In order to reveal more information about the toxicity caused by metals and furthermore their influence to the physiological metabolism of the cell, the hexapeptide model Ac-ThrTyrThrGluHisAla-am representing the C-terminal 71-76 fragment of histone H4 which lies into the nucleosome core, was synthesized. A combined pH-metric and spectroscopic UV-VIS, EPR, CD and NMR study of Ni(II) and Cu(II) binding to the blocked hexapeptide, revealed the formation of octahedral complexes involving imidazole nitrogen of histidine, at pH 5 and pH 7 for Cu(II) and Ni(II) ions respectively. In basic solutions a major square-planar 4 N Ni(II)-complex, adopting a {N(Im), 3N(-)} coordination mode, was formed. In the case of Cu(II) ions, a 3 N complex, involving the imidazole nitrogen of histidine and two deprotonated amide nitrogens of the backbone of the peptide, at pH 7 and a series of 4 N complexes starting at pH 6.5, were suggested. In addition Ni(II)-mediated hydrolysis of the peptide bond-Tyr-Thr was evident following our experimental data.  相似文献

Waglerin I is a 22 amino acid snake venom toxin. Its three fragments (GGKPDLRPCHP-NH2, PCHYIPRPKPR-NH2, PCHPPCHYIPR-NH2), due to the presence of two Cys and His residues, are potentially very attractive ligands for transition metal ions. The main aim of this work was to establish the impact of these two adjacent residues on Ni2+ ion binding, especially because this kind of motif is very common in nature, and the study of low molecular weight models could be helpful in understanding larger systems. In this work waglerin fragments and their N-protected analogues were studied with Ni2+ (and Cu2+ for peptides with disulfide bridges) ions using combined potentiometric and spectroscopic measurements (UV-Vis, CD, EPR and NMR). In all peptides, except PCHPPCHYIPR-NH2 with a disulfide bridge, the Cys-His motif was found to be crucial for the coordination of Ni2+ ions. In the case of the N-unprotected analogues, the N-terminal amino group participates in the coordination as well.  相似文献

Chromatin proteins are believed to represent reactive sites for metal ion binding. We have synthesized the 31 amino acid peptide Ac-NSFVNDIFERIAGEASRLAHYNKRSTITSRE-NH2, corresponding to the 63-93 fragment of the histone H2B and studied its interaction with Cu(II) and Ni(II). Potentiometric and spectroscopic studies (UV-vis, CD, NMR and EPR) showed that histidine 21 acts as an anchoring binding site for the metal ion. Complexation of the studied peptide with Cu(II) starts at pH 4 with the formation of the monodentate species CuH2L. At physiological pH values, the 3N complex (N(Im), 2N(-)), CuL is favoured while at basic pH values the 4N (N(Im), 3N(-)) coordination mode is preferred. Ni(II) forms several complexes with the peptide starting from the distorted octahedral NiH2L at about neutral pH, to a square planar complex where the peptide is bound through a (N(Im), 3N(-)) mode in an equatorial plane at basic pH values. These results could be important in revealing more information about the mechanism of metal induced toxicity and carcinogenesis.  相似文献

Four dithiooxalato (Dto) bridged one-dimensional Ni(ll) and Ni(ll)Cu(ll) complexes (Me6[14]dieneN4)Ni2(Dto)2) (1), (Me6[14]dieneN4)CuNi(Dto)2 (2), (Me6[14]aneN4)Ni2(Dto)2 (3), and (Me6[14]aneN4)CuNi(Dto)2 (4), were synthesized. These complexes have been characterized by elemental analysis, IR, UV and ESR spectra. The crystal structure of complex3 was determined. It crystallizes in the monoclinic system, space group C2/c with a = 2. 2425(4) nm,b = 1.0088(2) nm,c= 1.4665(3) nm, β= 125.32(3)δ Z = 4;R = 0.076, Rw = 0.079. In the complex, Ni(1) coordinates four sulphur atoms of two Dto ligands in plane square environment. Ni(2) lies in the center of macrocyclic ligand. For Dto ligand, two sulphur atoms coordinate Ni(1), and O(1) coordinates Ni(2) and forms weak coordination bond. O(2) is linked to N(2) of macrocyclic ligand through hydrogen bond.  相似文献

Tridentate ligands 2-hydroxyphenylsalicylaldimine (SAPH2), 2-hydroxyphenyl-2-hydroxy-1-naphtalaldimine (NAPH2) and Ni(II) complexes with multidentate ligand Bis-N·N′-(salicylidene)-1,3-propanediamine (LH2) as well as mononuclear complex of Cu(II) were prepared using the same multidentate ligand. Diethylamine (Et2NH), NH3 and H2O monodentate ligands were bound to these complexes coordinatively. The heat absorbed at the temperatures where these ligands thermally dissociated from the complexes were measured using the TG and DSC methods. It is assumed that the states both of the complexes with and without the monodentate ligands are solid and coordination bond energy for the monodentate ligand is calculated. It is seen that these calculated coordination bond energies are comparable with hydrogen bond energies.  相似文献

Cu(II) complexes have been prepared with 2-n-propylaminopyridine N-oxide (nP) employing the perchlorate, tetrafluoroborate, nitrate, chloride and bromide salts. Preparative molar ratios of 4:1 and 2:1 ligand to Cu(II) salt yielded the following unique salts: Cu(nP)4X2(X=ClO4, BF4, NO3staggeredt-) and Cu(nP)2X2)(X=NO3, Cl, Br). Characterization has been accomplished primarily by IR, electronic and electron spin resonance measurements of the solid state. nP binds as a monodentate ligand via its N-oxide oxygen in the complexes prepared from the Cu(II) salts of polyatomic anions and as a bidentate ligand in the halogen solids. Anion coordination occurs in the halogen complexes as well as in Cu(nP)2(NO3)2.  相似文献

A new asymmetric oligobipyridine ligand, 1- (5’-methyl-2, 2’-bipyridin-5-y1)-2- (6’-methyl-2, 2’-bipyridin-6-yl)ethane (L), in which the bipyridine units are bridged by CH2CH2 at 5,6’-position has been synthesized. The ligand L reacts with Cu(I) and Cu(I1) ions giving double-stranded helical complexes [Cu 2 1 L2](C104)2.Et20 (1) and [Cu 2 II L2,(OH)(H20) ] [ClO4]3(2), respectively. Complexes 1 and 2 were characterized by X-ray diffraction analyses, ES-MS, ESR and cyclic voltammetry, etc. Differing from the oligobipyridine ligands bridged by CH2CH2 at 6,6’-or 5,5’-position, the ligand L not only forms a double-stranded helicate with Cu(1) ion, but also gives a double-stranded helicate with Cu(I1) ion. The results show that the linkage mode of the spacer group to the bipyridine units exerts a great impact on the formation of helix. The stability constants of Ni(II) complexes with 2-methylhydrazinedithio-carboxylic and 3,3-dimethylhydrazinedithiocarboxylic acids and Ni(II) and Cu(II) complexes with 3-methyl-3-phenylhydrazinedithiocarboxylic acid have been determined in aqueous solution at 25°C and I = 0.01 M. The Bjerrum method of corresponding solutions has been applied, revealing the simultaneous presence of 1:1 and 1:2 complexes in solution.  相似文献

Imidazolinone and triazine herbicides are used in many countries and may have a great impact on metal biocycles in soil. This article deals with the dynamics of imidazolinone and triazine herbicides in soils related to the formation of complexes with Cu(II) ions, which can be very stable. The stability constants of the complexes formed by five imidazolinone herbicides and ten triazine herbicides with Cu(II) ions are determined by means of fast, easy, and inexpensive measurements performed by ultraviolet–visible spectroscopy, for imidazolinones, and voltammetry (cyclic and differential pulse), for triazines. Because of the occurrence of dissociation reactions, the determinations were performed at three pH values for imidazolinones and at one pH value for triazines. In aqueous solutions of 5 < pH < 10 (corresponding to the majority of soils of agricultural use), the herbicides form very stable complexes with the Cu(II) ions, the complexes being integrated by two ligands (herbicides) and one copper ion. In conclusion, crops treated with such herbicides in conjunction with Cu(II) salts experience a decrease in its persistence and effectiveness. In addition, the herbicides and the copper ions may pass to the phreatic layer of the soil, increasing the chance of pollution.  相似文献

Four new heterotrinuclear complexes have been synthesized and characterized, namely {[Ni(L)2]2[Cu(opba)]}(ClO4)2, where opba denotes o-phenylenebis(oxamato) and L stands for 1,10-phenanthroline(phen) (1), 5-nitro-l,10-phenanthroline(NO2-phen) (2), 2,2′-bipyridyl(bpy) (S) and 4,4′-dimethyl-2,2′-bipyridyl(Me2bpy) (4). The temperature dependence of the magnetic susceptibility of {[Ni(phen)2]2[Cu(opba)]}(ClO4)23H2O has been studied in the 4–300 K range, giving the exchange integral J—109 cm?1. The HMT vs. T plot exhibits a minimum at about 100 K, characteristic of this kind of coupled polymetallic complex with an irregular spin-state structure.  相似文献

The kinetics of the formation and dissociation of the Cu(2+) and Ni(2+) complexes with a series of N(2)S(2) macrocycles, in which the ring size and the geometry of the arrangement of the donor groups have been varied, have been measured at 25 [degree]C and I= 0.5 (KNO(3)). Both the deprotonated (L) and the monoprotonated (LH(+)) form of the ligands are reactive species in the formation step. In their deprotonated form, first bond formation, in some cases supported by an ICB effect, is rate determining, independently of the ring size. In the monoprotonated form, we find slower rates, due to the charge repulsion and/or conformation changes induced by hydrogen bonds. In contrast the mechanism of the dissociation is very dependent on the ring size. The complexes with the smaller rings react as flexible open-chain ligands directly with H(+). In contrast, the complexes with the larger rings react in a similar way as rigid ligands: first the metal amine bond slowly dissociates so that the free electron pair of the amine can take the "out conformation" and then it is protonated. The 14-membered macrocycle L(3) forms complexes in which the metal ions are ideally coordinated so that their dissociation becomes extremely slow.  相似文献

Tryptophan-containing N-acetylated peptides AcTrp-Gly, AcTrp-Ala, AcTrp-Val, and AcTrp-ValOMe bind to platinum(II) and undergo selective hydrolytic cleavage of the C-terminal amide bond; the N-terminal amide bond remains intact. In acetone solution, bidentate coordination of the tryptophanyl residue via the C(3) atom of indole and the amide oxygen atom produces complexes of spiro stereochemistry, which are characterized by (1)H, (13)C, and (195)Pt NMR spectroscopy, and also by UV-vis, IR, and mass spectroscopy. Upon addition of 1 molar equiv of water, these complexes undergo hydrolytic cleavage. This reaction is as much as 10(4)-10(5) times faster in the presence of platinum(II) complexes than in their absence. The hydrolysis is conveniently monitored by (1)H NMR spectroscopy. We report the kinetics and mechanism for this reaction between cis-[Pt(en)(sol)(2)](2+), in which the solvent ligand is water or acetone, and AcTrp-Ala. The platinum(II) ion as a Lewis acid activates the oxygen-bound amide group toward nucleophilic attack of solvent water. The reaction is unimolecular with respect to the metal-peptide complex. Because the tryptophanyl fragment AcTrp remains coordinated to platinum(II) after cleavage of the amide bond, the cleavage is not catalytic. Added ligand, such as DMSO and pyridine, displaces AcTrp from the platinum(II) complex and regenerates the promoter. This is the first report of cleavage of peptide bonds next to tryptophanyl residues by metal complexes and one of the very few reports of organometallic complexes involving metal ions and peptide ligands. Because these complexes form in nonaqueous solvents, a prospect for cleavage of membrane-bound and other hydrophobic proteins with new regioselectivity has emerged.  相似文献

A general molecular mechanics (MM) model for treating aqueous Cu2+ and Zn2+ ions was developed based on valence bond (VB) theory and incorporated into the atomic multipole optimized energetics for biomolecular applications (AMOEBA) polarizable force field. Parameters were obtained by fitting MM energies to that computed by ab initio methods for gas‐phase tetra‐ and hexa‐aqua metal complexes. Molecular dynamics (MD) simulations using the proposed AMOEBA‐VB model were performed for each transition metal ion in aqueous solution, and solvent coordination was evaluated. Results show that the AMOEBA‐VB model generates the correct square‐planar geometry for gas‐phase tetra‐aqua Cu2+ complex and improves the accuracy of MM model energetics for a number of ligation geometries when compared to quantum mechanical (QM) computations. On the other hand, both AMOEBA and AMOEBA‐VB generate results for Zn2+–water complexes in good agreement with QM calculations. Analyses of the MD trajectories revealed a six‐coordination first solvation shell for both Cu2+ and Zn2+ ions in aqueous solution, with ligation geometries falling in the range reported by previous studies. © 2012 Wiley Periodicals, Inc.  相似文献

The spectra of five aquoamino complexes have been established by resolving the observed absorption spectra of solutions containing salts of Ni(II) and ammonia. These spectra have been used to deduce the parameters of the bonds on the assumption of octahedral symmetry. The antibonding and nepheloxet effects are found to be nonadditive and nonlinear; the behavior can be explained on the basis of the interaction between the central ion and the water molecules.  相似文献

The preparation and characterization are described for four ruthenium(II) complexes containing hemilabile phosphine-ether ligand o-(diphenylphosphino)anisole (Ph2PC6H4OMe-o) and/or bidentate ligand diphenylphosphino-phenolate ([Ph2PC6H4O-o]) Ru(RCN)22-Ph2PC6H4O-o)2 (1a: R = Me; 1b: R = Et) and [Ru(RCN)22-Ph2PC6H4O-o)(κ2-Ph2PC6H4OMe-o)](PF6) (2a: R = Me; 2b: R = Et). The ruthenium(II) phosphine-ether complexes undergo mild methyl-oxygen bond cleavage. Two different kinds reaction mechanism are proposed to describe the methyl-oxygen bond cleavage, one involving attack of anionic nucleophiles and another involving the phosphine. The new reactions define novel routes to phosphine-phenolate complexes. The structures of complexes 1a, 1b and 2a were confirmed by X-ray crystallography.  相似文献

Potentiometric studies have been carried on complexes of sulphamethazine salicylaldimine (SUMTSA) with copper, nickel and cobalt at 25°C. The Calvin Bjerrum pH titration technique as used by Irving and Rossatti has been applied to determine the protonation constant of the ligand and the formation constants of the complexes. The log K values have been computed by least square method. Free energy changes and probable errors have also been evaluated.  相似文献

This communication will describe the electron doping effect into Ni(III) complexes by Cu(II) ions, [Ni(1-x)Cu(x)(chxn)(2)Br]Br(2-x) (x = 0.038 and 0.101) by using an electrochemical oxidation method. A drastic increase of electrical conductivity as well as a new absorption band around 0.5 eV in single crystal reflectance spectra was observed by doping Cu(II) ions, indicating the electron doping was successfully made. An ESR result shows unpaired electrons locate in the d(x2-y2) orbitals of Cu(II) and have almost no interaction with those of other ions.  相似文献

Four oxamato-bridged heterotrinuclear Ni(II)Cu(II)Ni(II) complexes of formula ([Ni(bispictn)](2)Cu(pba))(ClO(4))(2).2.5H(2)O (1), ([Ni(bispictn)](2)Cu(pbaOH))(ClO(4))(2).H(2)O (2), ([Ni(cth)](2)Cu(pba))(ClO(4))(2) (3), and ([Ni(cth)](2)Cu(opba))(ClO(4))(2).H(2)O (4) and a binuclear Ni(II)Cu(II) complex of formula [Cu(opba)Ni(cth)].CH(3)OH (5) have been synthesized and characterized by means of elemental analysis, IR, ESR, and electronic spectra, where pba = 1,3-propylenebis(oxamato), pbaOH = 2-hydroxyl-1,3-propylenebis(oxamato), opba = o-phenylenebis(oxamato), bispictn = N,N'-bis(2-pyridylmethyl)-1,3-propanediamine, and cth = rac-5,7,7,12,14,14-hexamethyl-1,4,8,11-tetraazacyclotetradecane. The crystal structures of 1, 3, and 5 have been determined. The structures of complexes 1 and 3 consist of trinuclear cations and perchlorate anions, and that of 5 consists of neutral binuclear molecules which are connected by hydrogen bonds and pi-pi interactions to produce a unique supramolecular "double" sheet. In the three complexes, the copper atom in a square-planar or axially elongated octahedral environment and the nickel atom in a distorted octahedral environment are bridged by the oxamato groups, with Cu.Ni separations between 5.29 and 5.33 A. The magnetic properties of all five complexes have been investigated. The chi(M)T versus T plots for 1-4 exhibit the minimum characteristic of antiferromagnetically coupled NiCuNi species with an irregular spin state structure and a spin-quartet ground state. The chi(M)T versus T plot for 5 is typical of an antiferromagnetically coupled NiCu pair with a spin-doublet ground state. The Ni(II)-Cu(II) isotropic interaction parameters for the five complexes were evaluated and are between 102 and 108 cm(-)(1) (H = -JS(Cu).S(Ni)).  相似文献
