A simple method was used to fabricate flavin adenine dinucleotide (FAD)/NiOx nanocomposite on the surface of glassy carbon (GC) electrode. Cyclic voltammetry technique was applied for deposition nickel oxide nanostructures onto GC surface. Owing to its high biocompatibility and large surface area of nickel oxide nanomaterials with immersing the GC/NiOx-modified electrode into FAD solution for a short period of time, 10–140 s, a stable thin layer of the FAD molecules immobilized onto electrode surface. The FAD/NiOx films exhibited a pair of well-defined, stable, and nearly reversible CV peaks at wide pH range (2–10). The formal potential of adsorbed FAD onto nickel oxide nanoparticles film, E o′ vs. Ag/AgCl reference electrode is −0.44 V in pH 7 buffer solutions was similar to dissolved FAD and changed linearly with a slope of 58.6 mV/pH in the pH range 2–10. The surface coverage and heterogeneous electron transfer rate constant (k s ) of FAD immobilized on NiOx film glassy carbon electrode are 4.66 × 10−11 mol cm−2 and 63 ± 0.1 s−1, indicating the high loading ability of the nickel oxide nanoparticles and great facilitation of the electron transfer between FAD and nickel oxide nanoparticles. FAD/NiOx nanocomposite-modified GC electrode shows excellent electrocatalytic activity toward S2O82− reduction at reduced overpotential. Furthermore, rotated modified electrode illustrates good analytical performance for amperometric detection of S2O82−. Under optimized condition, the concentration calibration range, detection limit, and sensitivity were 3 μM–1.5 mM, 0.38 μM and 16.6 nA/μM, respectively.  相似文献

A novel and sensitive biosensor was developed for the determination of nitrite. Firstly, multi-walled carbon nanotubes–poly(amidoamine)–chitosan (MWNT–PAMAM–Chit) nanocomposite along with the incorporation of DNA was used to modify the glassy carbon electrode. Then the immobilization of Cyt c was accomplished using electrochemical deposition method by consecutive cyclic voltammetry (CV) scanning in a neutral Cyt c solution. CV behaviors of the modified electrodes showed that the MWNT–PAMAM–Chit nanocomposite is a good platform for the immobilization of DNA and Cyt c in order, at the same time, an excellent promoter for the electron transfer between Cyt c and the electrode. At high potential, the immobilized Cyt c could be further oxidized into highly reactive Cyt c π-cation by two-step electrochemical oxidation, which could oxidize NO2 into NO3 in the solution. Therefore, a nitrite biosensor based on the biocatalytic oxidation of the immobilized Cyt c was fabricated, which showed a fast response to nitrite (less than 5 s). The linear range of 0.2–80 μM and a detection limit of 0.03 μM was obtained. Finally, the application in food analysis using sausage as testing samples was also investigated.  相似文献

One-step synthesis method was proposed to obtain the nanocomposites of platinum nanoclusters and multiwalled carbon nanotubes (PtNCs–MWNTs), which were used as a novel immobilization matrix for the enzyme to fabricate glucose biosensor. The fabrication process of the biosensor was characterized by cyclic voltammetry, electrochemical impedance spectroscopy, atomic force microscopy and scanning electron microscope. Due to the favorable characteristic of PtNCs–MWNTs nanocomposites, the biosensor exhibited good characteristics, such as wide linear range (3.0 μM–12.1 mM), low detection limit (1.0 μM), high sensitivity (12.8 μA mM−1), rapid response time (within 6 s). The apparent Michaelis–Menten constant ( Km\textapp K_m^{\text{app}} ) is 2.1 mM. The performance of the resulting biosensor is more prominent than that of most of the reported glucose biosensors. Furthermore, it was demonstrated that this biosensor can be used for the assay of glucose in human serum samples.  相似文献

A new H2O2 biosensor was fabricated on the basis of nanocomposite films of hemoglobin (Hb), silver nanoparticles (AgNPs), and multiwalled carbon nanotubes (MWNTs)–chitosan (Chit) dispersed solution immobilized on glassy carbon electrode (GCE). The immobilized Hb displayed a pair of well-defined and reversible redox peaks with a formal potential (E θ′) of −22.5 mV in 0.1 M pH 7.0 phosphate buffer solution. The apparent heterogeneous electron transfer rate constants (k s) in the Chit–MWNTs film was evaluated as 2.58 s−1 according to Laviron’s equation. The surface concentration (Γ*) of the electroactive Hb in the Chit–MWNTs film was estimated to be (2.48 ± 0.25) × 10−9 mol cm−2. Meanwhile, the Chit–MWNTs/Hb/AgNPs/GCE demonstrated excellently electrocatalytical ability to H2O2. Its apparent Michaelis–Menten constant (K Mapp) for H2O2 was 0.0032 mM, showing a good affinity. Under optimal conditions, the biosensors could be used for the determination of H2O2 ranging from 6.25 × 10−6 to 9.30 × 10−5 mol L−1 with a detection limit of 3.47 × 10−7 mol L−1 (S/N = 3). Furthermore, the biosensor possessed rapid response to H2O2 and good stability, selectivity, and reproducibility.  相似文献

A novel biocompatible acerate ZnO whiskers (AZW) has been prepared. We explored AZW and sodium alginate for the construction of electrochemical biosensors. The composition, morphology, and size were studied by scanning electron microscopy. UV–vis spectra revealed that hemoglobin (Hb) adsorbed in the acerate ZnO whiskers and sodium alginate retained its native structure. The amperometric response was measured as a function of H2O2 concentration at a fixed potential of −0.25 V in phosphate-buffered saline (pH 7.0). The electrochemical parameters of Hb in acerate ZnO whiskers and sodium alginate were calculated with the results of the electron transfer coefficient (α) and the apparent heterogeneous electron transfer rate constant (k s) as 0.5 and 2.5 s−1, respectively, indicating good facilitation of the electron transfer between Hb and the modified electrode, which may result from the unique nanostructures and larger surface area of acerate ZnO whiskers. The hydrogen peroxide biosensor showed a fast response of <5 s of linear range 2.1 μM–4.8 mM, with a detection of 0.7 μM (S/N = 3). The apparent Michaelis–Menten constant Kmapp K_m^{{app}} is 0.8 mM. The biosensor possesses high sensitivity, good reproducibility, and long-term stability.  相似文献

A magnetic mesoporous carbon material (i.e., mesoporous iron oxide/C, mesoFe/C) is synthesized for protein immobilization, using glucose oxidase (GOx) as model. Transmission electron microscopy images show that mesoFe/C has highly ordered porous structure with uniform pore size, and iron oxide nanoparticles are dispersed along the wall of carbon. After adsorption of GOx, the GOx-mesoFe/C composite is separated with magnet. The immobilized GOx remains its natural structure according to the reflection–absorption infrared spectra. When the GOx-mesoFe/C composite is coated on a Pt electrode surface, the GOx gives a couple of quasireversible voltammetric peaks at −0.5 V (vs. saturated calomel electrode) due to the redox of FAD/FADH2. The electron-transfer rate constant (k s) is ca. 0.49 s−1. The modified electrode presents remarkably amperometric response to glucose at 0.6 V. The response time (t 95%) is less than 6 s; the response current is linear to glucose concentration in the range of 0.2–10 mM with a sensitivity of 27 μA mM−1 cm−2. The detection limit is 0.08 mM (S/N = 3). The apparent Michaelis–Menten constant (K mapp) of the enzyme reaction is ca. 6.6 mM, indicating that the GOx immobilized with mesoFe/C has high affinity to the substrate.  相似文献

A sulfite oxidase (SOx) purified from leaves of Syzygium cumini (Jamun) was immobilized covalently onto a gold nanoparticles (AuNPs)/chitosan (CHIT)/carboxylated multiwalled carbon nanotubes (cMWCNTs)/polyaniline (PANI) composite that was electrodeposited onto the surface of a gold (Au) electrode. A novel and highly sensitive sulfite biosensor was developed that used this enzyme electrode (SOx/AuNPs/CHIT/cMWCNT/PANI/Au) as the working electrode, Ag/AgCl as the standard electrode, and Pt wire as the auxiliary electrode. The modified electrode was characterized by Fourier transform infrared (FTIR) spectroscopy, cyclic voltammetry (CV), scanning electron microscopy (SEM), and electrochemical impedance spectroscopy (EIS) before and after the immobilization of the SOx. The sensor produced its optimum response within 3 s when operated at 50 mVs−1 in 0.1 M phosphate buffer, pH 7.0, and at 35 °C. The linear range and detection limit of the sensor were 0.75–400 μM and 0.5 μM (S/N = 3), respectively. The biosensor was employed to determine sulfite levels in fruit juices and alcoholic beverages. The enzyme electrode was used 300 times over a period of three months when stored at 4 °C.  相似文献

Multilayer films of multiwalled carbon nanotubes (MWNTs) were homogeneously and stably assembled on a glassy carbon (GC) electrode using the layer-by-layer (LBL) method based on electrostatic interaction between MWNTs (negatively charged) and a biopolymer chitosan (CHIT) (positively charged). Scanning electron microscopy (SEM) image of the resulting {CHIT/MWNTs}9 film indicated that the substrate was mostly covered with MWNTs in the form of small bundles or single nanotubes. The multilayer film was used to study the electrocatalytic oxidation of NADH. The assembled {CHIT/MWNTs}9/GC electrode could decrease the oxidation overpotential of NADH by more than 350 mV. The {CHIT/MWNTs}9/GC electrode exhibited a wide linear response range of 8 × 10−7 to 1.6 × 10−3 mol · L−1 with a correlation coefficient of 0.997 for the detection of NADH. The response time and detection limit (S/N = 3) were determined to be 3 s and 0.3 × 10−6 mol · L−1, respectively. Another attractive characteristic was that the method was simple and the assembled {CHIT/MWNTs}9/GC electrode was highly stable.  相似文献

A sensitive molecularly imprinted electrochemical sensor was developed for selective detection of streptomycin by combination of mercaptoacetic acid-modified PbS nanoparticles with Au-coated Fe3O4 magnetic nanoparticles dispersed multi-walled carbon nanotubes doped chitosan film. The imprinted sensor was fabricated onto the Au electrode via stepwise modification of nanocomposites and an electrodeposited thin film of molecularly imprinted polymers via sol–gel technology. The morphologies and electrochemical behaviors of the imprinted sensor were characterized by scanning electron microscope, cyclic voltammetry, and differential pulse voltammetry, respectively. The prepared sensor showed very high recognition ability and selectivity for streptomycin. Under optimal conditions, the imprinted sensor displayed good electrocatalytic activity to the redox of streptomycin. And the differential voltammetric anodic peak current was linear to the logarithm of streptomycin concentration in the range from 1.0 × 10−6 to 1.0 × 10−3 mol L−1, and the detection limit obtained was 1.5 × 10−9 mol L−1. This proposed imprinted sensor was used successfully for streptomycin determination in different injection solution samples.  相似文献

A novel glucose oxidase (GOD) biosensor was fabricated with a protic ionic liquid (PIL) N-ethylimidazolium trifluoromethanesulfonate ([EIm][TfO]) as the modifier of a carbon electrode. Due to the excellent conductivity and the conformational changes of the microenvironment around the GOD, the electrochemical and biocatalytic properties of GOD immobilized on the PIL-based electrode were dramatically enhanced. A couple of well-defined redox peaks could be observed, with a formal potential of −0.476 V. The GOD biosensor presented good catalytic activity to the oxidation of glucose in oxygen-saturated phosphate buffer solutions. The cathodic peak currents of GOD decreased along with glucose concentrations. A linear response in the range 0.005–2.8 mM was obtained with a detection limit of 2.5 μM. The sensitivity and the apparent Michaelis–Menten constant (K m) were estimated to be 14.96 μA mM−1 and 1.53 μM, respectively. In addition, the biosensor remained stable over 30 days, indicating its good chemical and mechanical stability. The glucose content of several serum samples was determined by using the newly developed biosensor, and the results were in good agreement with those obtained by hospital measurements. All results suggested that PILs were a good media for supporting biocatalytic processes on the bioelectrode.  相似文献

Nanostructured bioelectrodes were designed and assembled into a biofuel cell with no separating membrane. The glassy carbon electrodes were modified with mediator-functionalized carbon nanotubes. Ferrocene (Fc) and 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulfonate) diammonium salt (ABTS) bound chemically to the carbon nanotubes were found useful as mediators of the enzyme catalyzed electrode processes. Glucose oxidase from Aspergillus niger AM-11 and laccase from Cerrena unicolor C-139 were incorporated in a liquid-crystalline matrix-monoolein cubic phase. The carbon nanotubes–nanostructured electrode surface was covered with the cubic phase film containing the enzyme and acted as the catalytic surface for the oxidation of glucose and reduction of oxygen. Thanks to the mediating role of derivatized nanotubes the catalysis was almost ten times more efficient than on the GCE electrodes: catalytic current of glucose oxidation was 1 mA cm−2 and oxygen reduction current exceeded 0.6 mA cm−2. The open circuit voltage of the biofuel cell was 0.43 V. Application of carbon nanotubes increased the maximum power output of the constructed biofuel cell to 100 μW cm−2 without stirring of the solution which was ca. 100 times more efficient than using the same bioelectrodes without nanotubes on the electrode surface.  相似文献

Cyclic voltammetry, chronoamperometry, and rotating disk electrode voltammetry were used to investigate the electrochemical behavior of thiobencarb (TB) at carbon paste electrode modified with an azo dye, 2-(4-((4-ethoxyphenyl)diazenyl)phenylamino)ethanol (EDPE), EDPE/modified carbon paste electrode (MCPE). The modified electrode showed high electrocatalytic activity toward thiobencarb. The current was enhanced significantly relative to the situation prevailing when a bare glassy carbon electrode was used. The kinetics parameters of this process were calculated, the apparent electron transfer rate constant k s and α (charge transfer coefficient between electrode and EDPE) were 14.6 s−1 and 0.48, respectively. The experimental parameters were optimized, and the mechanism of the catalytic process was discussed. The best defined cathodic peak was obtained with 0.1 M acetate buffer (pH 3.0). The response of the sensor was very quick, and response time was approximately 5 s. The differential pulse voltammetry response of the MCPE was linear against the concentration of TB in the range of 0.96 to 106 μg L−1. The limit of detection was found to be 0.025 μg L−1. The precision was examined by carrying out eight replicate measurements at a concentration of 25 μg L−1 TB; the relative standard deviation was 2.9%.  相似文献

A mid-infrared enzymatic assay for label-free monitoring of the enzymatic reaction of fructose-1,6-bisphosphatase with fructose 1,6-bisphosphate has been proposed. The whole procedure was done in an automated way operating in the stopped flow mode by incorporating a temperature-controlled flow cell in a sequential injection manifold. Fourier transform infrared difference spectra were evaluated for kinetic parameters, like the Michaelis–Menten constant (K M) of the enzyme and V max of the reaction. The obtained K M of the reaction was 14 ± 3 g L−1 (41 μM). Furthermore, inhibition by adenosine 5′-monophosphate (AMP) was evaluated, and the K MApp value was determined to be 12 ± 2 g L−1 (35 μM) for 7.5 and 15 μM AMP, respectively, with V max decreasing from 0.1 ± 0.03 to 0.05 ± 0.01 g L−1 min−1. Therefore, AMP exerted a non-competitive inhibition.  相似文献

Glassy carbon electrode modified with electrodeposited nickel oxide nanoparticles (NiOxNPs) was used as electrocatalyst for oxidation of omeprazole and pentoperazole in alkaline solution. The modified electrode exhibited efficient electrocatalytic activity for the oxidation of omeprazole and pentoperazole with relatively high sensitivity, excellent stability, and long lifetime. Hydrodynamic amperometric method is used for determination of selected analytes. Under optimized condition, the linear concentration range, detection limit, and sensitivity of modified electrode toward omeprazole detection are 4.5–120 μM, 0.4 μM (at signal to noise 3), and 40.1 nA μM−1 cm−2, respectively. For pantoperazole, hydrodynamic amperometric determination yielded calibration curve with linear range of 2.5–180 μM, detection limit of 0.2 μM, and sensitivity of 39.2 nA μM−1 cm−2, respectively. The proposed method was successfully applied to pentoperazole and omeprazole determination in drug samples.  相似文献

The electrochemical solid phase micro-extraction of salicylic acid (SA) at graphite-epoxy-composed solid electrode surface was studied by cyclic voltammetry. SA was oxidized electrochemically in pH 12.0 aqueous solution at 0.70 V (vs. saturated calomel electrode) for 7 s. The oxidized product shows two surface-controlled reversible redox couples with two proton transferred in the pH range of 1.0∼6.0 and one proton transferred in the pH range of 10.0∼13.0 and is extracted on the electrode surface with a kinetic Boltzman function of i p = 3.473–4.499/[1 + e(t − 7.332)/6.123] (χ 2 = 0.00285 μA). The anodic peak current of the extracted specie in differential pulse voltammograms is proportional to the concentration of SA with regression equation of i p = −5.913 + 0.4843 c (R = 0.995, SD = 1.6 μA) in the range of 5.00∼200 μM. The detection limit is 5.00 μM with RSD of 1.59% at 60 μM. The method is sensitive and convenient and was applied to the detection of SA in mouse blood samples with satisfactory results.  相似文献

A method to fabricate poly(3,4-ethylene dioxythiophene)-poly(4-styrene sulfonate)-Meldola Blue (PEDOT-PSS-MDB)-modified electrodes had been disclosed. Firstly, the PEDOT-PSS-film-modified electrode was electrochemically prepared. Then, the PEDOT-PSS was treated as a matrix to immobilize electroactive mediator, Meldola Blue (MDB), by means of an electrostatic interaction to form the proposed film, PEDOT-PSS-MDB. Electrochemical properties of the proposed film exhibited surface confinement and pH dependence. The PEDOT-PSS-MDB electrode could electrocatalytically reduce hydrogen peroxide (H2O2) with a low overpotential and showed a linear response to H2O2 in the concentration range of 5 to 120 μM, detection limit of 0.1 μM, and sensitivity of 353.9 μA mM−1 cm−2 (S/N = 3). By comparison, the electrocatalytic activity of PEDOT-PSS-MDB electrode was found superior to that of PEDOT-PSS and MDB-PSS electrodes. It also has competitive potential as compared with other mediators, through the use of HRP to determine H2O2. Moreover, the potential interferents such as ascorbic acid, dopamine, uric acid, and glucose were also studied for H2O2 determination by the proposed film.  相似文献

The potential application of ordered mesoporous carbon (OMC)-modified glassy carbon electrode (OMC/GCE) in electrochemistry as a novel electrode material was investigated. X-ray diffraction, transmission electron micrographs, and cyclic voltammetry were used to characterize the structure and electrochemical behaviors of this material. Compared to GC electrode, the peak currents of potassium ferricyanide (K3[Fe(CN)6]) increase and the peak potential separation (ΔE p) decreases at the OMC/GC electrode. These phenomena suggest that OMC-modified GC electrode possesses larger electrode area and faster electron transfer rate, as compared with bare GC electrode. Furthermore, riboflavin was detected using OMC/GC electrode in aqueous solutions. The results showed that, under an optimum condition (pH 7.0), the OMC/GC electrode exhibited excellent response performance to riboflavin in the concentration range of 4.0 × 10−7 to 1.0 × 10−6 M with a high sensitivity of 769 μA mM−1. The detection limit was down to around 2 × 10−8 M. With good stability and reproducibility, the present OMC/GC electrode was applied in the determination of vitamin B2 content in vitamin tablets, and satisfactory results were obtained.  相似文献

Solid composite polymer electrolytes consisting of polyethylene oxide (PEO), LiClO4, and porous inorganic–organic hybrid poly (cyclotriphosphazene-co-4, 4′-sulfonyldiphenol) (PZS) nanotubes were prepared using the solvent casting method. Differential scanning calorimetry and scanning electron microscopy were used to determine the characteristics of the composite polymer electrolytes. The ionic conductivity, lithium ion transference number, and electrochemical stability window can be enhanced after the addition of PZS nanotubes. The electrochemical impedance showed that the conductivity was improved significantly. Maximum ionic conductivity values of 1.5 × 10−5 S cm−1 at ambient temperature and 7.8 × 10−4 S cm−1 at 80 °C were obtained with 10 wt.% content of PZS nanotubes, and the lithium ion transference number was 0.35. The good electrochemical properties of the solid-state composite polymer electrolytes suggested that the porous inorganic–organic hybrid polyphosphazene nanotubes had a promising use as fillers in SPEs and the PEO10–LiClO4–PZS nanotube solid composite polymer electrolyte might be used as a candidate material for lithium polymer batteries.  相似文献

The electrocatalytic oxidation of quinine sulfate (QS) was investigated at a glassy carbon electrode, modified by a gel containing multiwall carbon nanotubes (MWCNTs) and room-temperature ionic liquid of 1-Butyl-3-methylimidazolium hexafluorophate (BMIMPF6) in 0.10 M of phosphate buffer solution (PBS, pH 6.8). It was found that an irreversible anodic oxidation peak of QS with E pa as 0.99 V appeared at MWCNTs-RTIL/glassy carbon electrode (GCE). The electrode reaction process was a diffusion-controlled one and the electrochemical oxidation involved two electrons transferring and two protons participation. Furthermore, the charge-transfer coefficient (α), diffusion coefficient (D), and electrode reaction rate constant (k f) of QS were found to be 0.87, 7.89 × 10−3 cm2⋅s−1 and 3.43 × 10−2 s−1, respectively. Under optimized conditions, linear calibration curves were obtained over the QS concentration range 3.0 × 10−6 to 1.0 × 10−4 M by square wave voltammetry, and the detection limit was found to be 0.44 μM based on the signal-to-noise ratio of 3. In addition, the novel MWCNTs-RTIL/GCE was characterized by the electrochemical impedance spectroscopy and the proposed method has been successfully applied in the electrochemical quantitative determination of quinine content in commercial injection samples and the determination results could meet the requirement.  相似文献

A novel method was developed for the determination of captan, folpet, and captafol in apples by dispersive liquid–liquid microextraction (DLLME) coupled with gas chromatography–electron capture detection (GC–ECD). Some experimental parameters that influence the extraction efficiency, such as the type and volume of the disperser solvents and extraction solvents, extraction time, and addition of salt, were studied and optimized to obtain the best extraction results. Under the optimum conditions, high enrichment factors for the compounds were achieved ranging from 824 to 912. The recoveries of fungicides in apples at spiking levels of 20.0 μg kg−1 and 70.0 μg kg−1 were 93.0–109.5% and 95.4–107.7%, respectively. The relative standard deviations (RSDs) for the apple samples at 30.0 μg kg−1 of each fungicide were in the range from 3.8 to 4.9%. The limits of detection were between 3.0 and 8.0 μg kg−1. The linearity of the method ranged from 10 to 100 μg kg−1 for the three fungicides, with correlation coefficients (r 2) varying from 0.9982 to 0.9997. The obtained results show that the DLLME combined with GC–ECD can satisfy the requirements for the determination of fungicides in apple samples.
