Reaction of 4-aryl-2-methylbut-3-yn-2-ols with thiourea as a new approach to the synthesis of 4-arylmethylidene-5,5-dimethyl-4,5-dihydrothiazole-2-amines

A reaction of thiourea with 4-aryl-2-methylbut-3-yn-2-ols in refluxing pyridine led to the synthesis of 4-arylmethylidene-5,5-dimethyl-4,5-dihydrothiazole-2-amines.     

Synthesis of 1-alkyl-2-methylazetidin-3-ones and 1-alkyl-2-methylazetidin-3-ols

Several 1-alkyl-2-methylazetidin-3-ones were prepared in good yield by the hydride-induced cyclization of the corresponding β-bromo-α,α-dimethoxyketimines, the resulting 3,3-dimethoxyazetidines being hydrolyzed by acid. Imination of these 1,2-disubstituted azetidin-3-ones, followed by alkylation under kinetic control conditions resulted in regioisomeric mixtures of 2,4- and 2,2-dialkylated compounds. Analytical samples of the major 2,4-disubstituted derivatives were obtained after extensive chromatographic separation. The cis stereochemistry of the major 2,4-dialkylated isomer was demonstrated on the basis of NMR data.     

Synthesis and structure of 5,5-dialkyl-3-arylamino-2-thiohydantoins

The reaction of arylhydrazides of -alkyl--chlorocarboxylic acids with thiocyanate ions gave intermediate thiocyanates which rearranged to isothiocyanates on heating and the latter cyclized to 5,5-dialkyl-3-arylamino-2-thiohydantoins.Institute of Technical Chemistry, Urals Branch, Russian Academy of Sciences, Perm' 614000. Synthesis of 4-azacyclopent-2-enones and 5,5-dialkyl-4-azacyclopent-2-enones

Three different methods are reported for the preparation of 4-azacyclo-2-enones 1, two of which allow the preparation of the compounds in optically active form. In addition, a facile route to 4-aza-5,5-dimethylcyclopent-2-enones 2 is disclosed.     

Reduction of Alkyl-2-amino-5,6-dialkyl-3-cyanopyridine-4-carboxylates

KO(t)Bu-promoted three-component coupling of aldehydes and alkynes: highly efficient synthesis of 1-en-4-yn-3-ols and 2-en-4-yn-1-ols

A KO(t)Bu-promoted three-component coupling reaction of aldehydes and alkynes without a transition-metal catalyst was developed in which the sequential addition/isomerization/addition processes take place in one pot. This reaction could be developed into a straightforward and effective method for rapid access to stereodefined (E)-1-en-4-yn-3-ol and (Z)-2-en-4-yn-1-ol compounds.     

Homopolymerization of 2-alkyl- and 2-aryl-2-oxazolines

2-Aryl- and 2-alkyl-2-oxazolines have been polymerized to poly-(N-aroyl)aziridines and poly(N-acyl)aziridines, respectively, in the presence of boron trifluoride. The polymers obtained were glassy, light yellow resins with molecular weights ranging from 3500 to 7500 (35–50 oxazoline units per chain). The polymerization rates have been determined for several of these monomers. A polymerization mechanism is proposed.     

Synthesis and x-ray structural analysis of 2-amino-4-cyanomethylene-5,5-dimethylthiazoline

The reaction of thiourea with 4-hydroxy-4-methyl-2-pentynenitrile in triethylamine is accompanied by heterocyclization and the formation of 2-amino-4-cyano-methylene-5, 5-dimethylthiazoline, whose structure was established by x-ray structural analysis (XSA).Translated from Izvestiya Akademii Nauk SSSR, Seriya Khimicheskaya, No. 10, pp. 2344–2346, October, 1989.     

Synthesis of 4-alkyl-3-pyridinols from 3-benzyloxypyridine

The addition of Grignard reagents to the 1-phenoxycarbonyl salt of 3-benzyloxypyridine and a catalytic amount of cuprous iodide afforded 4-alkyl-3-benzyloxy-1-phenoxycarbonyl-1,4-dihydropyridines in good yield. The crude dihydropyridines were aromatized with o-chloranil to give 4-alkyl-3-benzyloxypyridines, which were debenzylated with hydrogen and 10% palladium on carbon to provide 4-alkyl-3-pyridinols.     

Palladium-catalyzed one-pot synthesis of polysubstituted furans from alkynoates and 2-yn-1-ols

Easily accessible propargyl vinyl ethers are found to be acyclic precursors to construct furan derivatives. According to the reactive behavior of the substrates and the regulation of reaction conditions, three synthetic routes to polysubstituted furans via palladium-catalyzed reactions of propargyl vinyl ethers in situ-prepared from alkynoates and 2-yn-1-ols are presented herein. These methods could have potential application in synthetic and pharmaceutical chemistry for its facilitation and accessible starting materials.     

Synthesis of some 2-alkyl-4-quinolone and 2-alkyl-4-methoxyquinoline alkaloids

Acid-catalyzed condensation of a series of 3-oxoalkanoic acid esters with aniline affords the corresponding 2-alkyl-4-quinolones in good yield. Treatment of these compounds with methyl iodide affords 2-alkyl-4-methoxyquinolines and 2-alkyl-N-methylquinolones. Three of the compounds are naturally-occurring alkaloids which are synthesized here for the first time.     

Alkyl 2-amino-5,6-dialkyl-3-cyanopyridine-4-carboxylates in reactions with electrophilic reagents

In reaction of alkyl 2-amino-5,6-dialkyl-3-cyanopyridine-4-carboxylates with isocyanates formed unstable ureas, and with nitrous acid at 60–70°C alkyl 5,6-dialkyl-2-oxo-1,2-dihydro-4-pyidinecarboxylates were obtained. It was shown for the latter that their reactions with organic acids and amides occurred at the cyano group, and the alkaline hydrolysis involved the ester group.     

BF3-OEt2-mediated rearrangement of 4-substituted-5,5-diphenylazepan-4-ols

Synthesis of 2-(2-substituted-ethylidene)-3,3-diphenylpyrrolidines has been established starting from different 4-substituted-4-aryl-5,5-diphenylazepan-4-ols via boron trifluoride etherate-mediated rearrangement.     

An unusual synthetic way to 2-methyl-3-alkyl-6-ethoxy-heptan-2-ols from 2-ethoxy-5-alkyl-3,4-dihydro-2H-pyrans

A simple and swift preparation of 2-methyl-3-alkyl-6-ethoxyheptan-2-ols may be accomplished in two steps starting from 2-ethoxy-5-alkyl-3-,4-dihydro-2H-pyrans via chlorination of the heterocyclic compounds and subsequent reaction with MeMgBr.     

Synthesis of 4-alkyl-6-amino-3,5-dicyano-2(1H)-pyridinethiones

Condensation of aliphatic aldehydes with cyanothioacetamide has given 4-alkyl-6-amino-3,5-dicyano-2(1H)-pyridinethiones, which have also been synthesized by recyclization of 4-alkyl-2,6-diamino-4H-thiopyrans. Substituted 2-alkylthiopyridines and thieno[2,3-b]pyridines have been prepared from the pyridinethiones. 2,6-Diamino-4-isopropyl-3,5-dicyano-4H-thiopyran and 6-amino-4-isobutyl-2-methylthio-3,5-dicyanopyridine have been studied by x-ray crystallography.T. Synthesis of 3-alkyl-4-aminofurazans

A “one-pot” method for the synthesis of 3-alkyl-4-aminofurazans from ethyl β-alkyl-β-oxopropionates was developed. The multistep process involves hydrolysis of the ester, nitrosation at the activated methylene group, and treatment of the resulting intermediate with an alkaline solution of hydroxylamine in the presence of urea.
