 Experiments on a newly prepared (E)-configured ω-benzal-hypericin derivative using TLC and 1H NMR together with quantum chemical calculations revealed that in stilbenoid hypericin derivatives photodiastereomerization between the (E)- and (Z)-diastereomers occurs in principle. However, due to its low diastereomerization quantum yield and photo and thermal equilibria, which reside mostly on the side of the (E)-diastereomer, this photoreaction is only of marginal importance to the photochemistry of stilbenoid hypericin derivatives. Thus, photodiastereomerization does not appreciably interfere with the photoreactions important for photodynamic therapy. This was demonstrated by comparing the sensitized bilirubin photodestruction of hypericin and the ω-benzal-hypericin derivative.  相似文献

Summary.  A hypericin derivative containing ω,ω ′-4-dimethylaminobenzal residues was shown to undergo an intramolecular [2 + 2] cycloaddition upon irradiation leading to a cyclobutane derivative whose main absorption band is hardly shifted as compared to hypericin. The corresponding ω-substituted derivative displayed a 34 nm bathochromic shift and a strongly reduced fluorescence quantum yield rendering it a nice candidate for a photodynamic therapy agent. Summary.  The (E)- and (Z)-diastereomers of resveratrol were investigated with respect to their photochemical and thermal diastereomerization reactions. The free enthalpy difference between the two diastereomers was estimated to be in the order of common stilbenes, with the (E)-diastereomer more stable by about 11–14 kJ mol−1. The Arrhenius activation barrier of about 280 kJ mol−1 was found to be quite high and implies that thermal equilibration cannot account for the (Z)-diastereomer found in nature. A preparative access to the (Z)-diastereomer by photodiastereomerization is described. The 1H and 13C NMR spectra of the two diastereomers were assigned and their absorption spectra and fluorescence quantum yields of the neutral and monodeprotonated species were determined. Two diastereomeric tetraphenylporphyrinyl-ω-hypericinyl-ethylenes were prepared and their properties investigated. The (Z)-diastereomer displayed an even higher photosensitization of singlet oxygen and/or reactive oxygen species than hypericin, whereas the (E)-configured derivative showed a somewhat weaker effect. Accordingly, hybridization of hypericin and porphyrin chromophores seems to be a promising target for the development of novel sensitizers for photodynamic therapy. Summary.  Homochiral Nα-methyl-2,3-diaminopropionic and Nα-methyl-2,4-diaminobutyric acid derivatives 8a,b were obtained via a stereoconservative four-step synthesis starting from hexafluoroacetone protected L-aspartic and L-glutamic acid 2a,b, respectively. Hexafluoroacetone protected ω-isocyanato-α-methylamino acids 4a,b– the key intermediates of the synthesis – are versatile building blocks for amino acid and peptide modification and promising candidates for combinatorial chemistry. Upon reaction with alcohols, compounds 4 give activated N ω-urethane protected ω-amino-α-methylamino acid derivatives 57; upon reaction with amines, ω-ureido-α-methylamino acid derivatives 1012 and 3-methylamino-pyrrolidin-2-ones 13 are available. ω,ω′-Disubstituted hypericin derivatives with the nucleo-bases thymine, cytosine, and adenine in these positions were prepared starting from tri-O-methyl-ω-bromoemodin. The most promising derivative proved to be that with a thymine moiety. It displayed the best solubility of the three products together with a potency to produce singlet oxygen and/or reactive oxygen species comparable to the parent compound hypericin. In addition, although no specific interaction with DNA or poly(2′-deoxyadenylic acid) could be detected, it proved to be significantly better accumulating in the nucleus of prostatic cancer LNCaP cells than hypericin making it a promising candidate for a second-generation photodynamic hypericin agent.  相似文献

Summary.  The protonation and deprotonation behaviour and the assignment of pK a values of hypericin are reviewed and discussed. Three experiments (electrospray MS, 1H NMR, acid–base indicator equilibria) provided additional evidences for the assignment of pK a values of −5 and −6 to mono- and diprotonation at the carbonyl groups of hypericin, of pK a = 2 to monodeprotonation at the bay-region, and of pK a = 11 to dideprotonation at the bay- and peri-regions. The inductive and resonance effects of silicon-, germanium-, tin-, and lead-containing and some organic substituents on the HOMO energies (E HOMO) for 43 monosubstituted andp-disubstituted benzene derivatives were analyzed in the Koopmans approximation. A linear dependence between the perturbation energy δE and the resonanceσ R + parameters of the substituents was established. The Koopmans approximation is a rough approximation for the compounds studied, since to provide for its rigorous fulfilment, the δE values must depend on the σ R 0 parameters of the substituents. The principal regularities of increasing the σ,π-conjugation between the organoelement substituents and the π-system caused by a positive charge on the benzene ring were established. Summary.  The distribution of tetraalkylammonium ions (C n H 2n+1 )4N+ (R +, TAAn +, n = 4–7) with picrate ion (pic ) and inorganic anions X , (Cl, Br, ClO 4), into various inert organic solvents was studied at 25.0°C. The distribution data were analyzed by taking into consideration the distribution of ion pairs, R + · X , and the dimerization of the ion pairs, (R + · X )2, in the organic phase. The ion-pair, distribution constant, K dist, increases with increasing chain length of the tetraalkylammonium ion and with increasing ionic radius of the anion. The values of K dist show a good correlation with the E T value of solvent, i.e. the solvation ability with respect to the anion, and smoothly increase with increasing E T. The effect of the solvent on the dimerization constants, K dim, is markedly different between the ion pairs of picrate ion and inorganic anions. In the case of picrate, K dim significantly decreases with decreasing length of the alkyl chain of the tetraalkylammonium ion, but hardly changes by changing the solvent. On the other hand, in the case of ion pairs of inorganic anions the value of K dim decreases with decreasing E T and is almost constant for all anions. These results were reasonably explained by the difference of the solvation of the anion moieties of the monomeric and dimeric ion pairs. Received May 15, 2001. Summary.  The reaction of 2-amino-4,5-dihydro-3-furancarboxamides with morpholine in the presence of acetic acid in pyridine or under the influence of ammonium acetate gave the corresponding 3-diaminomethylene-4,5-dihydro-2(3H )-furanones; 4,5-dihydro-2-morpholino-3-furancarboxamides were not isolated. One of the former reacted with benzylamine to give (E )- and (Z )-3-(amino-(benzylamino)-methylene)-4,5-dihydro-4-phenyl-2(3H )-furanones and 2-benzylamino-4,5-dihydro-4-phenyl-3-furancarboxamide. Summary.   Kamlet-Taft’s α (hydrogen bond donor acidity) and π* (dipolarity/polarizability) values of various silica batches measured in various solvents are presented. The α and π* parameters for the various solid acids are analyzed by means of Fe(phen)2(CN)2 (cis-dicyano-bis-(1,10)-phenanthroline-iron(II), 1), Michler’s ketone (4,4′-bis-(dimethylamino)-benzophenone, 2), and two hydrophilic derivatives of 2, (4-(dimethylamino)-4′-(di-2-hydroxyethyl)-amino-benzophenone (3a) and 4,4′-bis-(di-(2-hydroxyethyl)-amino)-benzophenone (3b) as well as coumarin 153 (4) as solvatochromic surface polarity indicators. Apparent β (hydrogen bond acceptor basicity) parameters for bare silica have been evaluated by means of an aminobenzodifuranone dye (5) as solvatochromic probe. The chemical interpretation of the α and π* parameters and the nature of the solvent/surface interaction which they reflect are discussed. It can be shown that an increase of the HBA (hydrogen bond accepting) capacity of the solvent significantly decreases the HBD (hydrogen bond donating) capacity of the surface environment, whereas the dipolarity/polarizability value of the silica/solvent interface is a composite of many effects. The classification of the polarity of silica particles in organic solvents compared to pure liquids is outlined. Theoretical E T(30) values of the solid/solvent interfaces are calculated by applying linear solvation energy (LSE) relationships using the independently measured α and π* values of the solid acids according to Received February 2, 2001. Summary.  Ab initio calculations at the HF/6-31G* level of theory for geometry optimization and the MP2/6-31G*//HF/6-31G* level for a single point total energy calculation are reported for (Z,Z)-, (E,Z)-, and (E,E)-cycloocta-1,4-dienes. The C 2-symmetric twist-boat conformation of (Z,Z)-cycloocta-1,4-diene was calculated to be by 3.6 kJ·mol−1 more stable than the C S-symmetric boat-chair form; the calculated energy barrier for ring inversion of the twist-boat conformation via the C S-symmetric boat-boat geometry is 19.1 kJ·mol−1. Interconversion between twist-boat and boat-chair conformations takes place via a half-chair (C 1) transition state which is 43.5 kJ·mol−1 above the twist-boat form. The unsymmetrical twist-boat-chair conformation of (E,Z)-cycloocta-1,4-diene was calculated to be by 18.7 kJ·mol−1 more stable than the unsymmetrical boat-chair form. The calculated energy barrier for the interconversion of twist-boat-chair and boat-chair is 69.5 kJ·mol−1, whereas the barrier for swiveling of the trans-double bond through the bridge is 172.6 kJ·mol−1. The C S symmetric crown conformation of the parallel family of (E,E)-cycloocta-1,4-diene was calculated to be by 16.5 kJ·mol−1 more stable than the C S-symmetric boat-chair form. Interconversion of crown and boat-chair takes place via a chair (C S) transition state which is 37.2 kJ·mol−1 above the crown conformation. The axial- symmetrical twist geometry of the crossed family of (E,E)-cycloocta-1,4-diene is 5.9 kJ·mol−1 less stable than the crown conformation. Corresponding author. Relativistic energy-consistent small-core lanthanide pseudopotentials of the Stuttgart–Bonn variety and extended valence basis sets have been used for the investigation of the dimers La2 and Lu2. It was found that the ground states for La2 and Lu2 are most likely 1 g + g 2π u 4) and 3 g (4f 144f 14σ g 2σ u 2πu 2), respectively. The molecular constants including error bars were derived from multireference configuration interaction as well as coupled-cluster calculations, taking into account corrections for atomic spin–orbit splitting as well as possible basis set superposition errors. The theoretical values for La2 (R e=2.70±0.03 ?, D e=2.31±0.13 eV, ωe=186±13 cm−1) show good agreement with the experimental binding energy (D e=2.52±0.22 eV), but the experimental vibrational constant in an Ar matrix (ωe=236±0.8 cm−1) is significantly higher. For Lu2 the theoretical values (R e=3.07±0.03 ?, D e=1.40±0.12 eV, ωe=123±1 cm−1) are in overall excellent agreement with experimental data (D e=1.43±0.34 eV, ωe=122± 1 cm−1). The electronic structures of La2 and Lu2 are compared to those other lanthanide dimers and trends in the series are discussed. Summary.  The structure dependence of the total π-electron energy E (and therefore of the thermodynamic stability) of phenylenes is examined. Sharp and narrow upper and lower bounds for E are obtained. The gross part of E (above 99%) is determined by the size of the phenylene molecule, measured by the number h of hexagons. Summary.  A simple and rapid derivative spectrophotometric assay procedure is described for the analysis of caffeine (1), acetaminophen (2), and propyphenazone (3) in tablet formulations. The concentration range of application is 5.0–25.0 μg·cm−3 for 2 and 3 and 1.0–5.0 μg·cm−3 for 1. The method involves the extraction of the drugs from tablets with 0.1 N H2SO4, filtration, appropriate dilution, and measurement of the fourth derivative absorbance values at zero crossing wavelengths of 230.0, 263.2, and 256.6 nm for 1, 2, and 3. As a reference method, a reversed phase HPLC procedure was developed. Commercially available tablets were analyzed; statistical comparison of the results with those obtained from the reference method showed good agreement. Summary.  The two new compounds Mn(dien)2[MoS4] (1) and Mn(dien)2[Mo2O2S6] (2) (dien = diethylenetriamine) were prepared under solvothermal conditions. Both compounds were obtained as phase-pure products. The structures consist of new [Mn(dien)2]2+ cations and isolated tetrahedral [MoS4]2− (1) or [Mo2O2S6]2− (2) anions. Between the anions and the cations, hydrogen bonding is observed. Compound 1 crystallizes in the tetragonal space group I (a = 10.219(2), c = 9.259(2) ?, Z = 2), whereas 2 crystallizes in the monoclinic space group P21/c (a = 8.703(2), b = 18.390(4), c = 14.603(3) ?, β = 103.18(3)°, Z = 4). The thermal behaviour of the thiomolybdates was investigated using difference thermoanalysis (DTA) and thermogravimetry (TG). Both compounds decompose under argon with a single endothermic signal in the DTA curve (peak maximum: 252 (1) and 242°C (2)). Received November 5, 2001. Accepted December 27, 2001  相似文献

 The ground state and several low-lying excited states of the Mg2 dimer have been studied by means of a combination of the complete-active-space multiconfiguration self-consistent-field (CASSCF)/CAS multireference second-order perturbation theory (CASPT2) method and coupled-cluster with single and double excitations and perturbative contribution of connected triple excitations [CCSD(T)] scheme. Reasonably good agreement with experiment has been obtained for the CCSD(T) ground-state potential curve but the dissociation energy of the only experimentally known A1Σ u + excited state of Mg2 is somewhat overestimated at the CASSCF/CASPT2 level. The spectroscopic constants D e, R e and ωe deduced from the calculated potential curves for other states are also reported. In addition, some spin–orbit matrix elements between the excited singlet and triplet states of Mg2 have been evaluated as a function of internuclear separation. Summary.  A hypericin derivative was synthesized in which instead of the methyl groups two benzene rings were condensed to the chromophoric system in order to extend its conjugation. This derivative showed lowered fluorescence and concomitantly enhanced sensitized production of active oxygen species as compared to hypericin. However, in contrast to intuition its long wavelength band remained unshifted in comparison to its parent compound hypericin. Summary.  By reaction of O-protected 3-amino-1,6-anhydro-β-D-glucopyranose with succinic, glutaric, and tartaric anhydride, respectively, the corresponding 3-substituted succinimido, glutarimido, and tatarimido derivatives were obtained. Irradiation of the succinimido and glutarimido derivatives at 254 nm gave 1,4-diradical intermediates by γ-hydrogen abstraction (Norrish Type II reaction) which subsequently recombined (Yang cyclization) to form azetidinols. These in turn fragmented and led to azepanedione and azocanedione derivatives. In contrast, irradiation of the tartarimido derivative resulted in elimination and led to both isomeric enolethers.
