SYNTHESIS OF THE LEU-ENKEPHALIN GENE

The synthesis of Leu-enkephalin gene by an alternative approach was described in thispaper. The sequence of the synthetic gene, 26 base-pairs in length, was derived from the ami-no acid sequence of the hormone peptide Leu-enkephalin. It bears single-stranded cohesive ter-mini for the restriction endonucleases EcoR I and BamH I so that it may be inserted into apBR 322 plasmid. The 4 deoxyoligonucleotide fragments, varying in length from octanucleotideto octadecanucleotide, were synthesized by an improved phosphotriester method developed inour laboratory. All chemically synthetic fragments were pure and had the correct sequences.The assembly of the Leu-enkephalin gene was carried out in a one-step ligation reaction cata-lysed by T_4-DNA ligase with good yield, Finally, the purified products from polyacrylamidegel electrophoresis had the correct joining-points as predicted.

A precursor 9 of isosarcophytol-A(1)was prepared from(■)-farnesol(3)through 6 steps.Isosarcophytol-A(1),a cembrane-type diterpenoid,was first isolated fromustralian soft coral(Nephthea brassica)in 1982,and its structure was estab-shed as(1,3,7,11-all ■,13■)-cembra-1,3,7,11-tetraen-13-ol,an isomer ofarcophytol-A■.As far as we know,its bioactivity and synthesis have noteen reported yet.

The effects of the chain length and stereo-factor of substrates and solvents on the lipase catalysed lactonization of ω-hydroxy-ester were studied, And also the catalytic effects of various enzymes were compared.     

ETEREOSPECIFIC SYNTHESIS OF α-D-FRUCTOFURANOSIDES——SYNTHESIS OF THE OLIGO-FRUCTAN DIASTEREOISOMERS

α-fructofuranosides were stereospecifically synthesized from2-O-Ac-1,3,4,6-O-tetra-benzoylfructose by employing TMSOTf as a catalyst.Meanwhile,one diastereoisomer derivative of sucrose and two of fructanoligomers were synthesized.The structure of all products were confirmed by~(13)C-NMR and ~1H-NMR spectra.

THE USE OF MALONONITRILE IN THE SYNTHESIS OF NOVEL α-AMINO NITRILES

Continuing earlier studies designed to obtain compounds of pharmaceutical interest, we used acetyl acetone (1, R=CH₃) and ethyl acetoacetate (1, R=OEt) for the preparation of some novel α-amino nitriles (3, 6 and 7). EFFECTS OF L-PHENYLALANINE ON THE RADIATION SYNTHESIS OF POLY(N,N''''-METHYLENEBISACRYLAMIDE-co-4-VINYLPYRIDINE)(MICRO)GELS

The effects of L-phenylalanine(L-Phe)on the synthesis of poly(N,N'-methylenebisacrylamide-co-4-vinylpyridine) (poly(Bis-co-4-VP))(micro)gels by γ-ray irradiation were studied.The addition of L-Phe could not only decrease the gelation dose(D_g)of the synthesis obviously,but also transform the morphology of copolymer from microgel to gel.In addition,the swelling ability of the(micro)gels was also affected in the presence of L-Phe.The decrease of D_g was ascribed to the effect of pH,while the transformation of the morphology was ascribed to the effect of L-Phe on the stability of the poly(Bis-co-4-VP)microgel.Such an effect was confirmed further as compared with the effects of L-alanine,L-glutamic acid,L-arginine,sulfuric acid and aqueous ammonia.

STUDIES ON THE TOTAL SYNTHESIS OF MAYTANSINOIDS (Ⅳ)——SYNTHESIS OF MAYTANSINE

This paper reports the total synthesis of optically pure maytansinol and maytansine. The physical constants and spectral data including R_f, m.p., specific rotation, HPLC, UV, CD, IR, NMR, and MS, of synthetic maytansinol and maytansine are identical with those of natural products in all respects. The preliminary biological test showed that synthetic maytansine possesses the same inhibitory activity in vitro against carcinoma cells as the natural product.     

STUDIES ON THE SYNTHESIS OF THE SPIROKETAL FRAGMENT OF AZADIRACHTIN(Ⅰ)

The stereoselective synthesis of substituted [2,2,1]-bicyclo-heptan-2-one 12,a supposed intermediate to the spiroketal fragment of azadi-rachtin was described.When 12 was treated with mCPBA in CH_2Cl_2 in the pres-ence of excess amount of solid sodium bicarbonate,only 13 was obtained.Thespiroketal intermediate 20 was converted from 9 through 5 steps by an alterna-tive route.
