以取代芳醛(1a~1h),乙酰乙酸乙酯(2)和脲(3)为原料,MMT/CuCl2为催化剂,乙醇为溶剂,在超声条件下经Beginelli反应合成了8个3,4-二氢嘧啶-2(1H)-酮衍生物(4a~4h),其结构经1H NMR和IR确证。以4a为例,分别采用单因素法和正交实验法研究了催化剂、溶剂、反应温度、超声时间和物料比r[n(1a): n(2) :n(3)]对4a产率的影响。结果表明:在最优反应条件(1a 2.4 mol, r=1.2 : 1.0 : 1.0, MMT/CuCl220 mol%, EtOH 1 mL,于90 ℃超声15 min)下,4a产率88.4%。 MMT/CuCl2循环使用3次,产率基本不变。 相似文献

二氢嘧啶酮(DHPMs)及其衍生物具有广泛的生物活性,如抗病毒、抗肿瘤、抗癌、抗高血压及消炎等作用[1].此外,DHPMs及其衍生物作为钙通道阻滞剂、α1a-对抗剂和神经肽Y的对抗剂,显示出良好的药理活性[2].更重要的是,最近几种含二氢嘧啶酮-5-羧酸盐的海洋生物碱被成功分离出来,并表现出重要的生物学性质[3],某些可以阻止HIVgp-120-CD4键的形成,是一种潜在的HIV抑制剂[4].因此,对DHPMs及其衍生物的研究成为生物活性有机杂环化合物的研究热点之一. 相似文献

4-Alkynyl-3,4-dihydropyrimidin-2-(1H)-ones were synthesized by a one-pot reaction of propynals, ethyl acetoacetate, and urea. The yields of acetylenic dihydropyrimidinones depend significantly upon the propynal structure and catalyst type. A comparative study of the catalysts revealed an important advantage of polyphosphate ester in tetrahydrofuran in comparison with hydrochloric acid in methanol or trimethylchlorosilane in dimethylformamide, allowing the preparation of target compounds in good or moderate yields. 相似文献

Abstract A simple, efficient and environmentally friendly process for synthesis of 3,4-dihydropyrimidinones using a one-pot three-component condensation of aromatic aldehydes, β-dicarbonyl compounds, and urea/thiourea is discussed. The cost-effective, new generation ionic liquid (IL), tri-(2-hydroxyethyl) ammonium acetate was used as both solvent and catalyst. The reactions were carried out using both conventional heating and microwave energy. Application of IL technology with microwave energy represents an attractive and rapid alternative to the conventional acid–base catalyzed thermal processes. 相似文献

Recent advances in the synthesis of C-5-substituted analogs of 3,4-dihydropyrimidin-2-ones: A review
3,4-Dihydropyrimidin-2-ones act as a versatile scaffold in organic synthesis, which serves as a significant template for the development of various therapeutic agents and shows a wide spectrum of activities. The attractive application of 3,4-dihydropyrimidin-2-ones in organic synthesis is undoubtedly owing to C-5 ester group, which is responsible for the change in its bioactivity. Introduction of various groups like electron-withdrawing and electron-donating groups at positions 1, 2, 3, 5, and 6 greatly increased biological activity. Significant efforts have been undertaken to exploit different synthetic routes to synthesize various derivatives of 3,4-dihydropyrimidin-2-ones. This review article gives a comprehensive account of the synthetic utility of C-5 substitution of 3,4-dihydropyrimidin-2-ones used in the design and synthesis of different types of compounds with greater emphasis on recent literature. 相似文献

The Biginelli reaction, known for over 100 years, is an important multicomponent reaction for accessing dihydropyrimidinones (DHPMs). The individual enantiomers of DHPMs exhibit different or even opposite pharmaceutical activities, which require synthetic methods to easily access the optically pure DHPMs. In recent decades, many efforts have focused on developing procedures for the preparation of optically active Biginelli products. In this article, we will summarize the developments in the synthetic methods to access optically active DHPMs with an emphasis on the recent advances in the asymmetric catalytic Biginelli reactions, along with concepts to design the organocatalytic asymmetric variants. 相似文献
