Reaction of 5,6‐dihydro‐4H‐pyrrolo[3,2,1‐ij]quinoline‐1,2‐dione ( 1 ) with two equivalents of some 6‐aminouracils (or 6‐amino‐2‐thiouracil) generates spirocyclic tetrahydrobenzo[if]quinolizines ( 7 ). The one‐pot, three‐component reaction of amido ketone ( 1 ) with 6‐aminouracil (or 6‐amino‐2‐thiouracil) and a cyclic six‐membered 1,3‐diketone produces spirocyclic tetrahydropyrrolo[3,2,1‐ij]quinolinones ( 15 ).  相似文献

A synthesis of pyrrolo[3,2‐c]quinolines substituted in the 7‐ and 8‐ positions by methoxy groups and in the 3‐ position by an amido group is described. The structures were designed to have a crescent shape, a planar fused cyclic moiety with two ortho methoxy groups and ionisable amino or amidinic group at pH 7.  相似文献

Treatment of 2‐hydroxy‐, 2‐mercapto‐, and 2‐ethoxycarbonylamino‐benzonitriles 12 with 2‐fluoro‐ or 2‐nitrophenacylbromides 13 under alkaline conditions provided the corresponding benzofuran, benzothiophene, and indole intermediates 10 , respectivelly. Nucleophilic cyclization of these compounds led to the corresponding tetracyclic quinolinones 7a, 7b , and 3. Denitrocyclization reaction of compounds 10 (R = NO2) was found especially useful. Compounds 7a, 7b , and 3 were converted to their chloro derivatives 14a‐c , which were reduced with hydrogen and a catalyst to the corresponding compounds 8a, 8b , and 2. The presented pathway represents a new method of preparation of quindoline 2 and its O and S analogs 8. Chloro derivatives 14 are reactive enough to provide the corresponding methoxy derivatives 15 and dimethylamino derivatives 16. Methylation of compounds 7a and 7b with iodomethane providing mixtures of major N‐methyl derivatives 17 and minor O‐methyl derivatives 15 were also studied.  相似文献

A series of novel fluorinated 11H‐azaindolo[3,2‐c]isoquinolines ( 7 ) have been synthesized starting from 2(1H)pyridones ( 1 ) via azaindoles ( 5 ). Initially, compound 1 was treated with POCl3/DMF, and the resulting compound 2 was reacted with benzylamine to obtain compounds 3 that were subjected to cyclization after protecting the secondary amine to get azaindoles ( 5 ). Further, compounds 5 were subjected to cyclization as per Pictet–Spengler reaction condition. However, it was not successful. Subsequently, the azaindoles ( 5 ) were acetylated and then cyclized to give title compounds 7 . These compounds are new and well characterized by spectral data.  相似文献

Several new benzo[ij]pyrano[2,3‐b]quinolizine‐8‐ones 5 and 4H‐pyrano[2,3‐b]pyridine 8 derivatives were synthesized from 4‐hydroxyquinolines 1 . Reacting 3‐acetyl‐4‐hydroxy‐1‐phenyl‐1H‐quinoline‐2‐one with dimethylformamide dimethylacetal afforded 3‐(3‐Dimethylarnino‐acryloyl)‐4‐hydroxy‐1‐phenyl‐1H‐quinolin‐2‐one 9 . This reacted with hippuric acid and diethyl 3‐oxoglutarate to give 2H‐pyran‐2‐one 13 and pyranopyridoquinoline 17 respectively.  相似文献

A simple and efficient method for the cis‐selective synthesis of pyrano‐ and furano[3,2‐c]quinolines via gadolinium chloride catalyzed one‐pot aza–Diels–Alder reaction is described. The crystal structures of pyrano­quinolines 9‐fluoro‐5‐phenyl‐3,4,4a,5,6,10b‐hexa­hydro‐2H‐pyrano[3,2‐c]quinoline, C18H18FNO, and 9‐methyl‐5‐phenyl‐3,4,4a,5,6,10b‐hexa­hydro‐2H‐pyrano­[3,2‐c]quinoline, C19H21NO, are isomorphous. In both structures, the pyran ring is exo to the six‐membered N‐heterocyclic ring formed in the cyclo­addition step. The torsion angles across the phenyl linkage for the two structures are −91.2 (1) and −88.3 (2)°. The striking feature in both crystal packings is that they do not contain the expected conventional hydrogen bonds, in spite of the presence of good hydrogen‐bonding functionalities. Possible C—H⋯π inter­actions are, however, observed.  相似文献

Furocoumarins 3,5 and pyranocoumarin 7 were synthesized from the reaction of furonaphthalenediones 2,4 and pyranonaphthalenedione 6 respectively with carbethoxymethylene(triphenyl)phosphorane in refluxing DCM for 3‐6 hours or under microwave irradiation in toluene for a few minutes. Compounds 3,5,7 and their precursors were tested as anti‐inflammatory/antioxidant agents. They were found to compete significantly high DMSO for OH radicals, to scavenge O2? and to inhibit lipoxygenase to a high extent.  相似文献

8‐Chloro‐9‐fluoro‐5‐phen­yl‐3,4,4a,5,6,10b‐hexa­hydro‐2H‐pyrano[3,2‐c]quinoline and 10‐chloro‐9‐fluoro‐5‐phen­yl‐3,4,4a,5,6,10b‐hexa­hydro‐2H‐pyrano­[3,2‐c]quinoline, both C18H17ClFNO, are diastereo­isomers, formed as the result of the imino Diels–Alder reactions of N‐benzyl­ideneanilines with 3,4‐dihydro‐2H‐pyran. The crystal structures reveal the stereochemistry of the pyran ring, which is endo/exo to the quinoline ring system formed in the cyclo­addition step. In both structures, the pyran ring adopts a chair conformation, while the nitrogen‐containing heterocyclic ring prefers a half‐chair conformation. The structures differ essentially in the relative orientation of the ring junction H atoms.  相似文献

Comparable spiro condensation products are also obtained using 4‐hydroxy‐2H‐1‐benzopyran‐2‐one in place of cyclohexane‐1,3‐diones.  相似文献

A simple and efficient approach to synthesize a novel pyrrolocoumarin 9‐hydroxy‐8H‐pyrano[3,2‐f]indol‐2‐one ( 7 ) has been described. Starting from vanillin, the key intermediate 7‐methoxy‐1H‐indol‐6‐yl propiolate ( 6 ) was synthesized in six steps. Then, the target compound was obtained by forming pyrone‐ring and demethylation simultaneously in one step. A plausible mechanism invoking PtCl4 catalyzed one‐step reaction of cyclization and demethylation was also presented.  相似文献

The three‐component reaction of N‐phenacylbenzothiazolium bromides, aromatic aldehydes and indane‐1,3‐dione in ethanol at room temperature in the presence of triethylamine as base afforded functionalized spiro[benzo[d]pyrrolo[2,1‐b]thiazole‐3,2′‐indenes] in good yields and with high diastereoselectivity. The 1H NMR data and single crystal structure clearly indicated that the obtained spiro compounds predominately have one diastereoisomer.  相似文献

Heating of hexahydro‐5H‐pyrrolo[2,1‐c][1,4]benzodiazepine‐2,5,11‐trione in boiling phosphoryl chloride led to a rearranged product like 3,5‐dichlorobenzo[h][1,6]naphthyridine. This structure was established from X‐ray diffraction analysis.  相似文献

Two series of furo[3,2‐f ]quinoline‐2‐carboxylate were obtained via a three‐component reaction of aldehydes, 5‐aminobenzofuran‐2‐carboxylate, and 4‐hydroxy‐2H‐chromen‐2‐one or cyclopentane‐1,3‐dione in DMF under catalyst‐free condition in high yields. This one‐pot three‐component reaction provided an efficient method for the synthesis of fused polycyclic heterocycles for bioactive screening.  相似文献
