Dimethyl dihydro-1,2,4,5-tetrazine-3,6-dicarboxylate can exist either in 1,2- or 1,4-dihydro tautomeric forms. The 15N NMR spectra of dimethyl dihydro-1,2,4,5-tetrazine-3,6-dicarboxylate were measured at the 15N natural abundance level as well as in 15N doubly labelled selectively and in 15N completely labelled compounds (20% 15N). The J(15N,15N) value was determined in 15N completely labelled compounds (20% 15N) using 1D 15N INADEQUATE and was found to be 12.2 ± 0.2 Hz in deuteriochloroform, acetonitrile-d3, DMSO-d6 and CD3OH. Very similar 15N chemical shifts and 1J(15N,1H) values were also observed in all the solvents. This indicates that compound 1 exists completely in the 1,4-dihydro tautomeric form (i.e., as dimethyl 1,4-dihydro-1,2,4,5-tetrazine-3,6-dicarboxylate) in all the solvents tested.  相似文献

An analytical method for the determination of three polyamines (putrescine, spermidine, and spermine) and five acetylpolyamines [N1-acetylspermidine (N1AcSpd), N8-acetylspermidine (N8AcSpd), N1-acetylspermine, N1,N8-diacetylspermidine, and N1,N12-diacetylspermine] involved in the polyamine catabolic pathway has been developed using a hybrid tandem mass spectrometer. Heptafluorobutyryl (HFB) derivatives of these compounds and respective internal standards labeled with stable isotopes were analyzed simultaneously by TOF MS, based on peak areas appearing at appropriate m/z values. The isomers, N1AcSpd and N8AcSpd were determined from their fragment ions, the acetylamidopropyl and acetylamidobutyl groups, respectively, using MS/MS with 13C2-N1AcSpd and 13C2-N8AcSpd which have the 13C2-acetyl group as an internal standard. The TOF MS method was successfully applied to measure the activity of enzymes involved in polyamine catabolic pathways, namely N1-acetylpolyamine oxidase (APAO), spermine oxidase (SMO), and spermidine/spermine N1-acetyltransferase (SSAT). The following natural substrates and products labeled with stable isotopes considering the application to biological samples were identified; for APAO, [4,9,12-15N3]-N1-acetylspermine and [1,4,8-15N3]spermidine (15N3-Spd), respectively; for SMO, [1,4,8,12-15N4]spermine and 15N3-Spd, respectively; and for SSAT, 15N3-Spd and [1,4,8-15N3]-N1-acetylspermidine, respectively.  相似文献

A simple and efficient synthetic route to the novel 3a,4-dihydro-3H,7H- and 4H,7H-pyrazolo[4′,3′:5,6]pyrano[4,3-c][1,2]oxazole ring systems from 3-(prop-2-en-1-yloxy)- or 3-(prop-2-yn-1-yloxy)-1H-pyrazole-4-carbaldehyde oximes has been developed by employing the intramolecular nitrile oxide cycloaddition (INOC) reaction as the key step. The configuration of intermediate aldoximes was unambiguously determined using NOESY experimental data and comparison of the magnitudes of 1JCH coupling constants of the iminyl moiety, which were greater by approximately 13 Hz for the predominant syn isomer. The structures of the obtained heterocyclic products were confirmed by detailed 1H, 13C and 15N NMR spectroscopic experiments and HRMS measurements.  相似文献

A straightforward, two-step synthesis of fluoro substituted chromeno[2,3-c]pyrazol- and [1]benzothieno[2′,3′:5,6]pyrano[2,3-c]pyrazol-4(1H)-ones, respectively, is presented. Hence, treatment of 1-substituted or 1,3-disubstituted 2-pyrazolin-5-ones with fluoro substituted 2-fluorobenzoyl chlorides or 3-chloro-6-fluoro-1-benzothiophene-2-carbonyl chloride using calcium hydroxide in refluxing 1,4-dioxane gave the corresponding 4-aroylpyrazol-5-ols, which were cyclized into the fused ring systems. 5-Fluorochromeno[2,3-c]pyrazol-4(1H)-one was obtained upon treatment of the 1-(4-methoxybenzyl) protected congener with trifluoroacetic acid. Treatment of 5-fluorochromeno[2,3-c]pyrazol-4(1H)-ones with methylhydrazine afforded novel tetracyclic ring systems such as 2-methyl-7-phenyl-2,7-dihydropyrazolo[4′,3′:5,6]pyrano[4,3,2-cd]indazole. Detailed NMR spectroscopic investigations (1H, 13C, 15N, 19F) with the obtained compounds were undertaken.  相似文献

In the course of our program focused on the preparation of high-nitrogen content heterocyclic compounds, we wish to report an original synthesis of the tricyclic 7-azidofurazano[3,4-b]tetrazolopyrazine via an unprecedented reaction between 2,6-dimethoxy-3,5-dinitropyrazine and hydrazine hydrate. This compound was identified by an X-ray diffraction analysis. Further studies of its structure by 15N and 13C NMR spectroscopy were carried out in different solvents. This allowed us to observe a noteworthy equilibrium involving three forms resulting from the reversible opening of a tetrazole ring.  相似文献

The one-pot reaction between 2-aminobenzo[d]isothiazol-3-one and alkyl propiolates in presence of triphenylphosphine leads to the corresponding alkyl 4-amino-5-oxobenzo[f][1,4]thiazepine-3-carboxylates. A plausible mechanism of the reaction is proposed and unambiguous evidence for the structures is obtained from a detailed magnetic resonance spectral analysis. 1D and 2D NMR spectra such as COSY, 1H-13C and 1H-15N HSQC and HMBC heteronuclear correlations and an INADEQUATE experiment are reported and discussed.  相似文献

trans-[Co(py)4Cl2]Cl·6H2O, mer-[Co(py)3Cl3] and mer-[Co(py)3(CO3)Cl] were studied by UV-Vis, far-IR and 1H, 13C, 15N NMR. The formation of Co-N bonds lead to variable in sign and magnitude changes of 1H NMR chemical shifts, heavily dependent on proton position, coordination sphere geometry and character of auxiliary ligands. 13C nuclei were deshielded upon Co(III) coordination, while 15N NMR studies exhibited ca. 85–110 ppm shielding effects (ca. 15–25 ppm more expressed for nitrogens trans to N than trans to Cl or O). 13C and 15N CPMAS spectra revealed a slight inequivalency of formally identical Co-py bonds in trans-[Co(py)4Cl2]Cl·6H2O and mer-[Co(py)3Cl3], suggesting for the latter complex an existence of distortion isomers. In chloroform, a spontaneous trans-[Co(py)4Cl2]Cl → mer-[Co(py)3Cl3] + py reaction was monitored by 1H NMR and UV-Vis. This process of py → Cl substitution allowed the design of a more convenient and efficient method of mer-[Co(py)3Cl3] preparation.   相似文献

Nitropyrazoles     
The structures of substitutedN-aminonitropyrazoles and 1- and 2-amino-4-nitro-1,2,3-triazoles as well as the site of protonation of 1-aminopyrazole were determined based on the1H,13C, and15N (14N) NMR spectra. The13C NMR spectra were recorded under conditions of13C-{1H,14N} triple resonance. Effects of substituents in the pyrazole ring on the13C and14N chemical shifts were studied. The13C,1H and15N,1H spin-spin coupling constants, obtained using techniques of [1H]13C and [1H]15N polarization transfer (SPT, INEPT), were measured, fully assigned, and discussed.Translated fromIzvestiya Akademii Nauk. Cadmium(II) complexes of 3-hydroxypicolinic acid, namely [CdI(3-OHpic)(3-OHpicH)(H2O)]2 (1), [Cd(3-OHpic)2(H2O)2] (2) and [Cd(3-OHpic)2]n (3) were prepared and characterized by spectroscopic methods (IR, NMR) and their molecular and crystal structures were determined by X-ray crystal structure analysis. Complexes 1 and 2 were prepared in similar reaction conditions using different cadmium(II) salts: cadmium(II) iodide and cadmium(II) acetate dihydrate, respectively, while 3 was prepared by recrystallization of 2 from N,N-dimethylformamide solution. Various coordination modes of 3-OHpicH in 13 were established in the solid state: bidentate N,O-chelated mode in 1 and 2, monodentate mode through the carboxylate O atom from zwitterionic ligand in 1 and bidentate N,O-chelated and bridging mode in 3. In the DMF solution of all prepared complexes, only monodentate mode of 3-OHpicH binding to cadmium(II) through the carboxylate O atom was established by 1H, 13C, 15N and 113Cd NMR spectroscopy.  相似文献

The active ingredient of ecstasy, N-methyl-3,4-methyldioxyphenylisopropylamine (MDMA) can be manufactured by a number of easy routes from simple precursors. We have synthesised 45 samples of MDMA following the five most common routes using N-precursors from 12 different origins and three different precursors for the aromatic moiety. The 13C and 15N contents of both the precursors and the MDMA samples derived therefrom were measured by isotope ratio mass spectrometry coupled to an elemental analyser (EA-IRMS). We show that within-pathway correlation between the 15N content of the precursor and that of the derived MDMA can be strong but that no general pattern of correlation can be defined. Rather, it is evident that the δ15N values of MDMA are strongly influenced by a combination of the δ15N values of the source of nitrogen used, the route by which the MDMA is synthesised, and the experimental conditions employed. Multivariate analysis (PCA) based on the δ15N values of the synthetic MDMA and of the δ15N and δ13C values of the N-precursors leads to good discrimination between the majority of the reaction conditions tested.  相似文献

Novel acridine spirocompounds, spiro[dihydroacridine-9′(10′H),5-imidazolidine]-2-thiones have been prepared by the spontaneous cyclization of 1-substituted 3-(acridin-9-ylmethyl)thioureas, which were obtained from 1-(acridin-9-yl)methanamine, N-(acridin-9-ylmethyl)propan-1-amine, and N-(acridin-9-ylmethyl)benzylamine and alkyl/aryl isothiocyanates, as continuation of our previous studies on new acridine spirocycles. Imidazolidine-2-thiones thus obtained were subsequently transformed with mesitylnitrile oxide to imidazolidine-2-one analogues, some of which partly reopened to the corresponding (acridin-9-ylmethyl)ureas. An unusual spirocyclization via a CH carbanion instead of the N-1 nitrogen has been found for 3-(acridin-9-ylmethyl)-1-(acridin-9-yl)thioureas possessing two acridine rings. Structural modifications in positions 1, 3, and 4 of the spiro ring together with 1H, 13C, and 15N NMR spectroscopy and X-ray crystallography have been employed to rationalize a general propensity of various 9-substituted acridines to undergo easy spirocyclization.  相似文献

The 1H, 13C and 15N NMR spectroscopic properties of six thallium tris-(pyrazol-1-yl)borates, including a tetrakis derivative, were determined. The results in solution were necessary to understand those, more complicated, in the solid state. A collection of 205Tl-15N and 205Tl-13C couplings was measured in the latter state. Among those, a very large coupling constant (between 194 and 282 Hz) has been measured on the carbon at the position 4 of the pyrazole ring in several compounds and particularly for the cyclobutyl derivative [Tl(TpCbu)]. It has been assigned to a direct interaction of the 4-C-H ? Tl type and related to the X-ray structures, when available.  相似文献

The15N NMR chemical shifts and15N-1H SSCCs are presented for substituted N-methylpyrazoles with substituents such as CH3, NO2, Br, Cl, NH2, O=CNH2, O=CPh, and COOH at the carbon atoms. The complete 1H, 13C and 15N NMR analyses for a series of 25 diaryl-aldimines containing phenyl, pyridyl, pyrazolone and furanyl moieties are described herein. Detailed evaluation of substituent chemical shift and coupling constant effects showed that interaction between the lone pair of the pyrazolone carbonyl group or the nitrogen of 2-substitued pyridines with the aldimine hydrogen increases the value and shifts the resonance signal for this hydrogen to high frequency, in the 1H NMR spectra. The X-ray crystal structure analysis of pyrazolone substituted aldimines evidenced the planarity of the aryl groups which are conjugated with the CN double bond. In the case of the N-(2-pyridinemethylene)-1,5-dimethyl-2-phenyl-1,2-dihydro-pyrazol-3-one, two rotamers were observed in the same unit cell.  相似文献

The recently reported LR-HSQMBC experiment has been optimized for 1H–15N long-range heteronuclear couplings. Several previously unreported four-bond correlations, consistent with the predicted by DFT calculations (0.2–0.3 Hz 4JNH couplings), have been observed for strychnine using 2 Hz optimization of the LR-HSQMBC experiment. This experiment offers an advantage over accordion-optimized experiments such as IMPEACH and CIGAR for the observation of long-range 1H–15N correlations in that the experiment is refocused and employs a CLIP pulse sequence element to bring the long-range correlations into phase, allowing broadband X-decoupling to be employed during acquisition.  相似文献

The dibenzo[3n]crown-n were synthesised from1,2-bis(o-hydroxyphenoxy)ethane obtained from 1,2-bis(o-formylphenoxy)ethane via Bayer-Willigeroxidations with H2O2/CH3COOH in good yields. The cyclic condensation of 1,2-bis(o-hydroxyphenoxy)ethanewith dichlorides, and ditosylates of polyethylene glycols in DMF/Me2CO3 gave the macrocyclesdibenzo[15]crown-5, dibenzo[18]crown-6, dibenzo[21]crown-7 anddibenzo[24]crown-8. The structures were identified using IR, mass, 1H and 13C NMR spectroscopy. Therecognition of the molecules for the cations, Li+, Na+, K+, Rb+ and Zn2+were conducted quantitatively with steady state fluorescencespectroscopy. The 1:1 association constants in acetonitrileshowed a good relation of the appropriate size of the macrocyclic ether towards the fitting cationradii. Namely, dibenzo[15]crown-5 was the best for Li+ binding and more than 100 times better thanNa+ and K+. Dibenzo[21]crown-7 was excellent for Rb+ binding while K+ is 100 timesless preferred. The largest crown ether studied, dibenzo[24]crown-8, exhibited the order of binding power,Rb+ > K+ > Na+. Zn2+ displayed, however, a marked binding with only dibenzo[18]crown-6.p>  相似文献

Mercury cyanide complexes of alkyldiamines (16), [Hg(L)(CN)2] (where L?=?en (1,2-diaminoethane), pn (1,3-diaminopropane), N-Me-en, N, N′-Me2-en, N, N′-Et2-en, and N, N′-ipr2-en), have been synthesized and characterized by elemental analysis, IR, 13C, and 15N solution NMR in DMSO-d6, as well as 13C, 15N, and 199Hg solid-state NMR spectroscopy. Complexes 1 and 2 have been studied computationally, built and optimized by GAUSSIAN03 using DFT at B3LYP level with LanL2DZ basis set. Binding modes of en and bn (where bn?=?1,4-diaminobutane) toward Hg(CN)2 are completely different. Complexes with en and pn show chelating binding to Hg(II), while bn behaves as a bridging ligand to form a polymeric structure, [Hg(CN)2-bn] [B.A. Al-Maythalony, M. Fettouhi, M.I.M. Wazeer, A.A. Isab. Inorg. Chem. Commun., 12, 540 (2009).]. The solution 13C NMR of the complexes demonstrates a slight shift of the ?C≡N (0.9 to 2?ppm) and ?C–NH2 (0.25 to 6?ppm) carbon resonances, while the other resonances are relatively unaffected. 15N labeling studies have shown involvement of alkyldiamine ligands in coordination to the metal. The principal components of the 13C, 15N, and 199Hg shielding tensors have been determined from solid-state NMR data. Antimicrobial activity studies show that the complexes exhibit higher antibacterial activities toward various microorganisms than Hg(CN)2.  相似文献

Vasoactive intestinal peptide (VIP) receptors are expressed on various tumor cells in much higher density than somatostatin receptors, which provides the basis for radiolabeling VIP as tumor diagnostic agent. However, fast proteolytic degradation of VIP in vivo limits its clinical application. With the aim to develop and evaluate new ligands for depicting the VIP receptors with positron emission tomography (PET), the structure modified [R8,15,21, L17]-VIP analog was radiolabeled with 18F using two different methods. With the first method, N-4-[18F]fluorobenzoyl-[R8,15,21, L17]-VIP ([18F]FB-[R8,15,21, L17]-VIP 7) was produced in a decay-corrected radiochemical yield (RCY) of 33.6 ± 3%, a specific radioactivity of 255 GBq/μmol (n = 5) within 100 min in four steps. Similarly, N-4-[18F](fluoromethyl)-benzoyl-[R8,15,21, L17]-VIP ([18F]FMB-[R8,15,21, L17]-VIP 8) was synthesized in a RCY of 34.85 ± 5%, a specific radioactivity of 180 GBq/μmol (n = 5) within 60 min in only one step. The two products 7 and 8 were both shown good stability in HSA. Moreover, the low bone uptakes of 7 and 8 in vivo of mice showed good defluorination stability.  相似文献
