For a systematic exploration of structural relationships between molecular scaffolds, ~24,000 unique scaffolds were extracted from 458 different target sets. Substructure relationships between these scaffolds were systematically determined. The scaffold tree data structure was utilized to study structural relationships between original scaffolds and derivative scaffolds obtained by rule-based decomposition. Leaf-to-root substructure relationships that resulted from rule-based decomposition were compared to leaf-to-leaf relationships between original scaffolds most of which were not part of the scaffold tree hierarchy. Decomposed scaffolds not contained in active target set compounds were prioritized on the basis of hierarchical scaffold patterns and additional substructure relationships. For high-priority virtual scaffolds, activity predictions were carried out, and these scaffolds were often found in external test compounds having the predicted activity. Taken together, our results suggest that leaf-to-root substructure relationships in scaffold trees should best be complemented with additional substructure relationships to determine high-priority virtual scaffolds for activity prediction.  相似文献

To investigate the potential application of microcrystal cellulose (MCC) and cellulose whisker (CW) in the electrospun vascular tissue scaffolds, cellulose acetate (CA) and cellulose composite scaffolds containing MCC and CW were electrospun from CA solutions and deacetylation. Structure and morphology of MCC, CW and the fibrous composite scaffolds were investigated using FT-IR, SEM, TEM and AFM. The wettability of the scaffolds was evaluated by water contact angle analysis. The effect of MCC and CW on the biocompatibility of the scaffolds for vascular smooth muscle cells (VSMC) was assayed by MTT test, fluorescent imaging and SEM. The biocomposite scaffolds displayed multi-scaled structure and morphology. The scaffolds containing MCC and CW simultaneously exhibited significantly higher cell viability compared to those with only MCC or CW and no filler. Cell viability and morphology within the scaffolds become better with increasing content of MCC and CW. The composite scaffolds with both micro- and nano-scale organization could mimic the native extracellular matrix more closely, and further produce synergistic enhancement on VSMC viability, adhesion and proliferation. This study provides the potential applications of renewable cellulose-based particulates in biomedical field.  相似文献

Novel porous-conductive chitosan scaffolds were fabricated by incorporating conductive polypyrrole (PPy) particles into a chitosan matrix and employing a phase separation technique to build pores inside the scaffolds. Conductive polypyrrole particles were prepared with a microemulsion method using FeCl3 as a dopant. The preparation conditions were optimized to obtain scaffolds with controlled pore size and porosity. The conductivity of the scaffolds was investigated using a standard four-point probe technique. It was found that several kinds of scaffolds showed a conductivity close to 10(-3) S.cm(-1) with a low polypyrrole loading of around 2 wt.-%. The main mechanical properties, such as tensile strength, breaking elongation and Young's modulus of the scaffolds, were examined both in the dry and in the hydrated states. The results indicated that a few different kinds of scaffolds exhibited the desired mechanical strength for some tissue engineering applications. The miscibility of polypyrrole and chitosan was also evaluated using a dynamic mechanical method. The presence of significant phase separation was detected in non-porous PPy/chitosan scaffolds but enhanced miscibility in porous PPy/chitosan scaffolds was observed.  相似文献

Porous nano-hydroxyapatite/polycaprolactone (nHA/PCL) scaffolds with different composition ratios of nHA/PCL were fabricated via a melt-molding/porogen leaching technique. All scaffolds were characterized before and after degradation in vitro for six months. The original scaffolds had high porosity at around 70% and showed decreasing compressive modulus (from 24.48 to 2.69 MPa for hydrated scaffolds) with the introduction of nHA. It was noted that the scaffolds could retain relatively stable architecture and mechanical properties for at least six months, although some slight changes happened with the nHA/PCL scaffolds in the mass, the nHA content, the PCL molecular weight and the crystallinity. Moreover, during the 7 days culture of bone marrow stromal cells (BMSCs) on scaffolds, the cell adhesion and proliferation of BMSCs were presented well on both the surface and the cross-section of the scaffolds. All of these results suggested the nHA/PCL scaffolds to be promising in bone tissue engineering.  相似文献

In order to increase the biocompatibility and bioactivity of chitosan, hydroxyapatite had been in situ combined into chitosan scaffolds. The bioactivity of the composite scaffolds was studied by examining the apatite formed on the scaffolds by incubating in simulated body fluid and the activity of preosteoblasts cultured on them. The apatite layer was assessed using scanning electronic microscope (SEM), X-ray diffraction (XRD), Fourier-Transformed Infrared spectroscopy (FTIR) and weight measurement. Composite analysis showed that after incubation in simulated body fluid on both of the scaffolds carbonate hydroxyapatite was formed. With increasing nano-hydroxyapatite content in the composite, the quantity of the apatite formed on the scaffolds increased. Compared with pure chitosan, the composite with nano-hydroxyapatite could form apatite more readily during the biomimetic process, which suggests that the composite possessed better mineralization activity. Furthermore, preosteoblast cells cultured on the apatite-coated scaffolds showed different behavior. On the apatite-coated composite scaffolds cells presented better proliferation than on apatite-coated chitosan scaffolds. In addition, alkaline phosphatase activities of cells cultured on the scaffolds in conditioned medium were assessed. The cells on composite scaffolds showed a higher alkaline phosphatase activity which suggested a higher differentiation level. The results indicated that the addition of nano-hydroxyapatite improved the bioactivity of chitosan/nano-hydroxyapatite composite scaffolds. On the other hand, that is to say composition of substrates could affect the apatite formation on them, and pre-loaded hydroxyapatite can enhance the apatite-coating. It will also be significant in preparation of apatite-coating polymer scaffolds for bone tissue engineering.  相似文献

采用低热-高压法制备了聚(dl-丙交酯/乙交酯)75/25(PLGA75/25)组织工程多孔支架。该方法避免了使用有机溶剂,支架的孔隙率在90%以上,孔径大小分布均匀。多孔支架经过酒精处理后,支架表面产生许多微小的凹陷;用藻酸钙改性处理后,支架形态保持良好。两种处理都使支架的压缩强度有所增大,亲水性增强。虽然孔隙率高的支架降解速率稍慢,但其体外降解规律基本一致:特性粘数争力学强度衰减快,而质量损失较慢,降解6周后,支架的质量损失仅为3%左右;体外降解3周后,支架的形态保持良好,可望在细胞移植争组织修复的早期发挥支撑作用。  相似文献

The degradation behavior of porous scaffolds plays an important role in the synthesis of new tissue. In this study, three-dimensional hybrid porous scaffolds of chitosan (CS) comprised of nanostructured carbon (graphene oxide (GO) and single-walled carbon nanohorns (SWCNH)) were prepared by freeze-drying method. In-vitro degradation behavior of scaffolds was investigated up to 8 weeks in phosphate buffer saline (PBS) solution at 37 °C. The characteristics of scaffolds explored as a function of degradation time include crystalline structure, pore morphology, molecular weight, and wet/dry weight. The pH value of the PBS solution during degradation was also monitored. The study demonstrates for the first time that hybrid chitosan scaffolds with nanostructured carbon (GO and SWCNH) are potentially more stable than pure chitosan scaffolds during the time period required for tissue regeneration. The stability of hybrid scaffolds is attributed to nanostructured carbon that was processed with the objective that it is present in a robust manner via a highly cross-linked dense network structure. The chemical structure of chitosan was disrupted within a short period of two weeks, while disruption occurred in hybrid scaffolds after eight weeks. This was accompanied by a weight loss of ∼28% in pure chitosan and ∼20% in hybrid scaffolds. Furthermore, the degraded products were of low molecular weight in pure chitosan and high molecular weight in hybrid chitosan scaffolds. This led to significant decrease in the pH of solution to ∼6.2 in pure chitosan and to ∼7.2 in hybrid scaffolds. The observations clearly underscore that the introduction of GO and SWCNH via cross-link mechanism in CS is a potentially viable approach to tune the degradation rate of hybrid scaffolds in tissue engineering.  相似文献

A novel process was developed to fabricate biodegradable polymer scaffolds for tissue engineering applications, without using organic solvents. Solvent residues in scaffolds fabricated by processes involving organic solvents may damage cells transplanted onto the scaffolds or tissue near the transplantation site. Poly(L-lactic acid) (PLLA) powder and NaCl particles in a mold were compressed and subsequently heated at 180 degrees C (near the PLLA melting temperature) for 3 min. The heat treatment caused the polymer particles to fuse and form a continuous matrix containing entrapped NaCl particles. After dissolving the NaCl salts, which served as a porogen, porous biodegradable PLLA scaffolds were formed. The scaffold porosity and pore size were controlled by adjusting the NaCl/PLLA weight ratio and the NaCl particle size. The characteristics of the scaffolds were compared to those of scaffolds fabricated using a conventional solvent casting/particulate leaching (SC/PL) process, in terms of pore structure, pore-size distribution, and mechanical properties. A scanning electron microscopic examination showed highly interconnected and open pore structures in the scaffolds fabricated using the thermal process, whereas the SC/PL process yielded scaffolds with less interconnected and closed pore structures. Mercury intrusion porosimetry revealed that the thermally produced scaffolds had a much more uniform distribution of pore sizes than the SC/PL process. The utility of the thermally produced scaffolds was demonstrated by engineering cartilaginous tissues in vivo. In summary, the thermal process developed in this study yields tissue-engineering scaffolds with more favorable characteristics, with respect to, freedom from organic solvents, pore structure, and size distribution than the SC/PL process. Moreover, the thermal process could also be used to fabricate scaffolds from polymers that are insoluble in organic solvents, such as poly(glycolic acid). We investigated the in vitro degradation behavior of the electrospun silk fibroin (SF) scaffolds by protease XIV. Phosphate-buffered saline (PBS) without enzyme was used as a control. About 65% of the electrospun SF scaffolds were degraded within 24 d in protease XIV, while almost no scaffolds were degraded in PBS. A great deal of fragments was visible in protease XIV solution. SEM indicated surface erosion of the scaffolds increased during protease degradation with increasing exposure time. FTIR and XRD indicated the crystalline structure of the scaffolds decreased after protease degradation for 24 d, and a degradation mechanism was proposed. Furthermore, the results of the in vivo degradation by implantation in rats showed that the scaffolds were completely degraded in vivo after implantation for 8 weeks and well tolerated by the host animals. The insights gained in this study can serve as a guide to match desired degradation behavior with specific applications for the electrospun SF scaffolds, such as tissue engineering and drug delivery.  相似文献

The material surface must be considered in the design of scaffolds for bone tissue engineering so that it supports bone cells adhesion, proliferation and differentiation. A biomimetic approach has been developed as a 3D surface modification technique to grow partially carbonated hydroxyapatite (the bonelike mineral) in prefabricated, porous, polymer scaffolds using a simulated body fluid in our lab. For the rational design of scaffolding materials and optimization of the biomimetic process, this work focused on various materials and processing parameters in relation to apatite formation on 3D polymer scaffolds. The apatite nucleation and growth in the internal pores of poly(L-lactide) and poly(D,L-lactide) scaffolds were significantly faster than in those of poly(lactide-co-glycolide) scaffolds in simulated body fluids. The apatite distribution was significantly more uniform in the poly(L-lactide) scaffolds than in the poly(lactide-co-glycolide) scaffolds. After incubation in a simulated body fluid for 30 d, the mass of poly(L-lactide) scaffolds increased approximately 40%, whereas the mass of the poly(lactide-co-glycolide) scaffolds increased by about 15% (see Figure). A higher ionic concentration and higher pH value of the simulated body fluid enhanced apatite formation. The effects of surface functional groups on apatite nucleation and growth were found to be more complex in 3D scaffolds than on 2D films. Surprisingly enough, it was found that carboxyl groups significantly reduced the apatite formation, especially on the internal pore surfaces of 3D scaffolds. These findings are critically important in the rational selection of materials and surface design of 3D scaffolds for mineralized tissue engineering and may contribute to the understanding of biomineralization as well.SEM micrograph of a poly(L-lactide) scaffold.  相似文献

Results of the preparation of biodegradable porous scaffolds using an original modification of a wet phase inversion method were presented. Influence of gelatin non‐woven as a non‐classic pore precursor and polyvinylpyrrolidone, Pluronic as classic pore precursors on the structure of obtained scaffolds was analyzed. It was shown that the addition of gelatin non‐wovens enables the preparation of scaffolds, which allow for the growth of cells (size, distribution, and shape of pores). Mechanical properties of the obtained cell scaffolds were determined. The influence of pore precursors on mass absorption of scaffolds against isopropanol and plasma was investigated. Interaction of scaffolds with a T‐lymphocyte line (Jurkat) and with fibroblasts (L929) was investigated. Obtained scaffolds are not cytotoxic and can be used as implants, for example, the regeneration of cartilage tissue.  相似文献

3D porous scaffolds fabricated from binary and ternary blends of silk fibroin (SF), gelatin (G), and hyaluronan (HA) and crosslinked by the carbodiimide coupling reaction were developed. Water-stable scaffolds can be obtained after crosslinking, and the SFG and SFGHA samples were stable in cell culture medium up to 10 days. The presence of HA in the scaffolds with appropriate crosslinking conditions greatly enhanced the swellability. The microarchitecture of the freeze-dried scaffolds showed high porosity and interconnectivity. In particular, the pore size was significantly larger with an addition of HA. Biological activities of NIH/3T3 fibroblasts seeded on SFG and SFGHA scaffolds revealed that both scaffolds were able to support cell adhesion and proliferation of a 7-day culture. Furthermore, cell penetration into the scaffolds can be observed due to the interconnected porous structure of the scaffolds and the presence of bioactive materials which could attract the cells and support cell functions. The higher cell number was noticed in the SFGHA samples, possibly due to the HA component and the larger pore size which could improve the microenvironment for fibroblast adhesion, proliferation, and motility. The developed scaffolds from ternary blends showed potential in their application as 3D cell culture substrates in fibroblast-based tissue engineering.  相似文献

Nanostructured biocomposite scaffolds of poly(l-lactide) (PLLA) blended with collagen (coll) or hydroxyapatite (HA), or both for tissue engineering application, were fabricated by electrospinning. The electrospun scaffolds were characterized for the morphology, chemical and tensile properties by scanning electron microscopy (SEM), X-ray photoelectron spectroscopy (XPS), water contact angle (WCA), Fourier transform infrared (FTIR) measurement, and tensile testing. Electrospun biocomposite scaffolds of PLLA and collagen or (and) HA in the diameter range of 200-700 nm mimic the nanoscale structure of the extracellular matrix (ECM) with a well-interconnection pore network structure. The presence of collagen in the scaffolds increased their hydrophility, and enhanced cell attachment and proliferation, while HA improved the tensile properties of the scaffolds. The biocompatibility of the electrospun scaffolds and the viability of contacting cells were evaluated by 4',6-diamidino-2-phenylindole dihydrochloride (DAPI) nuclear staining and by fluorescein diacetate (FDA) and propidium iodide (PI) double staining methods. The results support the conclusion that 293T cells grew well on composite scaffolds. Compared with pure PLLA scaffolds a greater density of viable cells was seen on the composites, especially the PLLA/HA/collagen scaffolds.  相似文献

This paper reports dual-function (high cell attachment and cell viability) fibrous scaffolds featuring aligned fibers, displaying good biocompatibility and no cytotoxicity. These scaffolds are fabricated through the electrospinning of a co-polypeptide comprising molar equivalents of N6-carbobenzyloxy-l -lysine and γ-benzyl-l -glutamate, with the lysine moieties enhancing cell adhesion and the neural-stimulating glutamate moieties improving cell viability. These new scaffolds allow neural cells to attach and grow effectively without any special surface treatment or coating. Pheochromocytoma (PC-12) cells grown on these scaffolds exhibit better neuronal activity and longer neurite length, relative to those grown on scaffolds prepared from their respective homo-polypeptides. When the scaffolds are partially hydrolyzed such that they present net positive charge and increased hydrophilicity, the cell viability and neurite growth both increase further. Accordingly, these novel co-polypeptide fibrous scaffolds have potential applications in neural tissue engineering.  相似文献

In this study, orthogonal experiments were designed to explore the optimal process parameters for preparing polycaprolactone(PCL) scaffolds by the near-field direct-writing melt electrospinning(NFDWMES) technology. Based on the optimal process parameters, the PCL scaffolds with different thicknesses, gaps and structures were manufactured and the corresponding hydrophilicities were characterized. The PCL scaffolds were modified by chitosan (CS) and hyaluronic acid(HA) to improve biocompatibility and hydrophilicity. Both Fourier transform infrared spectroscopy(FTIR) analysis and antibacterial experimental results show that the chitosan and hyaluronic acid adhere to the surface of PCL scaffolds, sugges-ting that the modification plays a positive role in biocompatibility and antibacterial effect. The PCL scaffolds were then employed as a carrier to culture cells. The morphology and distribution of the cells observed by a fluorescence microscope demonstrate that the mo-dified PCL scaffolds have good biocompatibility, and the porous structure of the scaffolds is conducive to adhesion and deep growth of cells.  相似文献

In the current study, a porous 3D scaffold using Gallium-Apatite/chitin/pectin (Ga-HA/C/P) nanocomposites scaffolds (NCS) were fabricated by freeze-drying process with applications in orthopedics (bone tissue engineering). Various NCSs (0%, 30%, 50 and 70%) were prepared and characterized for its chemical structure, crystalline phase, surface texture by using various techniques such as FT-IR, XRD and SEM-EDX, respectively. The analyses of physicochemical properties proved that the formulated scaffolds were highly porous, and mechanically stable with superior density. The nanocomposite scaffolds also presented with increased swelling ability, lower biodegradation rate and higher mechanical strength. Further, biocompatibility and cytotoxicity of Ga-HA/C/P nanocomposite scaffolds were studied using NIH3T3 cells and MG-63 cells revealed no toxicity and cells attached and proliferated on scaffolds. Further implantation of prepared NCS showed mature bone formation through formation of new bone cells and osteoblast differentiation. Also, Ga-HA/C/P nanocomposites scaffolds proved to be more effective than chitin-pectin composite scaffolds. Taking results together it can be inferred that the prepared nanocomposite scaffolds possesses the prerequisites and showed great potential for treating orthopedic applications.  相似文献

The objective of this study is to design a novel kind of scaffolds for blood vessel and nerve repairs. Random and aligned nanofibrous scaffolds based on collagen-chitosan-thermoplastic polyurethane (TPU) blends were electrospun to mimic the componential and structural aspects of the native extracellular matrix, while an optimal proportion was found to keep the balance between biocompatibility and mechanical strength. The scaffolds were crosslinked by glutaraldehyde (GTA) vapor to prevent them from being dissolved in the culture medium. Fiber morphology was characterized using scanning electron microscopy (SEM) and atomic-force microscopy (AFM). Fourier transform infrared spectroscopy (FTIR) showed that the three-material system exhibits no significant differences before and after crosslinking, whereas pore size of crosslinked scaffolds decreased drastically. The mechanical properties of the scaffolds were found to be flexible with a high tensile strength. Cell viability studies with endothelial cells and Schwann cells demonstrated that the blended nanofibrous scaffolds formed by electrospinning process had good biocompatibility and aligned fibers could regulate cell morphology by inducing cell orientation. Vascular grafts and nerve conduits were electrospun or sutured based on the nanofibrous scaffolds and the results indicated that collagen-chitosan-TPU blended nanofibrous scaffolds might be a potential candidate for vascular repair and nerve regeneration.  相似文献

骨形态发生蛋白-2(BMP-2)的缓释载体一直是骨组织工程中的研究热点.本研究通过化学改性制备了两种肝素化丝素支架,并浸渍吸附BMP-2,研究了BMP-2在不同丝素支架样品上的吸附能力、体外释放性能及其对人骨肉瘤细胞MG-63碱性磷酸酶活性(ALP)的影响.结果表明,肝素化丝素支架对BMP-2具有较强的吸附能力,并能保持其体外缓慢释放性能;MG-63细胞在肝素化支架上生长状态良好,并具有显著的增殖能力,负载BMP-2后的肝素化支架能显著促进MG-63细胞的分化.因此,肝素化丝素支架是一种较理想的BMP-2缓释载体.  相似文献

Identification of meaningful chemical patterns in the increasing amounts of high-throughput-generated bioactivity data available today is an increasingly important challenge for successful drug discovery. Herein, we present the scaffold network as a novel approach for mapping and navigation of chemical and biological space. A scaffold network represents the chemical space of a library of molecules consisting of all molecular scaffolds and smaller "parent" scaffolds generated therefrom by the pruning of rings, effectively leading to a network of common scaffold substructure relationships. This algorithm provides an extension of the scaffold tree algorithm that, instead of a network, generates a tree relationship between a heuristically rule-based selected subset of parent scaffolds. The approach was evaluated for the identification of statistically significantly active scaffolds from primary screening data for which the scaffold tree approach has already been shown to be successful. Because of the exhaustive enumeration of smaller scaffolds and the full enumeration of relationships between them, about twice as many statistically significantly active scaffolds were identified compared to the scaffold-tree-based approach. We suggest visualizing scaffold networks as islands of active scaffolds.  相似文献
