以生物素化羊抗兔免疫球蛋白和量子点标记链霉亲和素为荧光探针,采用竞争免疫分析模式,建立了一种灵敏度高、选择性好的检测雌三醇的荧光免疫分析方法.对几种测量参数如温育时间和温度、缓冲溶液pH、试剂浓度等进行了优化;方法的线性范围为0.01~1000ng/mL,检出限0.0029 ng/mL,血清样品加标回收率在91%~117%之间.  相似文献

Homogeneous immunoassays are becoming more and more attractive for modern medical diagnosis because they are superior to heterogeneous immunoassays in sample and reagent consumption, analysis time, portability and disposability. Herein, a universal platform for homogeneous immunoassay, using human immunoglobulin (IgG) as a model analyte, has been developed. This assay relies upon the inner filter effect (IFE) of gold nanoparticles (AuNPs) on CdTe QDs fluorescence. The immunoreaction of antigen and antibody can induce the aggregation of antibody-functionalized AuNPs, and after aggregation the IFE of AuNPs on CdTe QDs fluorescence is greatly enhanced, resulting in a decrease of fluorescence intensity in the system. Based on this phenomenon, a wide dynamic range of 1-100 pg mL(-1) for determination of IgG can be obtained. The proposed method shows a detection limit of 0.3 pg mL(-1) for human IgG, which is much lower than the corresponding absorbance-based approach and compares favorably with other reported fluorescent methods. This immunoassay method is simple, rapid, cheap, and sensitive. The proposed method has been successfully applied to measuring IgG in serum samples, and the obtained results agreed well with those of the enzyme-linked immunosorbent assay (ELISA).  相似文献

We propose a homogenous multi-analyte immunoassay based on the quenching of quantum dot (QD) fluorescence by means of graphene. Two QDs with emission maxima at 636 and 607 nm were bound to antibodies selective for mouse or chicken immunoglobulins, respectively, and graphene functionalized with carboxylic moieties was employed to covalently link the respective antigen. The antibody-antigen interaction led graphene close enough to QDs to quench the QD fluorescence by resonance energy transfer. The addition of free antigens that competed with those linked to graphene acted as a “turn-on” effect on QD fluorescence. Fluorescence emitted by the two QDs could be recorded simultaneously since the QDs emitted light at different wavelengths while being excited at the same wavelength and proved to be linearly correlated with free antigen concentration. The developed assay allows measuring both antigens over 2–3 orders of magnitude and showed estimated limits of detection in the nanomolar range. This approach is thus a promising universal strategy to develop homogenous immunoassays for diverse antigens (cells, proteins, low-molecular-mass analytes) in a multi-analyte configuration.  相似文献

A novel strategy for “signal on” and sensitive one-spot simultaneous detection of multiple small molecular analytes based on electrochemically encoded barcode quantum dot (QD) tags is described. The target analytes, adenosine triphosphate (ATP) and cocaine, respectively, are sandwiched between the corresponding set of surface-immobilized primary binding aptamers and the secondary binding aptamer/QD bioconjugates. The captured QDs yield distinct electrochemical signatures after acid dissolution, whose position and size reflect the identity and level, respectively, of the corresponding target analytes. Due to the inherent amplification feature of the QD labels and the “signal on” detection scheme, as well as the sensitive monitoring of the metal ions released upon acid dissolution of the QD labels, low detection limits of 30 nM and 50 nM were obtained for ATP and cocaine, respectively, in our assays. Our multi-analyte sensing system also shows high specificity to target analytes and promising applicability to complex sample matrix, which makes the proposed assay protocol an attractive route for screening of small molecules in clinical diagnosis.  相似文献

A novel method for electrochemiluminescent (ECL) detection of nitrite was proposed based on its quenching effect on anodic ECL emission of CdSe quantum dots (QDs). The ECL emission could be greatly enhanced by sulfite and dissolved oxygen in a neutral system and occurred at a relatively low potential in comparison with traditional anodic ECL emitter, leading to high sensitivity and good selectivity. The quenching mechanism followed an “electrochemical oxidation inhibition” process, which was completely different from those of some analytes on the ECL emission of QDs. The coincidence of photoluminescence and ECL spectra of the QDs indicated that the ECL emission resulted from the redox process of QDs core and the sulfite acted as a coreactant. The nitrite quenched ECL emission could be analyzed according to the treatment of Stern-Volmer equation with a linear range from 1 μM to 0.5 mM for detection of nitrite. This work presented a new efficient ECL methodology for quencher-related detection.  相似文献

We report on a novel electrochemiluminescent (ECL) immunoassay for the ultrasensitive determination of morphine by making use of a gold electrode which was modified with a nanocomposite film containing self-assembled polyamidoamine (PAMAM) CdS quantum dots and electrodeposited gold nanoparticles (Au-NPs). The highly uniform and well-dispersed quantum dots were capped with PAMAM dendrimers. Due to the synergistic effect of the modified quantum dots and the electrodeposited Au-NPs, the ECL response is dramatically enhanced. Under optimal experimental conditions, the immunoreaction between morphine and anti-morphine antibody resulted in a decrease of the ECL signal because of steric hindrance. The calibration plot is linear in the morphine concentration range from 0.2 to 180 ng?mL?1, with a detection limit as low as 67 pg?mL?1. The sensor was successfully applied to the determination of morphine in blood plasma. This kind of assay is expected to pave new avenues in label-free drug assays.

Here, a creatinine-modified CdSe/ZnS quantum dots fluorescent probe has been prepared and used for sensing 2,4,6,-trinitrotoluene explosive (TNT). The proposed method is based on the selective interaction between creatinine and nitroaromatic compounds according to the well-known Jaffé reaction. The procedure for the synthesis of creatinine-CdSe/ZnS reagent is very simple and reproducible and its fluorescent characteristics are reported. We found that the presence of TNT quenches the original fluorescence of creatinine-QD according to the Stern–Volmer model. Under the working conditions, the calibration plot of Io/I versus concentration of TNT was linear in the range 10–300 μg L−1 (R2 = 0.996). The mechanism interaction is discussed. The selectivity of fluorescence quenching of creatinine-QD for TNT has been evaluated. Finally, the potential application of the proposed methodology for the determination of TNT in spiked soils is demonstrated. For the analysis of soil samples a solid-liquid extraction is carried out and a four-point standard addition protocol is used to correct the matrix effect. The method, which is simple and rapid, allows the detection of 0.057 μg g−1 of TNT in soil samples. This sensor could be a useful tool for environmental studies, a crucial topic for nanotechnology nowadays.  相似文献

In the study,we observed the strong adsorption of CdTe/CdS QDs to antibodies and the formation of QDs-antibodies conjugates. Capillary electrophoresis with laser-induced fluorescence detection(CE-LIF),fluorescence spectrometry and fluorescence correlation spectroscopy(FCS) were used to characterize the QDs conjugates with antibody.We found that the QDs-antibody conjugates possessed high fluorescence,small hydrodynamic radii and good stability in aqueous solution.  相似文献

Nanostructures of colloidal CdSe/ZnS core/shell quantum dots (QDs) surrounded by a discrete number of Au nanoparticles were generated via DNA hybridization and purified by gel electrophoresis. Statistics from TEM analysis showed a high yield of designed structures. The distance between Au particles and QD, the number of Au around the central QD, and the size of Au and QD can be adjusted. Rationally designed structures such as these hold great promise for researching the physical interactions between semiconductor and Au nanoparticles and for developing more efficient nanoprobes.  相似文献

以3-巯基丙酸为稳定剂,合成了具有特殊光学性质的水溶性CdTe量子点,其最大发射波长位于544 nm.利用荧光光谱、紫外可见光谱及圆二色光谱法系统的研究了CdTe量子点与肌红蛋白(Mb)二者结合前后体系光谱的变化,从而证实了CdTe量子点与Mb之间静电结合反应的特征.在pH 7.0的PBS缓冲液中,用CdTe量子点作为荧光探针研究了肌红蛋白与量子点的相互作用,并基于肌红蛋白对CdTe量子点有显著的荧光猝灭作用,建立了肌红蛋白的快速检测方法.在最佳实验条件下,该体系荧光强度的猝灭程度(△F)与肌红蛋白质量浓度呈良好的线性关系,线性范围为0.3~24 μg/mL,检出限为0.13 μg/mL.该方法已对合成样品中肌红蛋白进行检测,并用于人体尿样中肌红蛋白的测定.  相似文献

The synthesis of zinc sulfide (ZnS) quantum dots (QDs) by microwave heating in a water-ethanol medium is proposed. The effect of the synthesis temperature (80 °C, 100 °C, 120 °C, and 150 °C) on the QD characteristics is examined. Based on the analysis of X-ray diffraction profiles the conclusion is drawn that the hexagonal ZnS phase of wurtzite type with an average nanocrystal size of 2.6-3.7 nm forms in the synthesized QDs. The nanocrystallite size is found to increase with the QD synthesis heating temperature. The analysis of X-ray absorption spectra (XANES) at the zinc K-edge indicates a higher crystallinity of the QD samples prepared at higher synthesis temperatures. The combined analysis of X-ray diffraction profiles, optical diffuse reflectance spectra, and X-ray absorption spectra implies the following possible QD structure: the pure hexagonal ZnS phase of wurtzite type in the bulk of nanoparticles and the amorphous ZnO phase in the surface layer of nanoparticles.  相似文献

A universal sandwich-like immunoassay strategy based on quantum-dots immunoprobe (QD-labeled anti-mouse IgG antibody) was developed for rapid and ultrasensitive detection of small molecules. A portable and reusable optofluidic nano-biosensing platform was applied to investigate the sandwich-like immunoassay mechanism and format of small molecules, as well as the binding kinetics between QD immunoprobe and anti-small molecule antibody. A two-step immunoassay method that involves pre-incubation mixture of different concentration of small molecule and anti-small molecule antibody, and subsequent introduction of QD immunoprobe into the optofluidic cell was conducted for small molecule determination. Compared with the one-step immunoassay method, the two-step immunoassay method can obtain higher fluorescence signal and higher sensitivity index, thus improving the nano-biosensing performance. Based on the proposed strategy, two mode targets, namely, microcystin-LR (MC-LR) and Bisphenol A (BPA) were tested with high sensitivity, rapidity, and ease of use. A higher concentration of small molecules in the sample led to less anti-small molecule antibody bound with antigen-carrier protein conjugate immobilized onto the sensor surface, and less QD immunoprobes bound with anti-small molecule antibody. This phenomenon lowered the fluorescence signal detected by nano-biosensing platform. Under optimal operating conditions, MC-LR and BPA exhibited a limit of detection of 0.003 and 0.04 μg/L, respectively. The LODs were better than those of the indirect competitive immunoassay method for small molecules via Cy5.5-labeled anti-small molecule antibody. The proposed QD-based sandwich-like immunoassay strategy was evaluated in spiked water samples, and showed good recovery, precision and accuracy without complicated sample pretreatments. All these results demonstrate that the new detection strategy could be readily applied to the other trace small molecules in real water samples.  相似文献

We have demonstrated the design of a new type fluorescence assay based on the inner filter effect (IFE) of gold nanoparticles (AuNPs) on the fluorescence of quantum dots (QDs). With a high extinction coefficient, AuNPs are expected to be capable of functioning as powerful absorbers. QDs with tunable emission wavelength are ideal fluorophores because the emission spectra of the rationally synthesized QDs can perfectly overlap with the absorption band of the absorber. Aminothiols are chosen as the model analytes, and the IFE-based fluorescent method for detection of aminothiols was suggested. Under the optimum conditions, the response is linearly proportional to the concentration of cysteine in the range of 0.05-0.9 μg mL−1. The present IFE-based fluorescent strategy could be also used to detect glutathione and homocysteine. The linear concentration ranges were 0.05-1.0 μg mL−1 for glutathione and 0.01-1.0 μg mL−1 for homocysteine.  相似文献
