Tandem mass spectrometry has been applied to differentiate three sets of o-, m- and p-methyl, -methoxy and -nitro-substituted-6-phenyl-dibenzo(d,f)(1,3)dioxepines. Collision-induced dissociation (CID) experiments have been carried out on 2-phenylbenzo[b]furan fragment ions, which originate from the decomposition of the molecular ions after their EI-induced isomerization to spirocyclic structures. With the exception of m- and p-methylphenylbenzo[b]furan isomers, which display identical CID mass spectra, the three isomeric methoxy- and nitrophenylbenzo[b]furan fragment ions display very characteristic CID behavior which allows unequivocal differentiation of the 6-phenyl-dibenzo(d,f)(1,3)dioxepine isomers. 6-(o-nitrophenyl)-dibenzo(d,f)(1,3)dioxepine isomer, does not form a 2-(o-nitrophenyl)benzo[b]furan ion and, therefore, it can be differentiated from the m- and p- isomers based on the mere EI mass spectra. Furthermore, it shows a characteristic ion most likely due to an ortho effect between the nitro group and the dioxepine ring. Multiple stage mass spectrometric techniques (MSn), labeled derivatives and reference compounds were used in order to gain additional information on the structures of product ion from the CID fragmentation.  相似文献

Naphtho[2,1‐b]furan‐2‐yl)(8‐phenylpyrazolo[5,1‐c][1,2,4]triazin‐3‐yl)methanone, ([1,2,4]triazolo[3,4‐c][1,2,4]triazin‐6‐yl)(naphtho[2,1‐b]furan‐2‐yl)methanone, benzo[4,5]imidazo[2,1‐c][1,2,4]triazin‐3‐yl‐naphtho[2,1‐b]furan‐2‐yl‐methanone, 5‐(naphtho[2,1‐b]furan‐2‐yl)pyrazolo[1,5‐a]pyrimidine, 7‐(naphtho[2,1‐b]furan‐2‐yl)‐[1,2,4]triazolo[4,3‐a]pyrimidine, 2‐naphtho[2,1‐b]furan‐2‐yl‐benzo[4,5]imidazo[1,2‐a]pyrimidine, pyridine, and pyrazole derivatives are synthesized from sodium salt of 5‐hydroxy‐1‐naphtho[2,1‐b]furan‐2‐ylpropenone and various reagents. The newly synthesized compounds were elucidated by elemental analysis, spectral data, chemical transformation, and alternative synthetic route whenever possible. J. Variable benzo[b]furan derivatives having (E)- and (Z)-2-alkylcarbamoyl-1-methylvinyl groups at the 2-, 4- and 5-positions and a carboxylpropoxy or (1-phenyl)ethoxy group at the 7-position were prepared to find novel and selective leukotriene B4(LTB4) receptor antagonists. (E)-2-(2-diethylcarbamoyl-1-methylvinyl)-7-(1-phenylethoxy)benzo[b]furan (4v) showed selective inhibition to the human BLT2 receptor (hBLT2). On the other hand, (E)-2-acetyl-4-(2-diethylcarbamoyl-1-methylvinyl)-7-(1-phenylethoxy)benzo[b]furan (7v) inhibited both human BLT(1) receptor (hBLT1) and hBLT2. The (E)-2-(2-diethylcarbamoyl-1-methylvinyl) group lay on approximately the same plane as the benzo[b]furan ring, whereas the (E)-4-(2-diethylcarbamoyl-1-methylvinyl) group had the torsion angle (45.7 degree) from the benzo[b]furan ring plane. However, the (Z)-(2-alkylcarbamoyl-1-methylvinyl)benzo[b]furans were inactive. The inhibitory activity depended on the conformation of the 2-diethylcarbamoyl-1-methylvinyl group.  相似文献

The reaction of 2-lithiated benzo[b]thiophene with 8 equiv of elemental sulfur was found to give pentathiepino[6,7-b]benzo[d]thiophene. In contrast, treatment of 2-lithiated benzo[b]furan with sulfur under similar conditions produced the interesting ring system bis(benzo[4,5]furo)[2,3-e:3',2'-g][1,2,3,4]tetrathiocine. Both of these new cyclic polysulfides were studied by X-ray crystallography. Two polymorphic forms of pentathiepino[6,7-b]benzo[d]thiophene were found, displaying similar conformations but different packing schemes, which was also evident from powder diffraction data.  相似文献

Under acid catalyzed acylation 3-acetamidobenzo[b]furan, -benzo[b]tkiophene, and indole undergo heterocyclization to condensed 1,3-oxazinium salts. Novel 3-acetoacetylaminobenzo[b]furan derivatives having a modified triene system at the 3-position were synthesized starting with 3-aminobenzo[b]furans. The enol isomers, 3-[(3-hydroxybut-2-enonyl)amino]benzo[b]furans (), of the 3-acetoacetylaminobenzo[b]furans were obtained as stable isomers owing to formation of a hydrogen bonding between the enol hydroxyl group and the amidocarbonyl group. The planarity of the C-2 substituent through the C-3 side chain suggested the existence of a modified conjugational triene system in the enol compound. Cysteinyl leukotriene 1 and 2 receptor antagonistic activities for these compounds were evaluated. 2-(4-Cyanobenzoyl or ethoxycarbonyl)-3-[(2-cyano-3-hydroxybut-2-enonyl)amino]benzo[b]furans (, ) were moderately active.  相似文献

The photochemical behavior of benzo[b]thiophene is different from that of thiophene. The former does not undergo photoisomerization and is not converted into an indole when irradiated in the presence of a primary amine; its photochemical behavior resembles that of naphthalene. Thus photoexcited benzo[b]thiophene gives adducts with primary amines (1(H)), secondary amines (2), and pyrrole (3(H)): an exciplex is an intermediate. The existence of the exciplex is supported by the formation of aldehyde 7 when benzo[b]thiophene is irradiated in aqueous propylamine, the finding that photoexcited benzo[b]thiophene does not react with an alcohol and that photoexcited indole benzo[b]furan, 2-methylbenzo[b]thiophene, and 3-methylbenzo[b]thiophene do not react with propylamine or with piperidine. The results are interpreted in terms of spin densities calculated for the anion radicals of the compounds under study.  相似文献

Rates of the reversible deprotonation of benzo[b]-2,3-dihydrofuran-2-one (6H-O) and benzo[b]-2,3-dihydrothiophene-2-one (6H-S) by OH-, primary aliphatic amines, secondary alicyclic amines, and carboxylate ions have been determined in water at 25 degrees C. As noted earlier by Kresge and Meng, 6H-S (pKa = 8.82) is considerably more acidic than 6H-O (pKa = 11.68), which mainly reflects the greater aromatic stabilization of the conjugate base of 6H-S (thiophene derivative) compared to that of 6H-O (furan derivative). The main focus of this paper is to assess how the difference in the aromaticity of the two enolate ions affects the intrinsic barrier to the proton transfer. These intrinsic barriers were determined from Br?nsted plots for the reactions with the amines and carboxylate ions or calculated on the basis of the Marcus equation for the reactions with OH-. They are consistently somewhat higher for the reactions of 6H-S than for the reactions of 6H-O, implying that the aromaticity in the anion enhances the intrinsic barrier. This contrasts with a previous report on the deprotonation of some cyclic rhenium Fischer-type carbene complexes where the reaction that leads to the most aromatic conjugate base (thiophene derivative) has a lower intrinsic barrier than the reaction that leads to the less aromatic furan analogue. We are offering a detailed analysis of other potential factors that may affect the intrinsic barriers and which could explain these contradictory results.  相似文献

The reaction of 3-NHR-isoquinolin-1(2H)-ones (R = Ar) with aromatic aldehydes in the presence of Me3SiCl or in acetic acid leads to the formation of derivatives of dibenzo[b,f][1, 8]naphthyridin-5(6H)- one and benzo[f]isoquino[3,4-b][1, 8]naphthyridine-5,9(6H,7H)-dione. The reaction for R = Het in the presence of Me3SiCl gives derivatives of 5H-pyrido[1',2':1,2]pyrimido[4,5-c]isoquinolin-5-one, benzo[f]isoquinoline[3,4-b][1,8]naphthyridine-5,9[6H,7H]-dione, and derivatives of new heterocyclic systems, 5H-pyrazino[1',2':1,2]pyrimido[4,5-c]isoquinolin-5-one, 5H-[1,3]thiazolo[3',2':1,2]pyrimido- [4,5-c]isoquinolin-5-one, 5-H-benzo[f]pyrazolo[3,4-b][1,8]naphthyridin-5-one, and isoquino[3,4-b]- [1,5]naphthyridin-5(6H)-one. The effect of the structure of substituent R and nature of the substituent in the benzaldehydes on the structure of the reaction products was studied.  相似文献

The reaction of 6‐(4‐chlorophenyl)‐5‐nitrosoimidazo[2,1‐b][1,3]thiazole 1b , 6‐(4‐chlorophenyl)‐2‐methyl‐5‐nitrosoimidazo[2,1‐b][1,3]thiazole 1c , 6‐(4‐chlorophenyl)‐2,3‐dimethyl‐5‐nitrosoimidazo‐[2,1‐b][1,3]thiazole 1d and 2‐(4‐chlorophenyl)‐3‐nitrosobenzo[d]imidazo[2,1‐b][1,3]thiazole 1e with hydrochloric acid has been carried out in order to investigate the effect of substituents on the thiazole ring in a recently reported ring‐ring interconversion reaction. In every case the corresponding [1,4]‐thiazino[3,4‐c][1,2,4]oxadiazol‐3‐ones 2b‐e have been obtained. In particular, the benzoderivative 1e furnished the 4‐(4‐chlorophenyl)‐4‐hydroxy‐4H‐benzo[5,6][1,4]thiazino[3,4‐c][1,2,4]oxadiazol‐1‐one 2e , containing a new tricyclic system with a quasi‐planar geometry whose pharmacological potentialities appear promising.  相似文献

Condensation of 3-hydroxy-2-naphthalenecarboxylic acid with phloroglucinol afforded 1,3-dihydroxy-12H-benzo[b]xanthen-12-one. Construction of an additional dimethylpyran ring onto this skeleton, by alkylation with 3-chloro-3-methyl-1-butyne followed by Claisen rearrangement, gave access to a series of benzo[b]pyrano[2,3-i]xanthen-6-ones and benzo[b]pyrano[3,2-h]xanthen-7-ones related to psorospermine and benzo[b]acronycine. In contrast with what is observed in the pyridoacridone and benzopyridoacridone series, the linear benzo[b]-pyrano[2,3-i]xanthen-6-one derivatives were more potent than their angular benzo[b]pyrano[3,2-h]xanthen-7-one isomers. cis-3,4-Diacetoxy-5-methoxy-2,2-dimethyl-3,4-dihydro-2H,6H-benzo[b]pyrano[2,3-i]xanthen-6-one, the most active among the new compounds, was more potent than acronycine in inhibiting the proliferation of L1210 murine leukemia cells.  相似文献

Enzymatic cis-dihydroxylation of benzo[b]thiophene, benzo[b]furan and several methyl substituted derivatives was found to occur in both the carbocyclic and heterocyclic rings. Relative and absolute configurations and enantiopurities of the resulting dihydrodiols were determined. Hydrogenation of the alkene bond in carbocyclic cis-dihydrodiols and ring-opening epimerization/reduction reactions of heterocyclic cis/trans-dihydrodiols were also studied. The relatively stable heterocyclic dihydrodiols of benzo[b]thiophene and benzo[b]furan showed a strong preference for the trans configuration in aqueous solutions. The 2,3-dihydrodiol metabolite of benzo[b]thiophene was utilized as a precursor in the chemoenzymatic synthesis of the unstable arene oxide, benzo[b]thiophene 2,3-oxide.  相似文献

Several 2-alkylcarbamoyl-1-alkylvinylbenzo[b]furans were designed to find a selective leukotriene B4 (LTB4) receptor antagonist. 2-(2-Alkylcarbamoyl-1-alkylvinyl)benzo[b]furans having a substituent group at the 3-position, 4-(2-alkylcarbamoyl-1-methylvinyl)benzo[b]furans having a substituent group at the 3-position, and 7-(2-alkylcarbamoyl-1-methylvinyl)benzo[b]furans and 3-(2-alkylcarbamoyl-1-alkylvinyl)benzo[b]furans were prepared and evaluated for LTB4 receptor (BLT1 and BLT2) inhibitory activities. (E)-3-Amino-4-[2-[2-(3,4-dimethoxyphenyl)ethylcarbamoyl]-1-methylvinyl]benzo[b]furan ((E)-17c) showed potent and selective inhibitory activity for BLT2. On the other hand, (E)-7-(2-diethylcarbamoyl-1-methylvinyl)benzo[b]furan ((E)-27a) showed potent inhibitory activity for both BLT1 and BLT2.  相似文献

Neutral pentafluorophenyl benzoquinolinyl PtII [Pt(bzq)(HC^N−κN)(C6F5)] ( 1 a – g ) complexes, bearing nonmetalated N-heterocyclic HC^N ligands [HC^N=2,5-diphenyl-1,3,4-oxadiazole (Hoxd) a , 2-(2,4-difluorophenyl)pyridine (dfppy) b , 2-phenylbenzo[d]thiazole (pbt) c , 2-(4-bromophenyl)benzo[d]thiazole (Br-pbt) d , 2-phenylquinoline (pq) e , 2-thienylpyridine (thpy) f , 1-(2-pyridyl)pyrene (pypy) g ], and heteroleptic bis(cyclometalated) PtIV fac-[Pt(bzq)(C^N)(C6F5)Cl] ( 2 b – g , bzq: benzo[h]quinolinyl) derivatives, generated by oxidation of 1 b – g with PhICl2, are reported. The oxidation reaction of 1 a evolved with formation of the bimetallic PtIV complex syn-[Pt(bzq)(C6F5)Cl(μ-OH)]2 3 . The crystal structures of 1 a,d,f , 2 b,d,e and 3 were corroborated by X-ray crystallography. A comparative study of the absorption and photoluminescence properties of the two series of complexes PtII ( 1 ) and PtIV ( 2 ), supported by time-dependent DFT calculations (TD-DFT), is presented. The low-lying transitions (absorption and emission) of PtII complexes 1 a – e [solution and polystyrene (PS) films] were assigned to the IL/MLCT mixture located on the cyclometalated Pt(bzq) unit, with minor IL′/ML′CT/LL′CT contributions involving the non-metalated ligand. Complex 1 g , bearing the more delocalized pyridyl pyrene (Hpypy) as an ancillary ligand, shows dual 1ππ* and 3ππ* (Hpypy) emission in fluid CH2Cl2 and dual 3IL/3MLCT [Pt(bzq)] and [3ππ*, Hpypy] phosphorescence at 77 K. Upon oxidation, PtIV complexes 2 b – f display (solution, PS) ligand-based phosphorescence that arises from the bzq in 2 b (3LC) or from the second C^N ligand in 2 c – f (3L′C) with some 3LL′CT in 2 f . Despite metalation of the pyrenyl group, 2 g exhibits dual emission 1ππ*/3ππ* located on the pypy chromophore.  相似文献

Condensation of hetarene carboxaldehydes with phthalide gave 2-(3-hydroxy-1-oxoinden-2-yl)benzo[b]furan and 2-(3-hydroxy-1-oxoinden-2-yl)-5-ethylthieno[2,3-b]thiophene. Starting from hetaryl acetic acids gave 3-(3-hydroxy-1-oxoinden-2-yl)benzo[b]furan and 3-(3-hydroxy-1-oxoinden-2-yl)benzo[b]thiophene. Acylation of 3-hydroxy-1-oxoinden-2-yl-substituted heterocycles using acetic anhydride in the presence of 70% HClO4 leads to the formation of pentacyclic pyrilium salts. Pentacyclic indenopyridines are prepared by treating the pyrilium salts with ammonia. 针对二苯并[d,f][1,3]二噁烷传统制备方法中易产生二羟基联苯乙烯副产物的缺点,本文以路易斯酸性胍盐离子液体为催化剂,2,2'-二羟基联苯和端基炔为原料,高区域选择性的合成了二苯并[d,f][1,3]二噁烷,收率39%~84%。 本方法适用于各种端基炔。路易斯酸性胍盐离子液体循环利用5次,活性不变。  相似文献

Deprotonation of benzoxazole, benzothiazole, benzo[b]thiophene, benzo[b]furan, N-Boc-protected indole and pyrrole, and N-phenylpyrazole using an in situ mixture of ZnCl(2).TMEDA (0.5 equiv) and lithium 2,2,6,6-tetramethylpiperidide (1.5 equiv) in THF at room temperature is described. The reaction was evidenced by trapping with iodine, regioselectively giving the expected functionalized derivatives in 52-73% yields. A mixture of mono- and disubstituted derivatives was obtained starting from thiazole. Cross-coupling reactions of 2-metalated benzo[b]thiophene and benzo[b]furan with heteroaromatic chlorides proved possible under palladium catalysis. A reaction pathway where the lithium amide and zinc diamide present in solution behave synergically was proposed for the deprotonation reaction, taking account of NMR and DFT studies carried out on the basic mixture.  相似文献

A number of isomeric mercapto aldimines of benzo[b]furan have been obtained and their structure and properties have been studied in comparison with their benzo[b]-thiophene analogs. It has been shown that the nature of the heterocycle has no substantial influence on the structure and properties of these compounds, in contrast to the initial mercapto and hydroseleno aldehydes. In the present study, it has been demonstrated that polymers containing pendant hydroxyl groups can undergo photoinduced crosslinking upon irradiation in the presence of bifunctional benzodioxinone. Irradiation of the copolymers of 2‐hydroxyethyl methacrylate (HEMA) and methyl methacrylate (MMA) in dichloromethane and THF solutions or in films containing bisbenzodioxinone, namely 5‐[9‐(4‐oxo‐2,2‐diphenyl‐4H‐benzo[d][1,3]dioxin‐5‐yloxy) nonyloxy]‐2,2‐diphenyl‐4H‐benzo[d][1,3]dioxin‐4‐one resulted in the formation of insoluble networks. Model studies using monofunctional napthodioxinone, namely 7‐hydroxy‐2,2‐diphenyl‐4H‐naphtho[2,3‐d][1,3]dioxin‐4‐one, revealed that crosslinking occurs via inter‐chain ester formation between photochemically generated keten intermediates and pendant hydroxyl groups.
