Real-time functional magnetic resonance imaging: methods and applications   总被引:3,自引:0,他引:3  
Functional magnetic resonance imaging (fMRI) has been limited by time-consuming data analysis and a low signal-to-noise ratio, impeding online analysis. Recent advances in acquisition techniques, computational power and algorithms increased the sensitivity and speed of fMRI significantly, making real-time analysis and display of fMRI data feasible. So far, most reports have focused on the technical aspects of real-time fMRI (rtfMRI). Here, we provide an overview of the different major areas of applications that became possible with rtfMRI: online analysis of single-subject data provides immediate quality assurance and functional localizers guiding the main fMRI experiment or surgical interventions. In teaching, rtfMRI naturally combines all essential parts of a neuroimaging experiment, such as experimental design, data acquisition and analysis, while adding a high level of interactivity. Thus, the learning of essential knowledge required to conduct functional imaging experiments is facilitated. rtfMRI allows for brain-computer interfaces (BCI) with a high spatial and temporal resolution and whole-brain coverage. Recent studies have shown that such BCI can be used to provide online feedback of the blood-oxygen-level-dependent signal and to learn the self-regulation of local brain activity. Preliminary evidence suggests that this local self-regulation can be used as a new paradigm in cognitive neuroscience to study brain plasticity and the functional relevance of brain areas, even being potentially applicable for psychophysiological treatment.  相似文献

静息状态下脑功能连接的磁共振成像研究近年来取得了迅猛发展. 通过对fMRI信号低频涨落成分的同步性分析,可以得到大脑静息态任意脑区的功能连接和多套网络系统,其中“默认网络”的发现可能为人脑固有网络的研究提供新的思路. 而静息态网络与解剖连接之间可能存在的对应,以及在神经精神疾病患者脑中性质和连接的异常改变,使其具有重要的研究和临床应用价值. 该文总结了静息状态功能磁共振成像的主要研究成果,对静息状态脑功能网络的发现和发展、研究方法、各网络及其特点以及在临床方面的应用进行简单的介绍和分析.  相似文献

This paper investigates how well different kinds of fMRI functional connectivity analysis reflect the underlying interregional neural interactions. This is hard to evaluate using real experimental data where such relationships are unknown. Rather, we use a biologically realistic neural model to simulate both neuronal activities and multiregional fMRI data from a blocked design. Because we know how every element in the model is related to every other element, we can compare functional connectivity measurements across different spatial and temporal scales. We focus on (1) psycho-physiological interaction (PPI) analysis, which is a simple brain connectivity method that characterizes the activity in one brain region by the interaction between another region's activity and a psychological factor, and (2) interregional correlation analysis. We investigated the neurobiological underpinnings of PPI using simulated neural activities and fMRI signals generated by a large-scale neural model that performs a visual delayed match-to-sample task. Simulated fMRI data are generated by convolving integrated synaptic activities (ISAs) with a hemodynamic response function. The simulation was done under three task conditions: high-attention, low-attention and a control task ('passive viewing'). We investigated how biological and scanning parameters affect PPI and compared these with functional connectivity measures obtained using correlation analysis. We performed correlational and PPI analyses with three types of time-series data: ISA, fMRI and deconvolved fMRI (which yields estimated neural signals) obtained using a deconvolution algorithm. The simulated ISA can be considered as the 'gold standard' because it represents the underlying neural activity. Our main findings show (1) that evaluating the change in an interregional functional connection using the difference in regression coefficients (as is essentially done in the PPI method) produces results that better reflect the underlying changes in neural interrelationships than does evaluating the functional connectivity difference as a change in correlation coefficient; (2) that using fMRI and deconvolved fMRI data led to similar conclusions in the PPI-based functional connectivity results, and these generally agreed with the nature of the underlying neural interactions; and (3) the functional connectivity correlation measures often led to different conclusions regarding significance for different scanning and hemodynamic parameters, but the significances of the PPI regression parameters were relatively robust. These results highlight the way in which neural modeling can be used to help validate the inferences one can make about functional connectivity based on fMRI data.  相似文献

Machine learning and pattern recognition techniques are being increasingly employed in functional magnetic resonance imaging (fMRI) data analysis. By taking into account the full spatial pattern of brain activity measured simultaneously at many locations, these methods allow detecting subtle, non-strictly localized effects that may remain invisible to the conventional analysis with univariate statistical methods. In typical fMRI applications, pattern recognition algorithms "learn" a functional relationship between brain response patterns and a perceptual, cognitive or behavioral state of a subject expressed in terms of a label, which may assume discrete (classification) or continuous (regression) values. This learned functional relationship is then used to predict the unseen labels from a new data set ("brain reading"). In this article, we describe the mathematical foundations of machine learning applications in fMRI. We focus on two methods, support vector machines and relevance vector machines, which are respectively suited for the classification and regression of fMRI patterns. Furthermore, by means of several examples and applications, we illustrate and discuss the methodological challenges of using machine learning algorithms in the context of fMRI data analysis.  相似文献

The accurate mapping of functional magnetic resonance imaging (fMRI) activations to anatomical structures is critical for fMRI studies of brain organization. In the commonly used functional space analysis method, functional images are realigned to a functional reference image and processed in low-resolution functional space. The average functional activations are then projected into high-resolution anatomical space for visualization. Here, we describe a new technique, anatomical space analysis (ASA), whereby low-resolution functional images are first coregistered and resampled directly into high-resolution anatomical space with all subsequent data processing performed in high-resolution space. A major advantage of ASA is that minor scanner sampling instabilities and small head movements can increase spatial resolution by providing multiple samples of the relationship between functional and anatomical space. Both simulations and analyses of real fMRI data show that ASA improves the precision, objectivity and reproducibility of functional brain mapping.  相似文献

A new approach in studying interregional functional connectivity using functional magnetic resonance imaging (fMRI) is presented. Functional connectivity may be detected by means of cross correlating time course data from functionally related brain regions. These data exhibit high temporal coherence of low frequency fluctuations due to synchronized blood flow changes. In the past, this fMRI technique for studying functional connectivity has been applied to subjects that performed no prescribed task ("resting" state). This paper presents the results of applying the same method to task-related activation datasets. Functional connectivity analysis is first performed in areas not involved with the task. Then a method is devised to remove the effects of activation from the data using independent component analysis (ICA) and functional connectivity analysis is repeated. Functional connectivity, which is demonstrated in the "resting brain," is not affected by tasks which activate unrelated brain regions. In addition, ICA effectively removes activation from the data and may allow us to study functional connectivity even in the activated regions.  相似文献

Independent component analysis (ICA) has been proven to be effective for functional magnetic resonance imaging (fMRI) data analysis. However, ICA decomposition requires to optimize the unmixing matrix iteratively whose initial values are generated randomly. Thus the randomness of the initialization leads to different ICA decomposition results. Therefore, just one-time decomposition for fMRI data analysis is not usually reliable. Under this circumstance, several methods about repeated decompositions with ICA (RDICA) were proposed to reveal the stability of ICA decomposition. Although utilizing RDICA has achieved satisfying results in validating the performance of ICA decomposition, RDICA cost much computing time. To mitigate the problem, in this paper, we propose a method, named ATGP-ICA, to do the fMRI data analysis. This method generates fixed initial values with automatic target generation process (ATGP) instead of being produced randomly. We performed experimental tests on both hybrid data and fMRI data to indicate the effectiveness of the new method and made a performance comparison of the traditional one-time decomposition with ICA (ODICA), RDICA and ATGP-ICA. The proposed method demonstrated that it not only could eliminate the randomness of ICA decomposition, but also could save much computing time compared to RDICA. Furthermore, the ROC (Receiver Operating Characteristic) power analysis also denoted the better signal reconstruction performance of ATGP-ICA than that of RDICA.  相似文献

人体大脑活动的复杂度随年龄变化而变化, 并且和性别有一定的联系, 通过对功能磁共振成像复杂度的分析有助于发现人脑活动和性别年龄之间关系的规律. 本文提出需要根据年龄段的变化对基本尺度熵的参数做适当的调整, 以便获得良好的信号区分效果. 本文研究了人脑活动和性别年龄之间存在的关系. 结果证明, 同龄男女的基本尺度熵值存在一定的差异, 并且随年龄段的不同发生相应的变化, 另外基本尺度熵中的参数在数据分析中也随年龄变化存在一定规律的变化. 通过对fMRI数据的分析表明, 基本尺度熵能够有效地区分不同人群fMRI数据特征, 为进一步信号复杂度分析提供方便. 关键词: 功能磁共振成像 基本尺度熵 复杂度  相似文献

Wavelet methods for image regularization offer a data-driven alternative to Gaussian smoothing in functional magnetic resonance (fMRI) analysis. Their impact has been limited by the difficulties in integrating regularization in the wavelet domain and inference in the image domain, precluding the probabilistic decision on which areas are activated by a task. Here we present an integrated framework for Bayesian estimation and regularization in wavelet space that allows the usual voxelwise hypothesis testing. This framework is flexible, being an adaptation to fMRI time series of a more general wavelet-based functional mixed-effect model. Through testing on a combination of simulated and real fMRI data, we show evidence of improved signal recovery, without compromising test accuracy in image space.  相似文献

Independent component analysis (ICA) is a popular blind source separation technique that has proven to be promising for the analysis of functional magnetic resonance imaging (fMRI) data. A number of ICA approaches have been used for fMRI data analysis, and even more ICA algorithms exist; however, the impact of using different algorithms on the results is largely unexplored. In this paper, we study the performance of four major classes of algorithms for spatial ICA, namely, information maximization, maximization of non-Gaussianity, joint diagonalization of cross-cumulant matrices and second-order correlation-based methods, when they are applied to fMRI data from subjects performing a visuo-motor task. We use a group ICA method to study variability among different ICA algorithms, and we propose several analysis techniques to evaluate their performance. We compare how different ICA algorithms estimate activations in expected neuronal areas. The results demonstrate that the ICA algorithms using higher-order statistical information prove to be quite consistent for fMRI data analysis. Infomax, FastICA and joint approximate diagonalization of eigenmatrices (JADE) all yield reliable results, with each having its strengths in specific areas. Eigenvalue decomposition (EVD), an algorithm using second-order statistics, does not perform reliably for fMRI data. Additionally, for iterative ICA algorithms, it is important to investigate the variability of estimates from different runs. We test the consistency of the iterative algorithms Infomax and FastICA by running the algorithm a number of times with different initializations, and we note that they yield consistent results over these multiple runs. Our results greatly improve our confidence in the consistency of ICA for fMRI data analysis.  相似文献

Blood oxygenation level dependent (BOLD) contrast has been widely used for visualizing regional neural activation. Temporal filtering and parameter estimation algorithms are generally used to account for the intrinsic temporal autocorrelation present in BOLD data. Arterial spin labeling perfusion imaging is an emerging methodology for visualizing regional brain function both at rest and during activation. Perfusion contrast manifests different noise properties compared with BOLD contrast, represented by the even distribution of noise power and spatial coherence across the frequency spectrum. Consequently, different strategies are expected to be employed in the statistical analysis of functional magnetic resonance imaging (fMRI) data based on perfusion contrast. In this study, the effect of different analysis methods upon signal detection efficacy, as assessed by receiver operator characteristic (ROC) measures, was examined for perfusion fMRI data. Simulated foci of neural activity of varying amplitude and spatial extent were added to resting perfusion data, and the accuracy of each analysis was evaluated by comparing the results with the known distribution of pseudo-activation. In contrast to the BOLD fMRI, temporal smoothing or filtering reduces the power of perfusion fMRI data analyses whereas spatial smoothing is beneficial to the efficacy of analyses.  相似文献

Temporal clustering analysis (TCA) and independent component analysis (ICA) are promising data-driven techniques in functional magnetic resonance imaging (fMRI) experiments to obtain brain activation maps in conditions with unknown temporal information regarding the neuronal activity. Although comparable to ICA in detecting transient neuronal activities, TCA fails to detect prolonged plateau brain activations. To eliminate this pitfall, a novel derivative TCA (DTCA) method was introduced and its algorithms with different subtraction intervals were tested on simulated data with a pattern of prolonged plateau brain activation. It was found that the best performance of DTCA method in generating functional maps could be obtained if the subtraction interval is equal to or larger than the length of the rising time of the fMRI response. The DTCA method and its theoretical predication were further investigated and validated using in vivo fMRI data sets. By removing the limitations in the previous TCA, DTCA has shown its powerful capability in detecting prolonged plateau neuronal activities.  相似文献

Partial least squares (PLS) has been used in multivariate analysis of functional magnetic resonance imaging (fMRI) data as a way of incorporating information about the underlying experimental paradigm. In comparison, principal component analysis (PCA) extracts structure merely by summarizing variance and with no assurance that individual component structures are directly interpretable or that they represent salient and useful features. Oriented partial least squares (OrPLS) is a new PLS-like analysis paradigm in which extracted components can be oriented away from undesirable noise or confounds in the data and toward a desired targeted structure reflecting the fMRI experiment.  相似文献

Many informatics tools have emerged to process the voluminous and complex data generated by functional magnetic resonance imaging (fMRI). The interpretation of fMRI exams is largely determined by these tools. However, their performance is hard to evaluate because there is no independent means of calibration. A novel fMRI calibration system called SmartPhantom has been developed to simulate functional blood oxygen level dependent (BOLD) imaging. SmartPhantom contains a quadrature radio frequency coil, comprising two perpendicular planar loops that can be externally activated or deactivated. The system is able to produce reasonably uniform signal enhancements in a calibration sample surrounded by the two loops during an MRI scan. The enhancement is controlled well in both magnitude and predefined timing and produces BOLD-like signals. Characteristics of SmartPhantom are discussed in detail, followed by a comparison of fMRI informatics tools. Two fMRI data sets are acquired with the SmartPhantom. One with high signal-to-noise ratio provides the calibration. Another with lower SNR is input into three software packages (BrainVoyager, FSL and Statistical Parametric Mapping 2) for data preprocessing and statistical analysis. Results from the three packages are compared in both sensitivity of detecting the activation and correlation between the predicted activation and calibration.  相似文献

By measuring the changes of magnetic resonance signals during a stimulation, the functional magnetic resonance imaging (fMRI) is able to localize the neural activation in the brain. In this report, we discuss the fMRI application of the spatial independent component analysis (spatial ICA), which maximizes statistical independence over spatial images. Included simulations show the possibility of the spatial ICA on discriminating asynchronous activations or different response patterns in an fMRI data set. An in vivo visual stimulation fMRI test was conducted, and the result shows a proper sum of the separated components as the final image is better than a single component, using fMRI data analysis by spatial ICA. Our result means that spatial ICA is a useful tool for the detection of different response activations and suggests that a proper sum of the separated independent components should be used for the imaging result of fMRI data processing.  相似文献

Resting-state functional magnetic resonance imaging (fMRI) aims to measure baseline neuronal connectivity independent of specific functional tasks and to capture changes in the connectivity due to neurological diseases. Most existing network detection methods rely on a fixed threshold to identify functionally connected voxels under the resting state. Due to fMRI non-stationarity, the threshold cannot adapt to variation of data characteristics across sessions and subjects, and generates unreliable mapping results. In this study, a new method is presented for resting-state fMRI data analysis. Specifically, the resting-state network mapping is formulated as an outlier detection process that is implemented using one-class support vector machine (SVM). The results are refined by using a spatial-feature domain prototype selection method and two-class SVM reclassification. The final decision on each voxel is made by comparing its probabilities of functionally connected and unconnected instead of a threshold. Multiple features for resting-state analysis were extracted and examined using an SVM-based feature selection method, and the most representative features were identified. The proposed method was evaluated using synthetic and experimental fMRI data. A comparison study was also performed with independent component analysis (ICA) and correlation analysis. The experimental results show that the proposed method can provide comparable or better network detection performance than ICA and correlation analysis. The method is potentially applicable to various resting-state quantitative fMRI studies.  相似文献

The trust region method which originated from the Levenberg–Marquardt (LM) algorithm for mixed effect model estimation are considered in the context of second level functional magnetic resonance imaging (fMRI) data analysis. We first present the mathematical and optimization details of the method for the mixed effect model analysis, then we compare the proposed methods with the conventional expectation-maximization (EM) algorithm based on a series of datasets (synthetic and real human fMRI datasets). From simulation studies, we found a higher damping factor for the LM algorithm is better than lower damping factor for the fMRI data analysis. More importantly, in most cases, the expectation trust region algorithm is superior to the EM algorithm in terms of accuracy if the random effect variance is large. We also compare these algorithms on real human datasets which comprise repeated measures of fMRI in phased-encoded and random block experiment designs. We observed that the proposed method is faster in computation and robust to Gaussian noise for the fMRI analysis. The advantages and limitations of the suggested methods are discussed.  相似文献

Independent component analysis (ICA) is a widely accepted method to extract brain networks underlying cognitive processes from functional magnetic resonance imaging (fMRI) data. However, the application of ICA to multi-task fMRI data is limited due to the potential non-independency between task-related components. The ICA with projection (ICAp) method proposed by our group (Hum Brain Mapp 2009;30:417–31) is demonstrated to be able to solve the interactions among task-related components for single subject fMRI data. However, it still must be determined if ICAp is capable of processing multi-task fMRI data over a group of subjects. Moreover, it is unclear whether ICAp can be reliably applied to event-related (ER) fMRI data. In this study, we combined the projection method with the temporal concatenation method reported by Calhoun (Hum Brain Mapp 2008;29:828–38), referred to as group ICAp, to perform the group analysis of multi-task fMRI data. Both a human fMRI rest data-based simulation and real fMRI experiments, of block design and ER design, verified the feasibility and reliability of group ICAp, as well as demonstrated that ICAp had the strength to separate 4D multi-task fMRI data into multiple brain networks engaged in each cognitive task and to adequately find the commonalities and differences among multiple tasks.  相似文献

The cerebral cortex is the main target of analysis in many functional magnetic resonance imaging (fMRI) studies. Since only about 20% of the voxels of a typical fMRI data set lie within the cortex, statistical analysis can be restricted to the subset of the voxels obtained after cortex segmentation. While such restriction does not influence conventional univariate statistical tests, it may have a substantial effect on the performance of multivariate methods.

Here, we describe a novel approach for data-driven analysis of single-subject fMRI time series that combines techniques for the segmentation and reconstruction of the cortical surface of the brain and the spatial independent component analysis (sICA) of the functional time courses (TCs). We use the mesh of the white matter/gray matter boundary, automatically reconstructed from high-spatial-resolution anatomical MR images, to limit the sICA decomposition of a coregistered functional time series to those voxels which are within a specified region with respect to the cortical sheet (cortex-based ICA, or cbICA). We illustrate our analysis method in the context of fMRI blocked and event-related experimental designs and in an fMRI experiment with perceptually ambiguous stimulation, in which an a priori specification of the stimulation protocol is not possible.

A comparison between cbICA and conventional hypothesis-driven statistical methods shows that cortical surface maps and component TCs blindly obtained with cbICA reliably reflect task-related spatiotemporal activation patterns. Furthermore, the advantages of using cbICA when the specification of a temporal model of the expected hemodynamic response is not straightforward are illustrated and discussed. A comparison between cbICA and anatomically unconstrained ICA reveals that — beside reducing computational demand — the cortex-based approach improves the fitting of the ICA model in the gray matter voxels, the separation of cortical components and the estimation of their TCs, particularly in the case of fMRI data sets with a complex spatiotemporal statistical structure.  相似文献
