Summary.  Three new 4-quinolinone alkaloids (5,6-dimethoxy-2-(3-methoxyphenyl)-1H-quinolin-4-one, 5,6-dimethoxy-2-(3,4-dimethoxyphenyl)-1H-quinolin-4-one, 5,6-dimethoxy-2-(2,5,6-trimethoxyphenyl)-1H-quinolin-4-one) were isolated from the leaves of Casimiroa edulis Llave et Lex (Rutaceae) cultivated in Egypt. Their structures were determined by UV/Vis, IR, 1H and 13C NMR, and EI mass spectroscopy. The alkaloids were also detected in the kernels of the seeds. Received May 28, 2001. The extract of Casimiroa edulis was investigated for antihypertensive activity. The ethanol and total alkaloids (in chloroform) extracts were found to have antihypertensive properties at doses of 500 and 200 mg/kg, respectively. Four quinolinone alkaloids were isolated and identified as: 2-(2′-hydroxy-4′-methoxyphenyl)-5,8-dimethoxy-3-propyl-1H-quinolin-4-one (1), 5,8-dimethoxy-2-(3′-methoxyphenyl)-3-propyl-1H-quinolin-4-one (2), 5,8-dimethoxy-2-(3′,4′-dimethoxyphenyl)-3-propyl-1H-quinolin-4-one (3), and 5,6-dimethoxy-2-(2′,5′,6′-trimethoxyphenyl)-1H-quinolin-4-one (4). Interestingly, compounds 1, 2, and 3 were found to be new alkaloids. The four isolated alkaloids showed antihypertensive activity at doses of 50, 100, 200, and 300 mg/kg, respectively. O-Alkylation of 8-hydroxy-1H-quinolin-2-one ( 1 ) afforded 8-(2-oxopropoxy)-1H-quinolin-2-one ( 2 ) which was immediately cyclized to form the tricyclic 2,3-dihydro-3-hydroxy-3-methyl-5H-pyrido[1,2,3-de][1,4]benzoxazine,-5-one ( 3). The Reformatsky-type condensation of 3 furnished antiplatelet 8-[(2,3,4,5-tetrahydro-2-methyl-4-methylidene-5-oxofuran-2-yl)melhoxy]-1H-quinolin-2-one ( 4 ). Its counterparts 7a – f , Ph-substituted at C(2) of the furan ring, were obtained from 1 via alkylation and the Reformatsky-type condensation. Although compound 4 was less active against platelet aggregation than 7a – f , it was the only compound which exhibited significant inhibitory activity on high-K+ medium, Ca2+-induced vasoconstriction and was more active than most of its Ph-substituted counterparts against norepinephrine-induced vasoconstrictions.  相似文献

The quinaldyl ketone, 4-phenyl-3-(quinolin-2-yl)-butan-2-one was prepared by two methods: (a) benzylation of 1-(1H-quinolin-2-ylidene)propan-2-one in the presence of sodium hydride in dimethylformamide and (b) by the benzylative demethoxycarbonylation of methyl 2-(1H-quinolin-2-ylidene)-3-oxobutanoate in the presence of lithium bromide in hexamethylphosphoramide at 135°. In the absence of acid, the compound exists exclusively in the tautomeric form, 4-phenyl-3-(1H-quinolin-2-ylidene)butan-2-one.  相似文献

The present investigation describe the synthesis of 8-(benzyloxy)-5-(2-[1,3-diphenyl-1H-pyrazol-4-yl]thiazol-4-yl)quinolin-2(1H)-one derivatives. Quinolin-8-ol was transformed by five step synthetic procedures into 8-Benzyloxy-5-(2-bromo-acetyl)-1H-quinolin-2-one. Subsequently, 8-Benzyloxy-5-(2-bromo-acetyl)-1H-quinolin-2-one condensed with 1,3-Diphenyl-1H-pyrazole-4-carbothioic acid amide in the presence of acetonitrile to afford 8-(benzyloxy)-5-(2-[1,3-diphenyl-1H-pyrazol-4-yl]thiazol-4-yl)quinolin-2(1H)-one derivatives. Synthesized compounds were screened for their antimicrobial activity against gram-positive and gram-negative bacteria. Most of the synthesized compounds are found to be active against tested bacterial strains and fungal strain.  相似文献

A novel, efficient and convenient procedure has been developed for the synthesis of 2-(4-amino-substituted benzylidene)indanone derivatives. In the first step, the reaction of 4-fluorobenzaldehyde with 5, 6-dimethoxy-2, 3-dihydro-1H-inden-1-one in the presence of NaOH in EtOH was described. In the next step, a variety of aliphatic and aromatic amines were reacted with 2-(4-fluorobenzylidene)-5, 6-dimethoxy-2, 3-dihydro-1H-Inden-1-one via aromatic substitution (SNAr) reaction to produce 2-(4-aminobenzylidene)-5, 6-dimethoxy-2, 3-dihydro-1H-Inden-1-one derivatives as a novel class of 1-indanones. These products have been successfully prepared in good to excellent yields. 1?H and 13?C NMR, FT-IR spectroscopy and CHN analysis supported the proposed structures of the products.  相似文献

Base catalyzed cyclization of ethyl 4-[2-(2-amino-4,5-dimethoxyphenyl)ethyl]-2-phenylpyrimidine-5-carboxylate, derived from ethyl 4-methyl-2-phenyl-5-pyrimidinecarboxylate and 3,4-dimethoxy-6-nitrobenz-aldehyde, gave 5,6-dihydro-8,9-dimethoxy-3-phenylbenzo[b]pyrimido[4,5-f]azocine-12(11H)-one, the first reported example of this ring system.  相似文献

Summary 2-Benzoylamino-6,7-dimethoxy-4H-3,1-benzoxazin-4-one was prepared by thermal treatment of 2-(3-benzoylthioureido)-4,5-dimethoxybenzoic acid and by benzoylation of 2-amino-6,7-dimethoxy-4H-3,1-benzoxazin-4-one. The inactivation of chymotrypsin and human leukozyte elastase by the title compound and 2-benzoylamino-4H-3,1-benzoxazin-4-one is reported.

Über die Inaktivierung von Chymotrypsin und humaner Leukozyten-Elastase durch die Titelverbindung und 2-Benzoylamino-4H-3,1-benzoxazin-4-on wird berichtet.

Two dihydroxybenzofuroquinolinones, 3,9-dihydroxy-5H-benzofuro[3,2-c]quinolin-6-one (V) and 3,8-dihydroxy-6H-benzofuro[2,3-b]quinolin-11-one (VI), were obtained by the demethyl-cyclization of 3-(2,4-dimethoxyphenyl)-4-hydroxy-7-methoxy-1H-quinolin-2-one (IV). By the methylation with diazomethane, V gave a dimethylated compound (VII), while VI gave a trimethylated compound (VIII).  相似文献

The copper-catalyzed pyrolysis of 1-diazo-3-(pyrrol-1-yl)-2-propanone (1a) and 1-diazo-4-(pyrrol-1-yl)-2-butanone (1b) in benzene solution gave 1 H-pyrrolizin-2-(3H)-one (4a) and 5,6-dihydroindolizin-7 (8H)-one (4b) , respectively, in quantitative yield. Similar pyrolysis of 1-diazo-4-(3-methylindol-1-yl)-2-butanone (9) was less efficient giving 1-methylbenzo[b]-5,6-dihydroindroindolizin-7 (8H)-one (10) and 4-(3-methylindol-1-yl)-but-l-en-3-one (11) in 7% and 24% yield, respectively.  相似文献

The Schiff bases, 3-(indolin-2-one)hydrazinecarbothioamide (L1H), 3-(indolin-2-one)hydrazinecarboxamide (L2H), 5,6-dimethyl-3-(indolin-2-one)hydrazinecarbothioamide (L3H), and 5,6-dimethyl-3-(indolin-2-one)hydrazinecarboxamide (L4H), have been synthesized by the condensation of 1 H-indol-2,3-dione and 5,6-dimethyl-1H-indol-2,3-dione with the corresponding hydrazinecarbothioamide and hydrazinecarboxamide, respectively. The complexes of oxovanadium and ligands have been characterized by elemental analyses, melting points, conductance measurements, molecular weight determinations, and IR, 1H NMR and UV spectral studies. These studies showed that the ligands coordinated to the oxovanadium in a monobasic bidentate fashion through oxygen or sulfur and the nitrogen donor system. Thus, penta-and hexacoordinated environment around the vanadium atom has been proposed. All the complexes and their parent organic moieties have been screened for their biological activity on several pathogenic fungi and bacteria and were found to possess appreciable fungicidal and bactericidal properties. Condensation of 3,4-dihydro-6,7-dimethoxyisoquinoline ( 4 ) with 4-methylnicotinoyl chloride ( 12 ) in refluxing pyridine gives 5,6,13,13a-tetrahydro-2,3-dimethoxy-8H-isoquino[2,1-b][2,7] naphthyridin-8-one ( 11 ), along with some of its 13,13a-didehydro derivative 7 . A similar reaction of 4 with 4-(chloromethyl)nicotinoyl chloride ( 14 ) affords, in addition to 7 , the isomeric product 10,11-dihydro-7,8-dimethoxy-13H-pyrido[4′,3′:3,4]pyrrolo[2,1- b][3]benzazepin-13-one ( 3 ). Analogous pairs of products are obtained from 3,4-dihydro-6,7-(methylenedioxy)- and 3,4-dihydro-6,7,8-trimethoxy-isoquinolines ( 15 and 18 , resp.). The structure of 3 was established by extensive NMR data and confirmed by single-crystal X-ray studies. Structure 7 has the ring system of the Alangium alkaloids like alangimarinc ( 1 ), while the isomeric ring system 3 is predicted to be present in nature on biogenetic reasoning.  相似文献

Cycloaddition of dichloroketone to N,N-disubstituted (E)-4-aminomethylene-3,4-dihydro-1-benzoxepin-5(2H)-ones gave N,N-disubstituted 4-amino-3,3-dichloro-3,4,5,6-tetrahydro-2H-pyrano[3,2-d]-1-benzoxepin-2-ones II, which are derivatives of the new heterocyclic system 2H-pyrano[3,2-d]-1-benzoxepin. Dehydrochlorination with triethylamine of II afforded N,N-disubstituted 4-amino-3-chloro-5,6-dihydro-2H-pyrano-[3,2-d]-1-benzoxepin-2-ones III in good to moderate yields. In the triethylamine treatment of IIh (NR2 = diphenylamino), 3-chloro-5,6-dihydro-2H-pyrano[3,2-d]-1-benzoxepin-2-one was isolated in low yield near to IIIh, whereas IIc (NR2 = diisopropylamino) gave in low yield 4-diisopropylamino-5,6-dihydro-2H-pyrano(3,2-d)-1-benzoxepin-2-one.  相似文献

An efficient and novel route for the synthesis of 1H-indol-2-yl-(4-aryl)-quinolin-2(1H)-one 1 via palladium-catalyzed site-selective cross-coupling reaction and cyclization process was described. Reaction of 3-bromo-4-trifloxy-quinolin-2(1H)-one 3 with arylboronic acid catalyzed by PdCl2(PPh3)2 afforded 3-bromo-4-aryl-quinolin-2(1H)-one 4, which then reacted with 2-ethynylaniline 5 via Pd-catalyzed Sonogashira coupling and CuI-mediated cyclization leading to the desired 1H-indol-2-yl-(4-aryl)-quinolin-2(1H)-one 1 in good yields.  相似文献

1,1-Difluoro-3-(1H-quinolin-2-ylidene)propan-2-one 1a, 1,1,1-trifluoro-3-(1H-quinolin-2-ylidene)propan-2-one 1b, 1,1,1-trifluoro-3-(4-chloro-1H-quinolin-2-ylidene)propan-2-one 1c and 1,3-dibromo-1,1-difluoro-3-(2-quinolyl)propan-2-one 2 are prepared and characterized by various spectroscopic techniques. The crystal structure of 1a is determined by X-ray diffraction. Furthermore, a series of previously known non-halogenated (1H-quinolin-2-ylidene)propan-2-ones 1d-1h are oxidized with AgBrO3 in the presence of AlCl3. In all cases, 2-(1-bromo-1-chloromethyl)quinoline 3 is obtained in high yield. The bromination order and sites of 1a are analyzed based on ab initio MP2 and DFT calculations for the molecule and its anions.  相似文献

Ring Transformations of 3-Substituted 5-Trifluoromethyl-1,3,4-thiadiazol-2(3H)-one with Nucleophiles The 3-chlormethyl-5-trifluoromethyl-1,3,4-thiadiazolone 3 undergoes a ring transformation to 3-acylated 2,3-dihydro-1,3,4-thiadiazoles 4 with many nucleophiles. Upon formal replacement of the chloromethyl group in the 3-position of 3 by an extended bromoalkyl chain (→9a-c) , the reaction with nucleophiles yields 4-acylated 5,6-dihydro-4H-1,3,4-thiadiazines 10 (from 9a ), 4,5,6,7-tetrahydrothiadiazepines 13 (from 9b ) and 5,6,7,8-tetrahydro-4H-1,3,4-thiadiazocines 14 (from 9c ) by ring enlargement. The 3-propargyl-thiadiazolone 17 rearranges with nucleophiles to 4-acylated 6-methylidene-5,6-dihydro-4H-1,3,4-thiadiazines 18 . The structures of the new compounds were elucidated by 1H- and 13C-NMR spectroscopy.  相似文献

GLC. allowed the isolation of 1, 3, 6, 6-tetramethyl-5, 6, 7, 8-tetrahydro-isoquinolin-8-one (A) and 3,6,6-trimethyl-5,6-dihydro-7H-2-pyrindin-7-one (D) from Burley tobacco condensate (about 0.1% each). The structures and syntheses of these novel terpenoid alkaloids are described, and a possible way for their formation in tobacco is suggested.  相似文献

Pd/C-Cu catalyzed coupling reactions of 3-iodo-1H-quinolin-4-ones with a variety of terminal alkynes afforded furo[3,2-c]quinolines regioselectively in good to excellent yields. 3-Alkynyl quinolones were isolated under the same reaction conditions when the nitrogen of 3-iodo-1H-quinolin-4-one was substituted with an alkyl group.  相似文献

以芳亚甲基丙二腈或2-氰基-3-芳基丙烯酸酯和4-羟基喹啉-2-酮为原料, 水为溶剂, 在TEBA(三乙基苄基氯化铵)催化下90 ℃, 合成了2-氨基-4-芳基-5,6-二氢化-4H-吡喃[3,2-c]喹啉-5-酮衍生物. 和其它方法相比, 该反应具有反应条件温和, 产率高(87%~96%)和环境友好等优点. 产物的结构通过熔点, IR, 1H NMR和元素分析表征. 化合物3m的结构通过X单晶衍射分析确证.  相似文献

利用串联Aza-Wittig法合成了两类共计16个未见文献报道的噻吩并吡啶并嘧啶类化合物, 通过IR, 1H NMR, EI-MS, 元素分析等方法对所合成的化合物进行了结构表征, 化合物II-1还通过了X单晶衍射的进一步确证. 并初步测定了所合成化合物的杀虫、杀菌和除草活性. 结果表明: 所测化合物的杀虫活性较差, 但大部分化合物具有一定的除草活性和良好的杀菌活性.  相似文献
