The cyclization of 5-amino-3-methylthiopyrazole-4-carbonitriles or 4-carboxamides 3a-j , which were prepared by the reaction of ketene dithioacetals 1a,b [1a : bis(methylthiomethylenemalononitrile; 1b : bis(methylthio)methylenecyanoacetamide] with hydrazines (hydrazine hydrate, phenylhydrazine, p-chlorophenylhydrazine, p-nitrophenylhydrazine), with formamide or carbon disulfide proceeded to give the corresponding 4-amino- or 4-hydroxy-3-methylthiopyrazolo[3,4-d]pyrimidines 6a-h in good yields. 3-Aminopyrazolo[3,4-d]pyrimidine derivatives 6i-1 were also obtained by the application of the cyclization reaction of 3,5-diaminopyrazoles with formamide.  相似文献

Several new pyrazolo[3,4-d]pyrimidine, pyrazolo[3,4-e][1,4]diazepine, pyrazolo[3,4-d][1,2,3]triazine and pyrolo[4,3-e][1,2,4]triazolo[1,5-c]pyrimidine derivatives were prepared by the reaction of the corresponding 5-amino-pyrazole-4-carbonitrile derivative with different organic reagents under different reaction conditions. Using IR, 1H NMR, and mass spectra we have characterized all new compounds.  相似文献

1-(β-D-Ribofuranosyl)pyrazolo[3,4-b]pyrimidine-4-thione ( 6 ), obtained by a three-step synthesis from allopurinol riboside (3), was treated with certain alkyl and aryl halides to provide the corresponding 4-alkylthio derivatives. The nucleoside 4-methylthio-1-(β-D-ribofuranosyl)pyra-zolo[3,4-d]pyritnidine ( 7 ) has served as the precursor for the preparation of the 4-methylamino ( 11 ), 4-dimethylamino ( 12 ), 4-hydrazino ( 13 ), and the 4-hydroxylamino ( 14 ) analogs.  相似文献

Treatment of N-phenyl-substituted benzenecarbo-hydrazonoyl chlorides 1a - d with malononitrile in sodium ethoxide solution gave 5-amino-4-cyanopyrazole derivatives 2 - 5 . Compounds 2 - 5 were converted to formidate derivatives 6 - 9 upon treatment with TEOF in acetic anhydride. The reaction of the latter products 6 - 9 with hydrazine hydrate gave imino-amino derivatives 10 - 13 , which was converted to hydrazino derivatives 14 - 17 by refluxing with hydrazine hydrate. Hydrazino as well as imino-amino derivatives undergo condensation, cyclization, and cycloaddition reactions to give pyrazolo[3,4-d]pyrimidine 18 - 21 , pyrazolo[4,3-e][1,2,4]triazolo-[3,4-c]pyrimidine 22 - 27 , and pyrazolo[3′,4′:4,5]pyrimido[1,6-b][1,2,4]triazine 42 - 44 derivatives. Antimicrobial studies are performed using two Gram-positive bacteria and two Gram-negative bacteria. Data indicated that compounds 5 , 28D , 29B , and 31D are exploring elevated antibacterial effects against all strains tested. Compound 28D is the most promising antibacterial agent against the delicate bacterial strain Bacillus subtilis and Pseudomonas aeruginosa with high effectiveness (low minimum inhibitory concentration [MIC] value) 40 and 60 μg/mL, respectively.  相似文献

The condensation of 1-acyl-2-(morpholin-4-yl)cycloalkenes with 3-amino-1-phenyl-1H-pyrazol-5(4H)-ones gave the corresponding 2,3,6,7,8,9-hexahydropyrazolo[3,4-c]isoquinoline and 3,6,7,8,9,10-hexahydrocyclohepta[ d]pyrazolo[3,4-b]pyridine derivatives. Alkylation of 2,3,6,7,8,9-hexahydropyrazolo[3,4-c]-isoquinolines with alkyl halides occurred at the nitrogen atom in the 3-position. The structure of 7-methyl-2,5-diphenyl-2,3,6,7,8,9-hexahydro-1H-pyrazolo[3,4-c]isoquinolin-1-one was proved by X-ray analysis.  相似文献

The synthesis and characterization of a number of 3-aryl-6H-isoxazolo[3,4-d]pyrazolo[3,4-b]pyridines and 3-aryl-6H-isoxazolo[5,4-d]pyrazolo[3,4–6]pyridines from common precursors, 5-benzoyl-4-chloro-1H-pyrazolo-[3,4-b]pyridines, has been described. The structures were determined by unambiguous chemical synthesis and by isolation and 13C nmr analysis of some key, isolated, intermediates. The ability of these compounds to displace [3H]flunitrazepam from CNS binding sites was also observed.  相似文献

The synthesis of 1,2-dihydro-l-oxopyrazolo[1,5-d]-1,2,4-triaxine was achieved by rerrangment of 2-(5-pyrazolyl)-1,3,4-oxadiazole under alkaline condition. The cyclization of the N-carbethoxyhydrazone of the pyrazole-5-carboxaldehyde gave the 3,4-dihydro-4-oxopyrazolo[1,5-d]1,2,4-triazine. Electrophilic substitutions of the l-pyrazolotriazinome were made either on the lactam nitrogen with methylsulfate, benzylchloride and monochloracetic axid or on the pyrazole ring with bromine. The synthesis of pyrazolo[1,5-d]-1,2,4-triazine was made from teh l-pyrazolotrizinone via the l-pyrazolotrizinone. The methylation of l-pyrazolotrizinone and 8-bromo-l-pyrazolotrizinone afforded N-and S-methyl derivatives. The sysnthesis of 1,2,3,4-tetrahydropyrazolo[1,5-d]-1,2,4-triazine-1,4-diones was axhived by cyclising N-carbethoxyhydrazides of pyrrole-5-carboxylic acid. The structures of the derivatives were determined by 1H-nmr.  相似文献

A comparative study of alkylation by Grignard reagents of PSn(OH)2 (P = tetraphenylporphyrin (TPP), tetraphenylchlorine (TPC), tetraphenylisobacteriochlorine (TPiBC)) shows that dialkylstannylisobacteriochlorines are the most easily obtained. The presence of transition metals in the magnesium crystals directs the reaction towards reduction of the macrocycle instead of alkylation on tin. This is supplementary proof for the intervention of a single electron transfer mechanism (SET) in alkylation of a macrocycle by Grignard reagents.These results fit very well with earlier electrochemical experiments and the measurements of the reduction potentials of the Group IVB metalloporphyrins and their reduced forms.  相似文献

The synthesis of 1,3-disubstituted and 1,3,5-trisubstituted 1H-pyrazolo[3,4-d]pyridazin-4(5H)-ones is conveniently performed by photochemical cyclization. Functionalization of the former compounds leading to the formation of 1,4-disubstituted and 1,3,4-trisubstituted 1H-pyrazolo[3,4-d]pyridazines is smoothly effected through pertinent nucleophilic substitutions.  相似文献

The synthesis of several pyrimido[4,5-c]pyridazine (4-deazafervenulin) ( 1 ), pyrazolo[3,4-d]-pyrimidine ( 2 ) and 6-(pyrazino-1-yl)pyrimidine ( 9 ) analogs has been accomplished from 6-hydrazinouracil ( 3 ). 3-Cyano-4,6-bis(methylthio)pyrazolo[3,4-d]pyrimidine was synthesized by cyclization of 3,4-dicyano-5-aminopyrazole with CS2 in pyridine with subsequent Dimroth rearrangement and methylation of the resulting 3-cyano-4,6-dimercaptopyrazolo[3,4-d]-pyrimidine with methyl iodide. Glycosylation of the product by fusion with 1,2,3,5-tetra-O-acetyl--D-ribofuranose in the presence of iodine gave 1-(2,3,5-tri-O-acetyl--D-ribofuranosyl)-3-cyano-4,6-bis(methylthio)pyrazolo[3,4-d]pyrimidine, the deacetylation of which with a 1% solution of hydrogen chloride in methanol led to 3-cyano-4,6-bis(methylthio)pyrazolo[3,4-d]pyrimidine 1-fiboside.
