This study describes the development and use of a modified quick, easy, cheap, effective, rugged and safe (QuEChERS) method coupled with gas chromatography with mass spectrometry to determine 29 pesticide residues in green, red and dehydrated red peppers. Pesticides were extracted with acetonitrile (1% acetic acid), partitioned with sodium chloride and purified with primary secondary amino and octadecyl silane in acetone. The QuEChERS extraction conditions were optimized, and the matrix effects that might influence recoveries were evaluated and minimized using matrix‐matched calibration curves. Under the optimized conditions, the calibration curves for 29 pesticides showed good linearity in the concentration range of 0.1–10 μg/mL with determination coefficient R2 > 0.998. The limits of quantification of the 29 pesticides were 0.006–0.06 mg/kg for green pepper, 0.005–0.039 mg/kg for red pepper and 0.014–0.25 mg/kg for dehydrated red pepper. These values are below the suggested regulatory maximum residue limits. The mean recoveries ranged between 70.1 and 110%, and the relative standard deviations were <13%. The developed method was successfully applied to commercial samples. Some samples were found to contain the 29 pesticides with levels below the legal limits. Thirty‐three species of mosquitoes have been reported from the Kingdom of Saudi Arabia. Several of these mosquitoes, Anopheles gambiae Giles s.l., Anopheles stephensi Liston, Culex pipiens Linnaeus, Culex quinquefasciatus Say, Culex tritaeniorhynchus Giles, Stegomyia aegypti (Linnaeus) and Aedimorphus vexans arabiensis (Patton) are known vectors of human and animal diseases. In this study, the cuticular hydrocarbon profiles of eight mosquito species using gas chromatography–mass spectrometry were analyzed. Wild collected fourth‐instar larvae were reared, and single, newly emerged, unfed adult females were used for the analysis. A total of 146–160 peaks were detected from the cuticular extracts by gas chromatography. Repeated analysis of variance (ANOVA) and Tukey HSD Post Hoc test was used to test for quantitative differences in relative hydrocarbon quantity. In addition, a linear regression model was applied using Enter method to determine the diagnostic peaks for the eight mosquito specimens. The ANOVA test indicated that relative peaks were significant (P < 0.05) when selected pairs of peaks were compared. Also, seven compounds showed qualitative differences among the five mosquito vectors tested. The classes of constituents present were n‐alkanes, monomethylalkanes, dimethylalkanes, trimethylalkanes, alkenes, branched aromatic hydrocarbons, aldehydes and esters. These compounds have a carbon chain length ranging from 8 to 18 carbons. The most abundant compound in all adult mosquito specimens was n‐hexylacrylate [retention time (RT) 6.73 min], which was not detected in Cx. pipiens. In Cx. pipiens, the most abundant peak was benzaldehyde (RT 2.98 min). Gas chromatography–mass spectrometry is a suitable method to identify adult mosquitoes, especially from focal areas of public health concern such as Jazan Province, Saudi Arabia. An efficient method for the simultaneous analysis of seven 2‐hydroxy fatty acids (2‐HFAs) as tert‐butyldimethylsilyl (TBDMS) derivative was developed by gas chromatography–mass spectrometry in selected ion monitoring mode. New mass spectral data on 2‐hydroxycapric, 2‐hydroxypalmitic, 2‐hydroxystearic and 2‐hydroxybehenic acids as di‐TBDMS derivatives for hydroxyl and carboxyl groups were built. Under the optimal conditions, the present method showed a good correlation coefficient (r ≥ 0.999) in the range of 0.01–0.5 µg. The precision showed low relative standard deviation of <10%, and the accuracy (percentage relative error) varied from ?5.2 to 0.3 for the seven 2‐HFAs studied. Recovery rates of all 2‐HFAs were ≥ 93.2% with good precision. When applied to normal human plasma, seven 2‐HFAs were positively identified. This paper describes the application of gas chromatography–mass spectrometry (GC‐MS) for in vitro and in vivo studies of 6‐OXO in horses, with a special aim to identify the most appropriate target metabolite to be monitored for controlling the administration of 6‐OXO in racehorses. In vitro studies of 6‐OXO were performed using horse liver microsomes. The major biotransformation observed was reduction of one keto group at the C3 or C6 positions. Three in vitro metabolites, namely 6α‐hydroxyandrost‐4‐ene‐3,17‐dione (M1), 3α‐hydroxyandrost‐4‐ene‐6,17‐dione (M2a) and 3β‐hydroxyandrost‐4‐ene‐6,17‐dione (M2b) were identified. For the in vivo studies, two thoroughbred geldings were each administered orally with 500 mg of androst‐4‐ene‐3,6,17‐trione (5 capsules of 6‐OXO®) by stomach tubing. The results revealed that 6‐OXO was extensively metabolized. The three in vitro metabolites (M1, M2a and M2b) identified earlier were all detected in post‐administration urine samples. In addition, seven other urinary metabolites, derived from a further reduction of either one of the remaining keto groups or one of the remaining keto groups and the olefin group, were identified. These metabolites included 6α,17β‐dihydroxyandrost‐4‐en‐3‐one (M3a), 6,17‐dihydroxyandrost‐4‐en‐3‐one (M3b and M3c), 3β,6β‐dihydroxyandrost‐4‐en‐17‐one (M4a), 3,6‐dihydroxyandrost‐4‐en‐17‐one (M4b), 3,6‐dihydroxyandrostan‐17‐one (M5) and 3,17‐dihydroxyandrostan‐6‐one (M6). The longest detection time observed in urine was up to 46 h for the M6 metabolite. For blood samples, the peak 6‐OXO plasma concentration was observed 1 h post administration. A method for routinely determination of dimethyl sulfoxide (DMSO) and dimethyl sulfone (DMSO2) in human urine was developed using gas chromatography–mass spectrometry. The urine sample was treated with 2,2‐dimethoxypropane (DMP) and hydrochloric acid for efficient removal of water, which causes degradation of the vacuum level in mass spectrometer and shortens the life‐time of the column. Experimental DMP reaction parameters, such as hydrochloric acid concentration, DMP–urine ratio, reaction temperature and reaction time, were optimized for urine. Hexadeuterated DMSO was used as an internal standard. The recoveries of DMSO and DMSO2 from urine were 97–104 and 98–116%, respectively. The calibration curves showed linearity in the range of 0.15–54.45 mg/L for DMSO and 0.19–50.10 mg/L for DMSO2. The limits of detection of DMSO and DMSO2 were 0.04 and 0.06 mg/L, respectively. The relative standard deviations of intra‐day and inter‐day were 0.2–3.4% for DMSO and 0.4–2.4% for DMSO2. A novel derivatization method was developed for the simultaneous determination of six acidic metabolites of catecholamine and serotonin by gas chromatography–mass spectrometry (GC‐MS). The metabolites were converted to O‐ethoxycarbonyl/tert‐butyldimethylsilyl (EOC/TBDMS) derivatives for the direct GC‐MS analysis in selected ion monitoring mode. Their mass spectral pattern as EOC/TBDMS derivatives showed characteristic fragment ions of [M – 15]+ and [M – 57]+, which permitted rapid and accurate structural confirmation of acidic metabolites. The present method was linear (r ≥ 0.998), reproducible (percentage relative standard deviation = 1.0–10.0) and accurate (% relative error = ?9.7–9.8) with detection limits of 0.001–4.7 ng/mL. The thermal degradation mechanisms of liquid crystalline aromatic polyesters (LCPs) prepared from p-hydroxybenzoic acid (PHB), biphenol (BP), and terephthalic acid (TA) were studied by pyrolysis–gas chromatography/mass spectrometry (Py-GC/MS). The LCP containing deuterated terephthalate units and the LCPs that have different comonomer ratios were examined. On the basis of the pyrolysis products determined, the origin of the main pyrolysis products (benzene, phenol, biphenyl, phenyl benzoate, etc.) from the corresponding comonomer units were estimated and their thermal degradation mechanisms were eventually discussed in detail.  相似文献

Capillary gas chromatography in combination with atomic emission and electron impact-chemical ionizaion mass spectrometry detectors have been used to detect amitraz degradation products in honey storage, characterize their structure, and evaluate their occurrence over a 100 day peroid. To this end, honey samples were extracted with an 8:2 v/v n-hexane/acetone mixture. Amitraz was found to be rapidly decomposed into five related compounds, of which N-(2,4-dimethylphenyl)formamide was the most abundant and persistent.  相似文献

The present study describes a microextraction and determination method for analyzing residual solvents in pharmaceutical products using dynamic headspace–liquid phase microextraction technique followed by gas chromatography–flame ionization detection. In this method dimethyl sulfoxide (μL level) placed into a GC liner‐shaped extraction vessel is used as a collection/extraction solvent. Then the liner is exposed to the headspace of a vial containing the sample solution. The effect of different parameters influencing the microextraction procedure including collection/extraction solvent type and its volume, ionic strength, extraction time, extraction temperature and concentration of NaOH solution used in dissolving the studied pharmaceuticals are investigated and optimized. Under the optimum extraction conditions, the method showed wide linear ranges between 0.5 and 5000 mg L−1. The other analytical parameters were obtained in the following ranges: enrichment factors 240–327, extraction recoveries 72–98% and limits of detection 0.1–0.8 mg L−1 in solution and 0.6–3.2 μg g−1 in solid. Relative standard deviations for the extraction of 100 mg L−1 of each analyte were obtained in the ranges of 4–7 and 5–8% for intra ‐ day (n = 6) and inter ‐ day (n = 4) respectively. Finally the target analytes were determined in different samples such as erythromycin, azithromycin, cefalexin, amoxicillin and co‐amoxiclav by the proposed method.  相似文献

A rapid and sensitive method for the determination of 4-fluoro-3-phenoxybenzaldehyde cyanohydrin (FPBC) and 4-fluoro-3-phenoxy-benzaldehyde (FPB) in honey samples using ultrasonically assisted extraction and gas chromatography with electron capture detection (GC-ECD) has been developed. The different factors affecting the efficiency of the extraction were carefully optimized. The honey sample was extracted with a mixture of hexane and dichloromethane (v/v, 1:1) utilizing the ultrasonically assisted technique and cleaned up by solid-phase extraction on Oasis HLB cartridges. The eluate was evaporated to dryness and residues were reconstituted to 1.0 mL with hexane and determined by GC-ECD. The calibration curves of fortified samples showed acceptable linear response (R(2) >0.99) over a range of 3-100 ng/g for FPBC and FPB in seven replicate determinations of six concentrations, respectively, and analysis of variance (ANOVA) with a lack-of-fit test was performed to validate the regression data. Overall average recoveries ranged from 90.9 to 106.2% for honey samples. The detection limits were 0.9 ng/g for FPBC and 1.0 ng/g for FPB, respectively. This method can be successfully applied to routine determination of two degradation products of flumethrin in honey samples.  相似文献

This paper presents a fast and simple method for the extraction, preconcentration and determination of fluvoxamine, nortriptyline and maprotiline in urine using simultaneous derivatization and temperature‐assisted dispersive liquid–liquid microextraction (TA‐DLLME) followed by gas chromatography–flame ionization detection (GC‐FID). An appropriate mixture of dimethylformamide (disperser solvent), 1,1,2,2‐tetrachloroethane (extraction solvent) and acetic anhydride (derivatization agent) was rapidly injected into the heated sample. Then the solution was cooled to room temperature and cloudy solution formed was centrifuged. Finally a portion of the sedimented phase was injected into the GC‐FID. The effect of several factors affecting the performance of the method, including the selection of suitable extraction and disperser solvents and their volumes, volume of derivatization agent, temperature, salt addition, pH and centrifugation time and speed were investigated and optimized. Figures of merit of the proposed method, such as linearity (r2 > 0.993), enrichment factors (820–1070), limits of detection (2–4 ng mL?1) and quantification (8–12 ng mL?1), and relative standard deviations (3–6%) for both intraday and interday precisions (concentration = 50 ng mL?1) were satisfactory for determination of the selected antidepressants. The mass spectrometric behavior of four pairs of stereoisomers was investigated by liquid chromatography–electrospray ionization–mass spectrometry (LC–ESI–MS). The E‐ and Z‐isomers of the pesticides chlorfenvinphos, dimethomorph, mevinphos and phosphamidon—each with one double bond—were chosen for this study. The MS response of the individual isomers was investigated by infusing the isomers individually into the MS or after the separation of isomer mixtures via high‐performance liquid chromatography(HPLC). In the case of dimethomorph, the same MS response was found for the two isomers. In contrast to that, the individual isomers of chlorfenvinphos, mevinphos and phosphamidon showed different MS response both in the single ion monitoring (SIM) mode in single quadrupole MS and multiple reaction monitoring (MRM) mode in tandem MS. The MS response of the isomers partly depends on (1) the declustering potential of the precursor ion in the SIM mode, (2) the selected transition and (3) the collision energy in the MRM mode. Consequently, quantification by summation of the peak areas of the isomers is inaccurate due to over‐ or underestimating of one of the stereoisomers. Accurate quantitative results can only be achieved when the compound‐specific MS parameters are separately determined for each isomer. This can be done by using pure isomers or by the determination of the MS parameters after HPLC separation and the measurement of the actual isomer ratio with an independent technique. Ultraviolet irradiation of aqueous solutions of iodide/iodate ion containing low molecular weight organic acids generates volatile iodine species that are amenable to detection by atomic spectrometry. In the presence of formic, acetic or propionic acids, photo-chemical generation results in the formation of HI, methyl- and ethyl-iodide respectively, the latter two products being directly identified by gas chromatography–mass spectrometry. Deuterium and 13C-labeled reagents were employed to elucidate the provenance of the alkyl group. Use of 13CH3–COOH produced 13CH3–I; deuterated acetic acid (D3C-COOD) resulted in the formation of CD3–I. These observations indicate direct transfer of the alkyl group from the carboxylic acid to iodide, consistent with the suggestion that the mechanism of synthesis involves radical induced reactions.  相似文献

The aim of this study was to retrospectively diagnose and confirm inherited metabolic diseases (IMD), from a small population of IMD high‐risk patients, with the aid of gas chromatography–tandem mass spectrometry (GC–MS/MS), technologies yet to be popularized in Sichuan, China. Using GC–MS/MS coupled with clinical diagnosis, we retrospectively analyzed samples of dried blood spots and urine specimen from 183 IMD high‐risk infant patients, who visited the West China Second Hospital of Sichuan University between June 2013 and October 2015. Four out of 183 IMD high‐risk infant patients were finally diagnosed to be IMD positive, among which two patients were identified with phenylketonuria, one with maple syrup urine disease, and 1 with methylmalonic academia. Restrictive diets and other symptomatic treatments were employed to treat the confirmed infant patients whose conditions are still under tracking and there are zero cases of death so far. GC–MS/MS was found to be an efficient and reliable way to detect IMD. It is necessary to apply GC–MS/MS, in addition to other clinical approaches, for diagnosing candidate IMD patients so that the confirmed patients can get medical intervention and timely treatment.  相似文献

A sensitive and high‐throughput inhibition screening liquid chromatography–mass spectrometry (LC‐MS/MS) method was developed and validated for the simultaneous quantification of five probe metabolites (7‐hydroxycoumarin, CYP2A6; 4‐hydroxytolbutamide, CYP2C9; 4′‐hydroxymephenytoin, CYP2C19; α‐hydroxymetoprolol, CYP2D6; and 1‐hydroxymidazolam, CYP3A4) for in vitro cytochrome P450 activity determination in human liver microsome and recombinant. All the metabolites and the internal standard, tramadol, were separated on a Waters 2695 series liquid chromatograph with a Phenomenex Luna C18 column (150 × 2.0 mm, 5 µm). Quality control samples and a positive control CYP inhibitor were included in the method. The IC50 values determined for typical CYP inhibitors were reproducible and in agreement with the literature. The method was selective and showed good accuracy (99.13–103.37%), and inter‐day (RSD < 6.20%) and intra‐day (RSD < 6.13%) precision. Also, the incubation extracts of the sample were stable at room temperature (20 °C) for 48 h and for 96 h in the autosampler (4 °C). The presented method is the first HPLC‐MS/MS method of this combination for simultaneous detection of the five metabolites 7‐hydroxycoumarin, 4‐hydroxytolbutamide, 4′‐hydroxymephenytoin, α‐hydroxymetoprolol and 1‐hydroxymidazolam in a single‐run process. Antihistamines are excreted into breast milk in small amounts; however, there are no adequate published studies or data concerning their effects on newborns and safety during breastfeeding. Thus, the development of sensitive and specific methodologies for the determination of antihistamines in breast milk is critical. A simple and sensitive GC–MS method for the simultaneous determination of 11 antihistamines (diphenhydramine, orphenadrine, chlorpheniramine, dimethindene, meclozine, hydroxyzine, loratadine, desloratadine, cetirizine, rupatadine and ebastine) in breast milk was developed and validated. The antihistamines were solid‐phase extracted and derivatized with acetic anhydride and n‐propanol. Diazepam‐d5, hydroxyzine‐d4 and cetirizine‐d8 were used as internal standards. Absolute recovery values for all analytes ranged from 70.5 to 120.0%, while the limits of detection and quantification for all analytes were 1.50 and 5.00 ng/mL, respectively. All calibration curves were linear (R2 ≥ 0.990) within the range 5.00–1000.0 ng/mL. Accuracy (Er) ranged between −7.6 and 7.0%, while precision (RSD) was <12% for all antihistamines. The developed method is suitable for the investigation of antihistamine‐related clinical cases, as well as for pharmacokinetic and breastfeeding safety studies.  相似文献

Psoralea Corylifolia L. is a traditional Chinese medicine with many beneficial effects in medical therapies. Bakuchiol was the main active ingredient of Psoralea Corylifolia L., used for the treatment of various diseases and also as a natural food additive. A specific and reliable ultra‐high performance liquid chromatography–tandem mass spectrometry has been developed and fully validated for the quantification of bakuchiol in rat plasma. Chromatographic separation of bakuchiol and an internal standard, daidzein, was achieved on a Hypersil Gold C18 column with gradient elution that consisted of methanol and water at a flow rate of 0.2 mL/min. The compounds were detected at negative ionization mode using mass transition m/z 255.2 → 172.0 and 252.9 → 132.0 for bakuchiol and daidzein, respectively. Good linearity was obtained over the range of 2–1000 ng/mL and the lower limit of quantification was 2 ng/mL. The intra‐ and inter‐day accuracies ranged from 91.1 to 105.7% and precisions (relative standard deviations) were within 9.3%. Bakuchiol was found to be stable under three freeze–thaw cycles, short‐term temperature, post‐preparative and long‐term temperature conditions. A simple pretreatment method with liquid chromatography–tandem mass spectrometry (LC–MS/MS) was developed and validated to simultaneously determine dichlorvos and phoxim in tobacco and soil matrices. Satisfactory linearity (R2 ≥ 0.9991) of the method was obtained for both analytes. The limits of detection and limits of quantification for dichlorvos and phoxim in three matrices were 0.0015–0.006 and 0.005–0.02 mg/kg, respectively. Average recoveries were 78.24–92.21% for dichlorvos and 76.62–100.51% for phoxim in soil, green tobacco leaves and cured tobacco leaves. The intra‐ and inter‐day relative standard deviations were <6%. The established method was successfully applied for the residual analysis of dichlorvos and phoxim in real soil and tobacco samples. The results indicated that the established method could be used to detect trace amounts of dichlorvos and phoxim in tobacco. The data could also help the Chinese government establish maximum residue limits of dichlorvos and phoxim on tobacco and establish proper and safe use of dichlorvos and phoxim on tobacco plants in China.  相似文献

Supercritical fluid was used to extract volatile components from the rhizoma of Atractylode lancea (A. lancea). An orthogonal array design (OAD), L9 (3)4, was employed as a chemometric method for the optimization of the supercritical fluid extraction (SFE) of volatile compounds from the herbal medicine. Four parameters, namely, pressure, temperature, dynamic extraction time, and flow rate of CO2, were studied and optimized by a three‐level OAD in which the interactions between the parameters were neglected. These compounds were identified according to their retention times and mass spectra by GC–MS. A total of 30 compounds of SFE extracts were identified. Atractylon (8.63%), hinesol (1.44%), β‐eudesmol (6.64%), elemol (0.42%), and atractydin (13.92%) were the major sesquiterpenes identified in A. lancea SFE extracts.  相似文献

Systematic comparative research was conducted on essential oils from eight traditional Chinese medicines (TCM) of pungent flavor and cool nature because the essential oils are the main active ingredients of herbs of this kind. The work was based on their component analysis by gas chromatography–mass spectrometry (GC–MS), on their retention indices, as well as on chemometric resolution methods. A total of 144 compounds were tentatively identified, accounting for 69.0% to 91.8% of the total essential oils. It is worth noting that there are 67 compounds in at least three of these eight essential oils. Moreover, many biologically active compounds, such as hexanal, α‐pinene, camphene, β‐pinene, p‐cymene, limonene, eucalyptol, (Z)‐ocimene, γ‐terpinene, camphor, p‐menthone, 4‐terpineol, α‐terpineol, carvone, eugenol, caryophyllene, β‐farnesene, α‐curcumene, β‐selinene, δ‐cadinene, caryophyllene oxide, cedrol, n‐hexadecanoic acid, benzaldehyde, benzeneacetaldehyde, phthalic acid diisobutyl ester, linoleic acid, tetradecanoic acid, (Z,Z,Z)‐9,12,15‐octadecatrienoic acid, eucalyptol, pentadecanoic acid, hexadecanoic acid methyl ester, linoleic acid methyl ester, exist in at least four of the eight essential oils. These results might help us to understand why the eight herbs are all of pungent flavor and cool nature according to the theory of TCM, and may provide a useful chemical basis for future research on herbs of this kind.  相似文献
