)-2-[1-(dimethylamino)ethyl]phenyl-C,N}dipalladium(II) 3. The optically active BINASbs (S)-(+)-2 and (R)-(−)-2 have been shown to be effective chiral ligands for the rhodium-catalyzed asymmetric hydrosilylation of ketones.  相似文献

Inherently chiral anti-O,O′-dibenzyl-p-tert-butylcalix[4]arene 1 was resolved as the (S)-2-methoxy-2-(naphthalen-1-yl)propionic ester by flash chromatography. Conversely, the anti-O,O′-dibutyl analogue 2 was resolved as the (Sa)-2′-methoxy-1,1′-binaphthalene-2-carboxylic ester by crystallization combined with flash chromatography. CD analysis of these compounds indicated the absolute stereochemistries to be (Sa)-(+)-1 and (Sa)-(+)-2, respectively, the former of which was confirmed by X-ray crystallographic analysis.  相似文献

A new inherently chiral calix[4]arene ICC 1 has been disclosed. The dissymmetry of 1 is generated from a chirality plane in the quinol moiety of a 1,3-bridged bicyclic calix[4]arene. ICC 1 has been resolved by enantioselective HPLC, and the chiroptical properties of both isolated antipodes (pS)-1 and (pR)-1 confirm their enantiomeric nature. The absolute configuration of the (pS)-1/(pR)-1 enantiomeric pair was established through time-dependent density functional theory (TDDFT) calculations of electronic circular dichroism (CD) spectra.  相似文献

The synthesis of two new calixarene derivatives 4 and 5, functionalized at the lower rim with 4-amino-1-benzylpiperidine to give diamide and diamine derivatives of p-tert-butylcalix[4]arene, is described. They were obtained by the reaction of both the diester derivative of p-tert-butylcalix[4]arene (2) and the dialkyl bromide derivative of p-tert-butylcalix[4]arene (3) with 4-amino-1-benzylpiperidine. The 1H NMR spectra of calixarene derivatives show that 4 and 5 exist in the cone conformation. Moreover, these diamide and diamine derivatives of p-tert-butylcalix[4]arene (4 and 5) have been immobilized onto [3-(2,3-epoxypropoxy)-propyl]-trimethoxysilane-modified Fe3O4 magnetite nanoparticles to obtain calixarene-based magnetic nanoparticles M-DADBP-Calix (6) and M-DABP-Calix (7). The calix[4]arene immobilized materials were characterized by a combination of Fourier Transform Infrared Spectroscopy (FTIR), Transmission Electron Microscopy (TEM) and Thermogravimetric Analyses (TGA) and elemental analysis. Additionally, the studies regarding the removal of As(V)/Cr(VI) ions as well as U(VI) ion from aqueous solutions were also carried out by using these compounds in liquid-liquid/solid-liquid extraction experiments.  相似文献

Interaction of (Sp)-2-(diphenylphosphino)ferrocenecarboxylic acid [(Sp)-1] with N,N′-dicyclohexylcarbodiimide (DCC) and N-ethyl-N′-[3-(dimethylamino)propyl]carbodiimide (EDC) have been investigated in order to study the reacting system itself and to characterise side-products typically arising during the diimide-promoted condensation of acid (Sp)-1 with nucleophiles. The reaction between (Sp)-1 and DCC was found to give preferentially the respective urea derivative in the absence of a base, and (Sp)-2-(diphenylphosphino)ferrocenecarboxylic anhydride [(Sp,Sp)-3] when the same reaction was performed in the presence of 4-(dimethylamino)pyridine (DMAP). With EDC, the preference for a reaction pathway was less pronounced: whereas the reaction without the base afforded exclusively the corresponding urea, that in the presence of DMAP yielded a mixture of the urea and anhydride (Sp,Sp)-3.  相似文献

In the frame of substrate engineering, the steric effect of different N-protecting groups on the enantioselectivity and reaction rate of CAL-B-catalysed (S)-selective O-acylation of N-protected 1-hydroxymethyl-tetrahydro-β-carbolines was investigated. Excellent enantioselectivities (E?>?200) were observed when the acylation of N-Boc [(±)-1], N-Cbz [(±)-3], and N-Fmoc-protected [(±)-4] substrates was performed with the use of CAL-B and acetic anhydride in toluene at 60?°C. The resolution of N-acetyl-protected substrate (±)-2 showed excellent E (>200) after 30?min, but as the reaction progressed, E started decreasing after 2 days, because of NO and ON acyl migrations. Preparative resolutions of (±)-3 and (±)-4 resulted in unreacted amino alcohols (R)-3 and (R)-4 and esters (S)-7a and (S)-8a with good enantiomeric excesses (≥88%) and high yields (≥44%).  相似文献

This article describes the synthesis of a new calix[4]arene 1,3-distal glutaraldehyde derivative 4 as a cross-linker-reagent for immobilization of Candida rugosa lipase (CRL). p-tert-Butylcalix[4]arene 1,3-distal diaminoalkyl derivative (3) synthesized via aminolysis reaction of 5,11,17,23-tert-butyl-25,27-ethoxycarbonylmethoxy-26,28-hydroxycalix[4]arene (2) with 1,8-diaminooctane. Compound 3 was converted to its aldehyde derivative (4) by the treatment with glutaraldehyde solution. 4 was used in lipase immobilization in order to see the role of calix[4]arene binding site on the lipase activity and stability. It was observed that the immobilized lipase activity was maintained at levels exceeding 95% of its original activity after 40 min.  相似文献

Photoinduced electron transfer reactions of the title N-acyl-α-dehydronaphthylalaninamides [(Z)-1] with (S)-1-phenylethylamino and (S)-alaninamide auxiliary groups in methanol containing a tertiary amine were shown to form (R,S)- and (S,S)-3,4-dihydrobenzo[f]quinolinone derivatives (2) in excess at rt, respectively. The magnitude of diastereomeric excess (de) was varied in the range of −5-26% for (R,S)-2 and 16-92% for (S,S)-2, depending on the chiral auxiliary and reaction temperature. The mechanism of asymmetric induction in the photocyclization process eventually affording diastereomeric 2 was discussed based on solvent, tertiary amine, chiral auxiliary and temperature effects on the de value as well as on MM2 and PM5 calculations for the diastereomeric enol intermediates.  相似文献

Acyl groups are transferred from diverse N- and O-acyl derivatives of chiral 3,5-bis-(1-hydroxyethyl)-[1,2,4]-triazole to amino acid esters enantioselectively, with 7% to 68% ee, depending on the temperature conditions and nature of the reagents. Thionyl chloride replaced the hydroxyl groups of (S)-1-[4-amino-5-((S)-1-hydroxy-ethyl)-[1,2,4]-triazol-3-yl]-ethanol 3 stereospecifically with inversion, as confirmed by X-ray analysis, which also revealed unusual crystal structures with asymmetric units comprising three molecules of 4-amino-3,5-bis(R-1-chloroethyl)-1,2,4-triazole 5 and four of 3,5-bis((R)-1-chloroethyl)-1H-1,2,4-triazole 6.  相似文献

Promoted by catalytic amounts of Ni complexes tertiary α-hydroxyketones 1a, 3a5a undergo rearrangement, forming chiral isomers 1b, 3b5b. The best enantioselection was obtained with the model system 1-benzoylcyclopentanol 4a/2-hydroxy-2-phenylcyclohexanone 4b. In a ligand screening 2-[4-(S)-tert-butyloxazolin-2-yl]pyridine gave the highest enantiomeric excess of 46% (S)-4b. The analogous isomerisation reactions of α-hydroxyimines 6a, 7a forming chiral α-aminoketones 6b, 7b were established.  相似文献

We report herein, the novel enzymatic desymmetrization of 2-tert-butoxycarbonylamino-2-methyl-1,3-propanediol 1. This method makes it possible to prepare (S)-N-Boc-N,O-isopropylidene-α-methylserinal 3, which is a chiral building block for the synthesis of a variety of α-substituted alanine derivatives. Moreover, optically active (4R)-methyl-4-[2-(thiophen-2-yl)ethyl]oxazolidin-2-one 4, one of the key intermediates in the synthesis of a novel immunosuppressant, has been prepared by this methodology.  相似文献
