Bisabolane Sesquiterpenes from Ligularia thyrsoidea

Phytochemistry investigation of the whole plants of Ligularia thyrsoidea, has led to the isolation of five new bisabolane sesquiterpenes along with three known compounds. The structures of these new compounds were elucidated by spectroscopic methods and 2D NMR techniques.

Two New Eremophilane Sesquiterpene Lactones from Ligularia myriocephala Ling

Two new eremophilane sesquiterpene lactones, 1β-angeloyloxy-6β, 10β-dihydroxy-8β- methoxyeremophila-7（ll）en-8α,12-olide and 1β-angeloyloxy-6β,10α-dihydroxy-8α-methoxyeremo- phila-7（ 11）en-8β, 12-olide were isolated from the extract of the whole plant of Ligularia myriocephala Ling. Their structures and stereochemistry were elucidated by various spectroscopic methods including intensive 2D-NMR techniques （COSY, gHMQC, gHMBC and ^1H-^1H NOESY） and HR-ESIMS.     

A new eremophilane-type sesquiterpene from Ligularia virgaurea

A new eremophilane-type sesquiterpene,1α,8β,10β-trihydroxy-6β-(2-methylacryloyl)oxyeremophil-7(11)-en-8α,12-olide, was isolated from the roots of Ligularia virgaurea.Its structure was established on the basis of various spectroscopic analyses, including the 1D,2D NMR techniques and HR-ESI-MS.     

Sesquiterpene Coumarins from Ferula Foetida

A thorough investigation of the ethyl acetate soluble fraction from a methanol extract of the resin of Ferula foetida has resulted in the isolation of a new sesquiterpene coumarin namely epi‐conferdione ( 1 ), along with four known compounds, colladonin ( 2 ), karatavicinol ( 3 ), 8‐acetoxy‐5‐hydroxyumbelliprenin ( 4 ), and asacoumarin ( 5 ). These structures were elucidated on the basis of extensive spectroscopic techniques, including 1D and 2D NMR spectroscopy. The absolute configuration of the new compound was established by circular dichroism (CD).     

Lyciumaside and Lyciumate: A New Diacylglycoside and Sesquiterpene Lactone from Lycium shawii

One new diacylglycoside named lyciumaside ( 1 ) and a new sesquiterpene lactone named lyciumate ( 2 ) were isolated from Lycium shawii Roem . & Schult . The structures of the two new compounds were elucidated based on 1D‐ (¹H‐ and ¹³C‐NMR and NOE) and 2D‐NMR (COSY, HSQC, and HMBC) spectroscopic techniques, and mass spectrometry (ESI‐MS). Preliminary evaluations demonstrated lyciumaside ( 1 ) possesses strong antioxidant activity with an IC₅₀ = 30 μg/ml (80% inhibition) while it was inactive in α‐glucosidase and urease enzymes assays.     

A new guaianolide from the roots of Pterocypsela elata

In order to find more biologically active substances,we have carefully investigated the chemical constituents of Pterocypsela elata.One new guaianolide,(4S)-11-methoxycarbonyl-guaiane- 1(10),5(6),7(11),8(9)-tetraen-6,l2-olide was isolated from the EtOAc extracts of the roots of P.elata. Its structure was elucidated by various spectroscopic methods and confirmed by X-ray crystallographic analysis.     

Furanocoumarins and Sesquiterpene Ketone from Clausena Lansium Skeels

Three known furanocoumarins, imperatorin (1), wampetin (dehydroindicolactone) (2) and 8-geranoxypsoralen (3) and a new sesquiterpene ketone, (+)-oplopanone (4), were isolated from Clausena lansium Skeels. The stereochemistry of the last two coumarins was determined by NOESY experiments. The idendity of (+)-oplopanone was established by comparison of its physical constants and spectroscopic data with those reported for its enantiomer as well as a comparison of its' TLC behavior with that of an authentic sample.     

Sesquiterpene Lactones from Anthemis melanolepis

From the aerial parts of A. melanolepis, two new sesquiterpene lactones, melanolepin B ( = 1α,4β,8α‐trihydroxyeudesm‐11(13)‐en‐12,6α‐olide; 1 ) and melanolepin C (= 1α,10β‐epoxy‐8β‐hydroxygermacra‐4(15),11(13)‐dien‐12,6α‐olide; 2 ), were isolated, together with four known compounds, desacetyllaurenobiolide, dentatin A, taraxasterol, and (3S,5R)‐loliolide. Compound 1 was found to have the rare cis‐fused eudesmane skeleton. The structures of the isolated compounds were established by means of NMR (¹H, ¹H‐COSY, ¹H,¹³C‐HSQC, HMBC, NOESY) and MS analyses.     

Sesquiterpene lactones from Centaurea rhizantha C.A. Meyer

Two new sesquiterpene lactones, rhizantholide A (1) and rhizantholide B (2), together with five known compounds (3-7) have been isolated from the aerial parts of Centaurea rhizantha (Asteraceae). Sesquiterpene lactones belong to guaianolide class, and rhizantholide B is a rare guaianolide characterized by a free primary alcoholic function at C-10 along with a 3β,10β-epoxy function. Their structures have been established on the basis of 1D and 2D NMR experiments, as well as HR-ESIMS. The antimicrobial activity of compounds 1-7 has been evaluated against Gram-positive and Gram-negative strains. Only deacylcynaropicrin 8-O-[3′-hydroxy-2′-methylpropionate] (5) showed moderate antibacterial activity against Staphylococcus aureus with a MIC/MBC value of 500 μg/mL. All isolated compounds have been also evaluated for their cytotoxic activities against cancer cells. Among them, compound 5 showed the highest cytotoxic activity with IC₅₀ values in the range 5.02–16.76 μg/mL.     

Sesquiterpene Lactones from Elephantopus scaber

A new germacranolide sesquiterpene lactone, isoscabertopin, was isolated from Elephantopus scaber together with the known scabertopin. Their structures were determined by spectroscopic methods.     

杯菊中的倍半萜内酯

摘要杯菊(Cyathocline purpurea)是云南民间民族药, 全株用于治疗各种炎症和肺结核. 采用L1210细胞系对杯菊的粗提物、 分步萃取物的成分进行了细胞毒试验; 用柱层色谱法同步对活性组分进行化学成分分离纯化; 通过理化数据测定及波谱分析鉴定单体结构. 杯菊的氯仿及乙酸乙酯萃取物具有细胞毒活性, 其IC50分别为3.5和2.8 μg/mL. 从活性组分中分离出3种成分, 分别鉴定为Santamarin(1)、 9β-乙酰基广木香内酯(9β-acetoxycostunolide, 2)和9β-乙酰基小白菊内酯(9β-acetoxypathenolide, 3), 其IC50分别为0.41, 0.89, 0.59 μg/mL. 这三个化合物都有较强的抗癌活性, 其中化合物3为新化合物.
