Summary The reaction of malononitrile with -chloro acetanilides1 in presence of potassium carbonate yields the 1-aryl-5-oxo-2-pyrrolin-3-carbonitriles2, in presence of triethylamine the 4,6-diamino-1-aryl-2-oxo-2,3-dihydropyrrolo[2,3-b]pyridin-5-carbonitriles3 are formed. From 2-chloroacetylamino-benzencarbonitrile and malononitrile the 5-amino-1-oxo-1,2-dihydropyrrolo[1,2-a]chinazolin-3-carbonitrile (4) arise. Analogously from the 2-chloroacetylamino-thiophen-3-carbonitries5 the 7,8-dihydro-thieno[3,2-e]pyrrolo[1,2-a]pyrimidine derivatives6 are obtainable. Hydrolysis of2 a by treatment with hydroxide or acid, respectively, yields the 1,1,2-ethanetricarboxylic acid and derivatives9 a,b. Phenyldiazonium salt reacts with2 to form the triazene 7 only.

The Cyclization of 2-(1,1-dicyanovinylamino)-thiophenes2 by treatment with AlCl3 yield 4-amino-5-cyano-thieno[2,3-b]pyridines3. 2-(1-acylvinyl-amino)-3-cyano-thiophenes7, obtainable from 2-amino-3-cyano-thiophenes and -diketones, react in the presence of AlCl3 to form 4-acylamino-thieno[2,3-b] pyridines8. This reaction is connected with the transfer of the acyl group from C- to the N-atom. 4-Amino-5-cyano-thieno[2,3-b]pyridones-(6)11 are synthesized from 2-amino-3-cyano-thiophenes and ethyl cyano acetate in the presence of sodium ethoxide.

Summary 2,3-Dihydro-3-oxo-benzo[b]thiophen-1,1-dioxide (1) reacts as CH-acidic component with amidines of orthoesters and anilines resp. to give the anilinomethylene derivates3, 4, and5. With triethyl orthoformiate the hydroxymethylene-compound7 is obtained. Anilino- and phenylhydrazino derivates8 and9 prove the carbonyl activity of1, azo-coupling leads to10, whereas treatment of1 with sulfur and malononitrile yields the benzo[b]thieno[2,3-d]thiophenes11. Introduction of substituents with active NH-functions in position 2 of the dicyanomethylene-product2, such as azocoupling, reaction with phenyl isocyanate and formation of enamines, leads to ring closure reactions between a nitrile and the NH-group. Thereby the phenyl-benzo[4,5]thieno[2,3-c]pyridazines12, the phenyl-1H-benzo[4,5]thieno[2,3-c]pyridines13 and the phenylamino-benzo[4,5]thieno[2,3-c]pyridines14 are obtained.13C and15N-NMR spectroscopy was used as proof of the ring closure reactions.

Summary Different functionalized alkyl 3-oxo-butyrates (2) were reacted with 5-amino-3-Q-1H-1,2,4-triazoles (1) to yield3 and4 type 1,2,4-triazolo[1,5-a]pyrimidinones. In case of2 (R 1=methyl,R 2=1-ethoxycarbonylethyl,R 3=ethyl) beside the corresponding derivative4 the unexpected 5,6-dihydro-6,8-dimethyl-7-ethoxycarbonyl-3-methylthio-1,2,4-triazolo[4,3-a]-1,3-diazepin-5(9H)-one (7) was isolated, representing a novel ring system.

Zusammenfassung Verschiedene funktionalisierte 3-Oxo-buttersäurealkylester (2) wurden mit 5-Amino-3-Q-1H-1,2,4-triazolen (1) umgesetzt, wobei 1,2,4-Triazolo[1,5-a]pyrimidinone der Typen3 und4 erhalten wurden. Im Fall von2 (R 1=Methyl,R 2=1-Ethoxycarbonylethyl,R 3=Ethyl) wurde neben dem erwarteten Derivat4 das unerwartete 5,6-Dihydro-6,8-dimethyl-7-ethoxycarbonyl-3-me-thylthio-1,2,4-triazolo[4,3-a]-1,3-diazepin-5(9H)-on (7) isoliert, welches ein neues Ringsystem darstellt.

Methods for the preparation of synthons for syntheses of spiro[2.4]heptane analogues of prostaglandins are described. Two of them (1a and1b) enable the syntheses of 11-deoxy-type compounds and were prepared from spiro[2.4]heptan-4-one (3) which after transformation into the 5-phenylthio-,-unsaturated ketone5 was subjected to conjugate addition of organocuprate reagent6. The third synthon (2)-a potential intermediate in syntheses of complete spiro[2.4]heptane analogues of prostaglandins-was prepared from the bicyclic ketone10 byBaeyer-Villiger oxidation followed by epoxidation.

Zusammenfassung Es werden Synthesewege für Spiro[2.4]hepane als Analoge zu Prostaglandinen beschrieben. Zwei davon (1a und1b) ermöglichen die Synthese von Verbindungen des 11-Deoxy-Typs; sie wurden aus Spiro[2.4]heptan-4-on (3) dargestellt, das nach der Umwandlung zum 5-phenylthio-,-ungesättigten Keton5 einer konjugierten Addition von Organocuprat-Reagens6 unterworfen wurde. Das dritte (2), ein potentielles Zwischenprodukt in der Synthese von vollständigen Spiro[2.4]heptan-Analogen zu Prostaglandinen, wurde aus dem bicyclischen Keton10 durchBaeyer-Villiger-Oxidation gefolgt von einer Epoxidierung dargestellt.

Summary The enamino nitriles1 and2 react with phenyl isothiocyanate followed by cyclization with -haloketones3 and10 to afford in each case the thiazole5, thiophene11 and the thieno[2,3-b]pyridine derivatives19 and21. Chemical and spectroscopic evidences for the structures of the new compounds are described.

Chemische und spektroskopische Untersuchungen wurden als Strukturbeweise für die neuen Verbindungen herangezogen.

Abstract

The reaction of thionyl chloride with amidines 2, derived from N-benzimidazol-2-yl imidates 1, leads to [1,2,4,6]thiatriazino[2,3-a][1,3]benzimidazol-1(2H)-one 3 in good yields. [1,3,5]Thiadiazino[3,4-a][1,3]benzimidazol-2-imine 4 was prepared by condensation of NaSCN with benzimidazol-2-yl imidate 1. The isolated compounds 3 and 4 were identified by spectroscopic methods including IR, 1H NMR, and 13C NMR as well as elemental analyses and MS of 3d and 4b.

Summary The synthesis of compound3 by condensing 1H-benzimidazole-2-acetonitrile (1) with ethyl cyclopentanone-2-carboxylate (2) in the presence of ammonium acetate is described. Methylation of3 with trimethyl phosphate yielded the N-methyl derivative4. Methods for converting3 to some of its related derivatives in which the carbonyl function was replaced by Cl, N3 and amines are also reported.

Die Methylierung von3 mit Trimethylphosphat liefert das N-Methylderivat4. Die Sauerstoffunktion in3 kann durch Chlor, Azid und Aminogruppen ersetzt werden.

Summary The direct intramolecular acylation and alkylation of the phenylsulfonylmethyl-group in the -lactames5a and7 are not successful. The postulated intermediate8 of the acylating reaction could be quenched with trimethylchlorosilane as the stable silylketale10. Desulfonation and desilylation of10 leads to the title compound12. The reaction of7 with base does not result in a carbacephame. Instead, the benzo[b]-azepinones15c,d have been obtained.

8,10-Dimethyl-3-(unsubstituted, methyl, ethyl, n-butyl, phenyl)-4-hydroxypyrido[2′,3′:3,4]pyrazolo[1,5-a]pyrimidin-2(1H)-ones and 3-(2-hydroxyethyl)-2,8,10-trimethylpyrido[2′,3′:3,4]pyrazolo[1,5-a]pyrimidin-4-ol were synthesized by cyclocondensation of 3-amine-4,6-dimethyl-1H-pyrazolo[3,4-b]pyridine with ethyl malonates and α-acetyl-γ-butyrlolactone. Dichloro- and diazido- derivatives were obtained from the reaction of pyridopyrazlopyrimidine derivatives with POCl3 followed by NaN3. The tetrahetrocyclic systems were formed by cyclization of 4-chloro-3-(2-chloroethyl)-2,8,10-trimethylpyrido[2′,3′:3,4]pyrazolo[1,5-a]pyrimidine with the appropriate primary amines. The structures of all compounds were established by NMR and mass spectra.  相似文献

The binding properties of two phenylketones (2a and 3a) and two ethylesters (2b and {3b) derived from p-tert-butyldihomooxacalix[4]arene or from p-tert-butylcalix[4]arene, in the cone conformation, towards transition (Ag+, Ni2+, Cu2+, Co2+, Zn2+, Fe2+ and Mn2+) and heavy (Cd2+, Hg2+ and Pb2+) metal cations have been determined by extraction studies with metal picrates and liquid membrane transport experiments with the same salts. The affinity of these ligands for Ag+ has also been investigated by 1H NMR spectroscopy. Both ketones are better extractants than the esters, and show a strong preference for Ag+, while Cu2+ is the most extracted cation with the esters. 1H NMR titrations with AgSO3CF3 indicate 1 : 1 complexes for all ligands, those with ketones are more stable, on the NMR time scale, than those with esters. Both esters are good carriers for Ag+, and 2b exhibits the highest transport rate (4.7 mol h-1) found until now with dihomooxacalix[4]arene derivatives.  相似文献

Two routes to consecutive functionalization of 3-chloro-4-cyanobenzo[b][1,6]naphthyridine (1a) at positions 3 and 10 were developed. Oxidation of compound 1a with m-chloroperbenzoic acid in acetone leads to 3-chloro-4-cyano-10-oxobenzo[b][1,6]naphthyridine, while in acetic acid, the reaction gives 3-chloro-4-cyano-10-(3-chlorobenzoyloxy)-5-hydroxy-5,10-dihydrobenzo[b][1,6]naphthyridine. The reactions of 1a with some C-nucleophiles give -adducts at position 10. The reactions of N,N-dimethylamide acetals with chloride 1a leads to 4-cyano-3-dimethylaminobenzo[b][1,6]naphthyridine.  相似文献

The reactions of 2-chloro-1-cyano-1-diethoxyphosphoryl-2-trifluoromethylethylene (2a) and 2-chloro-2-chlorodifluoromethyl-1-cyano-1-diethoxyphosphorylethylene (2b) with arylamines, arylhydrazines, amidines, 2-aminopyridines, and 5-aminopyrazoles were studied. Alkenes 2a, b can serve as precursors of aminopyrazoles, pyrimidines, pyrido[1,2-a]pyrimidines, and pyrazolo[1,5-a]pyrimidines modified with the fluoroalkyl and diethoxyphosphoryl groups. Intermediates of some heterocyclization reactions were detected by NMR spectroscopy. The synthesis and reactions of methyl 2-[3-(trifluoromethyl)phenyl]-4H-furo[3,2-b]pyrrole-5-carboxylate (1a) are described. Upon reaction with methyl iodide, benzyl chloride, or acetic anhydride, this compound gave N-substituted products 1b-d. By hydrolysis of compounds 1a-c, the corresponding acids 2a-c were formed, or by reaction with hydrazine-hydrate, the corresponding carbohydrazides 3a-c were formed. By heating 2-[3-(trifluoromethyl)phenly]-4H-furo[3,2-b]pyrrole-5-carboxylic acid (2a) in acetic anhydride, 4-acetyl-2-[3-(trifluoromethyl)phenyl]furo[3,2-b]pyrrole (4) was formed. By hydrolysis of 4, 2-[3-(trifluoromethyl)phenyl]-4H-furo[3,2-b]pyrrole (5a) was formed, and reactions with methyl iodide or benzyl chloride gave N-substituted products 5b-c. The reaction of 4 with dimethyl butynedioate gave substituted benzo[b]furan 6. Compound 3a reacted with triethyl orthoesters giving 7a-c, which afforded with phosphorus (V) sulphide the corresponding thiones 8a-c. The thiones 8a-c reacted with hydrazine hydrate to form hydrazine derivatives 9a-c. The reaction of triethyl orthoformiate with compounds 9a-c led to furo[2′,3′: 4,5]pyrrolo[1,2-d][1,2,4]triazolo[3,4-f][1,2,4]triazines 10a-c. Hydrazones 11a-c were formed from 3a-c and 5-[3-(trifluoromethyl)phenyl]furan-2-carboxaldehyde. The effect of microwave irradiation on some condensation reactions was compared with “classical” conditions. The results showed that microwave irradiation shortens the reaction time while affording comparable yields.  相似文献

A new series of 1,2,4-triazolo[4,3-a]-quinoline derivatives were designed and synthesized to meet the structural requirements essential for anticancer activity. N-1-(5-Methylisoxazol-4-yl/4-fluoro-2,3-dihydro-1H-inden-1-yl/aryl)-N’-3-(4,5-dihydro-1-methyl-[1,2,4]triazolo[4,3-a]quinolin-7-yl) urea derivatives were accomplished in good yields by the reaction of 4,5-dihydro-1-methyl-[1,2,4]triazolo[4,3-a]quinolin-7-amine with 3-(3-chloro-5-fluoro-2-methoxyphenyl)-5-methyl-isoxazole-4-carboxylic acid, 4-fluoro-2,3-dihydro-1H-indene-1-carboxylic acid, and various simple aromatic carboxylic acids in the presence of diphenyl phosphoryl azide (DPPA). All the newly synthesized title compounds were characterized by elemental and spectral data. Furthermore, anticancer activity was screened for the title compounds (12a–j) in vitro against human neuroblastoma cell lines (SK N SH) and human colon carcinoma cell lines (COLO 205) by using the MTT cell viability method. A few of them (12a and 12b) possess significant cytotoxicity, and some other compounds 12d-f and 12j displayed moderate cytotoxicity against both cell lines.  相似文献

Summary Several new N-acyl derivatives of 1-(4-hydroxy-2,3-dihydrobenzofuran-5-yl)-7,8-di methoxy-1,9 b,2,3,3 a,4-hexahydro-[1]benzopyrano[3,4-c]pyrazole-1-ene have been prepared by appropriate chemical transformation of isohydrazones of rotenone and amorphigenin. A study of their1H- and13C-NMR spectra confirmed the presence of the twocis 3a, 9b, 2 and 3a, 9b, 2 diastereomers in the parent isohydrazones and revealed the strong predominance of the conformers withendo orientation of the 3-NAc group. The conformations due to rotation about the 1,5-bond between rings C and D in the 4-OH and 4-O-substituted compounds were also determined by taking into account the anisotropic effect of aromatic rings A and D, and the hydrogen bond between 4-OH and the 2-N atom, as well as by inspecting the Dreiding models.
Darstellung und stereochemische Charakterisierung einiger N-Acyl-[1]benzopyrano[3,4-c]pyrazol-Derivate von Rotenoiden
Zusammenfassung Mittels geeigneter chemischer Transformationen von Isohydrazonen von Rotenon und Amorphigenin wurden einige neue N-Acyl-Derivate von 1-(4-Hydroxy-2,3-dihydrobenzofuran-5-yl)-7,8-dimethoxy-1,9 b,2,3,3 a,4-hexahydro-[1]benzopyrano[3,4-c]pyrazol-1-en hergestellt. Eine Untersuchung ihrer1H und13C-NMR-Spektren zeigt die Gegenwart von zweicis 3a, 9b, 2- und 3a, 9b, 2-Diastereomeren in den Ausgangs-Isohydrazonen und eine starke Bevorzugung der Konformeren mitendo-Orientierung der 3-NAc-Gruppe. Die Konformationen bezüglich der Rotation um die 1,5-Bindung zwischen Ring C und D werden für die 4-OH und 4-O-substituierten Verbindungen unter Berücksichtigung von Anisotropie-Effekten der aromatischen A- und D-Ringe, der Wasserstoffbrücken zwischen 4-OH und dem 2-N Atom und auch der Betrachtung der entspechenden Dreiding-Modelle diskutiert.

Summary 5-Substituted 3,4-dihydro-pyrrolo[1,2-c]thiazole-6,7-dicarboxylic acid esters3 are obtained from 2-substituted-3-acyl-1,3-thiazolidine-4-carboxylic acids,1 in [3+2]-cycloaddition reactions via mesoionic oxazolone (münchnone) intermediates. The chirality at C-4 of the starting carboxylic acids1 is eliminated in the products3, and the chirality at C-3 (C-2 in the starting carboxylic acids1) can thus be determined through chiroptical measurements. Several representatives of the ring system3 have been characterised through1H- and CD-spectra and the molecular structure of (3S)-3da has been determined by X-ray crystallography.

Summary 3-Amino-2,4-pentadienthioamides, which were obtained from 6-dialkylamino-2H-thiopyrane-4(3H)iminium halides by alkali hydrolysis, react with hydroxylamine hydrochloride in a [C-C-C] + [N-O]-addition to give selectively 5-amino-3-alkenyl-1,2-oxazoles. These oxazoles have been identified by13C and1H NMR spectroscopic analysis. The configuration of these compounds was established on the basis of the coupling constants and 2D-NOESY-experiments. The position of the substituents has been determined by analysis of the fragmentation pattern.

4-Benzoyl-5-phenylthiophene-2,3-dione (1) reacts witho-phenylendiamine to give the quinoxaline derivative2 which cyclizes either to the furo[2,3-b]quinoxaline4 or to the thieno[2,3-b]quinoxaline5 depending on the reaction conditions. On the other hand, the pyrrol-2,3-diones6 ando-phenylendiamine combine yielding the pyrrolidino[2,3-b][1,5]benzodiazepine derivatives7. The structures of all compounds were confirmed by IR and13C NMR spectroscopic measurements based on X-ray structure determinations of4/5 (mixed crystal) as well as7 b.4/5 crystallize triclinically in the space group P1 (Nr. 2) with two molecules per cell, while7 b crystallizes monoclinically in space group P21/a (Nr. 14) with four molecules7 b and four moleculesDMSO per cell. The reaction pathways leading to the compounds2,4,5, and7 are briefly discussed.

Treatment of 1,2‐dideoxy‐4,5:6,7‐di‐O‐isopropylidene‐Dxylo‐hept‐1‐yn‐3‐uloses 4a,b with hydrazine hydrate and amidines yielded the 3‐(1,2:3,4‐di‐O‐isopropylidene‐Dxylo‐1,2,3,4‐tetrahydroxy‐butyl)‐5‐phenyl‐1H(2H)‐pyrazole 5 and the substituted 4‐(1,2:3,4‐di‐O‐isopropylidene‐Dxylo‐1,2,3,4‐tetrahydroxy‐butyl)pyrimidines 7a–f, respectively. Reaction of 4a,b with 2‐amino‐benzimidazol afforded the 2‐(1,2:3,4‐di‐O‐isopropylidene‐Dxylo‐1,2,3,4‐tetrahydroxy‐butyl)benzo[4,5]imidazo[1,2‐a]pyrimidines 9a,b. Compound 4a and 5‐amino‐pyrazole‐4‐carbonic acid derivatives yielded the 5‐(1,2:3,4‐di‐O‐isopropylidene‐Dxylo‐1,2,3,4‐tetrahydroxy‐butyl)pyrazolo[1,5‐a]pyrimidines 11a–d. Deprotection of pyrazole 5, pyrimidine 7a, and pyrazolo[1,5‐a]pyrimidine 11b yielded the acyclo‐C‐nucleosides 6, 8, and 12, respectively.  相似文献
