The mechanism for inhibition of the Pseudomonas aeruginosa arginine deiminase (PaADI) by the arginine analogue l-canavanine was investigated. Inhibition by this substance (kinact = 0.31 +/- 0.03 min-1 and Ki = 1.7 +/- 0.5 mM) is associated with the formation of a modestly stable S-alkylthiouronium intermediate, detected by using kinetic techniques and identified by using electrospray ionization mass spectrometry. The electronic and/or orientation effects, caused by oxygen-for-methylene substitution in l-canavanine, on the rate of enzyme regeneration from the S-alkylthiouronium intermediate could serve as the basis for a strategy for the rational design of new slow substrate inhibitors of ADI.  相似文献

Inactivation of prion deposits on medical devices or prion contamination in pharmaceutical raw materials is considered as impossible by using gamma irradiation. Early, the guideline WHO/CDS/CSR/APH/2000 has described irradiation as an ineffective process. But, in 2003, S. Miekka et al. noted radiation inactivation of prions in a particular application to purify human albumin, shown by the physical denaturation of the infectious protein (PrP). The aim of our study was to determine the inactivation of prions with a scrapie model (strain C506M3) by irradiating standardised preparations. Results: Gamma irradiation was partially effective, showing a 4–5 log reduction on exposure to 50 kGy. A characteristic effect–dose curve was not observed (25, 50 and 100 kGy), only an increase in the incubation period of the murine disease (229 days with 25 kGy to 290 days with 100 kGy) compared with 170 days without irradiation. Since the inactivation was not a total one, the observed effect is significant. It is proposed that further work be undertaken with the model to investigate the application of gamma radiation known levels of prion contamination.  相似文献

Influenza virus is readily transmitted by aerosols and its inactivation in the environment could play a role in limiting the spread of influenza epidemics. Ultraviolet radiation in sunlight is the primary virucidal agent in the environment but the time that influenza virus remains infectious outside its infected host remains to be established. In this study, we calculated the expected inactivation of influenza A virus by solar ultraviolet radiation in several cities of the world during different times of the year. The inactivation rates reported here indicate that influenza A virions should remain infectious after release from the host for several days during the winter "flu season" in many temperate-zone cities, with continued risk for reaerosolization and human infection. The correlation between low and high solar virucidal radiation and high and low disease prevalence, respectively, suggest that inactivation of viruses in the environment by solar UV radiation plays a role in the seasonal occurrence of influenza pandemics.  相似文献

The goal of this study was to determine the sensitivity of virulent Burkholderia pseudomallei to natural sunlight. We describe solar dosimetry calibrated to integrate radiation between 295 and 305 nm and an exposure system that minimizes thermal effects on bacterial cells. Burkholderia pseudomallei cells were either exposed to sunlight in UV transparent dishes or maintained in the dark covered by opaque foil. The cells maintained in the dark remained at constant levels for the duration of all experiments. The exposed cells nearby were killed with a kinetic studied through 5 Log10 inactivation. We found that cells in stationary phase of growth were nearly two-fold more resistant to sunlight than cells in lag or exponential growth. A virulent strain of B. pseudomallei that produced mucoid colonies showed sensitivity to sunlight similar to both a virulent strain that produced nonmucoid colonies and a strain of B. thailandensis . The inactivation of B. pseudomallei by sunlight in different types of water of environmental relevance or inside amoebae was investigated. The sensitivity of virulent B. pseudomallei was calculated and its comparison with previous studies employing monochromatic germicidal light (254 nm) is discussed . This may be the first report in the open literature of the inactivation of a virulent biological threat agent by natural sunlight. These data should assist in estimating the risk for contracting melioidosis and in predicting the time period during which B. pseudomallei remains infectious after an accidental or intentional release in the environment.  相似文献

Inactivation of bacterial endospores by photocatalytic nanocomposites   总被引:2,自引:0,他引:2  
A novel biocidal photocatalytic nanocomposite, composed of TiO(2) and multi-walled carbon nanotubes (MWNTs), was synthesized via wet chemistry followed by a heat treatment. Uniform anatase coatings on MWNTs were successfully obtained with a thickness of a few nanometers. The nanostructure of the composite was determined by high resolution transmission electron microscopy (HRTEM) and X-ray diffraction (XRD). The needle-like shape of the nanocomposite provided more than three times higher photocatalytic specific surface area than commercial TiO(2) nanoparticles (Degussa P25) when dispersed in water. Moreover, under ultraviolet (UV) radiation the excited electrons can be trapped at the interface between the TiO(2) layer and MWNTs and they can also be scavenged through the conductive graphitic layers. Thus, an intense photochemical reaction yielding a powerful biocide can be expected. Irradiating bacterial endospores (Bacillus cereus) with solar UV lamps in presence of the novel photocatalyst successfully inactivated the spores while solar UV lamps only or solar UV Lamps with Degussa P25 showed no significant inactivating behavior. Performance of photocatalytic nanocomposites was assessed based on time to achieve 90% inactivation of spores (LD(90)) and also in terms of time required to achieve a 1.0 log(10) reduction of spores in the tail region of the inactivation curve.  相似文献

Inactivation of horseradish peroxidase by phenoxyl radical attack   总被引:1,自引:0,他引:1  
To test the hypothesis that horseradish peroxidase (HRP) can be inactivated by phenoxyl radicals upon reaction with H(2)O(2)/phenol, we probed HRP-catalyzed phenol oxidation at various phenol/H(2)O(2) concentrations. To this end the total protein, phenolic product, active protein, and iron concentrations in the aqueous phase were determined by protein assay, phenol-(14)C isotopic labeling, resonance Raman and atomic absorption spectroscopy, respectively. Additionally, resonance Raman and FTIR measurements were carried out to probe possible structural changes of the enzyme during the reaction. The data obtained provide the first experimental support for the hypothesis that HRP can be inactivated by a phenoxyl radical attack. The heme macrocycle destruction involving deprivation of the heme iron occurs as a result of the reaction. An intermediate type of the active protein was observed by Raman difference spectra at low concentrations which features a stabilization of the quantum mixed state of the heme iron and a significant amount of phenoxylphenol-type oligomers in solution and probably also in the heme pocket. This work provides a basis for evaluating the relative contributions of different HRP inactivation mechanisms and is thus critical for optimizing engineering applications involving HRP reactions.  相似文献

This study determined the sensitivity of vaccinia virus, an orthopox virus commonly used as a surrogate for variola virus (etiological agent of smallpox), exposed to UVB radiation emitted by a solar simulator, or to direct natural sunlight. The data obtained indicate that: (1) the virucidal effect of natural sunlight can be mimicked adequately by an artificial light source with similar spectral characteristics in the UVB, (2) viral sensitivity to UVB or to solar radiation can be correlated with experimental data previously obtained with UVC, (3) the correlation factor between virus inactivation by solar radiation (measured at 300 ± 5 nm) and by UVC (254 nm) is between 33 and 60, and (4) the sensitivity of viruses either dry on glass surfaces or in liquid suspension is similar when in the presence of similar amounts of cellular debris and growth media. The findings reported in this study should assist in estimating the threat posed by the persistence of virus during epidemics or after an accidental or intentional release.  相似文献

Two recent crystal structures of acetyl-CoA synthase (ACS) from Moorella thermoacetica exhibited different metal contents and geometries at their active site, called the A-cluster. This led to the proposal of two catalytic mechanisms, one Ni-based, the other Cu-based. ACS was studied with respect to synthase activity, methyl group transfer activity, metal content, and EPR spectroscopy. Our results indicate that Cu is not required for catalysis and that it inactivates ACS by binding to the proximal site of the A-cluster. With Cu in this site, the A-cluster cannot accept a methyl group from the corrinoid-iron-sulfur protein, nor can it exhibit the NiFeC EPR signal after treatment with CO.  相似文献

The main goal of the present study was to investigate the effects of silver and gold ions on cholinesterases (ChEs) activity due to increasing application of these metals in a wide variety of nanomaterials. A chromogenic assay using the substrate o-nitrophenyl acetate/butyrate made it possible to conclude unmistakably that both metals inhibit ChEs. Addition of bovine serum albumin (BSA) indicates that binding of metal ions to albumin could serve to scavenge metals and consequently reduce their amount for reaction with ChEs. The effects of metal ions on ChE should be taken into consideration when using this enzyme as an environmental biomarker.   相似文献

The arginine electrode is based on a coupled enzymatic system consisting of arginase and urease with an ammonia gas sensor; conditions of immobilization are optimized. Arginine in the range 3 × 10?5–3 × 10?3 M gives a linear potential vs. log (concentration) plot with a response time of 5 min over the range specified. Several compounds structurally related to arginine do not interfere. The method is suitable for the determination of arginine in bovine insulin and human serum. Results compare well with values given in the literature or obtained by the conventional Sakaguchi method.  相似文献

The CN stretching vibrations of the guanidyl group in the arginine dipeptide side chain are examined by two-dimensional infrared spectroscopy. In D(2)O, the spectra display two distinct diagonal peaks. These nearly degenerate modes undergo ultrafast energy transfer. The energy-transfer rate was determined directly from the 2D-IR spectra to be 1/2.1 ps(-1). The cross peaks in 2D-IR arising from the energy transfer provide a definitive identification of arginine in larger proteins. An example of arginine in the transmembrane protein M2, found in influenza viruses, is given.  相似文献
